A natural gas hydrate-oil-gas system in the Qilian Mountain permafrost area, northeast of Qinghai-Tibet Plateau

Natural gas hydrate,oil and gas were all found together in the Qilian Mountain permafrost area,northeast of Qinghai-Tibet Plateau,*** are closely associated with each other in space,but whether they are in any genetic relations are unknown *** this paper,a hydrocarbon gas-generation series,gas-fluid migration series and hydrocarbon gas-accumulation series are analyzed to probe the spatial,temporal and genetic relationships among natural natural gas hydrate,oil and *** subsequent results show that natural gas hydrate,oil and gas actually form a natural gas hydrate-oil-gas *** on the Middle Jurassic and the Upper Triassic hydrocarbon gas-generation series,it is divided into four major sub-systems in the study area:(1)A conventional Upper Triassic gas-bearing sub-system with peak hydrocarbon gas-generation in the late Middle Jurassic;(2)a conventional Middle Jurassic oil-bearing sub-system with low to mature hydrocarbon gas-generation in the late Middle Jurassic;(3)a natural gas hydrate sub-system with main gas source from the Upper Triassic gas-bearing sub-system and minor gas source from the Middle Jurassic oil-bearing sub-system as well as little gas source from the Middle Jurassic coal-bed gas and the microbial gas;(4)a shallower gas sub-system with microbial alteration of the main gas source from the Upper Triassic gas-bearing *** natural gas hydrate-oil-gas system and its sub-systems are not only theoretical but also practical,and thus they will play an important role in the further exploration of natural gas hydrate,oil and gas,even other energy resources in the study area.

The coronavirus disease 2019(COVID-19)pandemic has devastated global *** key host factors essential for SARS-CoV-2 RNA replication is expected to unravel cellular targets for the development of broad-spectrum antiviral drugs which have been quested for the preparedness of future viral ***,we have identified host proteins that associate with nonstructural protein 12(nsp12),the RNA-dependent RNA polymerase(RdRp)of SARS-CoV-2 using a mass spectrometry(MS)-based proteomic *** the candidate factors,CDK2(Cyclin-dependent kinase 2),a member of cyclin-dependent kinases,interacts with nsp12 and causes its phosphorylation at T20,thus facilitating the assembly of the RdRp complex consisting of nsp12,nsp7 and nsp8 and promoting efficient synthesis of viral *** crucial role of CDK2 in viral RdRp function is further supported by our observation that CDK2 inhibitors potently impair viral RNA synthesis and SARS-CoV-2 *** together,we have discovered CDK2 as a key host factor of SARS-CoV-2 RdRp complex,thus serving a promising target for the development of SARS-CoV-2 RdRp inhibitors.

Background： Ambient aerosol fine particulate matter （PM2.5） is associated with male reproductive toxicity in experiments and may have adverse effects in the female. However, studies evaluating the protective effects and precise mechanisms of aspirin, Vitamin C, Vitamin E, or ozone against toxic effects of PM2.5 are sparse. This study was conducted to investigate the possible protective effects and mechanisms of aspirin, Vitamin C, Vitamin E, or ozone on fertility in female mice treated with PM2.5, Methods： Eighty-four ICR mice were divided into six groups： control group, PM2.5 group, PM2.5 ＋ aspirin group, PM2.5 ＋ Vitamin C group, PM2.5 ＋ Vitamin E group, and PM2.5 ＋ ozone group. PM2.5 was given by intratracheal instillation every 2 days for 3 weeks. Aspirin, Vitamin C, and Vitamin E were given once a day by oral gavage for 3 weeks, and ozone was administered by intraperitoneal injection once a day for 3 weeks. The levels ofanti-Mullerian honnone （AMH）, interleukin-6 （IL-6）, tumor necrosis factor-alpha （TNF-α）, and 8-hydroxy-2＇-deoxyguanosine （8-OHdG） were measured using enzyme-linked immunosorbent assay. Western blotting analysis was used to analyze the expressions ofBcl-2, Bax, and caspase-3 in ovaries. Changes in histological structure were examined by light microscope and electron microscopy was used to detect ultramicrostructure. Results： The results demonstrated that PM2.5 decreased AMH levels （P 〈 0.001）; however, aspirin （P 〈 0.001）, Vitamin C （P 〈 0.001）, Vitamin E （P = 0.001）, and ozone （P = 0.002） alleviated the decrease. Changes of IL-6, TNF-α, 8-OHdG, Bax/Bcl-2, and caspase-3 in PM2.5 group were increased compared to control group （P 〈 0.001）, while in PM2.5 ＋ aspirin, PM2.5 ＋ Vitamin C, PM2.5 ＋ Vitamin E, and PM2.5 ＋ ozone groups, they were statistically decreased compared to PM2.5 group （P 〈 0.001 or P 〈 0.05）. Identification of a NF-кB site in the negative regulatory element (ε-NRAII) of human ε-globin gene and its binding protein NF-кB p50 in the nuclei of K562 cells

The developmental control of the human e-globin gene expression is mediated by transcription regulatory elements in the 5’ flanking DNA of this gene. Sequence analysis has revealed a DNA motif (GGGGAATTTGCT) similar to NF-кB consensus sequence resides in the negative regulatory element (-3028bp～ -2902bp, termed ε-NRAII) 5’ to the cap site of this gene. NRF DNA fragment (-3010bp～ -2986bp) containing the NF-кB motif similar sequence was synthesized and used in electrophoresis mobility shift assay (EMSA) and competitive analysis. Data showed that a protein factor from nuclear extracts of K562 cells specifically interacted with NRF DNA fragment. The synthetic NF DNA fragment (containing NF-кB consensus sequence) could competed for the protein binding, but MNF DNA fragment (mutated NF-кB motif) could not, suggesting that the binding protein is a member of NF-кB/Rel family. Western blot assay demonstrated that the molecular weight of NF-кB protein in the nuclei of K562 cells is 50ku. We suggested that NF-кB p50 may play an important role in the regulation of human c-globin gene expression.

HEL cells, a human erythroleukemia cell line, mainly express the fetal (r)globin gene and trace amount of the embryonic (E)globin gene, but not adult (B) globin gene. Here we show that hydroxyurea (HU) can induce HEL cells to express adult (B) globin gene and lead these cells to terminal differentiation. Results showed in Gel mobility shift assays that GATA factors could specifically bind to the regulatory elements of human B- globin gene, including the proximal regulatory element (the B- promoter) and the distal regulatory elements (the DNase I hypersensitive sites in the LCR, HS2-HS4 core sequences). However, the DNA binding patterns of GATA factors were quite different between HU-induced and uninduced HEL cells. Western-blot analysis of nuclear extracts from both the uninduced and HU- induced HEL cells revealed that the level of GATA-2 transcription factor decreased, whereas the level of GATA-1 transcription factor increased following the time of hydroxyurea induction. Furthermore, using RT-PCR analysis the expression of human B-globin gene in HU-induced HEL cells could be blocked again when HEL cells were incubated in the presence of antisense oligonucleotides for hGATA-1, suggesting that the upregulation of hGATA-1 transcription factor might be critical for the expression of human β- globin gene in HU-induced HEL cells.

利用脉冲电压法在自组装制得的柔性石墨烯纸表面聚合聚苯胺,以该复合材料作为甲醇燃料电池的阳极电极基体,采用循环伏安法在其表面电沉积纳米Pd制备出无需外加粘接和支撑的催化电极。通过扫描电子显微镜、X射线光电子能谱和红外光谱分析该材料的表面形貌和化学组成,并通过电化学测试该电极在甲醇氧化过程中的催化性能。结果表明,该阳极材料柔韧性好,原料利用率高,聚苯胺在石墨烯纸表面分散良好,形貌均一,聚苯胺的存在提高了催化剂的催化效率,使得甲醇的氧化峰电流密度从3 mA·mg^(-1)提高到67 mA·mg^(-1),且延长了催化剂的使用寿命,正向与反向扫描的阳极峰值电流密度的比值(jf/jb值)达到5.7。

地震前兆异常性质的准确判断是提高地震预测水平的前提。据国内外相关统计,真正具有地震预测意义的地震前兆异常数量十分有限。因此,厘清震前异常与地震的关系对今后异常识别意义重大。2017年8月8日九寨沟M_S7.0地震前4个月,甘肃清水温泉水氡出现大幅波动异常变化。为了判断这次异常与九寨沟地震的关系,在异常持续期间,通过抽水实验、水化学组分以及氢氧同位素组成分析,并结合九寨沟地震发震构造以及其他前兆异常空间分布特征,对水氡异常成因机理进行了深入分析。结果表明:对于水氡与水位的变化关系,实验与实际观测结果一致,都呈正相关变化;实验中水氡变化幅度与实际观测的相当;水化学组分以及氢氧同位素测试结果显示清水温泉水氡异常期间,来自地壳深部的流体并未增加;清水温泉区其他前兆没有出现明显的短期异常变化;在空间上清水温泉与九寨沟地震震源区被两条深大断裂隔开。因此,清水温泉水氡大幅异常变化并非九寨沟地震的短期前兆异常,而是企业抽水开始时间、持续时间以及用水量不固定引起的。该实例为地震前流体前兆异常性质的准确判定提供了更为可靠的方法和思路。

Ideal Cardiovascular Health Metrics and Coronary Artery Calcification in Northern Chinese Population:A Cross-sectional Study

Objective Coronary artery calcification（CAC） is a well-established risk predictor of coronary heart disease events and is recognized as an indicator of subclinical *** A cross-sectional study consisting of 2999 participants aged ≥40 years from the Jidong community of Tangshan City,an industrial and modern city of China,was conducted between 2013 and 2014 to examine the association between the ideal cardiovascular health（CVH） metrics and *** ideal CVH metrics were determined based on the definition of the American Heart Association（AHA）.The participants were then grouped into 4 categories according to the quartiles of their CVH metric scores as follows：first quartile（0-2）,second quartile（3）,third quartile（4）,and fourth quartile（5-7）.CAC was assessed by using high-pitch dual-source CT,and patients were identified based on thresholds of 0,10,100,or 400 Agatston units,as per common *** The prevalence of subclinical atherosclerosis was 15.92%,13.85%,6.76%,and 1.93%,determined by using the CAC scores at thresholds of 0,10,100,and 400 Agatston units,*** with the group in the first quartile,the other three CVH groups had a lower odds ratio of CAC 0 after adjusting for age,sex,income level,education level,and alcohol use in the logistic regression *** odds ratios in these groups were 0.86 [95% confidence interval（CI）,0.63-1.17;P0.05],0.75（95% CI,0.55-1.02;P0.05）,and 0.49（95% CI,0.35-0.69;P0.05）,*** associations of CAC with the CVH metrics were consistent when different CAC cutoff scores were used（0,10,100,or 400）.Conclusion The participants with more-ideal cardiovascular metrics had a lower prevalence of subclinical atherosclerosis determined according to CAC *** an ideal cardiovascular health may be valuable in the prevention of atherosclerosis in the general population.
