As an important part of tumor microenvironment,neutrophils are poorly understood due to their spatiotemporal heterogeneity in *** we defined,at single-cell resolution,CD44-CxCr2-neutrophils as tumor-specific neutrophils(tsNeus)in both mouse and human gastric cancer(GC).We uncovered a Hippo regulon in neutrophils with unique YAp signature genes(e.g.,ICAM1,CD14,EGr1)distinct from those identified in epithelial and/or cancer ***,knockout of YAp/TAz in neutrophils impaired their differentiation into CD54+tsNeus and reduced their antitumor activity,leading to accelerated GC ***,the relative amounts of CD54+tsNeus were found to be negatively associated with GC progression and positively associated with patient ***,GC patients receiving neoadjuvant chemotherapy had increased numbers of CD54+***,pharmacologically enhancing YAp activity selectively activated neutrophils to suppress refractory GC,with no significant inflammation-related side ***,our work characterized tumor-specific neutrophils in GC and revealed an essential role of YAp/TAZ-CD54 axis in tsNeus,opening a new possibility to develop neutrophil-based antitumor therapeutics.

骨组织细胞主要有成骨细胞、破骨细胞、骨细胞3种类型,与细胞外基质共同维持骨的结构完整性和功能性。破骨细胞专门负责骨吸收,通过释放酸性物质和蛋白水解酶降解骨基质,发挥骨吸收作用;通过与成骨细胞相互协调,共同维持骨稳态。破骨细胞增多,导致骨吸收增加,从而引起骨质疏松症等骨骼疾病;破骨细胞生成过少,将导致骨吸收减弱等相关疾病,例如骨硬化症。因此,对于破骨细胞功能的精确调控,在维持骨稳态的平衡中发挥极为关键的作用。既往研究多从生物化学角度解释和阐述各种生物因素对破骨细胞的调控。然而越来越多的研究证实,力学刺激在破骨细胞分化过程中发挥着重要的作用。本文着眼于力学刺激对破骨细胞分化的影响,讨论力学刺激在其中可能发挥的作用,并对该领域的新发现以及未来的发展进行探讨。

Endodontic diseases are a kind of chronic infectious oral disease. Common endodontic treatment concepts are based on the removal of inflamed or necrotic pulp tissue and the replacement by gutta-percha. However, it is very essential for endodontic treatment to debride the root canal system and prevent the root canal system from bacterial reinfection after root canal therapy(rCT). recent research, encompassing bacterial etiology and advanced imaging techniques, contributes to our understanding of the root canal system’s anatomy intricacies and the technique sensitivity of rCT. success in rCT hinges on factors like patients, infection severity, root canal anatomy, and treatment techniques. Therefore, improving disease management is a key issue to combat endodontic diseases and cure periapical lesions. The clinical difficulty assessment system of rCT is established based on patient conditions, tooth conditions, root canal configuration, and root canal needing retreatment, and emphasizes pre-treatment risk assessment for optimal outcomes. The findings suggest that the presence of risk factors may correlate with the challenge of achieving the high standard required for rCT. These insights contribute not only to improve education but also aid practitioners in treatment planning and referral decision-making within the field of endodontics.

Background:prenatal and postnatal factors may have joint effects on cardiovascular health,and we aimed to assess the joint association of birth weight and ideal cardiovascular health metrics(ICVHMs)prospectively in adulthood with incident cardiovascular disease(CVD).Methods:In the UK Biobank,227,833 participants with data on ICVHM components and birth weight and without CVD at baseline were *** ICVHMs included smoking,body mass index,physical activity,diet information,total cholesterol,blood pressure,and hemoglobin *** Cox proportional hazards model was used to estimate the hazard ratios(Hrs)and 95%confidence intervals(CIs)in men and ***:Over a median follow-up period of 13.0 years(2,831,236 person-years),we documented 17,477 patients with incident *** with participants with birth weights of 2.5-4.0 kg,the Hrs(95%CIs)of CVD among those with low birth weights was 1.08(1.00-1.16)in men and 1.23(1.16-1.31)in *** association between having a birth weightrisk in men was more prominent for those agedrs than for those of older age(p for interaction=0.026).Lower birth weight and non-ideal cardiovascular health metrics were jointly related to an increased risk of *** with birth weightss score 0-1 had the highest risk of incident CVD(Hr[95%CI]:3.93[3.01-5.13]in men;4.24[3.33-5.40]in women).The joint effect(Hr[95%CI]:1.36[1.17-1.58])could be decomposed into 24.7%(95%CI:15.0%-34.4%)for a lower birth weight,64.7%(95%CI:56.7%-72.6%)for a lower ICVHM score,and 10.6%(95%CI:2.7%-18.6%)for their additive interaction in ***:Birth weight and ICVHMs were jointly related to CVD *** a normal birth weight and ideal ICVHMs may reduce the risk of CVD,and a simultaneous improvement of both prenatal and postnatal factors could further prevent additional cases in women.

目的:调查口腔颌面部肿瘤皮瓣移植患者围术期症状群和生活质量状况,分析两者的相关性。方法:对2022年12月—2023年12月就诊于上海交通大学医学院附属第九人民医院的211例口腔颌面部肿瘤皮瓣移植患者进行便利抽样,采用安德森症状评估量表-头颈部量表和华盛顿大学生存质量评估问卷量表第四版,在皮瓣移植术前、术后7天和术后1个月对患者进行纵向调查。采用spss 26.0软件包对数据进行统计学分析。结果:口腔颌面部肿瘤皮瓣移植患者在围术期3个时间点得出5个症状群,神经系统症状群在术前和术后1个月被提取出,胃肠道症状群只在术后7天被提取出,吞咽-发音障碍症状群、疾病特异性症状群以及疲乏-睡眠-情绪症状群持续存在于整个围术期。3个时间点生活质量总分比较有统计学差异(ppr^(2)=0.613,pr^(2)=0.614,pr^(2)=0.577,p<0.05)。结论:口腔颌面部肿瘤皮瓣移植患者围术期存在多个稳定的症状群,症状群越严重,患者生活质量越差。针对症状群采取有效的干预措施,可降低症状群严重程度,提高患者生活质量。

We has synthesized the biocompatible gelatin reduced graphene oxide(GOG)in previous research,and in this study we would further evaluate its effects on bone remodeling in the aspects of osteoclastogenesis and angiogenesis so as to verify its impact on accelerating orthodontic tooth *** mouse orthodontic tooth movement(OTM)model tests in vivo showed that the tooth movement was accelerated in the GOG local injection group with more osteoclastic bone resorption and neovascularization compared with the pBs injection *** analysis on the degradation of GOG in bone marrow stromal stem cells(BMsCs)illustrated its good biocompatibility in vitro and the accumulation of GOG in spleen after local injection of GOG around the teeth in OTM model in vivo also didn’t influence the survival and life of *** co-culture of BMsCs with hematopoietic stem cells(HsCs)or human umbilical vein endothelial cells(HUVECs)in transwell chamber systems were constructed to test the effects of GOG stimulated BMsCs on osteoclastogenesis and angiogenesis in *** the GOG stimulated BMsCs co-culture in upper chamber of transwell,the HsCs in lower chamber manifested the enhanced ***,the co-culture of GOG stimulated BMsCs with HUVECs showed a promotive effect on the angiogenic ability of *** mechanism analysis on the biofunctions of the GOG stimulated BMsCs illustrated the important regulatory effects of pErK pathway on osteoclastogenesis and *** the results showed the biosecurity of GOG and the biological functions of GOG stimulated BMsCs in accelerating bone remodeling and tooth movement.

为探究抗原特异性T细胞功能动态变化与卡介苗(Bacillus Calmette-Guérin,BCG)保护时效的相关性,采用酶联免疫斑点法(enzyme-linked immunospot assay,ELIspOT)和流式细胞术对上海地区健康志愿者外周抗原特异性CD4^(+)T细胞应答水平进行评估。结果显示,IFN-γ水平随着个体年龄的增长而升高(pr T cell,T_(eff))比例显著升高(pry T cell,T_(M))比例显著下降(ppprium tuberculosis,Mtb)感染中发挥一定的保护作用。因此,设计特定的亚单位结核疫苗以加强长效免疫T_(M)的保护性将对结核病的防控具有重要意义。

BACKGrOUND studies suggested that remote ischemic preconditioning(rIpC)may effectively lessen the harmful effects of ischemia reperfusion injury during organ transplantation *** To investigate the protective effects of rIpC on living liver donors and recipients following pediatric liver *** From January 2016 to January 2019 at renji hospital Affiliated with shanghai jiao tong university school of medicine,208 donors were recruited and randomly assigned to four groups:s-rIpC group(no intervention;n=55),D-rIpC group(donors received rIpC;n=51),r-rIpC group(recipients received rIpC,n=51)and Dr-rIpC group(both donors and recipients received rIpC;n=51).We primarily evaluated postoperative liver function among donors and recipients and incidences of early allograft dysfunction,primary nonfunction and postoperative complications among *** rIpC did not significantly improve alanine transaminase and aspartate aminotransferase levels among donors and recipients or decrease the incidences of early allograft dysfunction,primary nonfunction,and postoperative complications among *** protective effects were observed,including a lower creatinine level in the D-rIpC group than in the s-rIpC group on postoperative day 0(pr,no significant improvements were found in donors who received ***,rIpC had no effects on the overall survival of *** The protective effects of rIpC were limited for recipients who received living liver transplantation,and no significant improvement of the prognosis was observed in recipients.
