Antitumor and anti-angiogenic activity of Ganoderma lucidum polysaccharides peptide

AIM: To investigate the antitumor and anti-angiogenic activity of Ganoderma lucidum polysaccharides peptide (GLPP). METHODS: Antitumor effect of GLPP was observed in tumor-bearing mice in vivo. At the same time, the effects of GLPP on proliferation of tumor cells and human umbilical cord vascular endothelial cell (HUVEC) were detected by MTTassay in vitro. Subsequently, spleen lymphocytes proliferation of nude mice was stimulated by LPS or ConA. To investigate the anti-angiogenic effect of GLPP, GLPP 80 μg per disc and GLPP-treated serum 10 μL per disc were added to the chick chorioallantoic membrane (CAM) respectively in vivo. RESULTS: GLPP 50, 100, and 200 mg/kg inhibited growth of Sarcoma 180 in BALB/c mice markedly by 35.2 %, 45.2 %, and 61.9 %, respectively. GLPP which was directly added to the cultured medium did not inhibit PG cell proliferation in vitro; but GLPP-treated serum 50, 100, 200 mg/kg potently inhibited PG cell proliferation by 22.5 %, 26.8 %, and 30.3 %, respectively; and reduced the xenograft (human lung carcinoma cell PG) in BALB/c nude mice greatly in vivo by 55.5 %, 46.0 %, and 46.8 %, respectively. Lymphocytes proliferation of nude mice could be stimulated by LPS 5 mg/Lbut not by ConA 2.5 mg/L, indicating that GLPP could not promote the T lymphocyte proliferation and neutral red phagocytosis of peritoneal macrophages of nude mice. The CAM assay showed that GLPP and GLPP-treated serum had anti-angiogenic effect. GLPP (1, 10, and 100 mg/L) inhibited HUVEC proliferation in vitro with the inhibitory rate of 9.4 %, 15.6 %, and 40.4 %, respectively. CONCLUSION: GLPP has antitumor and anti- angiogenic activity. The anti-angiogenesis of GLPP may be a new mechanism underlying its anti-tumor effects.

Ganoderma lucidum, has been widely used as a medicine to promote health and longevity in china for thousands of years. A series of experiments have demonstrated that the antitumor activity of Ganoderma lucidum polysaccharides (Gl-PS) may be mediated to some extent by activation of host immune functions. In this study, we investigate the anti-angiogenic activity of Ganoderma lucidum polysaccharide (Gl-PS) on tumor growth, which maybe a new pathway of antitumor effect by *** present study, we investigated the antitumor activity of Gl-PS. BALB/c mice which implanted with Sarcoma 180 were administrated intragastrically with Gl-PS 50,100,200mg·kg-1 respectively for 10 days. At the end of experiment, the tumor were cut and weighted. The effect of Gl-PS and Gl-PS-treated serum on proliferation of human lung carcinoma cell line PG were detected by MTT assay;BALB/c nude mice were implanted with xenograft (human lung carcinoma cell line PG) . Gl-PS were given at the dose of 50,100,200mg·kg-1 intragastrically for 32 days, then the tumor were cut and weighted. At the same time, spleen lymphocytes of the nude mice were also prepared to observe the effect of Gl-PS on lymphocytes proliferation induced by 2.5μg·ml-1 ConA or 5μg·ml-1 *** influence of Gl-PS on neutral red phagocytosis of nude mice peritoneal maerophages were also investigated in vitro. Gl-PS 40,80μg·ml-1 and Gl-PS-treated serum 10μl were added to the CAM (chick embryo chorioallantoic membrane) respectively to investigate the anti-angiogenesis effect of *** result showed that Gl-PS inhibited the growth of Sarcoma 180 significantly which had been implanted in BALB/c mice in vivo. Gl-PS inhibited the growth of Sarcoma 180 in a dose-dependent manner. The inhibitory rates were 35.20% ,45.25% , 61.88%, respectively. Gl-PS directly adding to the cultured medium did not inhibit PG cell proliferation in vitro, but 50, 100, 200mg ·kg- 1 Gl-PS-treated serum significantly inhibited PG cell proliferation, the inhib

Neutron resonance imaging(NRI)has recently emerged as an appealing technique for neutron *** complexity surpasses that of conventional transmission imaging,as it requires a high demand for both a neutron source and ***,the progression of NRI technology has been sluggish since its inception in the 1980s,particularly considering the limited studies analyzing the neutron energy range above *** white neutron source(Back-n)at the china Spallation Neutron Source(CSNS)provides favorable beam conditions for the development of the NRI technique over a wide neutron energy range from eV to ***-sensitive microchannel plates(MCP)have emerged as a cutting-edge tool in the field of neutron detection owing to their high temporal and spatial resolutions,high detection efficiency,and low *** this study,we report the development of a 10B-doped MCP detector,along with its associated electronics,data processing system,and NRI experiments at the *** heavy elements such as gold,silver,tungsten,and indium can be easily identified in the transmission images by their characteristic resonance peaks in the 1–100 eV energy range;the more difficult medium-weight elements such as iron,copper,and aluminum with resonance peaks in the 1–100 keV energy range can also be *** particular,results in the neutron energy range of dozens of keV(Aluminum)are reported here for the first time.

中子辐射俘获反应在反应堆运行、核装置设计及核天体物理研究中起重要的作用.4πBaF_(2)探测装置有着高时间分辨能力、低中子灵敏度、高探测效率等优点,适合开展中子辐射俘获反应截面数据的测量.中国原子能科学研究院核数据重点实验室建立了伽马全吸收装置(Gamma total absorption facility,GTAF),该装置用28块六棱BaF_(2)晶体和12块五棱BaF_(2)晶体构成了外径25 cm,内径10 cm的球壳,覆盖了95.2%的立体角.利用GTAF在中国散裂中子源Back-n束线上,测量了197Au(n,γ)的反应截面数据.测量数据通过能量筛选、PSD方法、晶体多重性筛选进行了初步本底扣除,随后结合对^(nat)C及空样品的测量数据对本底进行了分析及扣除,获得了197Au俘获反应的产额,利用SAMMY程序拟合得到了^(197)Au在1—100 e V的共振能量、中子共振宽度和伽马共振宽度参数.实验测量结果与ENDF/B-VIII.0数据库符合良好,其共振参数存在一定差异,分析原因可能与GTAF能量分辨率、Back-n的中子能谱测量精度、以及实验本底扣除方法相关,这也是下一步工作的重点.

●AIM:To describe the complex,overlapping phenotype of four Chinese patients with inherited retinal dystrophies(IRDs)who harbored two pathogenic genes simultaneously.●METHODS:This retrospective study included 4 patients affected with *** and ophthalmic histories were obtained,and clinical examinations were performed.A specific Hereditary Eye Disease Enrichment Panel(HEDEP)based on exome capture technology was used for genetic screening.●RESULTS:Four patients were identified to harbor disease-causing variants in two different *** retinitis pigmentosa(RP)01-II:1 exhibited both classical ABCA4-induced Stargardt disease(STGD)1 and USH2 Aassociated RP,patient RP02-III:2 exhibited both classical ABCA4-induced STGD1 and CDH23-associated RP,patient RP03-II:1 exhibited both USH2 A-induced autosomal recessive retinitis pigmentosa(arRP)syndrome and SNRNP200-induced autosomal dominant retinitis pigmentosa(adRP),and patient RP04-II:2 exhibited USH2 Ainduced arRP syndrome and EYS-induced arRP at the same time.●CONCLUSION:Our study demonstrates that genotype–phenotype correlations and comprehensive genetic screening is crucial for diagnosing IRDs and helping family planning for patients suffering from the disease.
