Tumor metastasis, responsible for approximately 90% of cancer-associated mortality, remains poorly understood. Here in this study, we employed a melanoma lung metastasis model to screen for metastasis-related genes. By sequential tail vein injection of mouse melanoma B16F10 cells and the subsequently derived cells from lung metastasis into BALB/c mice, we successfully obtained highly metastatic B16F15 cells after five rounds of in vivo screening. RNA-sequencing analysis of B16F15 and B16F10 cells revealed a number of differentially expressed genes, some of these genes have previously been associated with tumor metastasis while others are novel discoveries. The identification of these metastasis-related genes not only improves our understanding of the metastasis mechanisms, but also provides potential diagnostic biomarkers and therapeutic targets for metastatic melanoma.

Combining logical function and memory characteristics of transistors is an ideal strategy for enhancing computational efficiency of transistor devices.Here,we rationally design a tri-gate two-dimensional(2D)ferroelectric van der Waals heterostructures device based on copper indium thiophosphate(CuInP_(2)S_(6))and few layers tungsten disulfide(WS_(2)),and demonstrate its multi-functional applications in multi-valued state of data,non-volatile storage,and logic operation.By co-regulating the input signals across the tri-gate,we show that the device can switch functions flexibly at a low supply voltage of 6 V,giving rise to an ultra-high current switching ratio of 107 and a low subthreshold swing of 53.9 mV/dec.These findings offer perspectives in designing smart 2D devices with excellent functions based on ferroelectric van der Waals heterostructures.

The differentiation imbalance in bone marrow mesenchymal stem cells(BMMSCs)is critical for the development of bone density diseases as the population ages.BMMSCs are precursor cells for osteoblasts and adipocytes;however,the chromatin organization landscapes during BMMSC differentiation remain elusive.In this study,we systematically delineate the four-dimensional genome and dynamic epigenetic atlas of BMMSCs by RNA sequencing,assay for transposase-accessible chromatin sequencing,and highthroughput chromosome conformation capture.The structure analyses reveal 17.5%common and28.5%-30%specific loops among BMMSCs,osteoblasts,and adipocytes.The subsequent correlation of genome-wide association studies and expression quantitative trait locus(e QTL)data with multi-omics analysis reveal 274 genes and 3634 single nucleotide polymorphisms(SNPs)associated with bone degeneration and osteoporosis(OP).We hypothesize that SNP mutations affect transcription factor(TF)binding sites,thereby affecting changes in gene expression.Furthermore,26 motifs,260 TFs,and 291SNPs are identified to affect the e QTL.Among these genes,DAAM2,TIMP2,and TMEM241 are found to be essential for diseases such as bone degeneration and OP and may serve as potential drug targets.

Enrichment characteristics of normal-pressure marine shale gas in the complex tectonic zone: a case study of the Lower Cambrian Niutitang Formation in northern Guizhou

The Lower Cambrian Niutitang Formation is a crucial stratum for the exploration of normal-pressure marine shale gas in southern China， with great development potential. To research the normal-pressure shale gas accumulation of the Niutitang Formation in the complex tectonic zone， the shale cores in northern Guizhou were subjected to X-ray diffraction (XRD) and field emission scanning electron microscopy (FE-SEM) observation， and tests were conducted to determine total organic content (TOC)， bitumen reflectance， and nitrogen isotopes. The results indicate that the equivalent vitrinite reflectance of cores varies from 2.30% to 3.73%， and the TOC content of cores varies from 3.8% to 7.8%. The total gas content of three wells in northern Guizhou is 0.5 m3/t， 0.3 m3/t， and 1.4 m3/t， respectively. The TOC content of the shale in the syncline structure is higher than that of the shale in the anticline structure. Nitrogen in shale gas originates primarily from the thermal ammoniation of organic matter， the atmosphere， and the deep crust. Shale gas preservation is favourable in regions with a large range of lifting， and small lifting differences. The syncline is broad and gentle with few faults have better gas accumulation conditions. The Niutitang Formation in the residual syncline of the study area has better gas content than that of other structures. Exploration of shale gas accumulation sweet spots in northern Guizhou should focus on the broad-gentle synclines with minimal changes in stratigraphic lifting， few faults， and no hydrothermal activity at the edges of paleo-uplifts.

背景与目的:食管癌(esophageal carcinoma,ESCA)是死亡率较高的恶性肿瘤之一,其发生、发展的机制不明。CDC20被认为具有癌基因功能,其表达失调与肿瘤的发生、发展密切相关。CDC20在多种肿瘤中表达升高,敲低CDC20能够抑制肿瘤细胞增殖。NLRP3是炎症小体的主要成分之一,炎症小体失调与肿瘤的发生、发展也密切相关。本研究旨在探究CDC20是否通过NLRP3促进ESCA细胞增殖,同时分析其调控机制。方法:通过癌症基因组图谱(The Cancer Genome Atlas,TCGA)数据库和GTEx公共数据库分析ESCA患者中CDC20和NLRP3基因的表达水平。收集新乡医学院第一附属医院收治的80例ESCA患者的临床病理学资料和组织,通过免疫组织化学染色检测NLRP3在ESCA患者中的蛋白表达水平。本研究通过新乡医学院第一附属医院伦理委员会的审批(编号:EC-021-137)。通过短发夹RNA(short hairpin RNA,shRNA)技术检测敲低CDC20和NLRP3基因后对食管鳞状细胞癌细胞系EC9706和KYSE150增殖能力的影响。通过免疫共沉淀(Co-immunoprecipitation,Co-IP)、蛋白酶体抑制剂和泛素化实验检测CDC20是否与NLRP3相互作用,以及阐明CDC20是否通过泛素化途径调控NLRP3表达。结果:TCGA数据库分析结果显示,ESCA组织中CDC20和NLRP3 mRNA表达水平明显高于癌旁组织。免疫组织化学结果也进一步显示,与癌旁组织相比,CDC20、NLRP3在ESCA组织中蛋白表达水平升高。敲低CDC20和NLRP3基因可抑制ESCA细胞增殖。Co-IP、蛋白酶体抑制剂和泛素化实验证实CDC20通过NLRP3的LRR区与NLRP3相互作用,且CDC20通过促进NLRP3泛素化稳定其表达。结论:CDC20和NLRP3在ESCA癌组织中表达上调,且CDC20通过泛素化NLRP3稳定其表达,从而促进ESCA细胞增殖,提示CDC20和NLRP3可能是ESCA潜在的诊断靶向标志物。

A framework of compositionally graded Ti-Al alloys was proposed to elucidate the Al alloying dependent deformation mechanism at room temperature.Slip trace analysis demonstrated low-alloyed Ti-Al model materials were plastically deformed by prismatic slip,pyramidal slip,stacking faults,and deformation twins.Increasing Al concentration promoted chemical ordering,thus suppressing the dislocation motion and deformation twinning.The mechanism behind such a kind of composition-mediated deformation physics was discussed.Our findings are expected to be applicable in the design of next-generation high-performance structural materials through the tailoring of the chemical ordering of solute atoms in substitutional solid solution.

研究轮胎-路面之间的三维作用力,对预测路面早期损坏、优化道路设计具有指导作用,同时也是分析车辆动力学性能、研制和改进整车控制系统的有效手段.文中利用ABAQUS有限元仿真软件,建立了轮胎与路面之间相互作用力与胎冠相对轮辋中心位移之间的力-位移多元非线性回归模型;该模型通过分析在三维力作用下轮胎不同位置处节点的变形和位移,实现了不同节点处的轮胎与路面之间作用力的求解,并通过改变充气断面尺寸、不同载荷下沉量等指标使模型有效性得到了验证,为设计并优化位移传感器的布局、提高测量结果的可信度,提供了理论依据.采用带有二次项的多元非线性回归模型对11.00R20载重子午线轮胎进行仿真计算.结果表明,在接地中心角θ不超过±15°范围内,回归模型的三维力预测值相对于预设值的误差不超过±2%.为在兼顾回归模型准确度的前提下,提高该模型的解算效率、降低由位移解算力过程的繁琐性,通过分析完整模型中各子项对力预测结果的贡献度,进一步提出了简化回归模型;当θ≤±15°时,各个节点处完整模型与简化模型均方根误差(RMSE)的最大值相差0.43 kN;当θ≤±9°时,两者差值减小至0.27 kN.分析结果表明,通过对安装在轮胎上的位移传感器所采集的数据进行处理,完整模型和简化模型均能实现轮胎与路面间相互作用三维力的预测和计算,但简化模型的适用范围比完整模型要窄.

研究重载卡车对道路的破坏机制能够指导道路的设计与养护,有效延长道路的使用寿命.其中,针对复杂重载荷作用下道路病害研究对轮胎路面作用力测量的需求,本文提出了一种复合工况下胎路三向力的建模估算方法.以11.00R20型全钢载重子午线轮胎为研究对象,经过对结构的合理简化,利用ABAQUS有限元仿真软件以参数化建模方法建立了轮胎有限元模型,并验证了模型的有效性;以不同的垂向载荷、侧偏角、纵滑率、轮速和胎压设置了多种复合工况,并对轮胎模型进行了稳态滚动分析,获得了轮胎同一断面内衬层上多个观测点的加速度响应信号;对采集的加速度信号进行了滤波处理,并对比了不同信号的响应程度,选取其中特征较为明显的信号作为观测加速度信号;提取了观测加速度信号的特征,并结合胎压和轮速作为输入,建立了基于卷积神经网络(CNN)的复合工况下胎路三向力的回归解析模型;以模型对15个测试工况的估算结果,近似地描述了该模型的泛化能力,该模型下胎路三向力估算的平均绝对百分比误差均在7%以内,均方根误差不超过1 kN.结果表明,轮胎内衬层上的加速度信号与胎路作用力具有较高的相关性,基于加速度信号与卷积神经网络建立的模型能够有效地实现胎路三向力的同时估算,为道路病害研究提供了有价值的载荷信息.
