Reduced expression of semaphorin 3A in osteoclasts causes lymphatic expansion in a Gorham-Stout disease(GSD)mouse model

Gorham-Stout disease(GSD)is a sporadic chronic disease characterized by progressive bone dissolution,absorption,and disappearance along with lymphatic vessel infiltration in bone-marrow cavities.Although the osteolytic mechanism of GSD has been widely studied,the cause of lymphatic hyperplasia in GSD is rarely investigated.In this study,by comparing the RNA expression profile of osteoclasts(OCs)with that of OC precursors(OCPs)by RNA sequencing,we identified a new factor,semaphorin 3A(Sema3A),which is an osteoprotective factor involved in the lymphatic expansion of GSD.Compared to OCPs,OCs enhanced the growth,migration,and tube formation of lymphatic endothelial cells(LECs),in which the expression of Sema3A is low compared to that in OCPs.In the presence of recombinant Sema3A,the growth,migration,and tube formation of LECs were inhibited,further confirming the inhibitory effect of Sema3A on LECs in vitro.Using an LEC-induced GSD mouse model,the effect of Sema3A was examined by injecting lentivirus-expressing Sema3A into the tibiae in vivo.We found that the overexpression of Sema3A in tibiae suppressed the expansion of LECs and alleviated bone loss,whereas the injection of lentivirus expressing Sema3A short hairpin RNA(shRNA)into the tibiae caused GSD-like phenotypes.Histological staining further demonstrated that OCs decreased and osteocalcin increased after Sema3A lentiviral treatment,compared with the control.Based on the above results,we propose that reduced Sema3A in OCs is one of the mechanisms contributing to the pathogeneses of GSD and that expressing Sema3A represents a new approach for the treatment of GSD.

Objective: Rheumatoid arthritis(RA) is a chronic inflammatory and destructive arthritis, characterized by inflammatory infiltration and bone destruction. Huangqi Guizhi Wuwu Decoction(HGWD) is traditional Chinese medicine, which has been applied in the treatment of RA in clinical. The aim of this study was to investigate the therapeutic effect of HGWD on collagen-induced arthritis(CIA) mouse model.Methods: DBA/1J female mice were used to establish the collagen-induced arthritis(CIA) model. HGWD was administered intragastrically once a day for four weeks starting on the 22nd day after the first immunization. The body weight, hind paw thickness and clinical score were measured every five days. Gait analysis, histopathological staining, enzyme-linked immunosorbent assay(ELISA), ultrasound imaging and micro-computed tomography imaging were performed to determine the effects of HGWD treatment on inflammation and bone structure in this model. Moreover, Real-time PCR and Western blot analysis were used to detect inflammatory factors m RNA and protein levels after HGWD intervention in RAW264.7 cells.Results: HGWD attenuated symptoms of arthritis, suppressed inflammatory synovium area and the serum levels of inflammatory factors, inhibited joint space enlargement in the knee and ankle joints,reduced numbers of osteoclasts, protected bone destruction, as well as improved motor function.HGWD decreased the expression of m RNA for inflammatory factors and the protein expression levels of p-NF-ΚB and IL-17.Conclusion: These results suggested that HGWD suppresses inflammation, attenuates bone erosion and maintains motor function in collagen-induced arthritis mice.

Triazole backbone ligand in an unprecedented efficient manganese catalyst for use in transfer hydrogenation

Noble metal catalysts are generally expensive,and thus,abundant 3d metals recently received significant attention as catalysts in catalytic hydrogenation.Mn catalysts are widely applied in transfer hydrogenations,but the reported catalyst loadings remain up to three orders of magnitude higher than noble metals.Thus,catalyst consumption should be overcome before 3d metal catalytic systems may be utilized practically in industry.Here,a catalytic system based on novel,scalable triazole N5-ligands coordinated to Mn is presented for use in transfer hydrogenations.Based on pre-activation via dehydrohalogenation,an unprecedented,efficient catalytic system operating via synergistic H-bond auxiliary activation was established.The Mn catalysts are practical at metal concentrations 0.0001 mol%,generating alcohol with turnover number(TON)up to 857,200,thus approaching loadings more conventionally observed in precious-metal-based systems.Notably,using this protocol,several pharmaceuticals may be easily synthesized in one pot.

Danggui Niantong decoction ameliorates joint inflammation and cardiopulmonary injury in TNF-Tg mice

Objective:Rheumatoid arthritis(RA)is a common autoimmune disease characterized by multiple joint lesions and systemic complications.Danggui Niantong decoction(DGNTT)has been clinically used for RA treatment;however,its beneficial effect on cardiopulmonary complications has not been reported.Methods:Female tumor necrosis factor-transgenic(TNF-Tg)mice were used to evaluate the therapeutic effects of DGNTT on arthritis and cardiopulmonary complications.Methotrexate(MTX)served as a positive control.Histopathological assessment of the joint sections was performed using hematoxylin and eosin(HE),Alcian Blue/Orange G,and tartrate-resistant acid phosphatase staining.Bone mass was assessed by micro-computed tomography,inflammatory infiltrates in the heart and lungs were evaluated by HE staining,cardiopulmonary fibrotic injury was identified by Masson’s trichrome staining,and hypertrophy of mouse cardiomyocytes was measured by wheat germ agglutinin(WGA)staining.Results:DGNTT mitigated the inflammation of the ankle joint synovium,decreased the number of osteoclasts,and increased the area of cartilage and bone mass in TNF-Tg mice.In addition,DGNTT decreased the infiltration of inflammatory cells into the lung and heart tissues,accompanied by a reduction in cardiopulmonary fibrosis and myocardial cell hypertrophy in TNF-Tg mice.As a positive control drug,MTX attenuated the pathological changes in joints,but had no beneficial effect on cardiopulmonary inflammation and fibrosis in TNF-Tg mice.Conclusions:DGNTT improved joint lesions and alleviated cardiopulmonary complications in TNF-Tg mice.

Mash1 efficiently reprograms rat astrocytes into neurons

To date, it remains poorly understood whether astrocytes can be easily reprogrammed into neurons. Mashl and Brn2 have been previously shown to cooperate to reprogram fibroblasts into neurons. In this study, we examined astrocytes from 2-month-old Sprague-Dawley rats, and found that Brn2 was expressed, but Mashl was not detectable. Thus, we hypothesized that Mashl alone could be used to reprogram astrocytes into neurons. We transfected a recombinant MSCV-MASH1 plasmid into astrocytes for 72 hours, and saw that all cells expressed Mashl. One week later, we observed the changes in morphology of astrocytes, which showed typical neuro- nal characteristics. Moreover, β-tubulin expression levels were significantly higher in astrocytes expressing Mashl than in control cells. These results indicate that Mashl alone can reprogram astrocytes into neurons.

在神经重症患者救治中,气管切开是常见的气道手术。目前使用的传统气管套管固定带易污染,且不易消毒,需要定期更换,易发生感染、皮肤损伤、非计划拔管等不良事件,无法满足临床治疗效果及患者的安全管理。为克服上述问题,河南省人民医院神经重症监护病房医护人员设计了一种可增加患者舒适度且能减少并发症的新型气管套管固定带,获得了国家实用新型专利(专利号:ZL 202220855188.8)。该装置的主要结构包括随行弯板、紧固带、锁定销和调距孔。随形弯板左右两侧均设置有附有透气防磨垫的紧固带,左侧紧固带的内侧面设有两组锁定销,右侧紧固带和透气防磨垫的外表面均开设有多组调距孔,且随形弯板的背面设有透气缓冲垫。通过紧固带可带动随形弯板与患者颈部贴紧,操作人员可根据"退一原则"将锁定销卡装到调距孔内,两侧紧固带在锁定销的配合下将装置固定,极大降低了气管套管在使用过程中出现过大位移的概率,有效提高了装置的固定效果,加强了装置对不同人员的适应性,从而增强了装置的实用性。新型气管套管固定带使用方便、安全、高效,能增加患者的舒适度,减少并发症的发生,有一定临床价值,适合在临床推广。

在城市集约化建设背景下,社区综合体成为供给社区服务设施的空间新载体,但目前存在空间规划系统性差、管理运营机制不清等诸多问题。本文以新加坡社区综合体为研究对象,系统梳理了其概念内涵、形成的驱动机制和空间形态演进,并从规划层级和空间结构、协同性功能配置模式、多样化空间设计策略三个层面探究其规划设计特征。研究发现,新加坡社区综合体的形成与演进源于社区服务设施层级的规划演进,社区服务设施在TOD模式和服务人口规模的综合影响下趋向综合化、紧凑化发展。基于其空间规划设计特征的深入分析,本文提炼出四方面针对性建议:系统构建社区综合体的空间层次和结构,更新兼容用地分类、设定条件和奖励机制,构建公共服务与商业功能的协同布局,以及健全政府主导、多元参与的可持续运营机制。

Dear Editor,Dilated cardiomyopathy(DCM)is a common form of inherited cardiomyopathy.In the past decades,single mutations in various genes encoding sarcomeric,cytoskeletal,and channel proteins etc.have been found to be associated with DCM(Hershberger et al.,2013;McNally and Mestroni,2017).However,the mechanisms how single mutations in sarcomeric or structural genes lead to the disease remain elusive.An interesting phenomenon often seen in familial cardiomyopathy is that different single mutations on the same gene can cause either DCM or hypertrophic cardiomyopathy(HCM)(Kathiresan and Srivastava,2012),which exhibit almost opposite disease phenotypes.DCM is characterized by thinned myocardium and septum,ventricular chamber dilation,and systolic dysfunction(Jefferies and Towbin,2010;McNally and Mestroni,2017),while HCM exhibits thickened myocardium and septum,reduced ventricular chamber,and diastolic dysfunction(Richard et al.,2003).At the cellular level,HCM cardiomyocytes exhibit concentric hypertrophy characterized by assembly of myofilaments in parallel and widening of the myocytes.In contrast,DCM cardiomyocytes show eccentric hypertrophy,with assembly of the myofilaments in series and myocyte elongation(Kehat and Molkentin,2010).
