Local and systemic inflammation triggers different outcomes of tumor growth related to infiltration of anti-tumor or pro-tumor macrophages

Background: Previous evidence suggests inflammation may be a double-edged sword with cancer-promoting and cancer suppressing function. In this study, we explore the impact of local and systemic inflammation on cancer growth.Methods: Female BALB/C mice were subcutaneously implanted with foreign body (plastic plates) to build up a local inflammation and intraperitoneally injected with PolyIC or lipopolysaccharides (LPS) to build up a systemic inflammation, followed by subcutaneous injection of5 × 105 colon cancer cells. Immunohistochemistry and enzyme linked immunosorbent assay were utilized to detect the Ki67 and interleukin (IL) 6, IL-1β, and monocyte chemoattractant protein-1 expression in the tumor tissues and serum, respectively. The distributions of immune cells and expression of toll-like receptors (TLRs) were evaluated by flow cytometry (FCM) and quantitative real time-polymerase chain reaction.Results: The results showed that local inflammation induced by foreign body implantation suppressed tumor growth with decreased tumor weight (P = 0.001), volume (P = 0.004) and Ki67 index (P < 0.001). Compared with the control group, myeloid-derived suppressive cells sharply decreased (P = 0.040), while CD4+ T cells slightly increased in the tumor tissues of the group of foreign body-induced local inflammation (P = 0.035). Moreover, the number of M1 macrophages (P = 0.040) and expression of TLRs, especially TLR3 (P < 0.001) and TLR4 (P < 0.001), were significantly up-regulated in the foreign body group. Contrarily, tumor growth was significantly promoted in LPS or PolyIC-induced systemic inflammation (P = 0.009 and 0.006). FCM results showed M1 type macrophages (P = 0.017 and 0.006) and CD8+ T cells (P = 0.031 and 0.023) were decreased, while M2 type macrophages (P = 0.002 and 0.007) were significantly increased in tumor microenvironment of LPS or PolyIC-induced systemic inflammation group. The intestinal immune function of chickens is limited during the early growing stage.Maternal nutritional intervention has been suggested to affect the innate immunity of offspring.The present study aimed to investigate the effects of maternal stevioside supplementation on the intestinal immune function of chicken offspring.A total of 120 Jinmao yellow-feathered breeder hens were fed a basal diet or a diet supplemented with 250 mg/kg stevioside for 5 weeks.During the last week,200 breeding eggs from each group were collected for incubation.After hatching,80 male offspring(40 chickens from each group)were randomly selected and fed the same basal diet for 28 d.In addition,90 well-shaped fertile eggs of non-treated breeder hens were incubated for the in ovo injection experiment.Steviol dissolved in 20%glycerol was injected at 7 d of incubation.The results showed that maternal stevioside supplementation could improve embryonic development,jejunal integrity and proliferation in the jejunal crypt(P0.05).Furthermore,maternal stevioside supplementation could induce hypo-methylation on the promoter region of suppressor of cytokine signaling 1(SOCS1).In conclusion,maternal stevioside supplementation could improve the intestinal immune function of chicken offspring potentially via modulating the gut microbiota and downregulating the promoter methylation level of SOCS1.

Effects of particle type on the particle fluidization and distribution in a liquid–solid circulating fluidized bed boiler

A liquid-solid circulating fluidized bed boiler is designed and built for visualization research by applying the fluidized bed heat transfer and fouling prevention technology to the water side of the boiler. Four types of engineering plastic particles with different physical properties are selected as the solid working media. The effect of particle types on the fluidization and distribution of particles in the boiler is investigated under different feedwater flow rates and amount of added particles by using the charge couple device image measurement and acquisition system. The results show that all kinds of particles can't be normally fluidized and accumulate in the drum at low amount of added particles and feedwater flow rate. The particles with great density and low sphericity are more likely to accumulate. The average solid holdup in the riser tubes increases with the increase in feedwater flow rate and the amount of added particles. The non-uniform degree of particle distribution in the riser tubes generally decreases with the increase in feedwater flow rate and the amount of added particles. The particles with small density and settling velocity have high average solid holdup in the riser tubes under close sphericity. In generally,the smaller the density and settling velocity, the more uniform the particle distribution in the riser tubes.Three-dimensional diagrams of the non-uniform degree of particle distribution in the riser tubes of the boiler are established.

A Planning Method for Operational Test of UAV Swarm Based on Mission Reliability

The unmanned aerial vehicle(UAV)swarm plays an increasingly important role in the modern battlefield,and the UAV swarm operational test is a vital means to validate the combat effectiveness of the UAV swarm.Due to the high cost and long duration of operational tests,it is essential to plan the test in advance.To solve the problem of planning UAV swarm operational test,this study considers the multi-stage feature of a UAV swarm mission,composed of launch,flight and combat stages,and proposes a method to find test plans that can maximize mission reliability.Therefore,a multi-stage mission reliability model for a UAV swarm is proposed to ensure successful implementation of the mission.A multi-objective integer optimization method that considers both mission reliability and cost is then formulated to obtain the optimal test plans.This study first constructs a mission reliability model for the UAV swarm in the combat stage.Then,the launch stage and flight stage are integrated to develop a complete PMS(Phased Mission Systems)reliability model.Finally,the Binary Decision Diagrams(BDD)and Multi Objective Quantum Particle Swarm Optimization(MOQPSO)methods are proposed to solve the model.The optimal plans considering both reliability and cost are obtained.The proposed model supports the planning of UAV swarm operational tests and represents a meaningful exploration of UAV swarm test planning.

Hypersonic vehicles have enormous military and economic value,while their power and thermal protection demands will increase substantially with the rise in Mach number and duration.Converting the tremendous high-quality heat on the vehicle surface and engine wall into electrical energy through heat-to-power technologies will not only play a role in thermal protection,but also supply power for the vehicle.This paper provides a comprehensive review of heat-to-power conversion technologies on hypersonic vehicles,including the indirect conversion of Brayton and Rankine cycles,direct conversion of thermoelectric materials,and combined conversion.For the open Brayton cycle with hydrocarbon fuel as the working fluid,the Power-to-Weight Ratio(PWR)can achieve the highest,at around 1.8,due to the high PWR of the hydrocarbon fuel turbine and the few components of the system.However,its work capacity is limited by the flow rate of the supplied fuel.The closed Brayton cycle can maintain a relatively high PWR,ranging from 0.2 to 0.8,while achieving relatively high output power and conversion efficiency.The Rankine cycle has a higher PWR,its range is close to that of the closed Brayton cycle,peaking at about 0.88.The thermoelectric materials technology has a small power generation level,making it more suitable for scenarios with low power demand.This review provides a basis for selecting and developing heat-to-power conversion technologies on hypersonic vehicles.

So far,there has been no safe and convenient method to weigh the largeﬁerce animals,like Amur tigers.To address this problem,we built models to predict the body weight of Amur tigers based on the fact that body weight is proportional to body measurements or age.Using the method of body measurements,we extracted the body measurements from 4 different kinds of the lateral body image of tigers,that is,total lateral image,central lateral image,ellipseﬁtting image,and rectangleﬁtting image,and then we respectively used artiﬁcial neural network(ANN)and power regression model to analyze the predictive relationships between body weight and body measurements.Our results demonstrated that,among all ANN models,the model built with rectangleﬁtting image had the smallest mean square error.Comparatively,we screened power regression models which had the smallest Akakai information criteria(AIC).In addition,using the method of age,weﬁtted nonlinear regression models for the relationship between body weight and age and found that,for male tigers,logistic model had the smallest AIC.For female tigers,Gompertz model had the smallest AIC.Consequently,this study could be applied to estimate body weight of captive,or even wild,Amur tigers safely and conveniently,helping to monitor individual health and growth of the Amur tiger populations.

A transient 3D model was established to investigate the effect of spatial interaction of ultrasounds on the dual-frequency ultrasonic field in magnesium alloy melt.The effects of insertion depth and tip shape of the ultrasonic rods,input pressures and their ratio on the acoustic field distribution were discussed in detail.Additionally,the spacing,angle,and insertion depth of two ultrasonic rods significantly affect the interaction between distinct ultrasounds.As a result,various acoustic pressure distributions and cavitation regions are obtained.The spherical rods mitigate the longitudinal and transversal attenuation of acoustic pressure and expand the cavitation volume by 53.7%and 31.7%,respectively,compared to the plate and conical rods.Increasing the input pressure will enlarge the cavitation region but has no effect on the acoustic pressure distribution pattern.The acoustic pressure ratio significantly affects the pressure distribution and the cavitation region,and the best cavitation effect is obtained at the ratio of 2:1(P15:P20).

The pushover method for underground structures is a seismic analysis method featured by high calculation accuracy and a simple implementation process.The method has been widely used in seismic design and other related scientific research;however,the influence of different soil-structure flexibility ratios on the accuracy of this method is still not well understood.In this study,we select the cross-section structures beneath the Daikai subway station as the research object and establish 12 finite element analysis models with different soil-structure flexibility ratios using ABAQUS.All models are computed by the dynamic time-history method or the pushover method.Furthermore,the dynamic time-history solution result is taken as the standard solution,and the precision and application of the pushover analysis method are discussed based on the parameters of peak interlayer displacement and peak internal force of the middle column section.The results show that the soil-structure flexibility ratio has a significant influence on the calculation accuracy of the pushover method,and the calculation accuracy of this method is the most ideal when the soil-structure flexibility is equal to 1.The research results can provide significant references for the seismic design of underground structures or the improvement of simplified seismic analysis methods.

Bone morphogenetic protein(BMP-2)has been approved by the FDA to promote bone regeneration,but uncertain osteogenic effect and dose-dependent side effects may occur.Osteoimmunomodulation plays an important role in growth factor-based osteogenesis.Here,we explored how proinflammatory signals affect the dose-dependent osteogenic potential of BMP-2.We observed that the expression level of local IL-1βdid not increase with the dose of BMP-2 in the mouse osteogenesis model.A low dose of BMP-2 could not promote new bone formation,but trigger the release of IL-1βfrom M1 macrophages.As the dose of BMP-2 increased,the IL-1βexpression and M1 infiltration in local microenvironment were inhibited by IL-1Ra from MSCs under osteogenic differentiation induced by BMP-2,and new bone tissues formed,even excessively.Anti-inflammatory drugs(Dexamethasone,Dex)promoted osteogenesis via inhibiting M1 polarization and enhancing BMP-2-induced MSC osteo-differentiation.Thus,we suggest that the osteogenic effect of BMP-2 involves macrophage-MSC interaction that is dependent on BMP-2 dose and based on IL-1R1 ligands,including IL-1βand IL-1Ra.The dose of BMP-2 could be reduced by introducing immunoregulatory strategies.
