FAM53B promotes pancreatic ductal adenocarcinoma metastasis by regulating macrophage M2 polarization

BACKGROUND Our study investigated the role of FAM53B in regulating macrophage M2 polarization and its potential mechanisms in promoting pancreatic ductal adenocarcinoma(PDAC)metastasis.AIM To further investigate the role of FAM53B in regulating macrophage M2 polarization and its potential mechanism in promoting PDAC metastasis.Our goal is to determine how FAM53B affects macrophage M2 polarization and to define its underlying mechanism in PDAC metastasis.METHODS Cell culture and various experiments,including protein analysis,immunohisto-chemistry,and animal model experiments,were conducted.We compared FAM53B expression between PDAC tissues and healthy tissues and assessed the correlation of FAM53B expression with clinical features.Our study analyzed the role of FAM53B in macrophage M2 polarization in vitro by examining the expression of relevant markers.Finally,we used a murine model to study the role of FAM53B in PDAC metastasis and analyzed the potential underlying mechanisms.RESULTS Our research showed that there was a significant increase in FAM53B levels in PDAC tissues,which was linked to adverse tumor features.Experimental findings indicated that FAM53B can enhance macrophage M2 polarization,leading to increased anti-inflammatory factor release.The results from the mouse model further supported the role of FAM53B in PDAC metastasis,as blocking FAM53B prevented tumor cell invasion and metastasis.CONCLUSION FAM53B promotes PDAC metastasis by regulating macrophage M2 polarization.This discovery could lead to the development of new strategies for treating PDAC.For example,interfering with the FAM53B signaling pathway may prevent cancer spread.Our research findings also provide important information for expanding our understanding of PDAC pathogenesis.

Microstructural Evolutions and Mechanical Properties of Energetic Al_(1) (TiZrNbTaMoCr)_(15) High-Entropy Alloys

The present study focuses on investigating the microstructural evolutions and mechanical properties of energetic Al_(1)(TiZrNbTaMoCr)_(15) refractory high-entropy alloys with the different heat treatments at low Al content state. It is found that even with a reduction in the Al content, the strength of these alloys remains unaffected at room temperature and high temperature, while the plasticity improves significantly. In particular, the coherent BCC/B_(2) microstructure with needle-like B_(2) nanoprecipitates dispersed into the BCC matrix is formed in 873 K-aged Al_(1)Ti_(6)Zr_(2)Nb_(3)Ta_(3)Mo_(0.5)Cr_(0.5) alloy. Therefore, this alloy exhibits the highest compression yield strength (σ_(YS) = 1333 and 717 MPa) at room temperature and 1073 K, respectively. After 973 K-aged, the coherent BCC/B_(2) microstructure underwent destabilization, and the B_(2) phase transforms into the brittle Zr5Al3 phase which then coarsens and dominates the microstructure of S3-AlTa_(3) alloy after 1073 K-aged. Moreover, these current alloys exhibit exceptionally high theoretical exothermic enthalpy (ΔH), surpassing 11606 J·g^(−1), which highlights their significant potential as innovative high-performance energetic structural materials.

The effects of various factors on spontaneous imbibition in tight oil reservoirs

Slickwater fracturing fluids have gained widespread application in the development of tight oil reservoirs. After the fracturing process, the active components present in slickwater can directly induce spontaneous imbibition within the reservoir. Several variables influence the eventual recovery rate within this procedure, including slickwater composition, formation temperature, degree of reservoir fracture development, and the reservoir characteristics. Nonetheless, the underlying mechanisms governing these influences remain relatively understudied. In this investigation, using the Chang-7 block of the Changqing Oilfield as the study site, we employ EM-30 slickwater fracturing fluid to explore the effects of the drag-reducing agent concentration, imbibition temperature, core permeability, and core fracture development on spontaneous imbibition. An elevated drag-reducing agent concentration is observed to diminish the degree of medium and small pore utilization. Furthermore, higher temperatures and an augmented permeability enhance the fluid flow properties, thereby contributing to an increased utilization rate across all pore sizes. Reduced fracture development results in a lower fluid utilization across diverse pore types. This study deepens our understanding of the pivotal factors affecting spontaneous imbibition in tight reservoirs following fracturing. The findings act as theoretical, technical, and scientific foundations for optimizing fracturing strategies in tight oil reservoir transformations.

One-step construction of strongly coupled Co_(3)V_(2)O_(8)/Co_(3)O_(4)/MXene heterostructure via in-situ Co-F bonds for high performance all-solid-state asymmetric supercapacitors

Co_(3)V_(2)O_(8)/Co_(3)O_(4)/Ti_(3)C_(2)T_(x) composite was easily synthesized via one-step succinct-operated hydrothermal process.The interconnected Co_(3)V_(2)O_(8)/Co_(3)O_(4) nanowires network can in-situ grow and anchor on the surface of Ti_(3)C_(2)T_(x) via the strong Co-F bonds and contribute tremendously to depress Ti_(3)C_(2)T_(x) self-restacking.Profiting from the synergistically interplayed effect among the multiple interfaces and high conductivity of Ti_(3)C_(2)T_(x) as well as outstanding stability of the as-designed nanostructure,the optimum Co_(3)V_(2)O_(8)/Co_(3)O_(4)/Ti_(3)C_(2)T_(x)electrode reaches a commendable specific capacitance(up to 3800 mF·cm^(−2)),great rate capability(80%capacitance retention after 20-times current increasing),and preeminent cycling stability(95.4%/85.5%retention at 7000th/20,000th cycle).Moreover,the all-solid-state asymmetric supercapacitor based on Co_(3)V_(2)O_(8)/Co_(3)O_(4)/Ti_(3)C_(2)T_(x) and active carbon can deliver a high energy density of 84.0μWh·cm^(−2) at the power energy of 3.2 mW·cm^(−2),and excellent cycling durability with 87.0%of initial capacitance retention upon 20,000 loops.This work provides a practicable pathway to tailor MXene-based composites for high-performance supercapacitor.

Collision risk assessment of reduced aircraft separation minima in procedural airspace using advanced communication and navigation

With the advancement of Communication,Navigation and Surveillance(CNS)technolo-gies such as space-based Automatic Dependent Surveillance-Broadcast/Contract(ADS-B/C),large separation minima may be reduced in procedural airspaces.It is of great significance to know the upper limit of the Reduced Separation Minima(RSM)for a procedural airspace and the corre-sponding consequences on collision risk with specifics of the advanced ADS-B and control interven-tion model.In this work,an interactive software is first developed for collision risk estimation.This software integrates the International Civil Aviation Organization(ICAO)collision risk models for lateral and longitudinal collision risk calculation for the Singapore procedural airspace.Results demonstrate that the lateral and longitudinal collision risk of Singapore procedural airspace with respect to current control procedures meets the ICAO Target Level of Safety(TLS)standard.Moreover,the feasibility of reducing the horizontal separations implemented in the Singapore pro-cedural airspace with respect to advanced CNS techniques is investigated.It is found that if advanced CNS technologies are applied,then the current 50-NM lateral and longitudinal separa-tion standards can be reduced to 22 NM(1 NM=1.825 km)and 20 NM,respectively,to meet the TLS standards based on current demand.A method is then devised to expand the traffic demand by p for p∈[10%,200%].It is found that the minimum lateral and longitudinal separa-tions can be reduced from 50 NM to be within the range of[23,31]NM,and 20 NM,respectively,for p∈[10%,200%],while the collision risk still meets the TLS standards.

Establishing and clinically validating a machine learning model for predicting unplanned reoperation risk in colorectal cancer

BACKGROUND Colorectal cancer significantly impacts global health,with unplanned reoperations post-surgery being key determinants of patient outcomes.Existing predictive models for these reoperations lack precision in integrating complex clinical data.AIM To develop and validate a machine learning model for predicting unplanned reoperation risk in colorectal cancer patients.METHODS Data of patients treated for colorectal cancer(n=2044)at the First Affiliated Hospital of Wenzhou Medical University and Wenzhou Central Hospital from March 2020 to March 2022 were retrospectively collected.Patients were divided into an experimental group(n=60)and a control group(n=1984)according to unplanned reoperation occurrence.Patients were also divided into a training group and a validation group(7:3 ratio).We used three different machine learning methods to screen characteristic variables.A nomogram was created based on multifactor logistic regression,and the model performance was assessed using receiver operating characteristic curve,calibration curve,Hosmer-Lemeshow test,and decision curve analysis.The risk scores of the two groups were calculated and compared to validate the model.RESULTS More patients in the experimental group were≥60 years old,male,and had a history of hypertension,laparotomy,and hypoproteinemia,compared to the control group.Multiple logistic regression analysis confirmed the following as independent risk factors for unplanned reoperation(P<0.05):Prognostic Nutritional Index value,history of laparotomy,hypertension,or stroke,hypoproteinemia,age,tumor-node-metastasis staging,surgical time,gender,and American Society of Anesthesiologists classification.Receiver operating characteristic curve analysis showed that the model had good discrimination and clinical utility.CONCLUSION This study used a machine learning approach to build a model that accurately predicts the risk of postoperative unplanned reoperation in patients with colorectal cancer,which can improve treatment decisions and prognosis.

A bifunctional wood membrane modified by MoS_(2)/covalent organic framework heterojunctions for effective solar-driven water evaporation and contaminant degradation

Interfacial solar evaporation technology is considered one of the most promising strategies for alleviating the scarcity of freshwater resources.However,solar-driven evaporation technology cannot eliminate the pollutants in the residual wastewater.To solve this problem,we have prepared a two-in-one solar-driven evaporation/photocatalysis system by decorating MoS_(2)/covalent organic framework(COF)heterojunctions on wood(MoS_(2)/COF-wood).Thanks to the unique porous structure of wood,it provides a strong guarantee for water transport and vapor release during the evaporation process.The introduction of MoS_(2) and COFs can promote the breaking of hydrogen bonds between water molecules,which leads to a significant decrease in the enthalpy of evaporation,achieving a water evaporation rate as high as 2.17 kg m^(-2)h^(-1)under 1 sun irradiation.Meanwhile,the resulting MoS_(2)/COF-wood exhibits good salt resistance and reusability.In addition,the heterojunctions formed between COFs and MoS_(2) can effectively inhibit charge carrier complexation and improve the photocatalytic degradation ability of pollutants(over 99%).This study highlights the construction strategy of bifunctional wood-based materials for freshwater production and wastewater remediation.

BACKGROUND Recently,a growing number of adolescents have been afflicted with mental disorders,with annual morbidity rates on the rise.This trend has been exacerbated by the global coronavirus disease 2019(COVID-19)pandemic,leading to a surge in suicide and self-harm rates among this demographic.AIM To investigate the impact of the COVID-19 pandemic on adolescent bipolar disorder(BD),along with the underlying factors contributing to heightened rates of suicide and self-harm among adolescents.METHODS A comprehensive statistical analysis was conducted utilizing clinical interviews and self-reports obtained from patients or their guardians.Diagnostic criteria for BDs were based on the Diagnostic and statistical manual of mental disorders,international classification of diseases-11,and the National institute of mental health research domain criteria.Statistical analyses were performed using SPSS 26.0 software,with significance set at P<0.05.RESULTS A cohort of 171 adolescents diagnosed with BD between January 1,2018,and December 31,2022,was included in the analysis.The gender distribution was 2.8:1(female to male),with ages ranging from 11 to 18 years old.Major factors contributing to adolescent BDs included familial influences,academic stress,genetic predisposition and exposure to school-related violence.Notably,a significant increase in suicide attempts and self-harm incidents was observed among adolescents with BD during the COVID-19 pandemic.Statistical analysis indicated that the pandemic exacerbated familial discord and heightened academic stress,thereby amplifying the prevalence of suicidal behavior and self-harm among adolescents.CONCLUSION The COVID-19 pandemic has exacerbated familial tensions and intensified the incidence of suicide and self-harm among adolescents diagnosed with BD.This study underscores the urgent need for societal,familial and educational support systems to prioritize the well-being of adolescents and offers valuable insights and guidelines for the prevention,diagnosis and treatment of ad

A Minimum Residual Based Gradient Iterative Method for a Class of Matrix Equations

In this paper, we present a minimum residual based gradient iterative method for solving a class of matrix equations including Sylvester matrix equations and general coupled matrix equations. The iterative method uses a negative gradient as steepest direction and seeks for an optimal step size to minimize the residual norm of next iterate. It is shown that the iterative sequence converges unconditionally to the exact solution for any initial guess and that the norm of the residual matrix and error matrix decrease monotonically. Numerical tests are presented to show the efficiency of the proposed method and confirm the theoretical results.

FLOWERING LOCUS T(FT)in Photosensitive Type Chrysanthemum Accelerates Flowering in Arabidopsis

FLOWERING LOCUS T(FT)is an important factor for integrating flowering signals through the photoperiod pathway,which significantly promotes flowering.Most chrysanthemum varieties are short-day plants,sensitive to the photoperiod,which limits their ornamental and annual production supply.In this study,we aim to analyze the nutrients and flowering-related genes of chrysanthemums with different photoperiod types and to clone and verify the function of the flowering-related gene CmFT.We found that the formation of floral buds requires the accumulation of starch while consuming soluble sugars and the expression patterns of flowering-related genes GIGANTEA(GI),CONSTANS(CO),and FT in C.morifolium‘Zilian’and C.morifolium‘Zihongtuogui’had a certain synchronization during floral buds differentiation according to our quantitative validation,and the expression levels of CmGI,CmCO and CmFT in C.morifolium‘Zihongtuogui’were higher than those in C.morifolium.‘Zilian’in the later stage of differentiation.CmFT was cloned from photosensitive chrysanthemums-C.morifolium‘Zihongtuogui’and polypeptide alignment and phylogenetic analysis showed that CmFT was clustered in FT-like subfamily.In further functional verification,we obtained two Arabidopsis transgenic lines.Our results showed that CmFT transgenic ft mutant lines can significantly accelerate flowering in Arabidopsis.Thus,we can initially confirm that CmFT plays an important role in promoting flowering,which may be the key reason for the photosensitivity of C.morifolium‘Zihongtuogui’.Overall,the results of this study are of great importance in revealing the flowering mechanism of different photoperiod types of chrysanthemums.
