Hypomethylation of Thyroid Peroxidase as a Biomarker for Hepatocellular Carcinoma with Tumor Thrombosis

Objective Thyroid hormones(THs)regulate multiple physiological activities in the liver,including cellular metabolism,differentiation,and cell growth,and play important roles in the pathogenesis of hepatocellular carcinoma(HCC).Thyroid peroxidase(TPO)is a key molecule involved in the THs synthesis and signaling pathway.As an epigenetic modification,DNA methylation has a critical role in tumorigenesis with diagnostic potential.However,the connection between THs and DNA methylation has been rarely investigated.Methods The methylation of key TH-related genes was analyzed by in-house epigenome-wide scanning,and we further analyzed the methylation levels of the TPO promotor in 164 sample pairs of HCC and adjacent non-cancerous tissues by Sequenom EpiTYPER assays,and evaluated their clinical implications.Results We identified that the methylation of the TPO promoter was downregulated in the HCC tissues(P<0.0001)with a mean difference ranging from 18.5%to 22.3%.This methylation pattern correlated with several clinical factors,including a multi-satellite tumor,fibrous capsule,and the presence of tumor thrombus.The receiver operator characteristic(ROC)curve analysis further confirmed that the percent methylated reference(PMR)values for TPO were predictive of the tumor[the area under the curve(AUC)ranged from 0.755 to 0.818]and the thrombosis in the HCC patients(the AUC ranged from 0.706 to 0.777).Conclusion These findings demonstrated that epigenetic alterations of TPO,as indicated by the PMR values,were a potential biomarker for HCC patients with tumor thrombosis.

Construction of a Three-dimensional Covalent Organic Framework via the Linker Exchange Strategy

Covalent organic framework(COF)is a porous crystalline material with a well-controlled structure and a wide range of potential applications.However,the construction of new COF faces huge challenges,including the design and synthesis of structural unit monomers,the choice of reaction solvent system,and the study of reaction time and temperature.So,it’s particularly important to widen the application scope of synthetic methods and further promote the development of COFs.Here,we performed structural transformations in a three-dimensional(3D)COF(COF-300),and Fourier transform infrared spectroscopy(FTIR),power X-ray diffraction analysis(PXRD)and nitrogen adsorption isotherms confirmed the chemical principles and the successful realization of these exchanges.At the same time,we found that the interpenetrating structure in 3D COF can be changed through the conversion of linkers.The structure simulation successfully proved the transformation of COF from five-fold to seven-fold interpenetration.In addition,in order to prove the versatility of this strategy,we used the same method to convert COF-300 into a high crystallinity 3D COF(TJNU-COF-302)that is also seven-fold interpenetrating and has not been reported.This simple strategy not only makes it easy to obtain a 3D COF connected with imines,which greatly promotes the development of COF,but also provides a new way to develop 3D COFs with complex interpenetrating structures.

To the Editor:Hand,foot,and mouth disease(HFMD)is well recognized as a pediatric infectious disease caused by a group of enteroviruses(EVs)with global distribution.Currently,the HFMD is still one among the major threats to the health of children in China,with estimated 20,537,199 cases,resulting in 3667 deaths in 2018.[1]Available data indicated that most of HFMD cases were caused by serotypes of enterovirus 71(EV-71)and coxsackievirus A16(CV-A16).EV-71 was found to be predominant among severe and fatal cases.Initiated from 2015,three inactivated monovalent EV-71 vaccines were licensed in China and proved with high protective efficacy against HFMD caused by EV-71 but no cross-protection for non-EV-71 serotypes,[2]and the EVs exhibit highly geographical diversity and serotype diversity.Recently,concerns have raised for the outbreaks of CV-A6 and CV-A10 with severe cases,as well as emerging of rare serotypes such as echo virus 11 and echo virus 30.The inadequate coverage of vaccine protection,the geographical diversity,and changing spectrum of HFMD etiology still challenge the comprehensive HFMD prevention and control in China.Information regarding the broad feature of HFMD epidemiology,and the shifting of viral genetic diversity and evolution dynamic as EV-71 vaccination progressing,are still limited.Thus,we conducted a comprehensive retrospective analysis on the epidemiological and etiological characteristics of HFMD in Sichuan Province from 2014 to 2019 before and after the introduction of EV-71 vaccines,which was expected to help better HFMD prevention and control in this area.

Bioactivity-based UPLC/Q-TOF/MS strategy for screening of anti-inflammatory components from Cimicifugae Rhizoma

In this study,the protective effects of Cimicifugae Rhizoma（CR） was demonstrated in Pseudomonas aeruginosa-induced pneumonia mouse model.To identify the anti-inflammatory ingredients,an ultraperformance liquid chromatography quadrupole time-of-flight mass spectrometry（UPLC/Q-TOF-MS）integrated nuclear factor κB（NF-κB） luciferase reporter assay screening system was carried out.As a result,some caffeic acid derivatives,including caffeic acid,ferulic acid/isoferulic acid,fukinolic acid,and cimicifugic acid ingredients were identified as the potential effective compounds from CR.For testing the anti-inflammatory capacity,caffeic acid was demonstrated to inhibit NF-kB and reduce the levels of IL-6 and IL-8 in TNF-α-treated BEAS-2B cells in a dose-dependent manner.Hence,CR preparations and its cinnamic acid derivatives have the possibility to be developed as a complementary therapy in the treatment of respiratory system infection in clinics.

Does labor off-farm employment inevitably lead to land rent out?Evidence from China

The focus of land economics is on how to decrease the misallocation of resources to achieve an optimal allocation of resources. Both the theories of new economics of labor migration(NELM) and the conclusions of empirical studies reveal that land resources will inevitably be reallocated(e.g., rented out) if the resources of family labor are reallocated(e.g., off-farm employment). However, this study reveals that off-farm employment does not inevitably lead to land rent out. More precisely, this study uses survey data on 8031 peasant households from 27 provinces in China and explores the relationship between off-farm employment and land rent out by describing spatial features and through empirical analysis(e.g., IV-Probit model and IV-Tobit model). The results show the following: 1) There is an indirect relationship between off-farm employment and land rent out regarding spatial area aggregation, i.e., regions with a higher ratio of off-farm employment also have a lower area of land rent out. 2) Off-farm employment is significantly positively correlated with the behavior of land rent out, but its square is significantly negatively correlated with the behavior, i.e., there is an inverted U-shaped relationship between off-farm employment and the behavior of land rent out, with the turning point being 55.55% offfarm employment. 3) Off-farm employment is significantly positively correlated with the area of land rent out, but its square is significantly negatively correlated with the area, i.e., there is an inverted Ushaped relationship between off-farm employment and the area of land rent out, with the turning point being 56.22% off-farm employment. This study helps explain why China has a high ratio of off-farm employment but a lower rate of land rent out.

Interventional Radionuclide Therapy of Hepatocellular Carcinoma： Assessment of Intratumoral Retention of HPMA Copolymers

To develop new radiopharmaceuticals for the interventional radionuclide therapy of recurrent hepatocellular carcinoma, poly（HPMA）-APMA-DTPA[HPMA=N-（2-hydroxypropyl） methacrylamide; APMA=N-（3-aminopropyl）methacrylamide; DTPA=diethylenetriaminepentaacetic acidl was synthesized by free radical precipitation polymerization in acetone/dimethylsulfoxide with N,N＇-azobis（isobutyronitrile） as the initiator. The copolymers were characterized with nuclear magnetic resonance（NMR） spectroscopy and gel permeation chromatography（GPC, Mn=2.2xl04, Mw/Mn=l.38）. Subsequently, poly（HPMA）-APMA-DTPA was conjugated with 99mTC radionuclide. Prolonged retention of poly（HPMA）-APMA-DTPA conjugate within the tumor tissues was demonstrated by single-photon emission computed tomography computed tomography（SPECT-CT） at 1, 2, 4 and 24 h following intra-tumoral injection of the conjugate to hepatocellular carcinoma xenografts in mice. DTPA-99mTc was also synthesized and characterized for comparison. The data suggest that the poly（HPMA）-APMA-DTPA conjugates might be useful for the interventional radionuclide therapy of recurrent hepatocellular carcinoma in humans.

Fresh-seed germination(FSG)impairs peanut production,especially in areas where the peanut harvest season coincides with rainy weather.Developing FSG-resistant cultivars by molecular breeding is expected to mitigate yield loss and quality impairment caused by FSG.However,the genetic control of FSG awaits elucidation.In this study,FSG at 1,3,5,7,and 9 days post-imbibition in three environments were tested,and quantitative-trait loci(QTL)associated with FSG were mapped in a peanut recombinant inbred line population by leveraging existing high-density peanut genetic maps.Of 24 QTL identified in 13 linkage groups,qFSGA04 was a stable major QTL on linkage group 04(LG04).It was consistently detected in five germination stages and three environments.By designing and validating DNA markers in the confidence interval of qFSGA04,we identified one single-nucleotide polymorphism and one In Del closely associated with FSG that could be used as linked markers for FSG resistance in peanut breeding.

电池故障诊断系统是电动汽车能够安全运行的重要保证,对其进行建模具有重要意义。采用自适应神经模糊网络(ANFIS),对磷酸铁锂电池组故障诊断系统进行建模。根据原有的专家经验确定电池的主要故障症状和原因;利用减法聚类算法确定模型的初始模糊规则和隶属度数量,减少网络中参数数量;利用由最小二乘估计算法和误差反向传播算法构成的混合学习算法对模糊神经网络的参数进行学习,完成诊断系统参数的训练和构建,该模型分类准确率可达92.7%。最后将ANFIS模型和传统BP神经网络模型的测试结果进行对比,结果表明:在针对48 V动力电池组的故障诊断中,ANFIS模型比传统BP神经网络模型分类准确率高出6.9%左右。

BACKGROUND Autoimmune pancreatitis(AIP)has been linked with elevated immunoglobulin(Ig)G4 levels.The characteristics and outcomes of AIP based on serum markers have not been fully evaluated.AIM To compare clinical features,treatment efficacy,and outcome of AIP based on serum IgG4 levels and analyze predictors of relapse.METHODS A total of 213 patients with AIP were consecutively reviewed in our hospital from 2006 to 2021.According to the serum IgG4 level,all patients were divided into two groups,the abnormal group(n=148)with a high level of IgG4[>2×upper limit of normal(ULN)]and the normal group(n=65).The t-test or Mann-Whitney U test was used to compare continuous variables.Categorical parameters were compared by theχ^(2) test or Fisher’s exact test.Kaplan-Meier curves Zhou GZ et al.Clinical characteristics and outcome of AIP WJG https://*** 5126 September 21,2023 Volume 29 Issue 35 and log-rank tests were established to assess the cumulative relapse rates.Univariate and multivariate analyses were used to investigate potential risk factors of AIP relapse.RESULTS Compared with the normal group,the abnormal group had a higher average male age(60.3±10.4 vs 56.5±12.9 years,P=0.047);higher level of serum total protein(72.5±7.9 g/L vs 67.2±7.5 g/L,P2×ULN),hazard ratio(HR):3.583;95%confidence interval(CI):1.218–10.545;P=0.020]and IgA(>1×ULN;HR:5.908;95%CI:1.199–29.120;P=0.029)and age>55 years(HR:2.383;95%CI:1.056–5.378;P=0.036)were independent risk factors of relapse.CONCLUSION AIP patients with high IgG4 levels have clinical features including a

Acupuncture is widely used to treat functional dyspepsia with satisfactory outcomes. Combination of the He and Mu acupoints is commonly used and has a synergistic effect on functional dyspepsia; however, its underlying mechanisms remain unclear. Therefore, a randomized controlled parallel clinical trial is currently underway at Chengdu University of Traditional Chinese Medicine, China. This trial is designed to explore the efficacy of and central responses to the He-Mu point combination in patients with functional dyspepsia using functional magnetic resonance imaging. A total of 105 patients with functional dyspepsia will be allocated into 3 groups： the low-He point group（puncturing at Zusanli（ST36））, Mu point group（puncturing at Zhongwan（CV12））, and He-Mu point combination group（puncturing at ST36 and CV12）. Every participant will receive 20 sessions of manual acupuncture for 4 weeks. The needles will be inserted perpendicularly to a depth of 1 to 2 cun. The angle of rotation and twisting will range from 90 to 180 degrees, while lifting and thrusting will range from 0.3 to 0.5 cm. The various manipulations will be performed 60 to 90 times per minute. The needles will remain in place for 30 minutes, during which manipulation will be applied every 10 minutes. Magnetic resonance imaging will be performed before and after 20 sessions of acupuncture. The primary outcome is symptom improvement according to the Chinese version of the Nepean Dyspepsia Index. Secondary outcomes include the Leeds dyspepsia questionnaire, Self-Rating Anxiety Scale, Self-Rating Depression Scale, Beck Anxiety Inventory, Beck Depression Inventory, and visual analogue scale scores before and after 10 and 20 sessions of acupuncture. Needle sensation and adverse events will be used to assess the therapeutic effects. This study will promote more widespread awareness of the benefits of acupoint combination in the clinical setting and provide a further explanation of the neuromechanism by which acupuncture at the He-Mu point combination for functio
