BPI_(700)-Fc∴1_(700) chimeric gene expression and its protective effect in a mice model of the lethal E. coli infection

Background Infections caused by gram-negative bacteria （GNB） often lead to high mortality in common clinical settings. The effect of traditional antibiotic therapy is hindered by drug-resistant bacteria and unneutralizable endotoxin. Few effective methods can protect high risk patients from bacterial infection. This study explored the protection of adeno-associated virus 2 （AAV2）-bacteriacidal permeability increasing protein 700 （BPI700） -fragment crystallizable gamma one 700 （Fcγ1700） chimeric gene transferred mice against the minimal lethal dose （MLD） of *** and application of gene therapy for bacterial infection. Methods After AAV2-BPI700-Fcγ1700 virus transfection, dot blotting and Western blotting were used to detect the target gene products in Chinese hamster ovary-K1 cells （CHO-Klcells）. Reverse transcription-polymerase chain reaction and immunohistochemical assay were carried out to show the target gene expression in mice. Modified BPI-enzyme linked immunosorbent assay was used to identify the target gene products in murine serum. The protection of BPI700-Fcγ1700 gene transferred mice was examined by survival rate after MLD E. coli challenge. Colony forming unit （CFU） count, limulus amebocyte lysate kit and cytokine kit were used to quantify the bacteria, the level of endotoxin, and proinflammatory cytokine. Results BPI1-99-Fcγ1 protein was identified in the CHO-K1 cell culture supernatant, injected muscles and serum of the gene transferred mice. After MLD E. coli challenge, the survival rate of AAV2-BPI700-Fcγ1700 gene transferred mice （36.7%） was significantly higher than that of AAV2-enhanced green fluorescent protein （AAV2- EGFP） gene transferred mice （3.3%） and PBS control mice （5.6%）. The survival rate of AAV2-BPI700-Fcγ1700 gene transferred mice treated with cefuroxime sodium was 65.0%. Effect of microRNA-208a on mitochondrial apoptosis of cardiomyocytes of neonatal rats

Objective: To explore the effect and mechanism of microRNA-208a(mi R-208a) in the mitochondrial apoptosis of cardiomyocytes of neonatal rats. Methods: The primary cultured cardiomyocytes of neonatal rats were added into the hypoxia incubator for the hypoxia induction. The overexpression system for mi R-208 a of cardiomyocytes of neonatal rats was built. The l ow cytometry assay was employed to detect the incidence of apoptosis in the overexpressed mi R-208 a. The mitochondrial staining technique was used to detect the change in the mitochondrial morphology of over-expressed mi R-208 a. The bioinformatic analysis was chosen to analyze and predict the target gene of mi R-208 a. Results: Firstly, the primary culture system of cardiomyocytes of neonatal rats was successfully built. The mi R-208 a was over-expressed in cardiomyocytes of neonatal rats by mi R-208 a Mimics. Results of flow cytometry assay showed that the over-expressed mi R-208 a could signii cantly reduce the incidence of apoptosis; while results of mitochondrial staining indicated the change in the mitochondrial morphology of over-expressed mi R-208 a and the mitochondrialfission process was inhibited. In conclusion, it was supposed that mi R-208 a could inhibit the activation of mitochondrialfission process to keep the cardiomyocytes from apoptosis. Conclusions: The over-expressed mi R-208 a can reduce the incidence of apoptosis in the cardiomyocytes of neonatal rats, signii cantly change the mitochondrial morphology and inhibit the mitochondrial fission process.

Real-time monitoring of raster temperature distribution and width anomalies in fused filament fabrication process

The aim of this study is to monitor the raster temperature distribution and width anomalies in a fused filament fabrication(FFF)process by an infrared(IR)array *** achieve this goal,two experiments were conducted on a desktop FFF *** the first experiment,three normal samples with different raster widths were fabricated,and thermal images of the newly deposited rasters were collected during the *** process the low-resolution images,a segmentation-based image processing method was *** temperature distributions along the horizontal direction of the raster section and along the raster length were *** temperature features that could indicate the raster widths were extracted and then fed to recognition models for training and *** classification performance of the models were evaluated based on the *** models with high F1-scores could be used to recognise width anomalies *** the second experiment,an abnormal sample with raster width anomalies was *** temperature features were extracted from the collected experimental *** obtained features were then fed to the built and evaluated models to recognise the width anomalies *** effectiveness of the monitoring method was validated by comparing the recognition results with the actual optical *** support vector machine(SVM)and k-nearest neighbour(KNN)were adopted to build the recognition *** F1-score and online recognition results of the models were *** comparison study shows that SVM is more suitable for our situation than KNN.A method for monitoring the temperature distribution and width anomalies of the FFF raster is provided in this *** the best of the authors’knowledge,this is the first study to explore the actual temperature distribution along the horizontal direction of the raster section,and the first study to monitor the width anomalies of the raster in the FFF process.

Lightweight Malicious Code Classification Method Based on Improved Squeeze Net

With the growth of the Internet,more and more business is being done online,for example,online offices,online education and so *** this makes people’s lives more convenient,it also increases the risk of the network being attacked by malicious ***,it is important to identify malicious codes on computer systems ***,most of the existing malicious code detection methods have two problems:(1)The ability of the model to extract features is weak,resulting in poor model performance.(2)The large scale of model data leads to difficulties deploying on devices with limited ***,this paper proposes a lightweight malicious code identification model Lightweight Malicious Code Classification Method Based on Improved SqueezeNet(LCMISNet).In this paper,the MFire lightweight feature extraction module is constructed by proposing a feature slicing module and a multi-size depthwise separable convolution *** feature slicing module reduces the number of parameters by grouping *** multi-size depthwise separable convolution module reduces the number of parameters and enhances the feature extraction capability by replacing the standard convolution with depthwise separable convolution with different convolution kernel *** addition,this paper also proposes a feature splicing module to connect the MFire lightweight feature extraction module based on the feature reuse and constructs the lightweight model *** malicious code recognition accuracy of LCMISNet on the BIG 2015 dataset and the Malimg dataset reaches 98.90% and 99.58%,*** proves that LCMISNet has a powerful malicious code recognition *** addition,compared with other network models,LCMISNet has better performance,and a lower number of parameters and computations.

Using the Cerenkov second harmonic generation, 478 nm blue and 402nm violet light are generated by direct frequency doubling of 956 and 804 nm diode lasers in a same proton exchanged MgO:LiNbO3 waveguide. Under the low fundamental guide wave power of less than 1mW, the estimated value of the output blue and violet light power is in the order of 0.1-0.01μW.

Dynamic CCT Diagram of Automobile Beam Steel With High Strength Produced by FTSR Technology

The dynamic continuous cooling transformation（CCT）diagram and phase transformation rules of 510 MPa automobile beam steel,which is produced by a continuous casting of thin slab of FTSR technology in Tangshan Iron and Steel ***.,are researched by thermal simulation *** microstructure characteristics of the beam steel under different test conditions are studied by means of optical microscope and scanning electron *** test results show that the critical temperatures of phase transformation Ar3 and Ar1 will all decrease with the increase of the cooling *** the cooling rate is lower than 20 ℃·s-1,the ferrite and pearlite phase transformations are the main parts;when the cooling rate is higher than 20 ℃·s-1,the bainite phase ***,the microstructures of 510 MPa automobile beam steel produced by FTSR technology are also studied,and the results are basically in accordance with the CCT diagram gained from the test.

全球性的气候变化严重地威胁着自然生态环境间的平衡。在影响生态系统功能的诸多因素中,太阳紫外辐射的增强逐渐成为最受关注的全球性环境问题之一。太阳紫外辐射对地球生物的生命过程有着举足轻重的影响。生物体一旦吸收了高能量的紫外辐射,则可对其各种生理过程产生影响,打破内稳态,尤其是紫外辐射对DNA的损伤作用,是诱导一系列生物效应的主要原因。本文综述了近年来与鱼类及水生无脊椎动物有关的紫外辐射研究。从紫外辐射对生物不同发育时期的影响、生物对紫外线响应方式、紫外辐射与其他因素复合影响三个层面进行总结。通过总结紫外辐射对鱼类及水生无脊椎动物的影响,可为预测紫外辐射在水生生态系统中的作用提供助力,为海洋生物对环境变化的生理响应研究提供参考。
