Identification of an immune classifier for predicting the prognosis and therapeutic response in triple-negative breast cancer

Triple-negative breast cancer(TNBC)poses a significant challenge due to the lack of reliable prognostic gene signatures and an understanding of its immune ***:We analyzed clinical information and mRNA expression data from 162 TNBC patients in TCGA-BRCA and 320 patients in *** weighted gene coexpression network analysis,we pinpointed 34 TNBC immune genes linked to *** least absolute shrinkage and selection operator Cox regression method identified key TNBC immune candidates for prognosis *** calculated chemotherapy sensitivity scores using the“pRRophetic”package in R software and assessed immunotherapy response using the Tumor Immune Dysfunction and Exclusion ***:In this study,34 survival-related TNBC immune gene expression profiles were identified.A least absolute shrinkage and selection operator-Cox regression model was used and 15 candidates were prioritized,with a concomitant establishment of a robust risk immune *** high-risk TNBC immune groups showed increased sensitivity to therapeutic agents like RO-3306,Tamoxifen,Sunitinib,JNK Inhibitor VIII,XMD11-85h,BX-912,and *** analysis of the Search Tool for Interaction of Chemicals database revealed the associations between the high-risk group and signaling pathways,such as those involving Rap1,Ras,and *** low-risk group showed a higher immunotherapy response rate,as observed through the tumor immune dysfunction and exclusion analysis in the TCGA-TNBC and METABRIC-TNBC ***:This study provides insights into the immune complexities of TNBC,paving the way for novel diagnostic approaches and precision treatment methods that exploit its immunological intricacies,thus offering hope for improved management and outcomes of this challenging disease.

Magnetic activated carbon for the removal of methyl orange from water via adsorption and Fenton-like degradation

Water pollution caused by organic dyes is a critical environmental *** activated carbon(AC)is commonly used for dye adsorption,its effectiveness is limited by challenges in separation and *** address these limitations,a convenient recyclable magnetic activated carbon(MAC)was fabricated via co-precipitation and calcination method,serving as adsorbent and catalyst for methyl orange(MO)removal through a Fenton-like degradation *** techniques,including XRD,FTIR,SEM and TEM,confirmed that Fe_(3)O_(4) nanoparticles(10–20 nm)were uniformly dispersed on AC *** MAC maintaining a high surface area(997 m^(2)/g)and pore volume(0.795 cm^(3)/g)and exhibited superparamagnetic properties with a saturated magnetization of 5.52 emu/g,enabling effective separation from aqueous solutions by *** adsorption studies revealed that MO adsorption onto MAC followed pseudo-second-order kinetic and Freundlich isotherm model,with a maximum adsorption capacity of 205 mg/g at 25℃.Thermodynamic analysis showed that the adsorption process was spontaneous and *** degradation of MO and in-situ regeneration of MAC were achieved via Fenton-like reaction using sodium persulfate(PS).Under a PS concentration of 9 mmol/L,the MO removal efficiency near 95%after 60 min,with a total organic carbon(TOC)reduction of 83.1%.The reaction of Fe_(3)O_(4) and oxygen functional groups on AC surface with PS facilitated the generation of SO_(4)^(·-),thereby enhancing catalytic degradation of *** degradation efficiency improved as the temperature increased from 25℃ to 45℃.Cycle tests demonstrated that the MO removal efficiency of MAC remained above 90%after 5 cycles of ***,this study highlights the potential of MAC for efficient removal of organic dyes from water through the coupling of adsorption and Fenton-like degradation,providing a promising solution for addressing water pollution challenges.

Paleogene marine deposition records of rifting and breakup of the South China Sea:An overview

A compilation of available marine deposition data from offshore S-SE China reveals evidence of rifting and breakup of the South China Sea(SCS) during the Paleogene. Marine deposition started earlier in the Paleocene in the East China Sea(ECS)-Taiwan region before expanding southwestward into the SCS region in the middle Eocene. Our data indicate the existence of an elongated Paleogene China Sea in these areas stretching along the northeasterly structural belts, probably as part of the marginal western paleo-Pacific. The southwestward shift of marine influence in the middle Eocene was responding to a period of intensive rifting and subsidence in the SCS region, while the sea in the ECS-Taiwan region started to shrink and shoal after the late Eocene, likely associated with local breakup and initial spreading in the Taiwan-Taixinan Basin area. The accumulation of hemipelagic sediments at ODP 1148 and IODP U1435 from near the continent-ocean boundary and at many other shelf-slope sites was in response to a large-scale breakup 34 to 33 Ma ago, subsequently leading to the birth of the SCS in the Oligocene.

A New DS-SS Signal Detection Trial Algorithm for False Alarm Rejection Based on Motion Parameters Constraint

To reject false alarm, a new detection trial algorithm named motion parameters constraint for Direct sequence spread spectrum(DS-SS) signal is proposed. A mathematical model based on uniform motion between two adjacent detections is established to analyze the overall performance of the proposed trial algorithm. Motion parameters constraint is applied to the conventional variable dwell time Tong trial algorithm. The expressions of the false alarm probability and detection probability are derived. Theoretical analysis and simulation results demonstrate that in comparison with the conventional M of N trial algorithm, Tong trial algorithm and Tong trial algorithm based on near neighbor constraint. The proposed trial algorithm can significantly reduce the false alarm probability and enhance the detection ability without increasing search speed and complexity.

Comprehensive Analysis of Differential Gene Expression Profile via RNA Sequencing in the Human Ovarian Cancer SKOV3 Cells Treated with Simvastatin

The experimental studies have demonstrated that some statins have an anticancer effect against ovarian cancer in vitro and in vivo, however, the potential molecular mechanism is still unclear. In the present study, high throughput RNA sequencing(RNA-seq) technology was applied to exploring the mRNA changes treated with Simvastatin in the human ovarian cancer SKOV3 cell line. The result of CCK-8 assay shows that Simvastatin inhibits significantly SKOV3 cell viability in a concentration-dependent manner, and cell cycle according to the flow cytometry analysis result shows cell cycle arrest of G1 phase is induced in SKOV3 cells with 10 μg/mL simvastatin added for 24 h. The differential expression genes(DEGs) analysis of RNA-seq data shows that there are a total of 372 DEGs found in the Simvastatin group, of which 150 mRNAs are up-regulated and 222 mRNAs are down-regulated(fold change>2.0, q value < 0.05). Real time reverse transcription-polymerase chain reaction(qRT-PCR) experiment has validated that the expression of KLF2, RHOB and CIDEB is up-regulated while that of DKK1, AMOTL2 and ANKRD1 is down-regulated. The DEGs are enriched in thirteen significant KEGG pathways including cell cycle, DNA replication, apoptosis, etc. This study provides supported useful information about transcriptome profiles of SKOV3 cell treated with simvastatin, and offers an application basis for the further researches on the mechanism of Simvastatin in the treatment of ovarian cancer.

QoE-Driven Big Data Management in Pervasive Edge Computing Environment

In the age of big data, services in the pervasive edge environment are expected to offer end-users better Quality-of-Experience(QoE) than that in a normal edge environment. However, the combined impact of the storage, delivery, and sensors used in various types of edge devices in this environment is producing volumes of high-dimensional big data that are increasingly pervasive and redundant. Therefore, enhancing the QoE has become a major challenge in high-dimensional big data in the pervasive edge computing environment. In this paper, to achieve high QoE, we propose a QoE model for evaluating the qualities of services in the pervasive edge computing environment. The QoE is related to the accuracy of high-dimensional big data and the transmission rate of this accurate data. To realize high accuracy of high-dimensional big data and the transmission of accurate data through out the pervasive edge computing environment, in this study we focused on the following two ***, we formulate the issue as a high-dimensional big data management problem and test different transmission rates to acquire the best QoE. Then, with respect to accuracy, we propose a Tensor-Fast Convolutional Neural Network(TF-CNN) algorithm based on deep learning, which is suitable for high-dimensional big data analysis in the pervasive edge computing environment. Our simulation results reveal that our proposed algorithm can achieve high QoE performance.

Prediction of Viscoelastic Behavior of Unidirectional Polymer Matrix Composites

To develop a novel method predicting the viscoelastic behavior of polymer matrix composites according to the viscoelasticity of the matrix, we used the viscoelastic model of the matrix to build new models for unidirectional composites in both 0° and 90° directions. Viscoelastic parameters for both new models were derived, and the obtained equations shared the same form as the viscoelastic constitutive equation of matrix material. The viscoelastic behaviors of matrix material and unidirectional composites were also tested. Results showed that fitting parameters of creep compliance equation were close to the theoretical values of viscoelastic constitutive parameters of the unidirectional composites, proving the validity of the models. A new method was obtained to predict the viscoelastic property of the unidirectional composites based on the viscoelastic property of composite matrix and elastic property of the unidirectional composites. This method provides a theoretical basis for future studies on the viscoelasticity of composite laminates.

为探究干香菇在复水过程中质地及风味的变化,本研究将干香菇于不同温度条件下进行恒温复水处理,采用电子舌、顶空固相微萃取-气相色谱-质谱联用技术和激光扫描共聚焦显微镜等技术对复水阶段香菇的质地及风味特性进行分析。结果表明,不同复水温度和时间对复水香菇的质地、挥发性风味和滋味均有影响。其中,40℃复水30 min时的复水比达到平衡,鲜味强度值较高,但与其他组相比持水率有所下降,硬度和咀嚼性适中,弹性达到最大值,其菌盖褐变指数相对较高,但内褶褐变不明显,微观结构显示其菌丝较为舒展,剪切力适中。此外,40℃复水的香菇其抗氧化性高于100℃复水的香菇,可溶性蛋白损失更少,游离氨基酸含量更高并在30 min达到最大值。同时以含硫化合物和酮类为主的特征风味物质得到较好保留,而100℃复水时特征性化合物含量下降严重。综上,复水过程中香菇的质地及风味会发生显著变化,将干香菇于40℃中复水30 min能够获得较好的品质。

Construction and expression of a humanized M_2 autoantigen trimer and its application in the diagnosis of primary biliary cirrhosis

AIM: To construct and express a humanized M2 autoantigen trimer designated as BPO and to apply it in the diagnosis of primary biliary cirrhosis (PBC).METHODS: cDNA fragments encoding M2-reactive epitopes of pyruvate dehydrogenase complex Ez (PDCE2), branched chain 2-oxo-acid dehydrogenase complex E2 (BCOADC-E2) and 2-oxo-glutarate dehydrogenase complex E2 (OGDC-E2) were amplified with PCR using total RNA extracted from human peripheral mononuclear blood cells. The fragments were cloned into the plasmid vector pQE-30 and then transferred into E. coliM15 (pREP4) for expression, which was induced by isopropylthio-β-Dgalactoside. The expressed recombinant BPO protein was demonstrated by SDS-PAGE, Western-blotting and Immunoabsorption test, its antigenic reactivity and specificity were identified with seven M2-positive sera confirmed at Euroimmun Research Center (Germany).Using the purified BPO, M2 antibodies in sera from patients with PBC and other liver related diseases were detected with ELISA.RESULTS: The expressed BPO was observed with both antigenic reactivity and specificity of M2 autoantigens. The determination of M2 antibodies by BPO with ELISA was more sensitive than using the Euroimmun's kit with the coefficients of variation less than 10 % in both interassay and *** the newly established method, M2 antibodies were found in 100 % (20/20) of patients with PBC. Six cases of liver disease with unknown etiology and 1 patient with drug induced liver injury had detectable levels of serum M2antibodies. There were also 2 patients with autoimmune cholangitis and 1 with autoimmune hepatitis showing M2-antibody positive.CONCLUSION: Compared with the routine immunofluorescenoe assay and commercially available assay kit using porcine heart mitochondrial protein as the antigen, the detection system established in the present study shows higher sensitivity and specificity and may be used as a powerful tool for the diagnosis of PBC.
