Bionic immunoactivator copresenting autophagy promoting and costimulatory molecules for synergistic cancer immunotherapy

Immunotherapy has great promise in improving malignant tumor ***,the efficacy of existing strategies is often limited by the immunosuppressive ***,w. demonstrate an in situ bionic immunoactivator,PLT-Bec1/DTA-1,w.th possessed natural advantages of platelets for tumor recruitment and activation,on w.ich DTA-1(CD357 monoclonal antibody)and Bec1 w.re tethered as combined immune ***-Bec1/DTA-1,as a self-triggered release repository,can deliver the pre-tethered Bec1 and DTA-1 deeply through the secretion of platelet microparticles(PMPs),thereby cooperate tacitly and exhibit superiority in immune activation of dendritic cells(DCs)and T cells via autophagy inducibility,coupled w.th glucocorticoid-induced tumor necrosis factor receptor(GITR)-triggered T_(Reg) suppression,remodeled the immunosuppressive netw.rk of tumor ***-Bec1/DTA-1 promoted antigen presentation and T cell proliferation,and alleviated the low.activity state of bone marrow.derived dendritic cells(BMDCs)in tumor suppressive ***-Bec1/DTA-1 inhibited tumor recurrence(5-and 13-fold low.r of control group in tumor volume)and CD8^(+)T/T_(Reg) ratio(6.3-and 8.8-fold *** group)in mouse tumor model after intravenous or subcutaneous ***,PLT-Bec1/DTA-1 prevented tumor colonization in lung through in situ immune activation,and w.s slightly superior to the combined of Bec1 and *** findings highlight the promise of delivering immunostimulatory payloads via bionic carriers,eliciting automatic in situ activation of effector immune cells in tumor microenvironment for tumor *** these results provide promising prospects into the application of immunoactivator in improving cancer synergistic immunotherapy to overcome the bottlenecks in clinic.

The identification of prototypical w.veforms, such as sleep spindles and epileptic spikes, is crucial for the diagnosis of neurological disorders. These prototypical w.veforms are usually recurrently presented in certain brain states, serving as potential biomarkers for clinical evaluations. Convolutional sparse coding(CSC) approaches have demonstrated strength in identifying recurrent patterns in time-series. How.ver,existing CSC approaches do not explicitly explore state-specific patterns, making it difficult to identify state-related biomarkers. To address this problem, w. propose state-sensitive CSC to learn state-specific prototypical w.veforms. Specifically, w. model signals of a certain state w.th specific w.veforms that only appear frequently in this state and background w.veforms that are independent of states. Based on this,state-sensitive CSC separates state-specific w.veforms from background ones explicitly by incorporating incoherence constraints into optimizations. Experiments w.th epilepsy brain signals demonstrate that our approach can effectively identify prototypical w.veforms in pre-ictal states, providing potential biomarkers for seizure prediction. Our approach provides a promising tool for automatic biomarker candidate identification.

Single-atom catalysts（SACs） hold great promise in addressing the sluggish kinetics of the sulfur reduction reaction（SRR） in lithium-sulfur（Li-S） batteries for their unique catalytic activity and maximum atom *** these SACs must be dispersed on solid substrates,the underlying support is usually limited to carbon materials that have a poor ability to modulate the coordination environment and electronic structures of single atoms,and consequently their catalytic activity tow.rd the SRR is *** w. propose tw.-dimensional（2D） graphene/electride heterostructu res as substrates to enhance the catalytic activities of SACs for Li-S batteries.2D electrides featuring the anionic electron gas on their surface enable efficient electron transfer to SACs,w.ich alters their electronic structures,resulting in the shifts of the d orbital and Fermi *** unique electronic structure decreases the filling of antibonding states such that the bonding w.th adsorbates at active sites is *** demonstrate the enhanced catalytic performance of SACs in terms of the Gibbs free energy of SRR and Li2S *** addition,a universal descriptor for the rapid screening of SACs is established by a linear regression fitting *** w.rk provides a new.design strategy to modulate SAC activity through electrides for Li-S batteries.

w.th the increasing presence of intermittent energy resources in microgrids,it is difficult to precisely predict the output of renew.ble resources and their load *** order to realize the economical operations of the system,an energy management method based on a model predictive control(MPC)and dynamic programming(DP)algorithm is *** method can reasonably distribute the energy of the battery,fuel cell,electrolyzer and external grid,and maximize the output of the distributed pow.r supply w.ile ensuring the pow.r balance and cost optimization of the *** on an ultra-shortterm forecast,the output pow.r of the photovoltaic array and the demand pow.r of the system load are *** offline global optimization of traditional dynamic programming is replaced by the repeated rolling optimization in a limited period of time to obtain pow.r values of each unit in the energy storage *** w.th the traditional DP,MILP-MPC and the logic based real-time management method,the proposed energy management method is proved to be feasible and effective.

To reveal the cavitation forms of tip leakage vortex(TLV)of the axial flow.pump and the flow.mechanism of the flow.field,this research adopts the partially-averaged Navier-Stokes(PANS)model to simulate the cavitation values of an axial flow.pump,follow.d by experimental *** experimental result show. that compared w.th the shear stress transport(SST)k-ωmodel,the PANS model significantly reduces the eddy viscosity of the flow.field to make the vortex structure clearer and allow.the turbulence scale to be more robustly *** cavitation area w.thin the axial flow.pump mainly comprises of TLV cavitation,clearance cavitation and tip leakage flow. combined effect of triangular cloud cavitation *** formation and development of cavitation are accompanied by the formation and evolution of vortex,and variations in vortex structure also generate and promote the development of *** addition,an in-depth analysis of the relationship betw.en the turbulent kinetic energy(TKE)transport equation and cavitation patterns w.s also conducted,finding that the regions w.th relatively high TKE are mainly distributed around gas/liquid boundaries w.th serious cavitation and evident gas-liquid *** phenomenon is mainly attributed to the combined effect of the pressure action term,stress diffusion term and TKE production term.
