Clinical outcomes of second-line chemotherapy in patients with advanced pancreatic adenocarcinoma: a real-world study

Objective: Little progress has been made in recent years using first-line chemotherapy, including gemcitabine combined with nabpaclitaxel, FOLFIRINOX, and NALIRIFOX, for advanced pancreatic adenocarcinoma(APC). In addition, the optimal second-line chemotherapy regimen has not been determined. This study aimed to compare the effectiveness of different types of second-line chemotherapy for ***: Patients with APC who received first-line treatment from January 2008 to January 2021 were considered eligible for this retrospective analysis. The primary and secondary endpoints were overall survival(OS) and progression-free survival(PFS), ***: Four hundred and thirty-seven and 617 patients were treated with 5-fluorouracil-and gemcitabine-based chemotherapy as first-line treatment, respectively. Demographic and clinical features, except age and liver metastasis, were comparable between the two groups(P < 0.05). The median OS was 8.8 and 7.8 months in patients who received a 5-fluorouracil-and gemcitabine-based combined regimen for first-line therapy, respectively(HR = 1.244, 95% CI = 1.090±1.419; P < 0.001). The median OS was 5.6 and 1.9 months in patients who received second-line chemotherapy and supportive care, respectively(HR = 0.766, 95% CI = 0.677±0.867; P < 0.001). The median PFS was not significantly differently between gemcitabine or 5-fluorouracil monotherapy and combination ***: A 5-fluorouracil-or gemcitabine-based combined regimen was shown to be as effective as a single 5-fluorouracil or gemcitabine regimen as second-line therapy for patients with APC.

Understanding and improving fairness in cognitive diagnosis

Intelligent education is a significant application of artificial intelligence. One of the key research topics in intelligence education is cognitive diagnosis, which aims to gauge the level of proficiency among students on specific knowledge concepts(e.g., Geometry). To the best of our knowledge, most of the existing cognitive models primarily focus on improving diagnostic accuracy while rarely considering fairness issues; for instance, the diagnosis of students may be affected by various sensitive attributes(e.g., region). In this paper,we aim to explore fairness in cognitive diagnosis and answer two questions:(1) Are the results of existing cognitive diagnosis models affected by sensitive attributes?(2) If yes, how can we mitigate the impact of sensitive attributes to ensure fair diagnosis results? To this end, we first empirically reveal that several wellknown cognitive diagnosis methods usually lead to unfair performances, and the trend of unfairness varies among different cognitive diagnosis models. Then, we make a theoretical analysis to explain the reasons behind this phenomenon. To resolve the unfairness problem in existing cognitive diagnosis models, we propose a general fairness-aware cognitive diagnosis framework, FairCD. Our fundamental principle involves eliminating the effect of sensitive attributes on student proficiency. To achieve this, we divide student proficiency in existing cognitive diagnosis models into two components: bias proficiency and fair *** design two orthogonal tasks for each of them to ensure that fairness in proficiency remains independent of sensitive attributes and take it as the final diagnosed result. Extensive experiments on the Program for International Student Assessment(PISA) dataset clearly show the effectiveness of our framework.

The Chinese pangolin(Manis pentadactyla,MP)has been extensively exploited and is now on the brink of extinction,but its population structure,evolutionary history,and adaptive potential are ***,we analyzed 94 genomes from three subspecies of the Chinese pangolin and identified three distinct genetic clusters(MPA,MPB,and MPC),with MPB further divided into MPB1 and MPB2 *** divergence of these populations was driven by past climate *** MPB2 and MPC,recent human activities have caused dramatic population decline and small population size as well as increased inbreeding,but not decrease in genomic variation and increase in genetic load probably due to strong gene flow;therefore,it is crucial to strengthen in situ habitat management for these two *** contrast,although human activities have a milder impact on MPA,it is at high risk of extinction due to long-term contraction and isolation,and genetic rescue is urgently ***1 exhibited a relatively healthy population status and can potentially serve as a source ***,our findings provide novel insights into the conservation of the Chinese pangolin and biogeography of the mammals of eastern Asia.

Therapeutic peptides contribute significantly to human health and have the potential for personalized medicine. The prediction for the therapeutic peptides is beneficial and emerging for the discovery of drugs. Although several computational approaches have emerged to discern the functions of therapeutic peptides, predicting multi-functional therapeutic peptide types is challenging. In this research, a novel approach termed TPpred-SC has been introduced. This method leverages a pretrained protein language model alongside multi-label supervised contrastive learning to predict multi-functional therapeutic *** framework incorporates sequential semantic information directly from large-scale protein sequences in TAPE. Then, TPpred-SC exploits multi-label supervised contrastive learning to enhance the representation of peptide sequences for imbalanced multi-label therapeutic peptide prediction. The experimental findings demonstrate that TPpred-SC achieves superior performance compared to existing related methods. Imidacloprid is a neonicotinoid insecticide used for managing the Asian citrus psyllid,Diaphorina citri Kuwayama,which serves as vector of phytopathogens causing citrus ***,development of resistance to neonicotinoids among populations of *** has coincided with occasional control failures in the *** objectives of this research were to(1)survey current levels of imidacloprid resistance in Florida citrus;(2)compare feeding behavior between imidacloprid-resistant and susceptible *** using electrical penetration graph recordings,and(3)investigate the possible amplification of insecticide hormoligosis associated with *** surveys confirmed that the susceptibility of *** populations to imidacloprid has decreased in commercial Florida citrus groves compared with a laboratory-susceptible *** 12 generations of selection,resistance to imidacloprid increased by 438 fold compared with the susceptible ***-susceptible *** feeding on citrus exhibited significantly more bouts associated with intercellular pathway(C),phloem penetration(D),phloem salivation(E1),and nonprobing(Np)activities than imidacloprid-resistant ***,there were no differences observed in the frequency or duration of phloem ingestion or xylem feeding between susceptible and resistant *** was no statistical difference in fecundity between resistant and susceptible ***,the fecundity of imidacloprid-susceptible female *** treated with a sublethal concentration of imidacloprid(LC_(25))increased significantly compared with controls,while such hormoligosis was less pronounced among imidacloprid-resistant *** results suggest that imidacloprid-resistant psyllids may cease feeding sooner than susceptible counterparts following sublethal exposure to this insecticide,indicative of a behavioral resistance mechanism.

The central nervous system, information integration center of the body, is mainly composed of neurons and glial cells. The neuron is one of the most basic and important structural and functional units of the central nervous system, with sensory stimulation and excitation conduction functions. Astrocytes and microglia belong to the glial cell family, which is the main source of cytokines and represents the main defense system of the central nervous system. Nerve cells undergo neurotransmission or gliotransmission, which regulates neuronal activity via the ion channels, receptors, or transporters expressed on nerve cell membranes. Ion channels, composed of large transmembrane proteins, play crucial roles in maintaining nerve cell homeostasis. These channels are also important for control of the membrane potential and in the secretion of neurotransmitters. A variety of cellular functions and life activities, including functional regulation of the central nervous system, the generation and conduction of nerve excitation, the occurrence of receptor potential, heart pulsation, smooth muscle peristalsis, skeletal muscle contraction, and hormone secretion, are closely related to ion channels associated with passive transmembrane transport. Two types of ion channels in the central nervous system, potassium channels and calcium channels, are closely related to various neurological disorders, including Alzheimer's disease, Parkinson's disease, and epilepsy. Accordingly, various drugs that can affect these ion channels have been explored deeply to provide new directions for the treatment of these neurological disorders. In this review, we focus on the functions of potassium and calcium ion channels in different nerve cells and their involvement in neurological disorders such as Parkinson's disease, Alzheimer's disease, depression, epilepsy, autism, and rare disorders. Fragrant rice has a high market value,and it is a popular rice type among consumers owing to its pleasant *** methods,nitrogen(N)fertilizers,and silicon(Si)fertilizers can affect the grain yield and fragrance of fragrant ***,the core commercial rice production attributes,namely the head rice yield(HRY)and 2-acetyl-1-pyrroline(2-AP)content of fragrant rice,under various nitrogen and silicon(N-Si)fertilization levels and different plantation methods remain *** field experiment in this study was performed in the early seasons of 2018 and 2019 with two popular indica fragrant rice cultivars(Yuxiangyouzhan and Xiangyaxiangzhan).They were grown under six N-Si fertilization treatments(combinations of two levels of Si fertilizer,0 kg Si ha^(−1)(Si0)and 150 kg Si ha^(−1)(Si1),and three levels of N fertilizer,0 kg N ha^(−1)(N0),150 kg N ha^(−1)(N1),and 220 kg N ha^(−1)(N2))and three plantation methods(artificial transplanting(AT),mechanical transplanting(MT),and mechanical direct-seeding(MD)).The results showed that the N-Si fertilization treatments and all the plantation methods significantly affected the HRY and 2-AP content and related parameters of the two different fragrant rice *** with the Si0N0 treatment,the N-Si fertilization treatments resulted in higher HRY and 2-AP *** rates of brown rice,milled rice,head rice,and chalky rice of the fragrant rice also improved with the N-Si fertilization *** N-Si fertilization treatments increased the activities of N metabolism enzymes and the accumulation of N and Si in various parts of the fragrant rice,and affected their antioxidant response *** key parameters for the HRY and 2-AP content were assessed by redundancy ***,the structural equation model revealed that the Si and N accumulation levels indirectly affected the HRY by affecting the N metabolism enzyme activity,N use efficiency,and grain quality of fragrant ***,high N
