Costunolide alleviates HKsA-induced acute lung injury via inhibition of macrophage activation

staphylococcus aureus(***)infection leads to a severe inflammatory response and causes acute lung injury(ALI),eventually threatening human ***,it is of importance to find an agent to inhibit inflammation and reduce ***,we found that costunolide,a sesquiterpene lactone,displays anti-inflammatory effects and ameliorates heat-killed ***(HKsA)-induced *** treatment attenuated HKsA-induced murine ALI in which pulmonary neutrophil infiltration was inhibited,lung edema was decreased,and the production of pro-inflammatory cytokines was significantly *** addition,costunolide dose-dependently inhibited the generation of IL-6,TNF-α,IL-1β,and keratinocyte-derived cytokine(KC),as well as the expression of iNOs,in HKsA-induced ***,costunolide attenuated the phosphorylation of p38 MApK and cAMp response element-binding protein(CrEB).Collectively,our findings suggested that costunolide is a promising agent for alleviating bacterialinduced ALI via the inhibition of the MApK signaling pathways.

Diabetic retinopathy(Dr)is one of the major causes of visual impairment in adults with *** coherence tomography angiography(OCTA)is nowadays widely used as the golden criterion for diagnosing ***,wide-field OCTA(WF-OCTA)provided more abundant information including that of the peripheral retinal degenerative changes and it can contribute in accurately diagnosing *** need for an automatic Dr diagnostic system based on WF-OCTA pictures attracts more and more attention due to the large diabetic population and the prevalence of retinopathy *** this study,automatic diagnosis of Dr using vision transformer was performed using WF-OCTA images(12 mm×12 mm single-scan)centered on the fovea as the ***-OCTA images were automatically classified into four classes:No Dr,mild nonproliferative diabetic retinopathy(NpDr),moderate to severe NpDr,and proliferative diabetic retinopathy(pDr).The proposed method for detecting Dr on the test set achieves accuracy of 99.55%,sensitivity of 99.49%,and specificity of 99.57%.The accuracy of the method for Dr staging reaches up to 99.20%,which has been proven to be higher than that attained by classical convolutional neural network *** show that the automatic diagnosis of Dr based on vision transformer and WF-OCTA pictures is more effective for detecting and staging Dr.

promoting white adipose tissue(WAT)browning and enhancing brown adipose tissue(BAT)activity are attractive therapeutic strategies for obesity and its metabolic *** sympathetic innervation in WAT and BAT represents a promising therapeutic ***,there are few reports on extracellular microenvironment remodeling,especially changes in nerve terminal *** the key molecules mediating the neuro-adipose synaptic junctions is a key *** this study,we used bioinformatics methods to identify the differentially expressed predicted secreted genes(DEpsGs)during WAT browning and BAT *** DEpsGs largely reflect changes of cytokines,extracellular matrix remodeling,vascularization,and adipocyteneuronal *** then performed functional enrichment and cellular distribution specificity *** upregulated and downregulated DEpDGs during WAT browning displayed a distinctive biological pattern and cellular *** listed a cluster of adipocyte-enriched DEpsGs,which might participate in the cross-talk between mature adipocytes and other cells;then identified a synaptogenic adhesion molecule,Clstn3,as the top gene expressed enriched in both mature white and brown *** Q-pCr and immunohistochemistry,we found significantly increased Clstn3 expression level during WAT browning and BAT activation in mice subjected to cold exposure(4℃).We further demonstrated that treatment with isoproterenol significantly increased Clstn3 and UCp1 expression in differentiated white and beige adipocytes in *** conclusion,our study demonstrates that the secretion pattern was somewhat different between WAT browning and BAT *** reveal that Clstn3 may be a key gene mediating the neuro-adipose junction formation or remodeling in WAT browning and BAT activation process.
