Sequentially immune-induced antibodies could cross-neutralize SARS-CoV-2 variants

Background:The Omicron(B.1.1.529)SARS-COV-2 variant has raised serious concerns because of its unprecedented rapid rate of spreading and the fact that there are 36 mutations in the spike protein.Since the vaccine-induced neutralizing antibody targets are the spike protein,this may lead to the possibility of vaccine-induced hu-moral immunity escape.Methods:We measured the neutralizing activity in vitro for Omicron and compared this with wild type(WH-09)and Delta variants in human and monkey sera from different types of immunity.The monkey sera samples were collected at 1 and 3 months post three-dose inactivated(PiCoVacc)and recombinant protein(ZF2001)vaccination.Human sera were collected from 1 month post three-dose inactivated vaccination.Results:In inactivated vaccine sera,at 1/3 months post three-dose,geometric mean titers(GMTs)of neutralization antibody(NAb)against the Omicron variant were 4.9/5.2-fold lower than those of the wild type.In recombinant protein vaccine sera,GMTs of NAb against Omicron were 15.7/8.9-fold lower than those of the wild type.In human sera,at 1 month post three-dose inactivated vaccination,GMTs of NAb against Omicron were 3.1-fold lower than those of the wild type.Conclusion:This study demonstrated that despite a reduction in neutralization titers,cross-neutralizing activity against Omicron and Delta variants was still observed after three doses of inactivated and recombinant protein vaccination.

Native promoters that can drive high and stable transgene expression are important tools for modifying plant traits.Although several such promoters have been reported in soybean(Glycine max),few of them function at multiple growth and development stages and during nodule development.Here,we report that the promoters of 40S RIBOSOMAL PROTEIN SMALL SUBUNIT S28(RPS28)and EUKARYOTIC TRANSLATION INITIATION FACTOR 1(EIF1)are ideal for high expression of transgene.Through bioinformatic analysis,we determined that RPS28 and EIF1 were highly expressed during soybean growth and development,nodule development,and various biotic and abiotic stresses.Fusion of both RPS28 and EIF1 promoters,with or without their first intron,with the reporter geneβ-GLUCURONIDASE(uidA)in transgenic soybean,resulted in high GUS activity in seedlings,seeds,and nodules.Fluorimetric GUS assays showed that the RPS28 promoter and the EIF1 promoter yielded high expression,comparable to the soybean Ubiquitin(GmUbi)promoter.RPS28 and EIF1 promoters were also highly expressed in Arabidopsis thaliana and Nicotiana benthamiana.Our results indicate the potential of RPS28 and EIF1 promoters to facilitate future genetic engineering and breeding to improve the quality and yield of soybean,as well as in a wide variety of other plant species.

Multifunctional binder designs for lithium-sulfur batteries

Lithium-sulfur(Li-S)batteries are promising next-generation high energy density batteries but their practical application is hindered by several key problems,such as the intermediate polysulfide shuttling and the electrode degradation caused by the sulfur volume changes.Binder acts as one of the most essential components to build the electrodes of Li-S batteries,playing vital roles in improving the performance and maintaining the integrity of the cathode structure during cycling,especially those with high sulfur loadings.To date,tremendous efforts have been devoted to improving the properties of binders,in terms of the viscosity,elasticity,stability,toughness and conductivity,by optimizing the composition and structure of polymer binders.Moreover,the binder modification endows them strong polysulfide trapping ability to suppress the shuttling and decreases the swelling to maintain the porous structure of cathode.In this review,we summarize the recent progress on the binders for Li-S batteries and discuss the various routes,including the binder combination use,functionalization,in-situ polymerization and ion cross-linking,etc.,to enhance their performance in stabilizing the cathode,building the high sulfur loading electrode and improving the cyclic stability.At last,the design principles and the problems in further applications are also highlighted.

Comparison of the replication and neutralization of different SARS-CoV-2 Omicron subvariants in vitro

Background:New Omicron subvariants are emerging rapidly from BA.1 to BA.4 and BA.5.Their pathogenicity has changed from that of wild-type(WH-09)and Omicron variants have over time become globally dominant.The spike proteins of BA.4 and BA.5 that serve as the target for vaccine-induced neutralizing antibodies have also changed compared to the previous subvariants,which is likely to cause immune es-cape and the reduction of the protective effect of the vaccine.Our study addresses the above issues and provides a basis for formulating relevant prevention and control strategies.Methods:We collected cellular supernatant and cell lysates and measured the viral titers,viral RNA loads,and E subgenomic RNA(E sgRNA)loads in different Omicron subvariants grown in Vero E6 cells,using WH-09 and Delta variants as a reference.Additionally,we evaluated the in vitro neutralizing activity of different Omicron sub-variants and compared it to the WH-09 and Delta variants using macaque sera with different types of immunity.Results:As the SARS-CoV-2 evolved into Omicron BA.1,the replication ability in vitro began to decrease.Then with the emergence of new subvariants,the replication ability gradually recovered and became stable in the BA.4 and BA.5 subvariants.In WH-09-inactivated vaccine sera,geometric mean titers of neutralization antibodies against different Omicron subvariants declined by 3.7~15.4-fold compared to those against WH-09.In Delta-inactivated vaccine sera,geometric mean titers of neutrali-zation antibodies against Omicron subvariants declined by 3.1~7.4-fold compared to those against Delta.Conclusion:According to the findings of this research,the replication efficiency of all Omicron subvariants declined compared with WH-09 and Delta variants,and was lower in BA.1 than in other Omicron subvariants.After two doses of inactivated(WH-09 or Delta)vaccine,cross-neutralizing activities against various Omicron subvariants were seen despite a decline in neutralizing titers.

Sensitivity of SARS-CoV-2 to different temperatures

This study was designed to investigate the sensitivity of SARS-CoV-2 to different temperatures,to provide basic data and a scientific basis for the control of COVID-19 epidemic.The virus was dispersed in 1 mL basal DMEM medium at a final concentration of 103.2 TCID 50/mL and then incubated at 4,22,30,35,37,38,39 and 40°C for up to 5 days.The infectivity of residual virus was titrated using the Vero E6 cell line.The results showed that the virus remained viable for 5 days at 4°C,and for 1 day only at 22 and 30°C.We found that the infectivity of the virus was completely lost after less than 12 hours at 37,38 and 39°C,while at 40°C,the inactivation time of the virus was rapidly reduced to 6 hours.We show that SARS-CoV-2 is sensitive to heat,is more stable at lower temperatures than higher temperature,remains viable for longer at lower temperatures,and loses viability rapidly at higher temperatures.

Instance-Specific Algorithm Selection via Multi-Output Learning

Instance-specific algorithm selection technologies have been successfully used in many research fields,such as constraint satisfaction and planning. Researchers have been increasingly trying to model the potential relations between different candidate algorithms for the algorithm selection. In this study, we propose an instancespecific algorithm selection method based on multi-output learning, which can manage these relations more directly.Three kinds of multi-output learning methods are used to predict the performances of the candidate algorithms：（1）multi-output regressor stacking;（2） multi-output extremely randomized trees; and（3） hybrid single-output and multioutput trees. The experimental results obtained using 11 SAT datasets and 5 Max SAT datasets indicate that our proposed methods can obtain a better performance over the state-of-the-art algorithm selection methods.

Improved Algorithm for Solving Discrete Logarithm Problem by Expanding Factor

The discrete logarithm problem(DLP)is to find a solution n such that g^n=h in a finite cyclic group G=,where h∈G.The DLP is the security foundation of many cryptosystems,such as RSA.We propose a method to improve Pollard’s kangaroo algorithm,which is the classic algorithm for solving the DLP.In the proposed algorithm,the large integer multiplications are reduced by controlling whether to perform large integer multiplication.To control the process,the tools of expanding factor and jumping distance are introduced.The expanding factor is an indicator used to measure the probability of collision.Large integer multiplication is performed if the value of the expanding factor is greater than the given bound.The improved algorithm requires an average of(1.633+o(1))q(1/2)times of the large integer multiplications.In experiments,the average large integer multiplication times is approximately(1.5+o(1))q(1/2).

Integration of root architecture,root nitrogen metabolism,and photosynthesis of‘Hanfu’apple trees under the cross-talk between glucose and IAA

Sugars and auxin have important effects on almost all phases of plant life cycle,which are so fundamental to plants and regulate similar processes.However,little is known about the effect of cross-talk between glucose and indole-3-acetic acid(IAA)on growth and development of apple trees.To examine the potential roles of glucose and IAA in root architecture,root nitrogen(N)metabolism and photosynthetic capacity in‘Hanfu’(Malus domestica),a total of five treatments was established:single application of glucose,IAA,and auxin polar transport inhibitor(2,3,5-triiodobenzoic acid,TIBA),combined application of glucose with TIBA and that of glucose with IAA.The combined application of glucose with IAA improved root topology system and endogenous IAA content by altering the mRNA levels of several genes involved in root growth,auxin transport and biosynthesis.Moreover,the increased N metabolism enzyme activities and levels of genes expression related to N in roots may suggest higher rates of transformation of nitrate(NO3--N)into amino acids application of glucose and IAA.Contrarily,single application of TIBA decreased the expression levels of auxin transport gene,hindered root growth and decreased endogenous IAA content.Glucose combined with TIBA application effectively attenuated TIBA-induced reductions in root topology structure,photosynthesis and N metabolism activity,and mRNA expression levels involved in auxin biosynthesis and transport.Taken together,glucose application probably changes the expression level of auxin synthesis and transport genes,and induce the allocation of endogenous IAA in root,and thus improves root architecture and N metabolism of root in soil with deficit carbon.
