Hypobaric hypoxia (HH) exposure can cause serious brain injury as well as life-threatening cerebral edema in severe cases. Previous studies on the mechanisms of HH-induced brain injury have been conducted primarily using non-primate animal models that are genetically distant to humans, thus hindering the development of disease treatment. Here, we report that cynomolgus monkeys (Macaca fascicularis) exposed to acute HH developed human-like HH syndrome involving severe brain injury and abnormal behavior. Transcriptome profiling of white blood cells and brain tissue from monkeys exposed to increasing altitude revealed the central role of the HIF-1 and other novel signaling pathways, such as the vitamin D receptor (VDR) signaling pathway, in co-regulating HH-induced inflammation processes. We also observed profound transcriptomic alterations in brains after exposure to acute HH, including the activation of angiogenesis and impairment of aerobic respiration and protein folding processes, which likely underlie the pathological effects of HH-induced brain injury. Administration of progesterone (PROG) and steroid neuroprotectant 5α-androst-3β,5,6β-triol (TRIOL) significantly attenuated brain injuries and rescued the transcriptomic changes induced by acute HH. Functional investigation of the affected genes suggested that these two neuroprotectants protect the brain by targeting different pathways, with PROG enhancing erythropoiesis and TRIOL suppressing glutamate-induced excitotoxicity. Thus, this study advances our understanding of the pathology induced by acute HH and provides potential compounds for the development of neuroprotectant drugs for therapeutic treatment.

AIM:To evaluate the short-and long-term outcomes of liver resection for caudate lobe hepatocellular carcinoma (HCC).METHODS:We retrospectively analyzed 114 consecutive patients with HCC,originating from the caudate lobe,who underwent resection between January 2001 and January *** and multivariate analyses were performed on several clinicopathologic variables to determine the factors affecting long-term outcome and intrahepatic ***:Overall mortality and morbidity were 0% and 18%,*** a median follow-up of 31 mo (interquartile range,11-66 mo),tumor recurrence had occurred in 76 patients (66.7%).The 1-,3-,and 5-year disease-free survival rates were 65.7%,38.1%,and 18.4%,*** 1-,3-,and 5-year overall survival rates were 76.1%,54.7%,and 31.8%,*** analysis showed that subsegmental location of the tumor (45.7% vs 16.2%,P=0.01),liver cirrhosis (12.3% vs 47.9%,P=0.03),surgical margin (18.5% vs 54.6%,P=0.04),vascular invasion (37.9% vs 23.2%,P=0.04) and extended caudate resection (42.1% vs 15.4%,P=0.04) were related to poorer long-term *** analysis showed that only subsegmental location of the tumor,liver cirrhosis and surgical margin were significant independent prognostic ***:Hepatectomy was an effective treatment for HCC in the caudate *** subsegmental location of the tumor,liver cirrhosis and surgical margin affected long-term survival.

BACKGROUND: Postoperative regional chemotherapy is one of the most effective methods to decrease the recurrent rate and improve the prognosis of primary hepatocarcinoma (PHC). This study was undertaken to assess the optimal pathway to implant the drug delivery system (DDS) in the different ways of resecting PHC so as to offer a valuable reference to clinical implantation of the DDS. METHODS: One hundred and ninety cases were divided into two groups according to whether the tumors were resected completely (A) or not (B). Groups A and B were subdivided into three groups a, b and c according to the pathway selected for DDS implantation. The patients in subgroup a received DDS implantation through both the hepatic artery and portal vein (A+P-implanted group), the patients in subgroup b received DDS implantation through the portal vein (P-implanted group), and the patients in subgroup c received DDS implantation through the hepatic artery (A-implanted group). RESULTS: The 1- and 3-year recurrent rates of subgroup c in group A were higher than those of subgroup b, and there was no significant difference between subgroups a and b. Compared with subgroups a and c, the 1- and 3-year survival rates of subgroup b were similar to those of group a but higher than those of group c. The 1- and 3-year survival rates between subgroups a and b in group B were significantly different. The prognosis of subgroup c was lower than that of subgroup a and no significant difference was observed between subgroups b and c. CONCLUSIONS: The DDS should be implanted into the portal vein when PHC is resected completely. It may be better to implant it into both portal vein and hepatic artery if the tumor cannot be completely resected.

Microglia,the primary immune cells within the brain,have gained recognition as a promising therapeutic target for managing neurodegenerative diseases within the central nervous system,including Parkinson’s *** perfluorocarbon droplets have been reported to not only possess a high oxygen-carrying capacity,but also exhibit remarkable anti-inflammatory ***,the role of perfluoropentane in microglia-mediated central inflammatory reactions remains poorly *** this study,we developed perfluoropentane-based oxygen-loaded nanodroplets(PFP-OLNDs)and found that pretreatment with these droplets suppressed the lipopolysaccharide-induced activation of M1-type microglia in vitro and in vivo,and suppressed microglial activation in a mouse model of Parkinson’s *** suppression led to a reduction in the inflammatory response,oxidative stress,and cell migration capacity in ***,the neurotoxic effects were mitigated,which alleviated neuronal ***,ultrahigh-performance liquid chromatography–tandem mass spectrometry showed that the anti-inflammatory effects of PFP-OLNDs mainly resulted from the modulation of microglial metabolic *** further showed that PFP-OLNDs regulated microglial metabolic reprogramming through the AKT-mTOR-HIF-1α***,our findings suggest that the novel PFP-OLNDs constructed in this study alleviate microglia-mediated central inflammatory reactions through metabolic reprogramming.

Peripheral nerve injury is a common neurological condition that often leads to severe functional limitations and *** on the pathogenesis of peripheral nerve injury has focused on pathological changes at individual injury sites,neglecting multilevel pathological analysis of the overall nervous system and target *** has led to restrictions on current therapeutic *** this paper,we first summarize the potential mechanisms of peripheral nerve injury from a holistic perspective,covering the central nervous system,peripheral nervous system,and target *** peripheral nerve injury,the cortical plasticity of the brain is altered due to damage to and regeneration of peripheral nerves;changes such as neuronal apoptosis and axonal demyelination occur in the spinal *** nerve will undergo axonal regeneration,activation of Schwann cells,inflammatory response,and vascular system regeneration at the injury *** damage to target organs can occur,including skeletal muscle atrophy and sensory receptor *** then provide a brief review of the research advances in therapeutic approaches to peripheral nerve *** main current treatments are conducted passively and include physical factor rehabilitation,pharmacological treatments,cell-based therapies,and physical ***,most treatments only partially address the problem and cannot complete the systematic recovery of the entire central nervous system-peripheral nervous system-target organ ***,we should further explore multilevel treatment options that produce effective,long-lasting results,perhaps requiring a combination of passive(traditional)and active(novel)treatment methods to stimulate rehabilitation at the central-peripheral-target organ levels to achieve better functional recovery.

通过批式吸附实验研究了溶液pH值、离子强度、温度、固液比等因素对^(125)I^-在高庙子膨润土上吸附行为的影响。结果表明,^(125)I^-在高庙子膨润土上的吸附受溶液pH值和离子强度影响显著,在中性和碱性条件下,^(125)I^-的吸附很弱,随着pH的降低吸附迅速增强并在pH=2.0出现最大值,之后随着pH继续降低,吸附迅速减小。在T=298K、pH=2.0、0.10mol/L NaClO_4背景电解质浓度下,^(125)I^-在高庙子膨润土上的最大吸附分配系数约为92mL/g。采用自动电位滴定技术测得高庙子膨润土的pH_(PZC)≈9.5,蒙脱石的pH_(PZC)≈10.5。温度对^(125)I^-在膨润土上的吸附无显著影响,Freundlich等温吸附模型更适合描述不同温度下^(125)I^-在高庙子膨润土上的吸附行为。

新型木霉氨基酸有机肥研制及其对番茄的促生效果

为提高贵州木霉NJAU4742固体发酵产孢能力,本研究拟用稻草秸秆和氨基酸水解液为原料进行固体发酵获得高分生孢子含量的固体菌种,将该固体菌种添加到含有不同比例氨基酸水解液的有机肥中进行二次发酵研制新型生物有机肥,并利用盆栽试验研究该生物有机肥对番茄的促生作用.结果表明:在利用稀释30倍的氨基酸水解液浸泡秸秆过夜后调节秸秆为初始p H 3.5、75%含水量、30%玉米粉添加量的条件下,贵州木霉NJAU4742所产分生孢子数量高达2.40×10^(10)CFU·g^(-1).接种2%固体木霉菌种于含有20%氨基酸水解液的腐熟有机肥后,该处理中木霉活菌数和吲哚乙酸(IAA)含量均达到最高,分别为6.40×10~9CFU·g^(-1)和38.66 mg·kg^(-1),与对照相比分别增加了1142.30和1.42倍.盆栽试验表明,木霉氨基酸生物有机肥(AT)与氨基酸有机肥(AA)处理跟对照(CK)相比株高分别增加了98.8%和23.8%,茎粗分别增加了58.9%和10.3%,叶绿素、叶长、叶宽等指标也显著增加.利用稻草秸秆和氨基酸水解液进行浅盘固体发酵生产木霉分生孢子,克服了工厂化生产固体木霉菌种的瓶颈,以此制备的新型木霉氨基酸有机肥(AT)与等养分的氨基酸有机肥(AA)和对照(CK)处理相比,对番茄的促生效果显著,在集约化农业生产中具有良好的应用前景.
