Peptidome profiling of human serum of uveal melanoma patients based on magnetic bead fractionation and mass spectrometry

AIM： To find new biomarkers for uveal melanoma （UM） by analyzing the serum peptidome profile. METHODS： Proteomic spectra in patients with UM before and after operation were analyzed and compared with those of healthy controls. Magnetic affinity beads were used to capture serum peptides and matrix-assisted laser desorption/ionization time-of-flight （MALDI-TOF） mass spectrometer were used to compile serum peptide profiles. RESULTS： A panel of 49 peptides were differentially expressed between UM patients and controls, of which 33 peptides were of higher intensities in patient group and 16 peptides were of higher intensities in control group. Based on combined use of these potential markers, peptides with mean molecular masses of 1467 and 9289.0 Da provide high sensitivity （83.3%）, specificity （100%） and accuracy rate （93.0%） together to differentiate melanoma patients from healthy controls. At the time point of 6mo postoperatively, the levels of many peptides differentially expressed before surgery showed no more statistical difference between the patients and the control group. Fibrinogen o-chain precursors were identified as potential UM markers. CONCLUSION： We have shown that a convenient and fast proteomic technique, affinity bead separation and MALDI- TOF analysis combined with bioinformatic software, facilitates the identification of novel biomarkers for UM.

The Rainbow Vertex-disconnection in Graphs

Let G be a nontrivial connected and vertex-colored graph. A subset X of the vertex set of G is called rainbow if any two vertices in X have distinct colors. The graph G is called rainbow vertex-disconnected if for any two vertices x and y of G, there exists a vertex subset S of G such that when x and y are nonadjacent, S is rainbow and x and y belong to different components of G-S;whereas when x and y are adjacent, S + x or S + y is rainbow and x and y belong to different components of(G-xy)-S. For a connected graph G, the rainbow vertex-disconnection number of G, denoted by rvd(G), is the minimum number of colors that are needed to make G rainbow vertexdisconnected. In this paper, we characterize all graphs of order n with rainbow vertex-disconnection number k for k ∈ {1, 2, n}, and determine the rainbow vertex-disconnection numbers of some special graphs. Moreover, we study the extremal problems on the number of edges of a connected graph G with order n and rvd(G) = k for given integers k and n with 1 ≤ k ≤ n.

Seasonal prediction of summer rainfall is crucial to reduction of regional disasters,but currently it has a low prediction *** developed a dynamical and machine learning hybrid(MLD)seasonal prediction method for summer rainfall in China based on circulation fields from the Chinese Academy of Sciences(CAS)Flexible Global Ocean-Atmosphere-Land System Model finite volume version 2(FGOALS-f2)operational dynamical prediction *** selecting optimum hyperparameters for three machine learning methods to obtain the best fit and least overfitting,an ensemble mean of the random forest and gradient boosting regression tree methods was shown to have the highest prediction skill measured by the anomalous correlation *** skill has an average value of 0.34 in the historical cross-validation period(1981-2010)and 0.20 in the 10-yr period(2011-2020)of independent prediction,which significantly improves the dynamical prediction skill by 400%.Both reducing overfitting and using the best dynamical prediction are important in applications of the MLD method and in-depth analysis of these warrants a further investigation.

Low soil carbon saturation deffcit limits the abundance of cbbL-carrying bacteria under long-term no-tillage maize cultivation in northern China

The responses of cbbL-carrying bacteria to different levels of soil carbon saturation deficits(SCSD)under tillage managements are largely *** assessed the influence of SCSD on the abundance and diversity of cbbLcarrying bacteria under long-term no-tillage with residue retention(NT)and conventional tillage without residue retention(CT)cultivation systems in *** found SCSD was smaller under NT than under CT in the 0-15 cm soil *** abundance and the Shannon diversity of cbbL-carrying bacteria in the NT treatment were lower than in the CT *** carbon saturation and cbbL gene abundance showed a significant positive correlation,but there was no correlation between soil carbon saturation and cbbL gene ***,the long-term NT practice decreased cbbL-carrying bacteria diversity and altered the community structure of the cbbL-carrying *** results indicated that low SCSD limited the abundance of cbbL-carrying bacteria,but there was no relationship between low SCSD and diversity of cbbLcarrying *** suggest that further studies of cbbL-carrying bacteria carbon sequestration rates and capacity should be based on the effect of management practices on cbbL-carrying bacteria abundance and *** study has important implications for the relationship between the biological and physicochemical mechanisms in CO_(2) fixation.

The crystal change and "excess heat" produced by long time electrolysis of heavy water with titanium cathode

According to the theory given in the paper , the long time electrolysis experiment with titanium cathode in heavy water (D 2O) were done for many times by using the open-loop multi-parameters electrolysis calorimetry system, which is established by us. The specialty is that the cathode is titanium rod and the anode is platinum wire. The early experiment result is still repeated in our recent experiment. The obvious "excess heat" phenomenon can take place only when the electrolysis last more than ten days and amount of "excess heat" increased with the electrolysis time. The "excess heat" can also be obtained from the "boiling to dry" experiment. In the recent experiment, we obtain the results that the amount of "excess heat" is about 3.6 times the input energy, the "excess heat" power is 76.5 W, and the "excess heat" power density is 121.7 W/cm 3. After the electrolysis, the crystal structure of the Ti cathode was measured with x-ray diffraction apparatus. We found that the crystal structure of Ti cathode was changed to face-centered cubic structure of TiD 2 from its hexagonal structure. This result is in agreement with the Gou’s theory mentioned in reference .

Effect of feeding transgenic cry1Ab/cry1Ac rice on indices of immune function in broilers

The safety of feeding transgenic crylAb/crylAc rice （a genetically modified （GM） rice variety） to broilers was examined from an immunological perspective. HatchlingArborAcres chickens （240） were assigned to two dietary treatments （diets containing GM or non-GM rice） with 12 replicates per group and 10 birds per replicate. Traits were measured on one randomly selected bird from each replicate at d 21 and 42. The 42-d feeding trial revealed that crylAb/crylAc rice had no significant effect relative to non-GM rice on body weight and the immune organ indices. No significant pathological lesion in the spleen and bursa of Fabricius was found in the GM rice group. There were no significant differences in serum concentrations of immunoglobulin Y （IgY）, IgM, interleukin 4 （IL-4） and IL-6 between the two groups at d 21 or 42, except for IL-6 being higher （P〈0.05） in the GM- fed chickens at d 42. There were no differences in the T and B lymphocyte transformation rate and CD4＋/CD8＋ ratio between the two groups at d 42. Additionally, there was no significant difference between the two diets in expression of relevant genes viz. the major histocompatibility complex class II beta chain （BLB2）, interferon beta 1 （IFNβ）, tumour necrosis factor alpha-like （TNFa） and toll-like receptor 4 （TLR4）in the spleen and bursa of Fabricius. All the data demonstrated that transgenic crylAb/ crylAc rice had no adverse effect on these aspects of immune function of broilers during 42-d feeding trial. Transgenic rice was therefore indistinguishable from non-GM rice in terms of short-term feeding in chickens.

Variability in moisture content is a common condition in natural *** influences soil properties significantly.A comprehensive understanding of the evolution of soil microstructure in wetting/drying process is of great significance for interpretation of soil macro hydro-mechanical *** this review paper,methods that are commonly used to study soil microstructure are *** them are scanning electron microscope(SEM),environmental SEM(ESEM),mercury intrusion porosimetry(MIP)and computed tomography(CT)***,progress in research on the soil microstructure evolution during drying,wetting and wetting/drying cycles is summarized based on reviews of a large body of research papers published in the past several *** compacted on the wet side of op-timum water content generally have a matrix-type structure with a monomodal pore size distribution(PSD),whereas soils compacted on the dry side of optimum water content display an aggregate structure that exhibits bimodal *** drying,decrease in soil volume is mainly caused by the shrinkage of inter-aggregate *** wetting,both the intra-and inter-aggregate pores increase gradually in number and *** in the characteristics of the soil pore structure significantly depend on stress state as the soil is subjected to *** wetting/drying cycles,soil structural change is not completely reversible,and the generated cumulative swelling/shrinkage deformation mainly derives from ***,based on this analysis and identified research needs,some important areas of research focus are proposed for future *** areas include innovative methods of sample preparation,new observation techniques,fast quantitative analysis of soil structure,integration of microstructural parameters into macro-mechanical models,and soil microstructure evolution charac-teristics under multi-field coupled conditions.
