Cancer therapies based on energy conversion,such as photothermal therapy(PTT,light-to-thermal energy conversion)and photodynamic therapy(PDT,light-to-chemical energy conversion)have attracted extensive attention in preclinical research.However,the PTT-related hyperthermia damage to surrounding tissues and shallow penetration of PDT-applied light prevent further advanced clinical practices.Here,we developed a thermoelectric therapy(TET)based on thermoelectric materials constructed p-n heterojunction(SrTiO_(3)/Cu_(2)Se nanoplates)on the principle of light-thermal-electricity-chemical energy conversion.Upon irradiation and natural cooling-induced the temperature gradient(35-45℃),a self-build-in electric field was constructed and thereby facilitated charges separation in bulk SrTiO_(3)and Cu_(2)Se.Importantly,the contact between SrTiO_(3)(n type)and Cu_(2)Se(p type)constructed another interfacial electric field,further guiding the separated charges to re-locate onto the surfaces of SrTiO_(3)and Cu_(2)Se.The formation of two electric fields minimized probability of charges recombination.Of note,high-performance superoxide radicals and hydroxyl radicals’generation from O_(2)and H_(2)O under catalyzation by separated electrons and holes,led to intracellular ROS burst and cancer cells apoptosis without apparent damage to surrounding tissues.Construction of bulk and interfacial electric fields in heterojunction for improving charges separation and transfer is also expected to provide a robust strategy for diverse applications.

Peptides-modified polystyrene-based polymers as high-performance substrates for the growth and propagation of human embryonic stem cells

As human stem cells with the special pluripotency play important roles in the innovative drug discovery and regenerative medicine,development of extracellular matrix(ECM)mimetics or functional materials that can support stem cell growth and propagation is of high significance.Despite numerous efforts spent,one major limitation restricting the wide applications of stem cells to the clinical translation is the lack of efficient strategies for low cost and large-scale stem cell production under xeno-free culture conditions.Herein,we reported a new strategy with peptides-modified polystyrene-based polymers coated onto the surface of coverslips for the growth and reproduction of human embryonic stem cells(h ESCs).The modified peptides are the active parts of proteins which has been shown to contribute to the pluripotent stem cell attachment or proliferation.The peptides were linked to the glass coverslips coated by the polymer materials via chemical crosslinking,and the composite substrates successfully maintain the longterm growth of HUES-7,H7 and DF699.Our study shows that the coating of polystyrene-derived polymer modified by our developed peptides is a good matrix for long-term growth and reproduction of stem cells.This polystyrene-derived polymer substrate can be produced in large scale and stored for a long time.The most important thing is that it can support the growth of undifferentiated human pluripotent stem cells(h PSCs)for more than ten passages,which could provide a new and relatively easy way to amplify h ESCs in vitro.

Dear Editor,Over-activation of the coagulation system in bacterial sepsis leads to disseminated intravascular coagulation(DIC),a lifethreatening pathophysiological syndrome.1 We previously verified that deficiency of the receptor of intracellular lipopolysaccharide,caspase-11,or its upstream,type I interferons(IFNs),significantly reduces endotoxin-mediated pore-forming and phosphatidylserine exposure,which dampens the activity of TF and subsequently the coagulation cascades.

Autophagy is one of essential cellular mechanisms which play a key role in cancer.It has been proven that induction autophagy is beneficial for cancer treatment.Herein,we designed and synthesized novel spherical nanoparticles named as MnO-MS with their sizes about 100 nm,which consist of manganese and mesoporous silica.Our studies showed that MnO-MS could obviously induce autophagy in GFP-LC3 Hela cell.Besides,to our surprise,MnO-MS cloud selectively inhibit lung cancer A549 cell growth at low dosages while little cytotoxicity on normal cell.Encouraged by this interesting discovery,the funher mechanism study was also performed and the result showed that Mn-MS activated reactive oxygen species(ROS),and MnO-MS can release free radicals,thus reducing ROS level after MnO-MS entering cell.At the same time,the increase of ROS level inhibited the mTOR production and then produced autophagosome.Collectively,these findings indicated that MnO-MS released free radicals lead to ROS activating induced cell death by autophagy pathway in cancer cells.It is envisioned that MnO-MS could be further developed as a potential autophagy inducer for the treatment of lung cancer.

Interferon gamma(IFN-γ) is an important cytokine, which regulate the transcription of immune related genes and coordinate the immune response of organisms.[1] It has been reported that IFN-γ was involved in antiviral, cell proliferation, cell apoptosis and immune regulation.[2] And it has developed as an important drug target for antiviral and tumor treatment and immune regulation. Research and development of IFN-γ drugs has always been a hot topic. Herein, we established a high throughput screening method to rapidly screen peptide drugs by selectively targeting IFN-γ protein. It was exciting that we have obtained several effective peptide drugs by using this method. And the affinity between these peptide drugs with IFN-γ protein was up to nanometer level. Further biological experiments showed that these peptide drugs have a powerful anticancer activity. However, these results revealed that this method is feasible and credible, and the IFN-γ peptide drugs by screening from this method can be used as potential anti-cancer drugs for cancer treatment in the future.

Fluticasone propionate, a new type of glucocorticoid developed by Glaxosmithkline in the United Kingdom, has strong efficacy and weak adverse reactions compared with other glucocorticoid drugs. We improved the synthesis of fluticasone propionate and studied its anticancer activity. First step in the synthesis of sodium periodate oxidation, silicone product load, the method of green environmental protection, economic efficiency, overall synthetic process is divided into seven steps, and of reactants, reaction is simple, from starting material fry at the end of the product, the overall yield of 90%, at the same time, in view of the fluticasone propionate functional group modification and optimization design of structure, through the study of the methylation of thiol, hydroxylation, selective oxidation of hydroxyl groups, the structure of modified research of anticancer activity, seen from the results, thiol methylation can effectively enhance its anticancer activity,A549,Hep G2, McF-7 and other cancer cells all showed good anticancer effect.

BRDs（Bromodomains） are a class of proteins that can specifically recognize and bind to acetyllysine[1]. Recognition of histone lysine acetylates by BRDs is an important step for histone acetylation to regulate gene expression. It can regulate chromatin regulatory factors to specific regions and complete gene expression regulation [2]. In this study, BRD4 in BRDs protein family was selected as the research target by OBOC（one-bead-one-compound screening） to screen polypeptide sequences with BRD4 binding capacity. Microarray screening were used to screen polypeptide sequences with selectivity and high affinity for two domains of BRD4（BD1, BD2）by two-color strategy. As a result, we have preliminarily identified more than 20 pentapeptides containing acetyllysine targeting BRD4, and selected 12 polypeptide sequences with strong binding power from them by measuring their binding capacity and binding dynamic curve, and then modified them and tested their cell activity. At present, a polypeptide sequence has been found to penetrate the cell membrane and specifically recognize BRD4 protein in the nucleus.

Signal transduction plays a vital role in cellular functions. It is well known that reversible tyrosine phosphorylation is also an early step in the transduction of many types of signals in multicellular organisms. Phosphorylated signaling proteins on tyrosine residues are usually specifically recognized by Src Homology 2（SH2） domain modular proteins. Since SH2 domain modular proteins are so important for the signal transduction inside the cells/human, mapping the SH2 domain binding specificity will help us predict/control the signaling state of the cell, thus facilitating us for human disease treatment caused by the aberrant signaling. A novel systematic phosphopeptide microarray platform with 144 members was developed to efficiently high throughput identification of some potent and selective binders for SH2 domain proteins, which were then validated by the lysates proteomes pull down and antibody detection on the microarray.

江油青林口油砂矿是四川盆地龙门山山前带非常规石油资源,受上寺-罗妙真-漆树槽逆冲断层南侧次一级庙子坝断层南东单斜构造带控制,区域上属下寺-金子山-厚坝油砂成矿带。对该地区已获取的地质、钻探工程、物探测井、采样测试等油砂矿成果资料进行综合研究,结果显示:油砂储集体发育在侏罗系中统沙溪庙组下段细-中粒岩屑石英砂岩中,油砂油矿体质量含油率平均值9.55%,含油饱和度平均值43.3%,含水饱和度平均值4.54%,有效孔隙度平均值15.9%,且具闪点高、密度大、粘度大及残碳、灰分、机械杂质高、高硫含量的典型稠油特点。油砂油储集体物性条件良好,水平方向渗透率低,垂直方向渗透率特低,具有较好的油砂油储存空间,显示出该地区有扩大资源储量的潜力,是值得进一步勘查的远景区。
