Enhanced carbon capture with motif-rich amino acid loaded defective robust metal-organic frameworks

The use of metal-organic frameworks(MOFs)as solid adsorption materials for carbon capture is promising,but achieving efficient and reversible adsorption with a balance of capacity and selectivity for carbon dioxide(CO_(2))over N_(2) remains a challenge.To take full advantage of the strong channel traffic and robustness of MOFs with relatively small pores,it is highly necessary to employ a defect-engineering strategy to construct a broader channel structure that can facilitate the loading of functional motif-rich amino acids(AAs).This strategy can greatly enhance the CO_(2) adsorption performance of MOF.In this study,motif-rich amino acids are loaded into the defective and robust porous frameworks via combined defect-engineering and post-synthetic methods.The defective Zr/Hf-MOF-808s modified with AAs,especially for the 18 mol%4-nitroisophthalic acid,generated defective products allowing for the loading of L-serine(L-Ser).This modification resulted in a significant improvement in both the adsorption capacity(248%improvement at 298 K,100 kPa)and the selectivity of CO_(2)/N_(2) using the ideal adsorbed solution theory(IAST),with the selectivity increasing to 120.55 and 38.27 at 15 and 100 kPa,respectively,while maintaining good cycling performance.Density functional theory(DFT)simulation,CO_(2) temperature-programmed desorption(CO_(2)-TPD),and in situ Fourier transform infrared spectroscopy(FTIR)were further employed to have a better understanding of the enhanced CO_(2) adsorption capacity.Interestingly,unlike the AAs loaded pristine MOF-808s that showed the best CO_(2) adsorption capacity with the loading of short and small glycine(Gly),the broadened channel size in our work enables the loading of functional motif-rich L-serine,which brings more active binding sites,improving CO_(2) adsorption.

PTEN-induced putative kinase 1(PINK1),a mitochondrial kinase that phosphorylates Parkin and other proteins,plays a crucial role in mitophagy and protection against neurodegeneration.Mutations in PINK1 and Parkin can lead to loss of function and early onset Parkinson's disease.However,there is a lack of strong in vivo evidence in rodent models to support the theory that loss of PINK1 affects mitophagy and induces neurodegeneration.Additionally,PINK1 knockout pigs(Sus scrofa)do not appear to exhibit neurodegeneration.In our recent work involving non-human primates,we found that PINK1 is selectively expressed in primate brains,while absent in rodent brains.To extend this to other species,we used multiple antibodies to examine the expression of PINK1 in pig tissues.In contrast to tissues from cynomolgus monkeys(Macaca fascicularis),our data did not convincingly demonstrate detectable PINK1expression in pig tissues.Knockdown of PINK1 in cultured pig cells did not result in altered Parkin and BAD phosphorylation,as observed in cultured monkey cells.A comparison of monkey and pig striatum revealed more PINK1-phosphorylated substrates in the monkey brain.Consistently,PINK1 knockout in pigs did not lead to obvious changes in the phosphorylation of Parkin and BAD.These findings provide new evidence that PINK1expression is specific to primates,underscoring the importance of non-human primates in investigating PINK1function and pathology related to PINK1 deficiency.

Effect of covalent-binding modes of osteogenic-related peptides with artificial carriers on their biological activities in vivo

Covalent binding between bioactive substances and materials in different ways can significantly improve the bone inductivity and biological activity of bone repair materials.However,there is a lack of systematic understanding of how these binding modes affect biological activities of the active substances.In this study,four kinds of functionalized Multi-walled carbon nanotubes(MWCNTs)were prepared,ensuring the same grafting rate of different functional groups.Subsequently,two kinds of osteogenic-related peptides,bone morphogenetic protein-2 mimicking peptides and osteogenic growth mimicking peptides,were covalently bound to functionalized MWCNTs,ensuring the same molar mass of peptides bound to different functionalized MWCNTs in this process.Then the same amount of functionalized MWC-NTs/Peptides composites were introduced into the scaffolds,and through the ectopic osteogenesis model in rats and calvarial defect model in rabbits,ectopic osteogenesis and bone repair ability of the composites were analyzed.Furthermore,the effects of different covalent binding modes on peptide-induced osteogenesis and bone repair were studied.The results showed that the negative influencing trend of different covalent binding modes of osteogenic-related peptides with artificial carriers on their biological activities was in the order as follows:amide binding(carboxyl)>silane coupling>dopamine bind-ing>amide binding(amino),whose mechanism might be mainly that the covalent binding of peptides with different functional groups resulted in different charges.We believe that the results of this study have important guiding significance for the research and development of bone repair materials covalently bound with bioactive substances.

IRE1α regulates the PTHrP-IHH feedback loop to orchestrate chondrocyte hypertrophy and cartilage mineralization

Cartilage development is controlled by the highly synergistic proliferation and differentiation of growth plate chondrocytes,in which the Indian hedgehog(IHH)and parathyroid hormone-related protein-parathyroid hormone-1 receptor(PTHrP-PTH1R)feedback loop is crucial.The inositol-requiring enzyme 1a/X-box-binding protein-1 spliced(IRE1α/XBP1s)branch of the unfolded protein response(UPR)is essential for normal cartilage development.However,the precise role of ER stress effector IRE1α,encoded by endoplasmic reticulum to nucleus signaling 1(ERN1),in skeletal development remains unknown.Herein,we reported that loss of IRE1α accelerates chondrocyte hypertrophy and promotes endochondral bone growth.ERN1 acts as a negative regulator of chondrocyte proliferation and differentiation in postnatal growth plates.Its deficiency interrupted PTHrP/PTH1R and IHH homeostasis leading to impaired chondrocyte hypertrophy and differentiation.XBP1s,produced by p-IRE1α-mediated splicing,binds and up-regulates PTH1R and IHH,which coordinate cartilage development.Meanwhile,ER stress cannot be activated normally in ERN1-deficient chondrocytes.In conclusion,ERN1 deficiency accelerates chondrocyte hypertrophy and cartilage mineralization by impairing the homeostasis of the IHH and PTHrP/PTH1R feedback loop and ER stress.ERN1 may have a potential role as a new target for cartilage growth and maturation.

Review on microbial metabolomics of probiotics and pathogens:Methodologies and applications

In recent years,microbial metabolomics,a new field that has attracted wide attention,provides a map of metabolic pathways and clarifies the interaction mechanism between microorganisms and hosts.Many microorganisms are found in the human intestine,oral cavity,vagina,etc.Probiotics could maintain the good health of the host,while pathogens and an imbalance of bacterial flora lead to a series of diseases of the body and mind.Metabolomics is a science for qualitative and quantitative analysis of all metabolites in an organism or biological system,which could provide key information to understand the related metabolic pathways and associated changes.This approach analyzes the final products of cellular regulatory processes,the level of which can be regarded as the ultimate response of the biological system to genetic or environmental changes.Microbial metabolomics has been widely used in different research fields,such as microbial phenotypic classification,mutant screening,metabolic pathways,microbial metabolic engineering,fermentation engineering monitoring and optimization,microbial environmental pollution,and so on.However,there are only a few reviews on microbial metabolomics of probiotics and pathogens.This review summarizes the main methodologies,including sample preparation,identification of metabolites,data processing,and analysis.Recent applications in microbial metabolomics of probiotics and pathogens are also described.This paper first summarized the research progress and application of microbial metabolomics from two aspects:probiotics and pathogenic bacteria.Probiotics and pathogenic bacteria do not exist independently most of the time;hence,these were reviewed in the research field of coexistence of probiotics and pathogenic bacteria,which was subdivided into important microbial research fields closely related to human health,including the human gut,oral cavity,food,and nutrition-related microorganisms.Then,the main problems and trends associated with microbial metabolomics are discussed.

The Five-hundred-meter Aperture Spherical radio Telescope（FAST） has the largest aperture and a 19-beam L-band receiver,making it powerful for investigating the neutral hydrogen atomic gas（H?） in the universe.We present HiFAST（https://hifast.***.io）,a dedicated,modular,and self-contained calibration and imaging pipeline for processing the H?data of FAST.The pipeline consists of frequency-dependent noise diode calibration,baseline fitting,standing wave removal using an FFT-based method,flux density calibration,stray radiation correction,and gridding to produce data cubes.These modules can be combined as needed to process the data from most FAST observation modes:tracking,drift scanning,On-The-Fly mapping,and most of their variants.With HiFAST,the root-mean-square（RMS） noises of the calibrated spectra from all 19 beams were only slightly（～5%） higher than the theoretical expectation.The results for the extended source M33 and the point sources are consistent with the results from Arecibo.The moment maps（0,1 and 2） of M33 agree well with the results from the Arecibo Galaxy Environment Survey（AGES） with a fractional difference of less than 10%.For a common sample of 221 sources with signal-to-noise ratio S/N>10 from the Arecibo Legacy Fast ALFA（ALFALFA） survey,the mean value of fractional difference in the integrated flux density,Sint,between the two datasets is approximately 0.005%,with a dispersion of 15.4%.Further checks on the integrated flux density of 23 sources with seven observations indicate that the variance in the flux density of the source with luminous objects(Sint>2.5 Jy km s-1) is less than 5%.Our tests suggest that the FAST telescope,with the efficient,precise,and user-friendly pipeline HiFAST,will yield numerous significant scientific findings in the investigation of the H? in the universe.

Microwave-assisted continuous flow phytosynthesis of silver nanoparticle/reduced graphene oxide composites and related visible light catalytic performance

The creation of an environmentally friendly synthesis method for silver nanomaterials(Ag-NPs)is an urgent concern for sustainable nanotechnology development.In the present study,a novel straightforward and green method for the preparation of silver nanoparti-cle/reduced graphene oxide(AgNP/rGO)composites was successfully developed through the combination of phytosynthesis,continuous flow synthesis and microwave-assistance.Oriental persimmon(Diospyros kaki Thunb.)extracts were used as both plant reducing and capping agents for fast online synthesis of AgNP/rGO composites.The experimental param-eters were optimized and the morphologies of the prepared materials were investigated.The characterization results reveal that spherical AgNPs were quickly synthesized and uni-formly dispersed on rGO sheets using the proposed online system.Fourier transform in-frared spectroscopy analysis confirmed that phenols,flavonoids,and other substances in the plant extracts played a decisive role in the synthesis of AgNP/rGO composites.Using sodium borohydride(NaBH4)degradation of p-nitrophenol(4-NP)as a model,the catalytic activity of the prepared AgNP/rGO materials was evaluated.The complete degradation of 4-NP was achieved within 12 min through the use of AgNP/rGO materials,and the compos-ite had a much better catalytic activity than the bare AgNPs and rGO had.Compared with the conventional chemical method,our online method is facile,fast,cost-efficient,and en-vironmentally friendly.

Abiotic stress reduces plant yield and quality.WRKY transcription factors play key roles in abiotic stress responses in plants,but the molecular mechanisms by which WRKY transcription factors mediate responses to drought and osmotic stresses in apple(Malus×domestica Borkh.)remain unclear.Here,we functionally characterized the apple GroupⅢWRKY gene MdWRKY115.qRT-PCR analysis showed that MdWRKY115 expression was up-regulated by drought and osmotic stresses.GUS activity analysis revealed that the promoter activity of MdWRKY115 was enhanced under osmotic stress.Subcellular localization and transactivation assays indicated that MdWRKY115 was localized to the nucleus and had a transcriptional activity domain at the N-terminal region.Transgenic analysis revealed that the overexpression of MdWRKY115 in Arabidopsis plants and in apple callus markedly enhanced their tolerance to drought and osmotic stresses.DNA affinity purification sequencing showed that MdWRKY115 binds to the promoter of the stress-related gene MdRD22.This binding was further verified by an electrophoretic mobility shift assay.Collectively,these findings suggest that MdWRKY115 is an important regulator of osmotic and drought stress tolerance in apple.

1.Objective In the past decade,a group of medium to giant lead-zinc deposits,represented by Huoshaoyun,Sachakou,and Yuanbaoling,have been discovered in the Aksai Chin region of Karakoram,Xinjiang.They are all located in the Mesozoic carbonate and clastic rock formations.The Sachakou leadzinc mining area is adjacent to the northwest of the Huoshaoyun lead-zinc mining area and is in the same stratigraphic layer as Huoshaoyun.Although many scholars have been arguing about the type and age of Huoshaoyun lead-zinc mineralization,few scholars have paid attention to the classification of the ore-bearing strata in the area.The stratigraphy of the Lower Permian Shenxianwan Group to the Upper Cretaceous Tielongtan Group is exposed in the Sachakou area of Karakorum,Xinjiang,however,the Late Permian-Early Triassic stratigraphy is missing(Fig.1a).Due to the harsh natural conditions in the area and the low level of work,the stratigraphic delineation is not exhaustive,and the regional lithology is dominated by carbonates and clastic rocks,which makes it difficult to identify the age of the regional lithology and causes problems for the exploration and research of lead-zinc in the area.

Coronaviruses RNA interacts with host miR-500a-5p and miR-501-5p to regulate multiple pathways

The Coronavirus disease is the contagious disease. Clinical manifests vary from no symptoms, upper and lower respiratory tract infections, and even severe acute respiratory distress syndrome and multiple organs failure. Recently, three of new beta-coronaviruses have emerged to cause serious and widespread illness and death, such as the Severe Acute Respiratory Syndrome Coronaviruses (SARS-CoV), Middle East Respiratory Syndrome Coronaviruses (MERS-CoV), and 2019-nCoV (the causative pathogen of Coronavirus Disease 2019 (COVID-19). The COVID-19 is continuing to spread globally. However, the detailed molecular mechanisms have not yet been fully clarified. Since the similarity of the pathological features among 2019-nCoV, SARS-CoV and MERS-CoV, we could speculate the biological features of current COVID-19 by researching those of MERS-CoV and SARS-CoV. Therefore, we analyzed expression levels of miRNA and mRNA profiles of MERS-CoV, SARS-CoV and 2019-nCoV from five public GEO datasets (Table S1). As a result, we found 16 miRNAs potentially target a group of hub genes clustering together to tune the immune response. Notably, viral RNAs have the ability to sponge miR-500a-5p and miR-501-5p, indicating viral RNAs sequestering these two miRNAs to trigger genes expression and contribute to pathogenesis. We hope this study will pave the way to better understand 2019-nCoV pathogenesis and develop the innovative therapeutics.
