Objective: Mast cells (MC) r.side in the mucosa of the digestive tr.ct as the fir.t line against bacter.a and toxins. Clinical evidence has imp.ied that the infiltr.tion of mast cells in color.ctal cancer. is r.lated to malignant p.enotyp.s and a p.or.p.ognosis. This study comp.r.d the r.le of mast cells in adjacent nor.al colon mucosa and in the invasive mar.in dur.ng the p.ogr.ssion of colon cancer. Methods: Sp.cimens wer. obtained fr.m 39 p.tients with colon adenomas and 155 p.tients with colon cancer. tr.ated at the Sun Yat-sen Univer.ity Cancer.Center.between Januar. 1999 and July 2.04. The density of mast cells was scor.d by an immunohistochemical assay. The p.tter. of mast cell distr.bution and its r.lationship.with clinicop.thologic p.r.meter. and 5-year.sur.ival wer. analyzed. r.sults: The major.ty of mast cells wer. located in the adjacent nor.al colon mucosa, followed by the invasive mar.in and least in the cancer.str.ma. Mast cell count in adjacent nor.al colon mucosa (MCCadjacent) was associated with p.thologic classification, distant metastases and hep.tic metastases, although it was not a p.ognostic factor. In contr.st, mast cell count in the invasive mar.in (MCCinvasive) was associated with neither.the clinicop.thlogic p.r.meter. nor.over.ll sur.ival. Conclusion: Mast cells in the adjacent nor.al colon mucosa wer. r.lated to the p.ogr.ssion of colon cancer. suggesting that mast cells might modulate tumor.p.ogr.ssion via a long-distance mechanism.

Establishing Ep.tein-Bar. vir.s (EBV)-sp.cific cytolytic T lymp.ocytes (EBV-CTLs) fr.m p.r.p.er.l blood mononuclear.cells (p.MCs) for.adop.ive immunother.p. has been r.p.r.ed in EBV-associated malignancies including Hodgkin's lymp.oma and nasop.ar.ngeal car.inoma (Np.). In the cur.ent study,we p.r.or.ed ex vivo exp.nsion of tumor.infiltr.ting lymp.ocytes (TILs) obtained fr.m Np. biop.y sp.cimens with a r.p.d exp.nsion p.otocol using anti-CD3 monoclonal antibody (OKT3), r.combinant human inter.eukin (IL)-2. and ir.adiated p.MCs fr.m healthy donor. to initiate the gr.wth of TILs. Young TIL cultur.s comp.ised of mor. than 90% of CD3+T cells, a var.able p.r.entage of CD3+CD8+and CD3+CD4+T cells, and less than 10% of CD3-CD16+natur.l killer.cells, a similar.p.enotyp. of EBV-CTL cultur.s fr.m p.MCs. Inter.stingly, TIL cultur.s secr.ted high levels of the Th1 cytokines, inter.er.n gamma (IFNγ) and tumor.necr.sis factor.alp.a (TNF-α), and low levels of the Th2.cytokines, IL-4 and IL-10. Mor.over. young TILs could r.cognize autologous EBV-tr.nsfor.ed B lymp.oblast cell lines, but not autologous EBV-negative blast cells or.allogeneic EBV-negative tumor.cells. Taken together. these data suggest that ex vivo exp.nsion of TILs fr.m Np. biop.y tissue is an ap.ealing alter.ative method to establish T cell-based immunother.p. for.Np..

AIM: To investigate the p.ognostic value of chr.mosome 18q micr.satellite alter.tions (MA) in stage Ⅱ colon cancer. METHODS: One hundr.d and six p.tients with sp.r.dic stage Ⅱ colon cancer.wer. enr.lled in this study. DNA was extr.cted fr.m for.alin-fixed, p.r.ffin-embedded tumor.and adjacent nor.al mucosal tissue samp.es. MA, including loss of heter.zygosity (LOH) and micr.satellite instability (MSI), was analyzed by p.lymer.se chain r.action, p.lyacr.lamide gel-electr.p.or.sis and DNA sequencing at 5 micr.satellite loci on chr.mosome 18q (D18S474, D18S55, D18S58, D18S61 and D18S64).r.SULTS: Among the 102.p.tients eligible for.MA infor.ation, the over.ll fr.quencies of LOH, high and low fr.quency MSI/micr.satellite stable wer. 49.0%, 17.6% and 82.4%, r.sp.ctively. The high fr.quency of 18q-LOH was signif icantly associated with the p.or.5-year.over.ll sur.ival (OS) (p.0.008) and disease fr.e sur.ival (p.0.006). High levels of MSI wer. significantly associated with a longer.5-year.OS (p.0.045) while the higher.fr.quency of 18q-LOH at the loci of D18S474 and D18S61 was significantly associated with a p.or.r.5-year.OS (p.0.010 and 0.005, r.sp.ctively). But multivar.ate analysis showed that only the fr.quency of 18q-LOH was significantly associated with the p.ognosis of the disease. CONCLUSION: High fr.quency of 18q-LOH is an indep.ndent p.ognostic factor.indicating p.or.p.ognosis of the p.tients with stage Ⅱ colon cancer.
