Enhanced drug loading efficiency of contact lenses via salt-induced modulation

Low drug loading efficiency is one of the main obstacles hindering the application of contact lenses(CLs) as the carrier for extended ocular drug delivery. Here in this study, a simple and effective drug loading method based on salt induced modulation was proposed and demonstrated with mechanism elucidation. First of all, using poly(2-hydroxyethyl methacrylate)(p-HEMA) as the contact lens material, betaxolol hydrochloride, Diclofenac Sodium and Betaxolol Base as the model drugs with different solubility, influence of salt concentration,salt type(sodium salts of sulfate, chloride, and sulfocyanate) and drug properties in the loading solution on drug loading efficiency was investigated. Mechanism of enhanced drug loading in contact lens was further explored via studying the influence of salt on the absorption isotherm, drug solubility and water content of CLs. Applicability of this method to other CLs materials was also investigated. It was demonstrated that adjusting the ionic strength of loading solutions resulted in significant increase of drug loading in CLs. Type and concentration of the salts and solubility of the drug were the main factors influencing enhancement ratio of drug loading. The mechanism for improved drug loading was related to the reduced drug solubility in loading solutions and the reduced bound water content in contact lenses. Modulation of drug loading by adjusting ionic strength was also applicable to other CLs and the light transmittance was not affected. This method was more suitable for salt-form drugs with high solubility. In summary, adjusting ionic strength of loading solution is an economical and effective way to improve drug loading in CLs, and this simple method may also find application in other hydrogel based drug delivery systems.

Nickel-based single crystal superalloys have become the main structural materials of the aero-engines due to excellent high-temperature strength.The micro defects evolution of nickel-based single crystal superalloys under shear deformation was investigated by molecular dynamics(MD)simulations in the present study.It is found that the interfacial dislocations decompose into Shockley dislocations under low shear stress,resulting in the plastic deformation of the Ni phase.The initial plastic deformation of the Ni3Al phase is caused by Shockley dislocations cutting into the Ni3Al phase.The following deformation from low temperature to medium temperature is controlled by dislocation slip,but the deformation at high temperature is changed.It is also found that the microvoid evolution can be divided into void growth and coalescence during shear deformation.The microvoid could prevent dislocation entanglement,accelerate dislocation decomposition,and promote earlier plastic deformation under relatively low temperatures.

Flexible polymeric patches find widespread applications in biomedicine because of their biological and tunable features including excellent patient compliance,superior biocompatibility and biodegradation,as well as high loading capability and permeability of drug.Such polymeric patches are classified into microneedles(MNs),hydrogel,microcapsule,microsphere and fiber depending on the formed morphology.The combination of nanomaterials with polymeric patches allows for improved advantages of increased curative efficacy and lowered systemic toxicity,promoting on-demand and regulated drug administration,thus providing the great potential to their clinic translation.In this review,the category of flexible polymeric patches that are utilized to integrate with nanomaterials is briefly presented and their advantages in bioapplications are further discussed.The applications of nanomaterials embedded polymeric patches in non-cancerous diseases were also systematically reviewed,including diabetes therapy,wound healing,dermatological disease therapy,bone regeneration,cardiac repair,hair repair,obesity therapy and some immune disease therapy.Alternatively,the limitations,latest challenges and future perspectives of such biomedical therapeutic devices are addressed.

Uncovering the miRNA-mediated regulatory network involved in Ma bamboo(Dendrocalamus latiflorus)de novo shoot organogenesis

Bamboo is an important non-timber forest product and is well-known for its reluctance to regenerate.Recently we have established a de novo shoot organogenesis(DNSO)protocol in Ma bamboo(Dendrocalamus latiflorus)and revealed the transcriptomic dynamics during Ma bamboo regeneration,which suggested the potential roles of Ma bamboo microRNAs(DlamiRNAs)in this process.However,how DlamiRNAs regulate bamboo DNSO is poorly understood.Here we performed integrated analysis with sRNAome,degradome,and transcriptome sequencing by using samples covering the four stages of the bamboo DNSO process.A total of 727 DlamiRNAs showed differential expression during the bamboo DNSO process,and the core DlamiRNA-DlamRNA-mediated regulatory networks for bamboo DNSO were constructed.Based on the results,DlamiR156 was selected for further functional characterization of its potential roles in bamboo DNSO.Transgenic bamboos with increased DlamiR156 levels exhibited an enhancement in their regeneration efficiency.Conversely,when DlamiR156 levels were downregulated,the regeneration efficiencies of transgenic bamboos decreased.Our findings show that the DlamiRNA-mediated regulatory pathways are significant in the process of bamboo regeneration and will contribute to our understanding of the molecular mechanisms governing plant organogenesis in a more comprehensive manner.

Nomogram for predicting axillary lymph node pathological response in node-positive breast cancer patients after neoadjuvant chemotherapy

Background:Pathological complete response(pCR)of axillary lymph nodes(ALNs)is frequently achieved in patients with clinically node-positive breast cancer after neoadjuvant chemotherapy(NAC),and ALN status is an important prognostic factor for breast cancer patients.This study aims to develop a new predictive clinical model to assess the ALN pCR rate after NAC.Methods:This was a retrospective series of 467 patients who had biopsy-proven positive ALNs at diagnosis and underwent ALN dissection from 2007 to 2014 at the National Cancer Center/Cancer Hospital of the Chinese Academy of Medical Sciences.We analyzed the clinicopathologic features of the patients and developed a nomogram to predict the probability of ALN pCR.A multivariable logistic regression stepwise model was used to construct a nomogram to predict ALN pCR in node-positive patients.The adjusted area under the receiver operating characteristic curve(AUC)was calculated to quantify the ability to rank patients by risk.Internal validation was performed using the 50/50 hold-out validation method.The nomogram was externally validated with prospective cohorts of 167 patients from 2016 to 2018 at the Cancer Hospital of the Chinese Academy of Medical Sciences and 114 patients from 2018 to 2020 at Beijing Tiantan Hospital.Results:In this retrospective study,115(24.6%)patients achieved ALN pCR after NAC.Multivariate analysis showed that clinical tumor stage(Odds ratio[OR]:0.321,95%confidence interval[CI]:0.121-0.856;P=0.023);primary tumor response(OR:0.189;95%CI:0.123-0.292;P<0.001),and estrogen receptor status(OR:0.530,95%CI:0.304-0.925;P=0.025)were independent predictors of ALN pCR.The nomogram was constructed based on the result of multivariate analysis.In the internal validation of performance of nomogram,the AUCs for the training and test sets were 0.719 and 0.753,respectively.The nomogram was validated in external cohorts with AUCs of 0.720,which demonstrated good discriminatory power in these data sets.Conclusion:We developed a nomogram to predict the likel

Asymptotic analysis of a nonlinear stochastic eco-epidemiological system with feedback control

This paper proposes a new stochastic eco-epidemiological model with nonlinear incidence rate and feedback controls.First,we prove that the stochastic system has a unique global positive solution.Second,by constructing a series of appropriate stochastic Lyapunov functions,the asymptotic behaviors around the equilibria of deterministic model are obtained,and we demonstrate that the stochastic system exists a stationary Markov process.Third,the conditions for persistence in the mean and extinction of the stochastic system are established.Finally,we carry out some numerical simulations with respect to different stochastic parameters to verify our analytical results.The obtained results indicate that the stochastic perturbations and feedback controls have crucial effects on the survivability of system.

Universal workflow-based high performance geo-computation service chain platform

Based on web services and the Activiti 5.0 workflow engine,we built a workflow-based web service chain processing flow platform to simplify the operations of complex geospatial information processing applications in the area of high performance geocomputation.The proposed framework includes a web portal and a convenient mode for similar specific applications.We also designed a general service publishing template for high performance commutating(HPC)that can reduce platform differences among different HPC systems and facilitate HPC techniques to improve processing efficiency.Finally,we used a digital elevation model to verify and test the platform.The results indicated that the proposed service chain platform offered convenience and efficiency when processing massive data with HPC.

Life-cycle assessment and techno-economic analysis of the production of wood vinegar from Eucommia stem: a case study

This research undertook a case study of the life-cycle assessment and techno-economic analysis of the slow pyrolysis of Eucommia stem for the production of wood vinegar and activated carbon.The results showed that the production of one ton of wood vinegar via the slow pyrolysis of Eucommia stem show comparatively low global warming potential(2.37×10^(2) kg CO_(2) eq),primary energy demand(3.16×10^(3) MJ),acidification potential(2.19 kg SO2 eq),antimony depletion potential(3.86×10^(–4) kg antimony eq),and ozone depletion potential(7.46×10^(–6) kg CFC-11 eq)and was more environmentally friendly than the production of dilute acetic acid(12 wt%)via petrochemical routes.Meanwhile,the total capital investment,total product cost,and cash flowsheet were provided in the techno-economic analysis.Then,the net present value,internal rate of return,and dynamic payback period of the production process were evaluated.The findings indicated that while this production process is cost-effective,it might not be economically attractive or could generate investment risks.An increase in the added value of the wood vinegar and the activated carbon could remarkably improve the economic feasibility of this production process.

Anti-drilling ability of Ziliujing conglomerate formation in Western Sichuan Basin of China

The conglomerate rock is usually featured by strong heterogeneity,high abrasiveness,and poor drillability due to its complex composition and texture,which brought a huge challenge for drilling efficiency.In order to guide the drill bit selection and high-efficiency drilling,the physical,mechanical,and drillability characteristics were investigated for conglomerate rock that collected from the lower Jurassic Ziliujing formation in the Western Sichuan Basin of China.The mineral composition,SEM micro-structure,P-and S-wave velocities,uniaxial and triaxial compressive testing,drillability,abrasiveness were systematically tested and analyzed.The mechanical properties and anti-drilling ability of Ziliujing formation were proposed for a typical deep well of S-07,and the distribution characteristics were analyzed.The results indicated that the Ziliujing rock is rich-in quartz and clay minerals,due to the co-existing of strong quartz gravel and weak argillaceous cement,the Ziliujing rock shows strong heterogeneity.The relationships are roughly linear among UCS,drillability,and grinding weight loss with P-wave velocity.The Young's modulus,UCS,internal friction angle,drillability,and abrasiveness meet the Weibull distribution pattern,while only the Poisson's ratio meets the Kernel Smooth distribution pattern.Logging interpretation results reval that the Ziliujing formation has the Young's modulus of 38.61±17.08 GPa,the Poisson's ratio of 0.327±0.006,the internal friction angle of 49.21±11.00°,the drillability of 8.04±1.54,and the abrasiveness grade of 4.32±1.94.The mechanical properties and anti-drilling ability of logging interpretation are in good agreement with the experimental data.The Ziliujing formation is a kind of hard rock with strong heterogeneity,high strength,poor drillability,and medium abrasiveness.Based on the characteristics of Ziliujing formations,the SV516TAUL PDC bit with non-planar cutters was selected for the field application due to the good abrasion resistance,impact resistance,self-sharpening and the

An unprecedented tandem carbonylation/aminocarbonylation triggered by palladium-catalyzed enantio-selective Heck-type exocyclopalladation delivering chiral heterocyclicα-ketoamides has been developed.The uncommon double CO insertion into theσ-alkylpalladium intermediate takes place selectively under atmospheric pressure of CO using alkylamine as the nucleophile.Unique structures hybridizing alkylα-ketoamides with indolin-2-one containing a C3 all-carbon quaternary stereogenic center and 5,7-dihydro-6H-dibenzo[b,d]azepin-6-one containing a thermodynamically stabilized stereogenic biaryl axis are produced in good yields with excellent enantio-and/or diastereoselectivities.
