Bacterial biofilms present a significant challenge in treating drug-resistant infections,necessitating the development of innovative nanomedicines.In this study,we introduce triclosan-conjugated,lipase-responsive polymeric micelles designed to exploit biofilm properties and serve as a responsive drug delivery platform.The micelles were created using an amphiphilic block polymer synthesized via ring-opening polymerization ofε-caprolactone(CL)and triclosan-containing cyclic trimethylene carbonate(MTC-Tri).Poly(ethylene glycol)(PEG-OH)acted as the macro-initiator,resulting in micelles with a PEG shell that facilitated their penetration into bacterial biofilms.An important advantage of our micelles lies in their interaction with local bacterial lipases within biofilms.These lipases triggered rapid micelle degradation,releasing triclosan in a controlled manner.This liberated triclosan effectively eliminated bacteria embedded in the biofilms.Notably,the triclosan-conjugated micelles displayed minimal toxicity to murine fibroblasts,indicating their biocompatibility and safety.This finding emphasizes the potential application of these micelles in combatting drug resistance observed in bacterial biofilms.Our triclosan-conjugated,lipase-responsive polymeric micelles exhibit promising characteristics for addressing drug resistance in bacterial biofilms.By harnessing biofilm properties and implementing a responsive drug delivery system,we seek to provide an effective solution in the fight against drug-resistant bacteria.

Polymeric partners breathe together: using gas to direct polymer self-assembly via gas-bridging chemistry

The quest for a general and facile way to regulate polymer self-assembled nanostructures with low-to high-order ergodicity is an eternal theme in soft nanoparticle fabrication. Here we present an unprecedented gas-bridging strategy that allows to use gas to direct polymer self-assembly in continuous and tunable manners. Such system comprises a partner of frustrated Lewis polymers with bulky Lewis acid and base groups. They can together “breathe in” external gases to form gas-bridged structures between the two complementary moieties, which drive their mutual complexation and assemble into polymer nanoparticles of diverse geometries and dimensionalities. This strategy is applicable to a broad family of gas substances including but not limited to carbon oxides, nitrogen oxides, sulfur oxides, and even olefins;moreover, tailoring gas types and levels can dictate distinct assembling evolutionary pathways and deformable behaviors among spherical, fibrous, polymersomal, tubesomal and cubosomal morphologies. We also discover that the gas-based bonding chemistry is the mechanistic basis underlying the phase transitional control and phase window regulation. This will open a new direction of making bespoke polymer nanostructures with gas.

Safety and efficacy of a programmed cell death 1 inhibitor combined with oxaliplatin plus S-1 in patients with Borrmann large type III and IV gastric cancers

BACKGROUND Gastric cancer(GC)is the fifth most common and the fourth most lethal malignant tumour in the world.Most patients are already in the advanced stage when they are diagnosed,which also leads to poor overall survival.The effect of posto-perative adjuvant chemotherapy for advanced GC is unsatisfactory with a high rate of distant metastasis and local recurrence.AIM To investigate the safety and efficacy of a programmed cell death 1(PD-1)inhibitor combined with oxaliplatin and S-1(SOX)in the treatment of Borrmann large type III and IV GCs.METHODS A retrospective analysis(IRB-2022-371)was performed on 89 patients with Borrmann large type III and IV GCs who received neoadjuvant therapy(NAT)from January 2020 to December 2021.According to the different neoadjuvant treatment regimens,the patients were divided into the SOX group(61 patients)and the PD-1+SOX(P-SOX)group(28 patients).RESULTS The pathological response(tumor regression grade 0/1)in the P-SOX group was significantly higher than that in the SOX group(42.86%vs 18.03%,P=0.013).The incidence of ypN0 in the P-SOX group was higher than that in the SOX group(39.29%vs 19.67%,P=0.05).The use of PD-1 inhibitors was an independent factor affecting tumor regression grade.Meanwhile,the use of PD-1 did not increase postoperative complications or the adverse effects of NAT.CONCLUSION A PD-1 inhibitor combined with SOX could significantly improve the rate of tumour regression during NAT for patients with Borrmann large type III and IV GCs.

Optimization of Charging/Battery-Swap Station Location of Electric Vehicles with an Improved Genetic Algorithm-Based Model

The joint location planning of charging/battery-swap facilities for electric vehicles is a complex problem.Considering the differences between these two modes of power replenishment,we constructed a joint location-planning model to minimize construction and operation costs,user costs,and user satisfaction-related penalty costs.We designed an improved genetic algorithm that changes the crossover rate using the fitness value,memorizes,and transfers excellent genes.In addition,the present model addresses the problem of“premature convergence”in conventional genetic algorithms.A simulated example revealed that our proposed model could provide a basis for optimized location planning of charging/battery-swapping facilities at different levels under different charging modes with an improved computing efficiency.The example also proved that meeting more demand for power supply of electric vehicles does not necessarily mean increasing the sites of charging/battery-swap stations.Instead,optimizing the level and location planning of charging/battery-swap stations can maximize the investment profit.The proposed model can provide a reference for the government and enterprises to better plan the location of charging/battery-swap facilities.Hence,it is of both theoretical and practical value.

The most common occurring sites of schwannomas are the head, neck and extremities [3]. In the abdominal cavity, the most frequently involved sites are the retroperitoneum and stomach [4].

BACKGROUND The pathogenicity of Helicobacter pylori is dependent on factors including the environment and the host.Although selenium is closely related to pathogenicity as an environmental factor,the specific correlation between them remains unclear.AIM To investigate how selenium acts on virulence factors and reduces their toxicity.METHODS H.pylori strains were induced by sodium selenite.The expression of cytotoxin-associated protein A(CagA)and vacuolating cytotoxin gene A(VacA)was determined by quantitative PCR and Western blotting.Transcriptomics was used to analyze CagA,CagM,CagE,Cag1,Cag3,and CagT.C57BL/6A mice were infected with the attenuated strains subjected to sodium selenite induction,and H.pylori colonization,inflammatory reactions,and the cell adhesion ability of H.pylori were assessed.RESULTS CagA and VacA expression was upregulated at first and then downregulated in the H.pylori strains after sodium selenite treatment.Their expression was significantly and steadily downregulated after the 5th cycle(10 d).Transcriptome analysis revealed that sodium selenite altered the levels affect H.pylori virulence factors such as CagA,CagM,CagE,Cag1,Cag3,and CagT.Of these factors,CagM and CagE expression was continuously downregulated and further downregulated after 2 h of induction with sodium selenite.Moreover,CagT expression was upregulated before the 3rd cycle(6 d)and significantly downregulated after the 5th cycle.Cag1 and Cag3 expression was upregulated and downregulated,respectively,but no significant change was observed by the 5th cycle.C57BL/6A mice were infected with the attenuated strains subjected to sodium selenite induction.The extent of H.pylori colonization in the stomach increased;however,sodium selenite also induced a mild inflammatory reaction in the gastric mucosa of H.pylori-infected mice,and the cell adhesion ability of H.pylori was significantly weakened.CONCLUSION These results demonstrate that H.pylori displayed virulence attenuation after the 10th d of sodium selenite treatment.Sodium s

Optically“silent”neptunium(Ⅴ)-nitrate complex in ionic liquid

The complexation of pentavalent neptunium,Np(Ⅴ),with nitrate ion in an ionic liquid solution has been studied spectroscopically for the first time.The characteristic f-f transition absorption band of Np(Ⅴ)in the NIR region changes significantly upon the titration of nitrate ion into the solution,revealing strong complexation of Np(Ⅴ)with nitrate ion in the ionic liquid.Most notably,the absorption band of Np(Ⅴ)almost disappears when a sufficiently high concentration of nitrate ion is present in the solution.Such a rare optically“silent”species can be assigned to the 1:2 Np(Ⅴ)/nitrate complex with a centrosymmetric coordination environment where Np sits at the inversion center.

基于技术组合创新理论,通过对华为无线网络技术后发追赶的纵向案例研究,建立复杂核心技术创新的五阶段模型,从复杂性和核心性两个方面建构复杂核心技术,分析复杂核心技术创新及后发追赶的主要困境与动力机制。研究发现,根据技术创新范围、规模、复杂性与核心性形成过程,复杂核心技术创新过程分为产品原型、市场准入、边缘市场、核心技术及核心市场5个阶段;复杂核心技术的高市场转换成本导致后发者面临巨大困境以及先发者存在路径依赖,后发追赶需要长期大规模的研发投入,分别解决技术复杂性和核心性;技术复杂性和核心性均来源于技术组合创新以及市场需求,技术复杂性为核心性提供整体优势和经济动力,技术核心性进一步增强技术复杂性,形成技术的多层次网络结构与整体优势。研究结论完善了复杂核心技术创新的后发追赶理论,对后发企业实现关键核心技术突破具有重要的实践意义。

Dynamic Macro-and Microgels Driven by Adenosine Triphosphate-Fueled Competitive Host-Guest Interaction

Supramolecular host–guest systems are generally at thermodynamic equilibrium states or in kinetically trapped states.Herein,we demonstrate the concept of a chemical fuel-driven competitive host–guest assembly featuring autonomous dynamics.The enabling key principle is to design a chemical fuel that possesses a high binding affinity to defeat the guest transiently,undergoes conversion to a waste product,and exhibit weak binding affinity in order to recover the original host–guest pair.By following this principle,adenosine triphosphate(ATP;chemical fuel and competitive guest),biguanidine-functionalizedβ-cyclodextrin(βCD;host),and an adamantine species(ADA;guest)have been engineered to construct dynamic adaptive macrogels or chemo-mechanochromic microgels with programmed time domains.Our reported methodology provides temporal control over the host–guest process,representing a conceptually new tool for the design of living supramolecular materials.

Photoregulated supramolecular hydrogels driven by polyradical interactions

Organic radical as a powerful tool has been extensively applied in synthetic chemistry. However, harnessing radical-mediated noncovalent interactions to fabricate soft materials remains elusive. Here we report a new category of supramolecular hydrogel system held by multiple radical-radical(polyradical) interactions, and its photosensitive cross-linking structure. A simple polyacrylamide with triarylamine(TAA)pendants is designed as the precursor. The TAA units in polymer can be converted into active TAA^(·+)radical cations with light and further associate each other via TAA^(·+)–TAA^(·+)stacking interactions to form stable supramolecular network. Temporal control of the light irradiation dictates the degree of radical stacks, thus regulating the mechanical performance of the resulting hydrogel materials on-demand. Moreover, the reversible collapse of this hydrogels can be promoted by adding radical scavenger or exerting reduction voltage.
