Pheromone-binding proteins (PBPs) have been extensively investigated in lepidopteran moths, but their evolution and function in hemipteran species remain unclear. Our previous study demonstrated that an odorant-binding protein, OBP4, of the mirid bug Adelphocoris lineolatus functions as a candidate hemipteran PBP but clustered with lepidopteran antennae-binding proteins (ABPs) rather than in the PBP/general odorant-binding protein (GOBP) clade. In this study, we hypothesized that origin and function of PBPs in hemipteran bugs may differ from those of lepidopteran moths. To test this hypothesis, we first constructed a phylogenetic tree using insect OBPs from sister hemipteran and holometabolous lineages, and the results indicated that neither OBP4 nor other types of candidate PBPs of mirid bugs clustered with the lepidopteran PBP/GOBP clade. Then, a fluorescence competitive binding assay was employed to determine binding affinities of recombinant OBP4 protein to host plant volatiles, with functional groups different from A. lineolatus sex pheromone components. The results revealed that OBP4 highly bound the female adult attractant 3-hexanone and 15 other mirid bug biologically active plant volatiles. Finally, we examined cellular expression profiles of OBP4 in putative antennal sensilla that are related to female A. lineolatus host plant location. The fluorescence in situ hybridization and immunocytochemical labeling assay showed that the OBP4 gene was highly expressed in the multiporous olfactory sensilla medium-long sensilla basiconica rather than in the short sensilla basiconica or uniporous sensilla chaetica. These results, together with those of our previous studies, indicate that OBP4 not only functions in recognition of bug-produced sex pheromones in males, but is probably involved in detection of host plant volatiles in both A. lineolatus sexes. Coprinus comatus polysaccharide(CCP)has significant hepatoprotective effect.To explore hepatoprotective mechanism of CCP,the study analyzed preventive effect of CCP on acute alcoholic liver injury in mice by histopathological examination and biochemical analysis.Simultaneously,hepatoprotective mechanism was also analyzed in conjunction with metabolomics and proliferation of gut microbiota.The results showed that CCP significantly decreased alanine aminotransferase(ALT),aspartate aminotransferase(AST)and triglyceride(TG)levels in serum of alcoholic liver disease(ALD)mice.Histopathological examination showed that CCP can significantly improve liver damage.Metabolomics results showed that there were significant differences in the level of metabolites in liver tissue of control group,ALD group and CCP group,including taurine,xanthosine,fumaric acid and arachidonic acid,among others.Metabolites pathways analysis showed that hepatoprotective effect of CCP was related to energy metabolism,biosynthesis of unsaturated fatty acids,amino acids metabolism and lipid metabolism.Additionally,CCP inhibited an increase in the number of Clostridium perfringens,Enterobacteriaceae and Enterococcus,and a decrease in the number of Lactobacillus and Bifidobacterium in the gut of ALD mice.All these findings suggested that CCP treatment reversed the phenotype of ethanol-induced liver injury and the associated metabolites pathways.

For solving the dilemma of the short exothermic life-span of WO_(3)based metastable interstitial composites(MICs)with extensive application prospect,this paper has firstly designed the promising antiwetting Al/WO_(3)MICs via electrophoresis assembly of nano-Al and WO_(3)particles fabricated by hydrothermal synthesis method,followed by the subsequent fluorination treatment.A combination of X ray diffraction(XRD),field emission scanning electron microscope(FESEM),energy dispersive X-ray spectroscopy(EDX),and Fourier transform infrared spectroscopy(FT-IR)techniques were utilized in order to characterize the crystal structure,microstructure,and elemental composition distribution of target films after different natural exposure tests.The product with uniform distribution and high purity possesses a high contact angle of~170°and a minute sliding angle of~1°,and displays the outstanding anti-wetting property using droplets with different surface tensions.It also shows great moisture stability in high relative-humidity circumstances after one year of the natural exposure experiment.Notably,the heat output of a fresh sample can reach up to 2.3 kJ/g and retain 96%after the whole exposure test,showing outstanding thermo-stability for at least one year.This work further proposed the mechanism of antiwetting Al/WO_(3)MICs considering the variation tendency of their DSC curve,providing a valuable theoretical reference for designing other self-protected MICs with a long exothermic life-span applied in wide fields of national defense,military industry,etc.

An H-phosphonate strategy for the synthesis of 2',3'-dideoxynucleoside triphosphates and homodinucleotides

Chimonanthus nitens Oliv.(CN) is a species in the family Calycanthaceae.Its leaf is widely used to make traditional herbal tea in southern China and has a wide range of therapeutic effects.The profile of the ethanol extracts from CN leaves was identified by UPLC-QTOF-MS/MS.Forty seven compounds were determined including organic acids,phenolic acids and derivatives,flavonoids,coumarins,fatty acids and other compounds.The effect of the CN extracts on the inflammatory damage in zebrafish and in RAW264.7 cells was investigated.The extracts demonstrated a strong ability to inhibit the recruitment of neutrophils in LPS-stimulated zebrafish,but macrophage migration was not significantly affected.Pro-inflammatory cytokines(i.e.,TNF-a,IL-6 and IL-1β) were also determined by q-PCR.The extracts strongly reduced mRNA expression of TNF-α,IL-6 but not IL-1β in zebrafish model,while significantly inhibited the production of the factors in the RAW 264.7 cells.Therefore,our results suggest that the ethanol extracts of CN leaves may serve as a source of nutraceutical compounds with anti-inflammatory properties.

LIDAR point cloud-based 3D object detection aims to sense the surrounding environment by anchoring objects with the Bounding Box(BBox).However,under the three-dimensional space of autonomous driving scenes,the previous object detection methods,due to the pre-processing of the original LIDAR point cloud into voxels or pillars,lose the coordinate information of the original point cloud,slow detection speed,and gain inaccurate bounding box positioning.To address the issues above,this study proposes a new two-stage network structure to extract point cloud features directly by PointNet++,which effectively preserves the original point cloud coordinate information.To improve the detection accuracy,a shell-based modeling method is proposed.It roughly determines which spherical shell the coordinates belong to.Then,the results are refined to ground truth,thereby narrowing the localization range and improving the detection accuracy.To improve the recall of 3D object detection with bounding boxes,this paper designs a self-attention module for 3D object detection with a skip connection structure.Some of these features are highlighted by weighting them on the feature dimensions.After training,it makes the feature weights that are favorable for object detection get larger.Thus,the extracted features are more adapted to the object detection task.Extensive comparison experiments and ablation experiments conducted on the KITTI dataset verify the effectiveness of our proposed method in improving recall and precision.

Objective: To observe the effects of Guizhi Fuling Capsule(GZFLC) on myeloma cells and explore the mechanisms. Methods: MM1S and RPMI 8226 cells were co-cultured with different concentrations of serum and the cell experiments were divided into negative(10%, 20% and 40%) groups, GZFLC(10%, 20%, and 40%)groups and a control group. Cell counting kit-8(CCK-8) assays and flow cytometry were used to detect the viability and apoptosis levels of myeloma cells. The effects on mitochondria were examined by reactive oxygen specie(ROS) and tetrechloro-tetraethylbenzimidazol carbocyanine iodide(JC-1) assays. Western blot was used to detect the expression of B cell lymphoma-2(Bcl-2), Bcl-2-associated X(Bax), cleaved caspase-3,-9, cytochrome C(Cytc)and apoptotic protease-activating factor 1(Apaf-1). RPMI 8226 cells(2×107) were subcutaneously inoculated into 48 nude mice to study the in vivo antitumor effects of GZFLC. The mice were randomly divided into four groups using a completely randomized design, the high-, medium-, or low-dose GZFLC(840, 420, or 210 mg/kg per day,respectively) or an equal volume of distilled water, administered daily for 15 days. The tumor volume changes in and survival times of the mice in the GZFLC-administered groups and a control group were observed. Cytc and Apaf-1 expression levels were detected by immunohistochemistry. Results: GZFLC drug serum decreased the viability and increased the apoptosis of myeloam cells(P<0.05). In addition, this drug increased the ROS levels and decreased the mitochondrial membrane potential(P<0.01). Western blot showed that the Bcl-2/Bax ratios were decreased in the GZFLC drug serum-treated groups, whereas the expression levels of cleaved caspase-3,-9,Cytc and Apaf-1 were increased(all P<0.01). Over time, the myeloma tumor volumes of the mice in the GZFLCadministered groups decreased, and survival time of the mice in the GZFLC-administered groups were longer than that of the mice in the control group. Background:The assessment of change in forest ecosystems,especially the change of canopy heights,is essential for improving global carbon estimates and understanding effects of climate change.Spaceborne lidar systems provide a unique opportunity to monitor changes in the vertical structure of forests.NASA’s Ice,Cloud and Land Elevation Satellites,ICESat-1 for the period 2003 to 2009,and ICESat-2(available since 2018),have collected elevation data over the Earth’s surface with a time interval of 10 years.In this study,we tried to discover forest canopy changes by utilizing the global forest canopy height map of 2005(complete global coverage with 1 km resolution)derived from ICESat-1 data and the ATL08 land and vegetation products of 2019(sampling footprints with 17 m diameter)from ICESat-2.Results:Our study revealed a significant increase in forest canopy heights of China’s Beijing-Tianjin-Hebei region.Evaluations of unchanging areas for data consistency of two products show that the bias values decreased significantly from line-transect-level(−8.0 to 6.2 m)to site-level(^(−1).5 to 1.1 m),while RMSE values are still relatively high(6.1 to 15.2 m,10.2 to 12.0 m).Additionally,58%of ATL08 data are located in‘0m’pixels with an average height of 7.9 m,which are likely to reflect the ambitious tree planting programs in China.Conclusions:Our study shows that it is possible,with proper calibrations,to use ICESat-1 and-2 products to detect forest canopy height changes in a regional context.We expect that the approach presented in this study is potentially suitable to derive a fine-scale map of global forest change.
