Effects of simvastatin on hypertrophy and PTEN expression of rat cardiac myocytes

Objective To study the effects of simvastatin on the hypertrophy of cultured rat cardiac myocytes induced by serum and the role ofphosphatase and tensin homolog deleted on chromosome ten （PTEN） in the signal pathway. Methods Cultured neonatal Sprague- Dawley （SD） rat cardiac myocytes were treated with 15% fetal bovine serum, or without serum, or different consentrations of simvastatin. Image analysis system was used to measure the cardiac myocytes surface area. Protein synthesis of myocytes was measured via [3H]-leucine incorporation method. The expression level of atrial natriuretic peptide （ANP） mRNA in myocytes was determined with reverse transcription polymerase chain reaction （RT-PCR）. The mRNA and protein expression levels of PTEN in cardiac myocytes were investigated with RT-PCR and Western blot respectively. Results At 24 hours, cardiac myocytes surface area was significantly higher in 15% serum group （1611.16± 160.75 lam2） than in serum-free group （538.04±118.60 ±tm2, P〈0.01）. Simvastatin decreased the cell surface area in a concentration dependent manner. The cell surface area in 10-5 and 10-6 mol/L simvastatin groups were 799.84＋ 167.70 ±tm2 and 1076.88± 199.28 um2 respectively, which were both significantly lower than that in 15% fetal bovine serum group （P〈0.01）. Incorporation rate of [3H]-leucine was significantly higher in 15% fetal bovine serum group （2360± 106cpm/well） than that in serum-free group （1305±92 cpm/well, P〈0.01）. Incorporation rate of [3H]-leucine in 10.5 and 10.6 mol/L simvastatin groups were 1707±101 clam/well and 1962±125 cpm/well respectively, which were both lower than that in serum group （P〈0.01）. With the increase of simvastatin concentration, the expression level ofANP mRNA in cardiac myocytes was decreased gradually, which were 0.29±0.03 and 0.40-±0.03 respectively in 10.5 and 10-6 mol/L simvastatin groups, and significantly lower than that in serum group（0.60-±.03, P〈0.01）. AIM: To investigate the effect of N-desulfated heparin onhepatic/renal ischemia and reperfusion injury in ***: Using rat models of 60 minutes hepatic or renalischemia followed by 1 h,3 h,6 h and 24 h reperfusion,animalswere randomly divided into following groups,the shamoperated controls,ischemic group receiving only normalsaline,and treated group receiving N-desulfated heparin ata dose of 12 mg/kg at 5 minutes before reperfusion. P-selectin expression was detected in bepatic/renal tissueswith immunohistochemistry methodRESULTS: P-selectin expression, serum ALT, AST, BUN andCr levels were significantly increased during 60 minuteischemia and 1 h, 3 h, 6 h and 24 h reperfusion,while theincrement was significantly inhibited,and hepatic/renalpathology observed by light microscopy was remarkablyimproved by treatment with the N-desulfated ***,the heparin was found no effects on PT and ***: P-selectin might mediate neutrophilinfiltration and contribute to hepatic/renal ischemia andreperfusion. The N-desulfated heparin might preventhepatic/renal damage induced by ischemia and reperfusioninjury without significant anticoagulant activity.

Small-volume chylous ascites after laparoscopic radical gastrectomy for gastric cancer:Results from a large population-based sample

AIM:To report the incidence and potential risk factors of small-volume chylous ascites(SVCA)following laparoscopic radical gastrectomy(LAG).METHODS:A total of 1366 consecutive gastric cancer patients who underwent LAG from January 2008 to June 2011 were enrolled in this *** analyzed the patients based on the presence or absence of ***:SVCA was detected in 57(4.17%)patients,as determined by the small-volume drainage(range,30-100 m L/24 h)of triglyceride-rich *** univariate and multivariate analyses revealed that the total number of resected lymph nodes(LNs),No.8 or No.9 LN metastasis and N stage were independent risk factors for SVCA following LAG(Phospital stay,there was a significant difference between the groups with and without SVCA(P3-year disease-free and overall survival rates of the patients with SVCA were 47.4%and 56.1%,respectively,which were similar to those of the patients without SVCA(P>0.05).CONCluSION:SVCA following LAG developed significantly more frequently in the patients with≥32harvested LNs,≥3 metastatic LNs,or No.8 or No.9LN ***,which was successfully treated with conservative management,was associated with a prolonged hospital stay but was not associated with the prognosis.

Association between folate intake, serum folate levels and the risk of lung cancer： a systematic review and meta-analysis

Background Folate plays a critical role in nucleotide synthesis and DNA methylation, and was considered to be associated with anti-carcinogenesis. Results from studies that concern the relationship between the folate intake or serum folate levels and lung cancer risk showed no consistency, which requires our further comprehensive meta- analysis. Methods Systematic literature search was conducted to identify the relevant studies （published prior to February 2013） according to standard protocol. Estimated effects were calculated under both random-effects and fixed-effects models. Heterogeneity between studies and publication bias were also evaluated. Results A total of 4390 cases and 6138 controls from 6 case-control studies revealed a significant overall inverse association between folate intake and lung cancer risk （OR = 0.74, 95% CI = 0.65-0.84, P〈 0.001）. Summary of 1438 cases and 2582 controls from 4 case-control studies and 44 cases out of a cohort of 1988 participants suggested a marginal association without significance （OR = 0.78, 95% CI = 0.60-1.02, P = 0.075） between high serum folate levels and less lung cancer susceptibility; however, subgroup analysis about population-based case-control studies showed that high serum folate levels significantly associated with the reduced lung cancer risk （OR = 0.76, 95% CI = 0.58-1.00, P = 0.048）. Conclusion Higher folate intake can be a protective factor against lung cancer risk, and higher serum folate level is probably associated with reduced lung cancer risk in marginal manner, though more studies are warranted to confirm these associations.

Activation and reconstruction of the spinal cord circuitry is important for improving motor function following spinal cord *** conducted a case series study to investigate motor function improvement in 14 patients with chronic spinal cord injury treated with 4 weeks of unilateral(right only)cortical intermittent theta burst stimulation combined with bilateral magnetic stimulation of L3-L4 nerve roots,five times a *** resting motor evoked potential amplitude was increased,central motor conduction time on the side receiving cortical stimulation was significantly decreased,and lower extremity motor score,Berg balance score,spinal cord independence measure-III score,and 10 m-walking speed were all increased after *** resting motor evoked potential amplitude was positively correlated with lower extremity motor score after 4 weeks of *** findings suggest that cortical intermittent theta burst stimulation combined with precise root stimulation can improve nerve conduction of the corticospinal tract and lower limb motor function recovery in patients with chronic spinal cord injury.

AIM: To assess the effects of dihydromyricetin(DHM) as a hepatoprotective candidate in reducing hepatic injury and accelerating hepatocyte proliferation after carbon tetrachloride(CCl4) ***: C57 BL/6 mice were used in this study. Mice were orally administered with DHM(150 mg/kg) for 4 d after CCl4 treatment. Serum and liver tissue samples were collected on days 1, 2, 3, 5 and 7 after CCl4 treatment. The anti-inflammatory effect of DHM was assessed directly by hepatic histology detection and indirectly by serum levels of aspartate aminotransferase(AST), alanine aminotransferase(ALT), albumin, and superoxide dismutase(SOD). Inflammatory cytokines, such as interleukin(IL)-1β, IL-6 and tumor necrosis factor-α(TNF-α), were detected using ELISA kits. Proliferating cell nuclear antigen(PCNA) staining was used to evaluate the role of DHM in promoting hepatocyte proliferation. Objective To compare and analyze the therapeutic effect of temozolomide (TMZ)chemotherapy with radiotherapy and that of MV (VM-26+Me-CCNU)chemotherapy with radiotherapy in the synchronous treatment of postoperative malignant brain *** A total of 104 postoperative malignant brain glioma patients were randomly assigned to the treatment group (TMZ chemotherapy with radiotherapy synchronous treatment group)or the control group (MV chemotherapy with radiotherapy synchronous treatment group).There were 52 cases in the treatment group (30 cases of class III and 22 cases of class IV)and 52 cases in the control group (32 cases of class III and 20 cases of class IV).With the head conformal radiotherapy DT 60 Gy/30 fx/42 d,the treatment group was also administrated with TMZ 75 mg/m2 daily until the end of radiotherapy,followed by adjuvant TMZ chemotherapy 150-200 mg/m2/d x 5 d every 28 days for 6 treatment *** control group received 1 treatment course of MV (VM-26 70 mg/m2/d x 3 d,Me-CCNU 100-150 mg/m2 x 1 d)following 30 Gy radiotherapy,and then adjuvant MV chemotherapy every 28 days for 6 treatment courses after *** The 1-year,2-year and 3-year survival rates were 86.5% (45/52),26.9 % (14/52)and 13.5% (7/52)respectively in the treatment group and 80.8% (42/52),17.3% (9/52)and 7.7% (4/52)in the control *** median survival time was 20 months and 15 months in the treatment and control groups *** were significant differences in the survival curve and median survival time between the two groups (P nant brain glioma patients,which is significantly different from the survival rate in patients with MV chemotherapy and radiotherapy.
