Myasthenia gravis(MG) is a prototypical antibody-mediated neurological autoimmune disease with the involvement of humoral immune responses in its pathogenesis. T follicular helper(Tfh) cells have been implicated in many autoimmune diseases. However, whether and how Tfh cells are involved in MG remain ***, we established and studied a widely-used and approved animal model of human MG, the rat model with acetylcholine receptor alpha(AChRa) subunit(RAChR97–116)-induced experimental autoimmune myasthenia gravis(EAMG). This model presented mild bodyweight loss 10 days after the first immunization(representing the early stage of disease) and more obvious clinical manifestations and body-weight loss 7 days after the second immunization(representing the late stage of disease). AChR-specific pre-Tfh cells and mature Tfh cells were detected in these two stages, respectively. In cocultures of Tfh cells and B cells, the number of IgG2 bsecreting B cells and the level of anti-AChR antibodies in the supernatant were higher in the cultures containing EAMG-derived Tfh cells. In immunohistochemistry and immunofluorescence assays, a substantial number of CD4^+/Bcl-6^+ T cells and a greater number of larger germinal centers were observed in lymph node tissues resected from EAMG rats. Based on these results, wehypothesize that an AChR-specific Tfh cell-mediated humoral immune response contributes to the development of EAMG.

Experimental allergic encephalomyelitis(EAE) can be elicited by injecting the MOG peptide *** cells(DCs) located at the immunization site phagocytize the MOG peptide,mature,migrate into the nearest draining lymph node(LN),and present antigen resulting in the activation of naive T *** and Thl7 cells represent the primary cells involved in EAE ***-trans retinoic acid(AT-RA) has been shown to have beneficial effects on EAE progression,however,whether AT-RA exerted its beneficial effect by influencing DC maturation or mediating other functions remains to be *** the present study,we showed that AT-RA lead to the down regulation of MHC-II,CD80(B7-1),and CD86(B7-2) expressed on the surface of DCs isolated from the draining LNs or spleen 3 days post immunization in an EAE *** to DC function influenced Th subset ***,the number of CD44 monocytes(which might trigger EAE progression) was also significantly decreased in draining LNs,spleen,subarachnoid space,and the spinal cord parenchyma post AT-RA *** findings demonstrated for the first time that the DC antigen presenting capacity was impaired by AT-RA resulting in the down regulation of pathogenic Thl and Thl7 inflammatory cell responses associated with reduced EAE severity.

It has been reported that peripheral Natural killer cells（NK cells）and their cytokine decreased during ischemic ***,whether NK cells and their pro-inflammation cytokines participate ischemic injury keep *** highly suspect the destination of reductive NK cells is central neural *** our research we found NK cells infiltration in human and mouse ischemic brain,but the role of NK cells and the destructive cytokine they release in mediating neuronal death is not well *** determine whether NK cells and its maily releasing cytokine IFN-γ were involved in post-ischemic cerebral cell death,mice permanant middle cerebral artery occlusion（pMCAO） model was used to investigate the expression of NK cells and IFN-γ in infarct *** observed that NK cells infiltrated the ischemic infarct and reach a peak at 12h after ischemia and there was a similar trend in IFN-γ/NK cells levels in the ischemic region starting from 6 h until 4 day which correlated with a similar trend of chemokines ***-culture experiments showed that NK cells promoted neural necrosis via IFN-γ.After immunofluorescence analysis and Mean fluorescence intensity detection by FACS of CXCR3,it was showed that NK cells were absorbed by IP-10 via the linkage of CXCR3 and has dose *** assay and permeability experiment showed the blood-brain barrier disintegration after NK cells addition and the chemotaxis of IP-10 can aggravate the injury *** on the above findings we support our hypothesis that NK cells participate in cerebral ischemia and promote neural cells necrosis vie IFN-γ and the chemotactic of IP-10 also intensify the injury caused by NK to blood-brain barrier with the linkage of CXCR3.

Background:Simiaowan(SMW),a well-known traditional Chinese medicine,has been employed to treat hyperuricemia(HUA)and gout for ***,the bioactive components and underlying mechanisms have not been *** objective of this study was to identify the active components and potential mechanisms of SMW by integrating pharmacological experimentation,serum pharmacochemistry,network pharmacology and molecular ***:HUA rats modelling by high-fat/high-sugar diet and potassium oxonate/adenine oral administration were used to evaluate the pharmacodynamic effects of ***-Q-Exactive-MS/MS was employed to detect the bioactive components present in SMW-containing *** pharmacology and molecular docking were utilized to elucidate the potential targets and underlying ***:SMW effectively ameliorated HUA rats via the inhibition of uric acid(UA)production,promotion of UA excretion,improvement of lipid and glucose metabolic abnormalities,antioxidant,anti-inflammatory and anti-insulin resistance effects.A total of 73 compounds detected in SMW-containing serum were identified as potential active components,with alkaloids,flavonoids,organic acids,and terpenoids emerging as the primary active *** 203 corresponding targets were obtained as SMW anti-HUA/gout targets,which mainly participated in apoptosis,insulin resistance,TNF,PI3K-Akt,HIF-1,NF-κB,MAPK,IL-17 and TLR signaling *** docking indicated that active compounds(***,phellodendrine,quercetin,formononetin,ferulic acid)had superior binding abilities to the key targets(*** carrier family 22 member 12(URAT1),solute carrier family 22 member 6(OAT1),ATP-binding cassette sub-family G member 2(ABCG2),solute carrier family 2,facilitated glucose transporter member 9(GLUT9),xanthine dehydrogenase/oxidase(XDH),transcription factor p65(RELA),toll-like receptor 4(TLR4),prostaglandin G/H synthase 2(PTGS2),caspase-3(CASP3),insulin(INS)).Conclusion:S

High-quality Zn-doped CdSe core-shell nanocrystals were successfully prepared by incorporating a stoichiometric amount of Zn precursor into the CdSe reaction system, in which the Se precursor was excess and an Se-rich surface was formed. By injecting different amounts of Zn precursor, the core-shell nanocrystals demonstrated by the emission spectra were formed. The obtained Zn-doped CdSe nanocrystals exhibit a photoluminescence efficiency from 30% to 85%, which is comparable to those for the reported CdSe/ZnS, CdSe/CdS in the literature. In particular, a shell ZnSe layer with different thicknesses of ZnSe can be formed in this experiment by only changing the amount of Zn precursor added, which is simple and effective.

WO2 and TiO2 colMds were synthesized by the hydrolysis technique and part of the TiO2 colloid was treated by means of the hydrothermal method. The photochromic performances of the resulting materials obtained via combining the WO3 colloid with the treated TiO2 colloid and the non-treated TiO2 colloid, respectively, are very different. The TiO2 colloid without hydrothermal treatment can effectively improve the photochromic performance of the WO3colloid. The TiO2 nanoparticles were investigated in detail by XRD, TEM, surface photovohage spectra（SPS） and field-induced surface photovoltage spectrometry（FISPS）. The photochromism mechanism of WO3 colloid is discussed.

The longitudinal ultrasonic velocity （V1）, attenuation （α1）, magnetization and resistivity of single phase polycrystalline La1/3Sr2/3CoO3 were measured as a function of temperature from 20 K to 300 K. The resistivity shows metallic behavior in the whole temperature range and a kink at 235 K was observed, which coincides with the ferromagnetic transition temperature （Tc）. As the temperature cools down from Tc, the V1 softens conspicuously at beginning and reaches a minimum at 120 K. After that the V1 dramatically stiffens below 120 K accompanied by a wide attenuation peak. The analysis of the results suggests that these ultrasonic anomalies ;nay correspond to local lattice distortions via the Jahn-Teller effect of intermediate spin Co^3＋.

The nature of the exergy consumption of district heating(DH) systems can not be explained clearly using the first law of *** analysis method was used.A case study based on a DH system in Inner Mongolia,China,was carried *** impact of operating parameters and design parameters on the energy quality of circulating water and exergy losses of DH systems during heat distribution was *** show that the energy quality of circulating water and exergy losses of DH systems during heat distribution could be reduced by decreasing the indoor temperature or increasing radiator *** with other factors,the outdoor temperature and indoor temperature have a greater impact on the energy quality of circulating water,exergy losses of circulating water,and total exergy losses during heat *** the outdoor temperature varied by 10.00%,the average variation rates of such parameters were 85.12%,90.02%,and 64.60%,*** the outdoor temperature was 273.00 K and indoor temperature varied by 50.00%,the average variation rates of such parameters were 83.88%,99.34% and 32.87%,*** can be observed that the energy quality and exergy losses of DH systems can be reduced in the operation process.

The scheduling of parallel machines and the optimization of multi-line systems are two hotspots in the field of complex manufacturing *** the two problems are considered simultaneously,the resulting problem is much more complex than either of *** sufficient training data for conventional data-based optimization approaches is difficult because of the high diversity of system ***,optimization of multi-line systems with alternative machines requires a simple mechanism and must be minimally dependent on historical *** define a general multi-line system with alternative machines,this study introduces the capability vector and matrix and the distribution vector and matrix.A naive optimization method is proposed in accordance with classic feedback control theory,and its key approaches are *** a reasonable target value is provided,the proposed method can realize closed-loop optimization to the selected objective *** studies are performed on a real 5/6-inch semiconductor wafer manufacturing facility and a simulated multi-line system constructed on the basis of the MiniFAB *** show that the proposed method can effectively and efficiently optimize various objective *** method demonstrates a potential for utilization in multi-objective optimization.

Detection of a.c. magnetic field is consequential for many developments in physical and biological sciences,and various designs of magnetometer have been proposed recently. However, the large size of sensor and the extreme measurement conditions required strongly limit their application. It remains a challenge to reconstruct the vector of a.c. field with nanoscale spatial resolution using a single spin under ambient conditions. In this work, we choose the radio-frequency(RF) field as a typical case and realize the measurement of RF field based on a nitrogen-vacancy(NV)center in diamond. We build a solid sensor through measuring the effect of RF field on NV electron spin energy levels and the transition between them. Both of the phase and amplitude(including the transverse and longitudinal components) are measured by this new approach.
