Differentiation of human amniotic epithelial cells into corneal epithelial-like cells in vitro

·AIM: To explore the feasibility that human amniotic epithelial cells (hAECs) have the potential to differentiate into corneal epithelial -like cells under the microenvironment replicated by spontaneously immortalized human corneal epithelial cells (S-ihCECs). ·METHODS: hAECs were isolated by enzyme digestion, and flow cytometry was used to analysis the expression of CD29/90/166/73/34 and HLA -DR. Recovered and cultured S -ihCECs, immunocytochemistry was used to detect the expression of CK3/12. The proliferation of S - ihCECs handled by different concentrations of mitomycin was detected by CCK -8. The proliferation of hAECs cultured by S-ihCECs culture media collected at different time was analyzed by CCK -8. After filtered out the optimal conditions, we collected S-ihCECs culture media for 5 days, then prepared conditioned medium to incubate hAECs, inverted phase contrast microscope and scanning electron microscope were used to observe the change of morphology in hAECs. Quantitative real -time reverse transcription -polymerase chain reaction (QRT - PCR) was carried out to evaluate the expression of Oct - 4, NANOG, PAX6, and CK12 in the differentiation period. Immunocytochemistry and western bloting were used to detect the expression of CK3/12. ·RESULTS: The culture media collected every 12h, from 20μg/mL mitomycin pretreatment S -ihCECs could significantly promote the proliferation of hAECs. In the period of differentiation, the morphology of differentiated hAECs was obviously different compared with the control group, and the distinctive CK3/12 for corneal epithelial cells was detected.·CONCLUSION: This study showed that hAECs can differentiate into corneal epithelial -like cells by replication of the corneal epithelial microenvironment, using the culture media collected from S -ihCECs, and it is possible that S -ihCECs culture media could be used in corneal tissue engineering. ·

Due to the heat pipes’ transient conduction,phase change and fluid dynamics during cooling/heating with high frequency charging/discharging of batteries,it is crucial to investigate in depth the experimental dynamic thermal characteristics in such complex heat transfer processes for more accurate thermal analysis and design of a BTMS. In this paper,the use of ultra?thin micro heat pipe(UMHP) for thermal management of a lithium?ion battery pack in EVs is explored by experiments to reveal the cooling/heating characteristics of the UMHP pack. The cooling performance is evaluated under di erent constant discharging and transient heat inputs conditions. And the heating e ciency is assessed under several sub?zero temperatures through heating films with/without UMHPs. Results show that the pro?posed UMHP BTMS with forced convection can keep the maximum temperature of the pack below 40 °C under 1 ~ 3 C discharging,and e ectively reduced the instant temperature increases and minimize the temperature fluctuation of the pack during transient federal urban driving schedule(FUDS) road conditions. Experimental data also indicate that heating films stuck on the fins of UMHPs brought about adequate high heating e ciency comparing with that stuck on the surface of cells under the same heating power,but has more convenient maintenance and less cost for the BTMS. The experimental dynamic temperature characteristics of UMHP which is found to be a high?e cient and low?energy consumption cooling/heating method for BTMSs,can be performed to guide thermal analysis and optimiza?tion of heat pipe BTMSs.

Background Lumiracoxib is a highly selective cyclooxygenase-2 （COX-2） inhibitor with antiinflammatory, analgesic and antipyretic activities comparable with class specific drugs, but with much improved gastrointestinal safety. No studies have examined lumiracoxib for antitumorigenic activity on human nonsmall cell lung cancer cell lines in vitro or its possible molecular mechanisms. Methods The antiproliferative effect of lumiracoxib alone or combined with docetaxol on A549 and NCI-H460 lines was assessed by 3-（4, 5-dimethylthiazol-2-yl）-2, 5-diphenyltetrazolium bromide （MTT） assay. Drug-drug interactions were analyzed using the coefficient of drug interaction （CDI） to characterize the interactions as synergism, additivity or antagonism. Morphological changes were observed by acridine orange fluorescent staining. Extent of apoptosis was determined by flow cytometry. Results Lumiracoxib （15-240 pmol/L） has an inhibitory effect on the proliferation of A549 and NCI-H460 cell lines in concentration- and time-dependent manners with the ICso values of 2597 pmol/L and 833 pmol/L, respectively. The synergistic effect was prominent when lumiracoxib （15-240 pmoVL） was combined with docetaxol （0.2-2 pmol/L）（CDI 〈1）. Fluorescent staining showed that lumiracoxib could induce apoptosis in A549 and NCI-H460 cells. Lumiracoxib treatment also caused an increase of the sub-G1 fraction in each cell line and resulted in an increase of G0/Gl-phase cells and a decrease of S-phase cells. Conclusions Lumiracoxib had antiproliferative effect on the human nonsmall cell lung cancer cell lines A549 and NCI-H460 and had a significant synergy with docetaxol, which may be related to apoptotic induction and cell cycle arrest.

Subsurface macro-inclusions and solidified hook character in aluminum-killed deep-drawing steel slabs

Subsurface macro-inclusions and hooks are detrimental to the surface quality of deep-drawing steel sheets. However, little is known about the relationship between macro-inclusions and hooks. Thus, in this work, two ultralow carbon （ULC） steel slabs and two low carbon （LC） aluminum-killed steel slabs were sampled to study the relationship between hooks and subsurface macro-inclusions, which were detected on the cross-sections of steel samples with an area of 56058 mm2 using an automated scanning electron microscopy/energy-disper-sive X-ray spectroscopy system. Results show that subsurface inclusions larger than 200 μm were almost entrapped by hook structures, whereas the location of other inclusions smaller than 200μm had no obvious dependence on the location of solidified hooks. Furthermore, the number density （ND） of subsurface inclusions larger than 200μm decreased from 0.02 to 0 cm-2 in ULC steel as the mean hook depth decreased from 1.57 to 1.01 mm. Similar trends were also observed in LC steel. In addition, the detected inclusions larger than 200μm were concentrated in the region near the slab center （3/8 width-5/8 width）, where hook depths were also larger than those at any other locations. Therefore, minimizing the hook depth is an effective way to reduce inclusion-induced sliver defects in deep-drawing steels.

It well known that vehicle detection is an important component of the field of object detection.However,the environment of vehicle detection is particularly sophisticated in practical processes.It is comparatively difficult to detect vehicles of various scales in traffic scene images,because the vehicles partially obscured by green belts,roadblocks or other vehicles,as well as influence of some low illumination weather.In this paper,we present a model based on Faster R-CNN with NAS optimization and feature enrichment to realize the effective detection of multi-scale vehicle targets in traffic scenes.First,we proposed a Retinex-based image adaptive correction algorithm(RIAC) to enhance the traffic images in the dataset to reduce the influence of shadow and illumination,and improve the image quality.Second,in order to improve the feature expression of the backbone network,we conducted Neural Architecture Search(NAS) on the backbone network used for feature extraction of Faster R-CNN to generate the optimal cross-layer connection to extract multi-layer features more effectively.Third,we used the object Feature Enrichment that combines the multi-layer feature information and the context information of the last layer after cross-layer connection to enrich the information of vehicle targets,and improve the robustness of the model for challenging targets such as small scale and severe occlusion.In the implementation of the model,K-means clustering algorithm was used to select the suitable anchor size for our dataset to improve the convergence speed of the model.Our model has been trained and tested on the UN-DETRAC dataset,and the obtained results indicate that our method has art-of-state detection performance.

With the shortage of lithium resources,sodiumion batteries（SIBs） are considered one of the most promising candidates for lithium-ion batteries.P2-type and O3-type layered oxides are one of the few cathodes that can access high energy density.However,they usually exhibit structural change,capacity decay,and slow Na ion kinetic.Herein,we present layered ternary-phase cathodes with P2,P3 and O3 phases by a lattice doping strategy,which is demonstrated by X-ray diffraction（XRD） refinement.Combining the characteristics of P2,P3 and O3 phases,the layered composites show performance improvement during long-term battery cycling.In particular,Na0.7Li0.1Co0.3-Fe0.3Mn0.3O2（NLCFM） delivers a reversible capacity of120.1 mAh·g-1at 0.1C(1.0C=175 mA·g-1) with a superior capacity retention of 72.5% after 1000 cycles at10.0C.This work offers insights into the development of advanced cathode materials for SIBs.

BACKGROUND Colorectal cancer(CRC)ranks among the most prevalent malignant tumors globally.Recent reports suggest that Fusobacterium nucleatum(F.nucleatum)contributes to the initiation,progression,and prognosis of CRC.Butyrate,a short-chain fatty acid derived from the bacterial fermentation of soluble dietary fiber,is known to inhibit various cancers.This study is designed to explore whether F.nucleatum influences the onset and progression of CRC by impacting the intestinal metabolite butyric acid.AIM To investigate the mechanism by which F.nucleatum affects CRC occurrence and development.METHODS Alterations in the gut microbiota of BALB/c mice were observed following the oral administration of F.nucleatum.Additionally,DLD-1 and HCT116 cell lines were exposed to sodium butyrate(NaB)and F.nucleatum in vitro to examine the effects on proliferative proteins and mitochondrial function.RESULTS Our research indicates that the prevalence of F.nucleatum in fecal samples from CRC patients is significantly greater than in healthy counterparts,while the prevalence of butyrate-producing bacteria is notably lower.In mice colonized with F.nucleatum,the population of butyrate-producing bacteria decreased,resulting in altered levels of butyric acid,a key intestinal metabolite of butyrate.Exposure to NaB can impair mitochondrial morphology and diminish mitochondrial membrane potential in DLD-1 and HCT116 CRC cells.Consequently,this leads to modulated production of adenosine triphosphate and reactive oxygen species,thereby inhibiting cancer cell prolif-eration.Additionally,NaB triggers the adenosine monophosphate-activated protein kinase(AMPK)signaling pathway,blocks the cell cycle in HCT116 and DLD-1 cells,and curtails the proliferation of CRC cells.The combined presence of F.nucleatum and NaB attenuated the effects of the latter.By employing small interfering RNA to suppress AMPK,it was demonstrated that AMPK is essential for NaB’s inhibition of CRC cell proliferation.CONCLUSION F.nucleatum can promote cancer progression through

We theoretically investigate the Ruderman–Kittel–Kasuya–Yosida(RKKY) interaction in helical higher-order topological insulators(HOTIs), revealing distinct behaviors mediated by hinge and Dirac-type bulk carriers. Our findings show that hinge-mediated interactions consist of Heisenberg, Ising, and Dzyaloshinskii–Moriya(DM) terms, exhibiting a decay with impurity spacing z and oscillations with Fermi energy εF. These interactions demonstrate ferromagnetic behaviors for the Heisenberg and Ising terms and alternating behavior for the DM term. In contrast, bulk-mediated interactions include Heisenberg, twisted Ising, and DM terms, with a conventional cubic oscillating decay. This study highlights the nuanced interplay between hinge and bulk RKKY interactions in HOTIs, offering insights into designs of next-generation quantum devices based on HOTIs.
