Targeted regulation of neuroinflammation via nanobiosignaler for repairing the central nerve system injuries

Neuroinflammation,commonly associated with various central nervous system(CNS)diseases such as postoperative cognitive dysfunction(POCD),is primarily mediated by the disruption of biological signals in microglia.However,the effective treatment of CNS diseases remains an ongoing challenge as biological signals show limited microglia-targeting effect.In this study,taking advantage of the highly expressed lipoprotein receptor-related protein-1(LRP1)on the microglia,a nanobiosignal delivery system modified by LRP1 high-affinity peptide ligand RAP12(RAP:receptor-associated protein)was constructed to specifically regulate neuroinflammation via targeting microglia.The uptake of the RAP12 modified-nanobiosignaler by microglia increased significantly,indicating its microglia-targeting ability.Both in vitro/vivo studies proved that the“nanobiosignaler”significantly reduced the secretion of pro-inflammatory cytokines,induced specific M2(anti-inflammatory type)microglia differentiation,and remarkably alleviated cognitive function impairment in the mice model when compared with unmodified groups.It was indicated that the“nanobiosignaler”could target microglia to deliver the biological signal and inhibit the excessive activation of microglia.Overall,the cell-targeted biological signal transmission system inspired by“nanobiosignaler”has broad application prospects in the future.

Graphene oxide(GO)has been widely used in the modification of membranes due to its excellent properties,i.e.,huge specific surface area,good electrical conductivity,good hydrophilicity and various functional groups.The addition of GO in membranes were proved to exhibit improved properties in water permeability,molecular selectivity,membrane fouling mitigation and contaminants decomposition.Recently,the development of laminated GO in membranes achieved both high selectivity and high water permeability,conquering the limitations of conventional polymeric or inorganic membranes.By analyzing the separation mechanisms and the performance of GO composite membranes,this review systematically summarized the applications of GO composite membranes in three highlighted areas of environmental fields:desalination,gas separation and wastewater treatment,with challenges discussed faced with GO composite membranes.

Selective growth of semiconducting single-walled carbon nanotubes solely from carbon monoxide

The selective growth of semiconducting single-walled carbon nanotubes(s-SWCNTs)is of great importance in many high-end applications represented by nanoelectronics.Here,we developed a general approach to grow horizontally aligned s-SWCNT arrays on stable temperature(ST)-cut quartz with bimetallic catalysts using carbon monoxide(CO)as both catalyst reductant and single component carbon feedstock under atmospheric pressure.The disproportionation of CO produces not only carbon species for SWCNT growth but also CO_(2),which could act as an in-situ etchant to remove both amorphous carbon and metallic tubes.The employment of bimetallic catalyst and quartz substrate facilitates the selective etching by narrowing the diameter distribution of as-grown SWCNT arrays.At the optimized conditions,we realized the selective growth of horizontally aligned s-SWCNT arrays with the content above 97%using CoCu catalysts,confirmed by Raman characterization and electrical measurements of the fabricated field effect transistor devices.This CO-based process in selective growth of s-SWCNTs has demonstrated its feasibility and universality by the broad growth window and applicability for other bimetallic catalysts,such as FeCu and CoMn.It possesses a practical potential in obtaining semiconducting channel materials for the scalable fabrication of CNT-based devices.

Multiscale design and biomechanical evaluation of porous spinal fusion cage to realize specified mechanical properties

Backgrou nd Dense titanium(Ti)fusion cages have been commonly used in transforaminal lumbar interbody fusion.However,the stiffness mismatch between cages and adjacent bone endplates increases the risk of stress shielding and cage subsidence.Methods The current study presents a multiscale optimization approach for porous Ti fusion cage development,including microscale topology optimization based on homogenization theory that obtains a unit cell with prescribed mechanical properties,and macroscale topology optimization that determines the layout of framework structure over the porous cage while maintaining the desired stiffness.The biomechanical performance of the designed porous cage is assessed using numerical simulations of fusion surgery.Selective laser melting is employed to assists with fabricating the designed porous structure and porous cage.Results The simulations demonstrate that the designed porous cage increases the strain energy density of bone grafts and decreases the peak stress on bone endplates.The mechanical and morphological discrepancies between the as-designed and fabricated porous structures are also described.Conclusion From the perspective of biomechanics,it is demonstrated that the designed porous cage contributes to reducing the risk of stress shielding and cage subsidence.The optimization of processing parameters and post-treatments are required to fabricate the designed porous cage.The present multiscale optimization approach can be extended to the development of cages with other shapes or materials and further types of orthopedic implants.

作为捕获风能的关键部件,风机叶片受制造及运行载荷影响,可能存在不同程度的损伤,这会直接影响风机运行可靠性。为防止风机叶片持续损伤发生质量安全事故,需要开发1种快速简便且非植入安装式的检测方法来识别风机叶片的损伤情况。根据叶片损伤和叶片运行噪声间存在的物理相关性,提出了一种基于声学信号和卷积神经网络(convolutional neural network,CNN)的风机叶片损伤检测方法,将时序声学信号转换成二维频谱图片,结合健康频谱图生成残差频谱图,并通过训练卷积神经网络来识别风机叶片是否存在损伤。分析结果表明:该算法消除了叶轮旋转产生的固有叶片扫风声音对损伤识别的影响,提高了识别精度;以某地风机的实测数据为例进行算法分析,结果表明该算法的分类精度达到了96.9%,验证了基于卷积神经网络的检测方法的有效性和精确性。

Nonlinear Modeling and Identification of Structural Joint by Response Control Vibration Test

Components of mechanical product are assembled by structural joints,such as bolting,riveting,welding,etc.Structural joints introduce nonlinearity to some engineering structures,and the nonlinearity need to be modeled precisely.To meet serious quality requirements,it is necessary to detect and identify nonlinearity of mechanical products for structural optimization.Modal test to acquire a dynamic response has been applied for decades,which provides reliable results for finite element(FE)model updating.Here response control vibration test for identification of nonlinearity is presented.A nonlinear system can be regarded as linearity for particular steady state response,and classical linear analysis tool is applicable to extract modal data for particular response.First,its applicability is illustrated by some numerical simulations.Subsequently,it is implemented on experimental setup with structural joints by shaking table.The stiffness and damping function dependent of relative displacement are fitted to describe its inherent nonlinearity.The spring and damping forces are identified by harmonic balance method(HBM)to predict output response.Based on the identified results,the procedure is recommended that it allows a reliable measurement of nonlinearity with a certain accuracy.

Performance and microbial community analysis of bioaugmented activated sludge for nitrogen-containing organic pollutants removal

Nitrogen-containing organic pollutants(quinoline,pyridine and indole)are widely distributed in coking wastewater,and bioaugmentation with specific microorganisms may enhance the removal of these recalcitrant pollutants.The bioaugmented system(group B)was constructed through inoculation of two aromatics-degrading bacteria,Comamonas sp.Z1(quinoline degrader)and Acinetobacter sp.JW(indole degrader),into the activated sludge for treatment of quinoline,indole and pyridine,and the non-bioaugmented activated sludge was used as the control(group C).Both groups maintained high efficiencies(>94%)for removal of nitrogen-containing organic pollutants and chemical oxygen demand(COD)during the long-term operation,and group B was highly effective at the starting period and the operation stage fed with raw wastewater.High-throughput sequencing analysis indicated that nitrogen-containing organic pollutants could shape the microbial community structure,and communities of bioaugmented group B were clearly separated from those of non-bioaugmented group C as observed in non-metric multidimensional scaling(NMDS)plot.Although the inoculants did not remain their dominance in group B,bioaugmentation could induce the formation of effective microbial community,and the indigenous microbes might play the key role in removal of nitrogen-containing organic pollutants,including Dokdonella,Comamonas and Pseudoxanthomonas.Phylogenetic Investigation of Communities by Reconstruction of Unobserved States(PICRUSt)analysis suggested that bioaugmentation could facilitate the enrichment of functional genes related to xenobiotics biodegradation and metabolism,probably leading to the improved performance in group B.This study indicated that bioaugmentation could promote the removal of nitrogen-containing organic pollutants,which should be an effective strategy for wastewater treatment.

Due to the increasing demand for microbially manufactured products in various industries,it has become important to find optimal designs for microbial cell factories by changing the direction of metabolic flow and its flux size by means of metabolic engineering such as knocking out competing pathways and introducing exogenous pathways to increase the yield of desired products.Recently,with the gradual cross-fertilization between computer science and bioinformatics fields,machine learning and intelligent optimization-based approaches have received much attention in Genome-scale metabolic network models(GSMMs)based on constrained optimization methods,and many high-quality related works have been published.Therefore,this paper focuses on the advances and applications of machine learning and intelligent optimization algorithms in metabolic engineering,with special emphasis on GSMMs.Specifically,the development history of GSMMs is first reviewed.Then,the analysis methods of GSMMs based on constraint optimization are presented.Next,this paper mainly reviews the development and application of machine learning and intelligent optimization algorithms in genome-scale metabolic models.In addition,the research gaps and future research potential in machine learning and intelligent optimization methods applied in GSMMs are discussed.

When carbon-containing species are involved in reactions catalyzed by transition metals at high temperature,the diffusion of carbon on or in catalysts dramatically influences the catalytic performance.Acquiring information on the carbon-diffusioninvolved evolution of catalysts at the atomic level is crucial for understanding the reaction mechanism yet also challenging.For the chemical vapor deposition process of single-walled carbon nanotubes(SWCNTs),we recorded in situ the catalyst state(solid and molten)composition as well as near-surface structural and chemical evolution at the cobalt catalyst-tube interface with carbon permeation using aberration-corrected environmental transmission electron microscopy and synchrotron X-ray absorption spectroscopy.The nucleation of SWCNTs was linked with an alternating dissolving and precipitating cycle of carbon in catalysts close to the nucleation site.Understanding the dynamics of carbon atoms in catalysts brings deeper insight into the growth mechanism of SWCNTs and facilitates inferring mechanisms of other reactions.The methodologies developed here will find broad applications in studying catalytic and other processes.
