Corneal nerve changes by anti-glaucoma medications examined by in vivo confocal microscopy

AIM:To evaluate the effects of antiglaucoma eye drops on corneal nerves by in vivo confocal microscopy(IVCM).METHODS:This study comprised 79 patients diagnosed with glaucoma and 16 healthy control *** the glaucoma patients,54 were treated with medication,while 25 remained *** corneal images were evaluated by IVCM,and then ACCMetrics was used to calculate the following parameters:corneal nerve fiber density(CNFD),branch density(CNBD),fiber length(CNFL),total branch density(CTBD),fiber area(CNFA),fiber width(CNFW),and fractal dimension(CNFrD).The correlation between IVCM parameters and drugs was evaluated using non-parametric measurements of Spearman’s rank correlation ***:The CNFD was reduced in glaucoma groups compared to healthy subjects(P<0.01).Patients using anti-glaucoma medications exhibited poorer confocal parameters compared to untreated *** the number of medications and usage count increased,CNFD,CNBD,CNFL,CTBD,CNFA,and CNFrD experienced a decline,while CNFW increased(all P<0.01).For the brinzolamide-therapy group,there was a significant decrease in CNFD and CNFL compared to the other monotherapy groups(P<0.001).In the absence of medication,CNFD in males was lower than that in females(P<0.05).Among patients under medication therapy,CNFD remained consistent between males and ***:Antiglaucoma eye drops affect the microstructure of corneal *** and ACCMetrics are useful tools that could be used to evaluate the corneal nerve changes.

Effect of hydrogen on the friction and wear of Ni-P coatings

The effects of charging and outgassing on the friction and wear of Ni-P amorphous and nanocrystalline coatings were studied under ambient humidity and *** results indicate that hydrogen charging can greatly decrease the volume loss during friction and increase the wear ***,the wear durability can be restored after *** are many cracks on the brittle nanocrystalline before charging,and no crack appears after charging,but there are only a few cracks on the outgassed *** indicates that hydrogen charging can inhibit the crack formation during friction for the brittle Ni-P *** the charged sample,after the surface layer was removed by polishing,the wear track and the friction coefficient are consistent with those of an uncharged *** friction coefficient of the charged amorphous coating is very small within the first 140 s and then increases suddenly to the value corresponding to the uncharged *** absorbed surface layer lubricates the surface,reduces the friction coefficient,and improves the wear durability.

Comparison of the therapeutic effects of extracts from Spirulina platensis and amnion membrane on inflammation-associated corneal neovascularization

AIM: To compare the therapeutic effects of polysaccharide extract from Spirulina platensis (PSP) and extract from amnion membrane CAME) on alkali burn-induced corneal neovascularization (CorNV). METHODS: PSP and AME were Extracted from dry powder of Spirulina platensis and human aminion membrane respectively. Murine CorNV was induced by applying 1N sodiumhydroxide (NaOH) solution directly on the mice corneas. PSP and AME extracts were administered topically on the corneas 4 times daily for 7 days. The therapy effects of PSP and AME extracts were evaluated daily using slit-lamp. At the end of the therapy, corneas were harvested for H&E staining, masson trichrome staining, immunohistochemical study, and semi-quantification reverse transcriptive PCR (RT-PCR) was utilized for measurement of inflammation-related molecules. RESULTS: Topical application of PSP extract had significant therapeutic effects on CorNV that could be shown in various assays of the corneas. Compared with AME Extract, PSP extract treatment was more effective in suppressing CorNV in berms of vessel length and levels of cluster of differentiation 31 (CD31) proteins or the angiogenesis related genes like vascular endothelial growth factor (VEGF), matrix metalloproteinase-2 (MMP2) and matrix metalloproteinase-9 (MMP9). PSP also inhibited inflammation more markedly by more effectively inhibiting mononuclear and polymorphonuclear cells infiltration into the corneal stroma and reducing levels of stromal cell-derived factor-1 (SDF1), tumor necrosis factor-alpha (TNF alpha) and macrophage inflammatory protein-3 (MIP3a). In addition, corneas of PSP group had a more regular and compact architecture of collagen with thinner corneal thickness than in the AME group. ' CONCLUSION: Polysaccharide extract from Spirulina platensis inhibited alkali burn-induced inflammation and CorNV more effectively than AME Extract at the studied doses, thus may be used for the therapy of corneal diseases involving neovascularizatio

MM： To develop a protocol for direct hepatic lineage differentiation from early developmental progenitors to a population of mature hepatocytes, METHODS： Hepatic progenitor cells and then mature hepatocytes from mouse embryonic stem （ES） cells were obtained in a sequential manner, induced by valproic acid （VPA） and cytokines （hepatocyte growth factor, epidermal growth factor and insulin）, Morphological changes of the differentiated cells were examined by phase-contrast microscopy and electron microscopy, Reverse transcription polymerase chain reaction and immunocytochemical analyses were used to evaluate the gene expression profiles of the VPA-induced hepatic progenitors and the hepatic progenitor-derived hepa- tocytes, Glycogen storage, cytochrome P450 activity, transplantation assay, differentiation of bile duct-like structures and tumorigenic analyses were performed for the functional identification of the differentiated cells, Furthermore, FACS and electron microscopy were used for the analyses of cell cycle profile and apoptosis in VPA-induced hepatic differentiated ***： Based on the combination of VPA and cytokines, mouse ES cells differentiated into a uniform and homogeneous cell population of hepatic progenitor cells and then matured into functional hepatocytes. The progenitor population shared several characteristics with ES cells and hepatic stem/progenitor cells, and represented a novel progenitor cell between ES and hepatic oval cells in embryonic development. The dif- ferentiated hepatocytes from progenitor cells shared typical characteristics with mature hepatocytes, including the patterns of gene expression, immunological markers,in vitro hepatocyte functions and in vivo capacity to restore acute-damaged liver function. In addition, the differentiation of hepatic progenitor cells from ES cells was accompanied by significant cell cycle arrest and selective survival of differentiating cells to-wards hepatic lineages. Content-Based Hierarchical Analysis of News Video Using Audio and Visual Information

A schema for content-based analysis of broadcast news video is presented. First, we separate commercials from news using audiovisual features. Then, we automatically organize news programs into a content hierarchy at various levels of abstraction via effective integration of video, audio, and text data available from the news programs. Based on these news video structure and content analysis technologies, a TV news video Library is generated, from which users can retrieve definite news story according to their demands.

Fibre-reinforced polymer(FRP)/rigid polyurethane foam(RPUF)interface is susceptible to moisture intrusion in addition to natural ageing during *** study investigated the influence of moisture intrusion on the FRP/RPUF interface under natural ageing using experiments and molecular dynamics(MD)*** absorption,shear strength,and leakage current tests were used to clarify the changes in the interface *** degradation mechanism of moisture intrusion at the interface under natural ageing was revealed by MD simulation and *** results show that the natural ageing of interfacial materials will slightly reduce the quality of the interface but will reduce the water resistance of the material and promote the process of water intrusion into the *** addition,water mainly invades the interface through *** water penetration,the interfacial bonding strength and insulation performance significantly *** and hydrolysis of interfacial materials are the main reasons for decreased interfacial *** can cause irreversible damage to the interface,increasing interface defects and water *** vicious cycle of material hydrolysis is the ultimate cause of interfacial *** polar molecules produced by this process and the water absorbed by the interface caused the degradation of the interface insulation performance.

Paeoniflorin,the Main Monomer Component of Paeonia lactiflora,Exhibits Anti-inflammatory Properties in Osteoarthritis Synovial Inflammation

Objective:To explore the mechanism of paeoniflorin(PF)on osteoarthritis(OA)synovial inflammation from network pharmacology to experimental ***:Targets of OA were constructed by detecting the database of network database platforms(Therapeutic Target database,Drug Bank and Gene Cards),and the targets of PF were constructed by Pub Chem and Herbal Ingredients'Targets *** Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)analysis of these co-targeted genes were conducted via Database for Annotation,Visualization,and Integrated Discovery(DAVID)database,and protein-protein interaction(PPI)networks were conducted via the search tool for the retrieval of interacting genes(STRING)*** counting kit-8(CCK-8)assay was performed to assess the potential toxicity of PF on human OA fibroblast-like synoviocytes(FLS),quantitative real-time polymerase chain reaction(q PCR),enzyme-linked immunosorbent assay(ELISA)and Western blot were used to verify the potential mechanism of PF in synovial ***:Twenty-six co-targeted genes were *** enrichment results showed that these co-targeted genes were most likely localized in the cytoplasm,and the biological processes mainly involved'cellular response to hypoxia''lipopolysaccharide(LPS)-mediated signaling pathway'and'positive regulation of gene expression'.KEGG pathway analysis indicated that these co-targeted genes may function through pathways associated with'hypoxia-inducible factor-1(HIF-1)signaling pathway'and'tumornecrosis factor(TNF)signaling pathway'.The PPI network showed that the top 3 hub genes were TP53,TNF,and *** docking results showed that PF was well docking with ***-8 showed no potential toxicity of 10,20 and 50μmol/L PF on human OA *** PF significantly decreased the expression levels of interleukin-1β,interleukin-6,TNF-αmatrix metalloproteinase 13(MMP13),and a disintegrin and metalloproteinase with thrombospondin motifs 5(ADAMTS5)and TNF

Efficacy of rhNGF-loaded amniotic membrane transplantation for rabbit corneal epithelial and nerve regeneration

AIM:To evaluate the efficacy of recombinant human nerve growth factor-loaded amniotic membrane(rh NGF-AM)on corneal epithelial and nerve regeneration in rabbit ***:Freshly prepared human amniotic membrane(AM)were immersed into PBS buffer containing 100 or 500μg/mL rh NGF for 15,30,and 60 min at 4℃.The in vitro release kinetics of rh NGF was measured with *** in vivo evaluation,the AM were immersed with 500μg/mL rh NGF for 30 ***-seven rabbits were selected to establish corneal epithelial defect *** addition to the 19 rabbits in control group,38 rabbits received AM transplantation with or without rh NGF after the removal of central *** epithelial defect area,sub-epithelial nerve fiber density,corneal sensitivity,rh NGF contents in resident AM and corneas were measured after the ***:rh NGF was sustained release from the AM within 14 d in vitro,with the positive correlation with initial immersion *** immersion of AM in 500μg/mL rh NGF for 30 min achieved the most stable release within 14 *** transplantation in rabbit cornea,a high concentration of rh NGF in resident rh NGF-AM and cornea was maintained within 8 *** epithelial healing,nerve fiber regeneration and the recovery of corneal sensitivity were significantly accelerated after the rh NGF-AM transplantation when compared to simple AM transplantation(all P<0.05).CONCLUSION:Simple immersion of AM achieves the sustained release of rh NGF,and promotes corneal epithelial wound healing and nerve regeneration,as well as the recovery of corneal sensitivity in rabbit.

Vapor Online Monitor Model of Vapor Power Station Based on UML

We presents a vapor online monitor system model of vapor power station developed by visual tool rational rose 2000. Use cases such as on line instrument (onlineinstr), control, query, report, real database (realdb) and alarm are generated according to the system requirements. Use case view and class view of the system are formed at the same time. As for all the UML models of the system, this paper focuses the discussion on the class view, the component diagram of the control class and the sequence diagram of the query class. Corresponding C++ codes are produced and finally transferred into the spot running software.
