3D eikonal equation is a partial differential equation for the calculation of first-arrival traveltimes and has been widely applied in many scopes such as ray tracing,source localization,reflection migration,seismic monitoring and tomographic *** recent years,many advanced methods have been developed to solve the 3D eikonal equation in heterogeneous ***,there are still challenges for the stable and accurate calculation of first-arrival traveltimes in 3D strongly inhomogeneous *** this paper,we propose an adaptive finite-difference(AFD)method to numerically solve the 3D eikonal *** novel method makes full use of the advantages of different local operators characterizing different seismic wave types to calculate factors and traveltimes,and then the most accurate factor and traveltime are adaptively selected for the convergent updating based on the Fermat *** with global fast sweeping describing seismic waves propagating along eight directions in 3D media,our novel method can achieve the robust calculation of first-arrival traveltimes with high precision at grid points either near source point or far away from source point even in a velocity model with large and sharp *** numerical examples show the good performance of the AFD method,which will be beneficial to many scientific applications.

Background： The interaction between activated microglia and T lymphocytes can yield abundant pro-inflammatory cytokines. Our previous study proved that thymus immune tolerance could alleviate the inflammatory response. This study aimed to investigate whether intrathymic injection of myelin basic protein （MBP） in mice could suppress the inflammatory response after co-culture ofT lymphocytes and BV-2 microglia cells. Methods： Totally, 72 male C57BL/6 mice were randomly assigned to three groups （17 - 24 in each）： Group A： intrathymic injection of 100 μl M BP （1 mg/ml）; Group B： intrathymic injection of 100 μ1 phosphate-buffered saline （PBS）; and Group C： sham operation group. Every eight mice in each group were sacrificed to obtain the spleen at postoperative days 3, 7, and 14, respectively. T lymphocytes those were extracted and purified from the spleens were then co-cultured with activated BV-2 microglia cells at a proportion of 1：2 in the medium containing MBP for 3 days. After identified the T lymphocytes by CD3, surface antigens oft lymphocytes （CD4, CD8, CD152, and CD154） and BV-2 microglia cells （CD45 and CD54） were detected by flow cytometry. The expressions of pro-inflammatory factors of BV-2 microglia cells （interleukin [1L]- 1β, tumor necrosis factor-o~ [TNF-α], and inducible nitric oxide synthase [iNOS]） were detected by quantitative real-time polymerase chain reaction （PCR）. One-way analysis of variance （ANOVA） and the least significant difference test were used for data analysis. Results： The levels of CD152 in Group A showed an upward trend from the 3rd to 7th day, with a downward trend from the 7th to 14th day （20.12 ± 0.71%, 30.71 ± 1.14%, 13.50 ± 0.71% at postoperative days 3, 7, and 14, respectively, P 〈 0.05）. Molting and metamorphosis are important physiological processes in insects that are tightly controlled by ecdysone receptor(EcR)through the 20-hydroxyecdysone(20E)signaling *** is a steroid nuclear receptor(SR).Several FK506-binding proteins(FKBPs)have been identified from the mammal SR complex,and are thought to be involved in the subcellular trafficking of ***,their roles in insects are poorly *** explore whether FKBPs are involved in insect molting or metamorphosis,we injected an FKBP inhibitor(FK506)into a lepidopteran insect,Spodoptera litura,and found that molting was inhibited in 61.11%of the larvae,and that the time for larvae to pupate was significantly extended.A total of 10 FKBP genes were identified from the genome of *** and were clustered into 2 distinct groups,according to their subcellular localization,with FKBP13 and FKBP14 belonging to the endoplasmic reticulum(ER)group and with the other members belonging to the cytoplasmic(Cy)*** the CyFKBPs were significantly upregulated in the prepupal or pupal stages,with the opposite being observed for the ER group ***506 completely blocked the transfer of EcR to the nucleus under 20E induction,and significantly downregulated the transcriptional expression of many 20E signaling genes.A similar phenomenon was observed after RNA interference of2 CyFKBPs(FKBP45 and FKBP12b),but not for *** together,our data indicate that the cytoplasmic FKBPs,especially FKBP45 and FKBP12b,mediate the nuclear localization of EcR,thereby regulating the 20E signaling and ultimately affecting molting and metamorphosis in insects.

The Central and Southern South China Sea(CSSCS) has a complex tectonic dynamic background and abundant oil and gas resources, which has always been a hot topic of academic and industrial ***, systematic analyses are still lacking regarding its sediment filling structure and evolution, mostly due to limited borehole penetration and poor quality of seismic reflection data for deeply buried sequences. No consensus has been reached yet on the sedimentary infilling processes, which impeded the reconstruction of the palaeogeography of Southeast Asia and the oil-and-gas exploration undertakings. Here, we illustrate the Cenozoic sedimentary evolution of the CSSCS region by synthesizing relevant data from previous literature and our own observations and displaying the evolution of depositional systems in sequential reconstructions. Besides, the controlling factors of preferred sedimentary scenarios in the CSSCS incorporate the latest interpretations of the spreading of South China Sea(SCS) as well as the demise of the hypothetical Proto-South China Sea(PSCS). The results show that there are three types of sedimentary basins in the CSSCS(foreland,strike-slip, and rift basins) with different sedimentary filling structures. The foreland basins formed a depositional pattern of ‘transition from deep water to shallow water environments', dominated by deep-water depositional systems which were formed before the Early Oligocene with submarine fans developed. Later,the foreland basins were gradually dominated by shallow-water depositional systems with deltas and shallow marine facies. The strike-slip basins showed the depositional architecture of ‘transition from lake to marine environments', i.e. the basins were dominated by lacustrine deposits during the Eocene and evolved into the marine depositional environment since Oligocene with delta developed in the western part of the basin. Intensive selection pressure constrains the evolutionary trajectory of SARS-CoV-2 genomes and results in various novel variants with distinct mutation *** mutations,particularly those within the receptor binding domain(RBD)of SARS-CoV-2 spike(S)protein,lead to the functional alteration in both receptor engagement and monoclonal antibody(mAb)***,we review the data of the RBD point mutations possessed by major SARS-CoV-2 variants and discuss their individual effects on ACE2 affinity and immune *** single amino acid substitutions within RBD epitopes crucial for the antibody evasion capacity may conversely weaken ACE2 binding ***,this weakened effect could be largely compensated by specific epistatic mutations,such as N501Y,thus maintaining the overall ACE2 affinity for the spike protein of all major *** predominant direction of SARS-CoV-2 evolution lies neither in promoting ACE2 affinity nor evading mAb neutralization but in maintaining a delicate balance between these two ***,this review interprets how RBD mutations efficiently resist antibody neutralization and meanwhile how the affinity between ACE2 and spike protein is maintained,emphasizing the significance of comprehensive assessment of spike mutations.

Originating but free from chromosomal DNA,extrachromosomal circular DNAs(eccDNAs)are organized in circular form and have long been found in unicellular and multicellular *** biogenesis and function are poorly understood as they are characterized by sequence homology with linear DNA,for which few detection methods are *** advances in high-throughput sequencing technologies have revealed that eccDNAs play crucial roles in tumor formation,evolution,and drug resistance as well as aging,genomic diversity,and other biological processes,bringing it back to the research *** mechanisms of eccDNA formation have been proposed,including the breakage-fusion-bridge(BFB)and translocation-deletion-amplification *** tumors and disorders of embryonic and fetal development are major threats to human reproductive *** roles of eccDNAs in these pathological processes have been partially elucidated since the first discovery of eccDNA in pig sperm and the double minutes in ovarian cancer *** present review summarized the research history,biogenesis,and currently available detection and analytical methods for eccDNAs and clarified their functions in gynecologic tumors and *** also proposed the application of eccDNAs as drug targets and liquid biopsy markers for prenatal diagnosis and the early detection,prognosis,and treatment of gynecologic *** review lays theoretical foundations for future investigations into the complex regulatory networks of eccDNAs in vital physiological and pathological processes.
