The necrotrophic fungus, Sclerotinia sclerotiorum, employs an array of cell wall-degrading enzymes(CWDEs), including cellulase, to dismantle host cell walls. However, the molecular mechanisms through which S. sclerotiorum degrades cellulose remain elusive. Here, we unveil a novel secretory cellobiohydrolase, SsdchA, characterized by a signal peptide and a Glyco_hydro_7(GH7) domain. SsdchA exhibits a robust expression of during early infection stages. Interestingly, colony morphology and growth rates remain unaffected across the wild-type, SsdchA deletion strains and SsdchA overexpression strains on potato dextrose agar(PDA) medium. Nevertheless, the pathogenicity and cellobiohydrolase activity decreased in the SsdchA deletion strains, but enhanced in the SsdchA overexpression strains. Moreover,the heterologous expression of SsdchA in Arabidopsis thaliana leads to reduced cellulose content and heightened susceptibility to S. sclerotiorum. Collectively, our data underscore the pivotal role of the novel cellobiohydrolase SsdchA in the pathogenicity of S. sclerotiorum.

Small cell lung cancer(SCLC)is a highly malignant and heterogeneous cancer with limited therapeutic options and prognosis prediction ***,we analyzed formalin-fixed,paraffin-embedded(FFPE)samples of surgical resections by proteomic profiling,and stratified SCLC into three proteomic subtypes(S-I,S-II,and S-III)with distinct clinical outcomes and chemotherapy *** proteomic subtyping was an independent prognostic factor and performed better than current tumor–node–metastasis or Veterans Administration Lung Study Group staging *** subtyping results could be further validated using FFPE biopsy samples from an independent cohort,extending the analysis to both surgical and biopsy *** signatures of the S-II subtype in particular suggested potential benefits from *** overexpressed proteins in S-III,the worst prognostic subtype,allowed us to nominate potential therapeutic targets,indicating that patient selection may bring new hope for previously failed clinical ***,analysis of an independent cohort of SCLC patients who had received immunotherapy validated the prediction that the S-II patients had better progression-free survival and overall survival after first-line ***,our study provides the rationale for future clinical investigations to validate the current findings for more accurate prognosis prediction and precise treatments.

The rational design of a specific co-drug delivery platform that can address the unavoidable resistance, toxic side effects and low targeting efficiency of traditional cancer treatments is of great ***, Zn-based MOF-zeolitic imidazole framework-90(ZIF-90) was selected as the drug delivery carrier, with the cancer therapeutic drug mercaptopurine(6-MP) and glucose oxidase(GOD) as the drug models, hyaluronic acid(HA) was used for protection and targeted delivery, which designed and fabricated an intelligent drug delivery platform(6-MP@ZIF-90@GOD@HA). This platform aims to reduce the toxic side effects of 6-MP and enhance its efficacy while improving the targeting ability of GOD,thereby further enhancing the treatment outcome. Notably,results from both in vitro and in vivo experiments demonstrate that the targeted synergistic chemo/reactive oxygen species(ROS)-mediated/starvation therapy inhibited the cancer cell growth while reducing the chemotherapy toxicity, which provides new possibilities for the development of more precise and effective treatment strategies.

Our primary objective is to mitigate the adverse effects of temperature fluctuations on the optical frequency transmission system by reducing the length of the *** optimization,the phase-temperature coefficient of the optical system is reduced to approximately 1.35 fs/*** applying a sophisticated temperature control to the remained“out-of-loop”optics fiber,the noise floor of the system has been effectively lowered to 10−21 *** on this performance-enhanced transfer system,we demonstrate coherent transmission of optical frequency through 500-km spooled fiber *** being actively compensated,the transfer instability of 4.5×10^(−16) at the averaging time of 1 s and 5.6×10^(−21) at 10000 s is *** frequency uncertainty of received light at remote site relative to that of the origin light at local site is achieved to be 1.15×10^(−19).This enhanced system configuration is particularly well suited for future long-distance frequency transmission and comparison of the most advanced optical clock signals.

The semi-hydrogenation of alkyne to form Z-olefins with high conversion and high selectivity is still a huge challenge in the chemical ***,flammable and explosive hydrogen as the common hydrogen source of this reaction increases the cost and danger of industrial ***,we connect the photocatalytic hydrogen evolution reaction and the semihydrogenation reaction of alkynes in series and successfully realize the high selective production of Z-alkenes using low-cost,safe,and green water as the proton *** the cascade reaction,a series of isomorphic metal–organic cage catalysts(Co_(x)Zn_(8−x)L_(6),x=0,3,4,5,and 8)are designed and synthesized to improve the yield of the photocatalytic hydrogen *** them,Co_(5)Zn_(3)L_(6) shows the highest photocatalytic activity,with a H_(2) generation rate of 8.81 mmol g^(−1) h^(−1).Then,Co_(5)Zn_(3)L_(6) is further applied in the above tandem reaction to efficiently reduce alkynes to Z-alkenes under ambient conditions,which can reach high conversion of>98%and high selectivity of>99%,and maintain original catalytic activity after multiple ***“one-pot”tandem reaction can achieve a highly selective and safe stepwise conversion from water into hydrogen into Z-olefins under mild reaction conditions.

Permafrost is degrading globally,particularly those with low thermal stability on the Qinghai-Tibet Plateau,owing to climate ***,the inadequacy of direct research on permafrost degradation based on in-situ monitoring limits the prediction of permafrost degradation and engineering *** study explored the processes and modes of permafrost degradation into talik by analyzing ground temperature data from five points in the hinterland of the Qinghai-Tibet Plateau from 2006 to *** results showed that the degradation of the warm permafrost layer with a geothermal gradient of zero occurred simultaneously in the top and bottom *** rate of permafrost degradation from the top down and bottom up increase during the degradation process,but the increase of the former is more drastic after the formation of thawed ***,the construction of the Qinghai-Tibet Railway changed the degradation modes of the permafrost in adjacent natural sites through horizontal heat transfer,particularly after through talik formation under the *** findings suggest that taking countermeasures before or immediately after forming thawed interlayer is more *** evaluating the thermal impact of projects in warm permafrost regions,special attention should be given to the horizontal heat transfer process that may result from the formation of a through talik.

Prediction of specific spoilage organisms in smoked chicken legs with modified atmosphere packaging at 4 ℃ using multivariate statistical analysis

The present study monitored bacterial succession， physicochemical properties， and volatile organic compounds (VOCs) changes in smoked chicken legs with modified atmosphere packaging (MAP， 60% CO2and 40% N2) during a 25-day storage period at 4 ℃. After 15 days of storage， Serratia proteamaculans and Pseudomonas fragi became the predominant bacteria. Furthermore， physicochemical properties changed significantly， as evidenced by an increase in thiobarbituric acid reactive substances and b* (yellowness) value， and a decrease in hardness. A total of 65 VOCs were identified during storage. Correlation between bacterial succession and quality indicators (including VOCs and physicochemical properties) allowed the identification of 26 core dominant bacteria， including S. proteamaculans， Psychrobacter alimentarius， Pseudomonas putide， and Pseudomonas poae， which were positively related to spoilage VOCs (e.g.， 1-octen-3-ol， 1-pentanol， and 3-methyl-1-butanol) and could be defined as specific spoilage organisms (SSOs). The results of this study provide a systematic approach to predict SSOs in smoked chicken legs during storage， which can also provide a basis for product safety.

With the exploding prevalence of obesity,many children are at risk of developing nonalcoholic fatty liver *** anthropometric and laboratory parameters,our study aimed to develop a model to quantitatively evaluate liver fat content(LFC)in children with *** A well-characterized cohort of 181 children between 5 and 16 years of age were recruited to the study in the Endocrinology Department as the derivation *** external validation cohort comprised 77 *** assessment of liver fat content was performed using proton magnetic resonance *** and laboratory metrics were measured in all subjects.B-ultrasound examination was carried out in the external validation *** Kruskal-Wallis test,Spearman bivariate correlation analyses,univariable linear regressions and multivariable linear regression were used to build the optimal predictive *** The model was based on indicators including alanine aminotransferase,homeostasis model assessment of insulin resistance,triglycerides,waist circumference and Tanner *** adjusted R^(2)of the model was 0.589,which presented high sensitivity and specificity both in internal[sensitivity of 0.824,specificity of 0.900,area under curve(AUC)of 0.900 with a 95%confidence interval:0.783–1.000]and external validation(sensitivity of 0.918 and specificity of 0.821,AUC of 0.901 with a 95%confidence interval:0.818–0.984).Conclusions Our model based on five clinical indicators was simple,non-invasive,and inexpensive;it had high sensitivity and specificity in predicting LFC in ***,it may be useful for identifying children with obesity who are at risk for developing nonalcoholic fatty liver disease.

Objective:To explore the balance of peripheral blood T helper 17 cells/regulatory T cell(Th17/Treg)ratio and the polarization ratio of M1 and M2 macrophages in lower extremity arteriosclerosis obliterans(ASO).Methods:A rat model of lower extremity ASO was established,and blood samples from patients with lower extremity ASO before and after surgery were *** was used to detect interleukin 6(IL-6),IL-10,and ***-time RCR and Western blot analyses were used to detect Foxp3,IL-6,IL-10,and IL-17 ***,flow cytometry was applied to detect the Th17/Treg ratio and M1/M2 ***:Compared with the control group,the iliac artery wall of ASO rats showed significant hyperplasia,and the concentrations of cholesterol and triglyceride were significantly increased(P<0.01),indicating the successful establishment of ***,the levels of IL-6 and IL-17 in ASO rats were pronouncedly increased(P<0.05),while the IL-10 level was significantly decreased(P<0.05).In addition to increased IL-6 and IL-17 levels,the mRNA and protein levels of Foxp3 and IL-10 in ASO rats were significantly decreased compared with the control *** Th17/Treg and M1/M2 ratios in the ASO group were markedly increased(P<0.05).These alternations were also observed in ASO *** endovascular surgery(such as percutaneous transluminal angioplasty and arterial stenting),all these changes were significantly improved(P<0.05).Conclusions:The Th17/Treg and M1/M2 ratios were significantly increased in ASO,and surgery can effectively improve the balance of Th17/Treg,and reduce the ratio of M1/M2,and the expression of inflammatory factors.
