Decreased PSD95 expression in medial prefrontal cortex(mPFC) was associated with cognitive impairment induced by sevoflurane anesthesia

Objective： Though sevoflurane has been widely used as an anesthetic in surgery, recent studies have shown that exposure to sevoflurane alone could lead to postoperative cognitive dysfunction （POCD）, of which the mechanisms still remain largely unknown. The medial prefrontal cortex （mPFC） is known to be implicated in various cognitive impairments, including working memory and attentional processes. In the present study, we tried to identify dysregulated gene expression in mPFC and investigate the underlying mechanisms involved in POCD. Methods： Behavioral tests, including elevated plus-maze, O-maze, and Y-maze tests, were performed on Wistar rats exposed to sevoflurane. Whole-genome mRNA profiling of mPFC from Wistar rats after exposure to sevoflurane was carried out. Real-time polymerase chain reaction （PCR） was done to verify the differentially expressed genes. Results： Significant impairment of working memory of rats after exposure to sevoflurane was observed. A total of 119 of 7319 detected mRNAs showed significantly different expression between rats with and without sevoflurane exposure （fold change （FC）〉2.0, P〈0.05, and false discovery rate （FDR）〈0.05）, among which 74 mRNAs were down-regulated and 45 mRNAs were up-regulated. Postsynaptic density-95 （PSD95, also named DLG4） showed the most significantly de- creased expression in mPFC and further investigation indicated that PSD95 expression level was correlated with spatial working memory performance. Conclusions： Our study revealed that PSD95 might be involved in the mecha- nism of POCD, which could provide clues for preventing POCD in clinical operations.

Protective effects of parecoxib on rat primary astrocytes from oxidative stress induced by hydrogen peroxide

Objective： To investigate the protective effects of parecoxib from oxidative stress induced by hydrogen peroxide （H202） in rat astrocytes in vitro. Methods： All experiments included 4 groups：（1） negative control （NC） group, without any treatment; （2） H202 treatment group, 100 μmol/L H202 treatment for 24 h; （3） and （4） parecoxib pre- treatment groups, 80 and 160 μmol/L parecoxib treatment for 24 h, respectively, and then treated with 100 μmol/L H202. Several indices were investigated, and the expressions of Bax, Bcl-2, and brain-derived neurotrophic factor （BDNF） were quantified. Results： Compared to the NC group, exposure to H202 resulted in significant morphological changes, which could be reversed by pretreatment of parecoxib. In addition, H202 treatment led to loss of viability （P=0.026） and increased intracellular reactive oxygen species （ROS） levels （P〈0.001）, and induced apoptosis （P〈0.01） in the primary astrocytes relative to the NC group. However, in the parecoxib pretreatment groups, all the above changes reversed significantly （P〈0.05） as compared to the H202 treatment group, and were nearly unchanged when compared to the NC group. Mechanical investigation showed that dysregulated Bax, Bcl-2, and BDNF could be im- plicated in these changes. Conclusions： Our results indicated that parecoxib provided a protective effect from oxidative stress induced by exposure to H202.

Fractal model of spontaneous imbibition in low-permeability reservoirs coupled with heterogeneity of pore seepage channels and threshold pressure

SiGe-based thermoelectric(TE)materials have gained increasing interests due to their low maintenance costs,environmental friendliness and long ***,the intrinsically high thermal conductivity of Si-based materials also results in poor TE *** this investigation,a zirconia(ZrO_(2))composite strategy was applied to an n-type SiGe alloy,tremendously elevating its TE *** mechanical alloying and spark plasma sintering(SPS)processes,the ZrO_(2)induced the formation of nanopores in the SiGe matrix via phosphorus ***,such increase in porosity enhanced the phonon scattering and dramatically suppressed lattice thermal conductivity,from 2.83 to 1.59 W·m^(-1)·K^(-1)at 873 ***,reduced phosphorus doping led to an increase in Seebeck coefficients and a relatively minor decrease in electrical conductivity,The power factor didn't deteriorate significantly,either,as its maximum of~3.43 mW·m^(-1-)K^(-2)was achieved at 873 K with(Si_(0.8)Ge_(0.2))_(0.097)P_(0.03)(ZrO_(2))_(0.003).In short,a peak figure of merit(ZT)of~1.27 at 873 K and an average ZT~0.7 from 323 to 873 K were *** study demonstrates that the electrical and thermal transportation of SiGe material can be synergistically tuned by compositing ZrO_(2),illustrating a novel strategy to optimize the TE properties of bulk materials.

Monotropein Induced Apoptosis and Suppressed Cell Cycle Progression in Colorectal Cancer Cells

Objective: To determine whether monotropein has an anticancer effect and explore its potential mechanisms against colorectal cancer(CRC) through network pharmacology and molecular docking combined with experimental verification. Methods: Network pharmacology and molecular docking were used to predict potential targets of monotropein against CRC. Cell counting kit assay, plate monoclonal assay and microscopic observation were used to investigate the antiproliferative effects of monotropein on CRC cells HCT116, HT29 and LoVo. Flow cytometry and scratch assay were used to analyze apoptosis and cell cycle, as well as cell migration, respectively in HCT116, HT29, and LoVo cells. Western blotting was used to detect the expression of proteins related to apoptosis, cell cycle, and cell migration, and the expression of proteins key to the Akt pathway. Results: The Gene Ontology and Reactome enrichment analyses indicated that the anticancer potential of monotropein against CRC might be involved in multiple cancer-related signaling pathways. Among these pathways, RAC-beta serine/threonine-protein kinase(Akt1, Akt2), cyclin-dependent kinase 6(CDK6), matrix metalloproteinase-9(MMP9), epidermal growth factor receptor(EGFR), cell division control protein 42 homolog(CDC42) were shown as the potential anticancer targets of monotropein against CRC. Molecular docking suggested that monotropein may interact with the 6 targets(Akt1, Akt2, CDK6, MMP9, EGFR, CDC42). Subsequently, cell activity of HCT116, HT29 and LoVo cell lines were significantly suppressed by monotropein(P<0.05). Furthermore, our research revealed that monotropein induced cell apoptosis by inhibiting Bcl-2 and increasing Bax, induced G_1–S cycle arrest in colorectal cancer by decreasing the expressions of CyclinD1, CDK4 and CDK6, inhibited cell migration by suppressing the expressions of CDC42 and MMP9(P<0.05), and might play an anticancer role through Akt signaling pathway. P-type Mg_(3)Sb_(2)-based Zintls have attracted considerable interest in the thermoelectric(TE)field due to their environmental friendliness and low ***,compared to their n-type counterparts,they show relatively low TE performance,limiting their application in TE *** this work,we simultaneously introduce Bi alloying at Sb sites and Ag doping at Mg sites into the Mg_(3)Sb_(2)to coopera-tively optimize the electrical and thermal properties for the first time,acquiring the highest ZT value of∼0.85 at 723 K and a high average ZT of 0.39 in the temperature range of 323-723 K in sample Mg_(2.94)Ag_(0.06)Sb_(1.9)Bi_(0.1).The first-principle calculations show that the codoping of Ag and Bi can shift the Fermi level into the valence band and narrow the band gap,resulting in the increased carrier concentration from 3.50×10^(17)cm^(-3)in the reference Mg 3 Sb 0.9 Bi 0.1 to∼7.88×10^(19)cm^(-3)in sample Mg 2.94 Ag 0.06 Sb 0.9 Bi *** a result,a remarkable power factor of∼778.9μW m^(-1)K^(-2)at 723 K is achieved in sample Mg 2.94 Ag 0.06 Sb 0.9 Bi ***,a low lattice thermal conductivity of∼0.48 W m^(-1)K^(-1)at 723 K is also obtained with the help of phonon scattering at the distorted lattice,point defects,and nano-precipitates in sample Mg 2.94 Ag 0.06 Sb 0.9 Bi *** synergistic effect of using the multi-element co-doping/-alloying to optimize electrical properties in Mg_(3)Sb_(2)holds promise for further improving the TE performance of Zintl phase materials or even others.

Full waveform inversion based on hybrid gradient

The low-wavenumber components in the gradient of full waveform inversion(FWI)play a vital role in the stability of the ***,when FWI is implemented in some high frequencies and current models are not far away from the real velocity model,an excessive number of low-wavenumber components in the gradient will also reduce the convergence rate and inversion *** solve this problem,this paper firstly derives a formula of scattering angle weighted gradient in FWI,then proposes a hybrid *** hybrid gradient combines the conventional gradient of FWI with the scattering angle weighted gradient in each inversion frequency band based on an empirical formula derived *** weighted hybrid mode,we can retain some low-wavenumber components in the initial lowfrequency inversion to ensure the stability of the inversion,and use more high-wavenumber components in the high-frequency inversion to improve the convergence *** results of synthetic data experiment demonstrate that compared to the conventional FWI,the FWI based on the proposed hybrid gradient can effectively reduce the low-wavenumber components in the gradient under the premise of ensuring inversion *** also greatly enhances the convergence rate and inversion accuracy,especially in the deep part of the *** the field marine seismic data experiment also illustrates that the FWI based on hybrid gradient(HGFWI)has good stability and adaptability.
