Bionic immunoactivator copresenting autophagy promoting and costimulatory molecules for synergistic cancer immunotherapy

Immunotherapy has great promise in improving malignant tumor treatment.However,the efficacy of existing strategies is often limited by the immunosuppressive environment.Here,we demonstrate an in situ bionic immunoactivator,PLT-Bec1/DTA-1,with possessed natural advantages of platelets for tumor recruitment and activation,on which DTA-1(CD357 monoclonal antibody)and Bec1 were tethered as combined immune boosters.PLT-Bec1/DTA-1,as a self-triggered release repository,can deliver the pre-tethered Bec1 and DTA-1 deeply through the secretion of platelet microparticles(PMPs),thereby cooperate tacitly and exhibit superiority in immune activation of dendritic cells(DCs)and T cells via autophagy inducibility,coupled with glucocorticoid-induced tumor necrosis factor receptor(GITR)-triggered T_(Reg) suppression,remodeled the immunosuppressive network of tumor microenvironment.PLT-Bec1/DTA-1 promoted antigen presentation and T cell proliferation,and alleviated the low activity state of bone marrow-derived dendritic cells(BMDCs)in tumor suppressive environment.PLT-Bec1/DTA-1 inhibited tumor recurrence(5-and 13-fold lower of control group in tumor volume)and CD8^(+)T/T_(Reg) ratio(6.3-and 8.8-fold vs.control group)in mouse tumor model after intravenous or subcutaneous administration.Also,PLT-Bec1/DTA-1 prevented tumor colonization in lung through in situ immune activation,and was slightly superior to the combined of Bec1 and PD-L1.Our findings highlight the promise of delivering immunostimulatory payloads via bionic carriers,eliciting automatic in situ activation of effector immune cells in tumor microenvironment for tumor eradication.All these results provide promising prospects into the application of immunoactivator in improving cancer synergistic immunotherapy to overcome the bottlenecks in clinic.

The growth in biomedical data resources has raised potential privacy concerns and risks of genetic information leakage. For instance, exome sequencing aids clinical decisions by comparing data through web services, but it requires significant trust between users and providers. To alleviate privacy concerns, the most commonly used strategy is to anonymize sensitive data. Unfortunately, studies have shown that anonymization is insufficient to protect against reidentification attacks. Recently, privacy-preserving technologies have been applied to preserve application utility while protecting the privacy of biomedical data. We present the PICOTEES framework, a privacy-preserving online service of phenotype exploration for genetic-diagnostic variants (https://***:3000/). PICOTEES enables privacy-preserving queries of the phenotype spectrum for a single variant by utilizing trusted execution environment technology, which can protect the privacy of the user's query information, backend models, and data, as well as the final results. We demonstrate the utility and performance of PICOTEES by exploring a bioinformatics dataset. The dataset is from a cohort containing 20,909 genetic testing patients with 3,152,508 variants from the Children's Hospital of Fudan University in China, dominated by the Chinese Han population (>99.9%). Our query results yield a large number of unreported diagnostic variants and previously reported pathogenicity.

Compared to other organisms,plants have evolved a greater number of aquaporins with diverse substrates and functions to adapt to ever-changing environmental and internal stimuli for growth and development.Although aquaporins were initially identified as channels that allow water molecules to cross biological membranes,progress has been made in identifying various novel permeable substrates.Many studies have characterized the versatile physiological and biophysical functions of plant aquaporins.Here,we review the recent reports that highlight aquaporin-facilitated regulation of major physiological processes and stress tolerance throughout plant life cycles as well as the potential prospects and possibilities of applying aquaporins to improve agricultural productivity,food quality,environmental protection,and ecological conservation.

An efficient N–H and C(sp~3)-H functionalization of aryl ketones with benzylamines/amino acids was developed under mild conditions by virtue of anodic oxidation. A variety of functionalized 2,5-diaryloxazoles were obtained with good to excellent yields. Moreover, some important natural products can be prepared by this method. The reaction features a broad substrate scope, scalability, metal-free and chemical oxidant-free.

Liquid-metal microgrid stretchable electronics based on bionic leaf veins with ultra-stretchability and high conductivity

Stretchable electronics that monitor joint activity and treat diseases based on liquid metal could be used in the development of healthcare applications.Such devices can be seamlessly integrated with human skin.However,most high-precision microstructures and complex patterns are difficult to fabricate due to the limitations of conventional fabrication solutions,resulting in suboptimal performance under practical conditions.Here,a liquid-metal stretchable system utilizing natural leaf veins was reported as microstructures,which was based on a biomimetic concept and utilized an all-solution process for the preparation of complex microstructures.The systems are ultra-high tensile(800% tensile strain),environmentally stable(20 days) and mechanically durable(300-cycle).The system can accurately recognize the wearer's finger bending level as well as simple gesture signals.At the same time,the system acts as a wearable heater,which can realize the fast heating behavior of heating up to 50℃in 3 min under the human body-safe voltage(1.5 V).The tensile stability is demonstrated by the heterogeneous integration of lasers(405 nm) with the system interconnects for a stretchable and wearable light source.

Event-triggered robust MPC of nonlinear cyber-physical systems against DoS attacks

This paper proposes an event-triggered robust nonlinear model predictive control(NMPC) framework for cyber-physical systems(CPS) in the presence of denial-of-service(Do S) attacks and additive disturbances. In the framework, a new robustness constraint is introduced to the NMPC optimization problem in order to deal with additive disturbances, and a packet transmission strategy is designed for NMPC such that Do S attacks can be tackled. Then, an event-triggered mechanism, which accommodates Do S attacks occurring in the communication network, is proposed to reduce the communication cost for resource-constrained CPSs. Besides, we prove that the NMPC algorithm is recursively feasible and the closed-loop system is input-to-state practical stable under some sufficient conditions. Simulation examples and comparisons are conducted to show the effectiveness of the proposed NMPC algorithm.

Experimental and mechanistic study of dispersed micrometer-sized particle resuspension in a square straight duct with rough walls

The resuspension of graphite dust is an important phenomenon in the release of radioactivity and the safety of nuclear reactors during severe accidents.In this study,a visualization experimental platform is constructed to study effects of particle size,flow velocity,and wall roughness on the resuspension characteristics of graphite particles.A statistical model of particle resuspension applicable to monolayer dispersed particles is developed based on the moment equilibrium of the particles and the flow field characteristics,as calculated by the large-eddy simulation framework.The results show that particle resuspension can be divided into short-and long-term resuspension stages.Most particle resuspension occurs during the short-term stage.With increases in flow velocity and particle diameter,the aerodynamic or adhesion force acting on the particles increases,and corresponding particle resuspension fraction increases.The influence of rough walls on particle resuspension is related to both the force on the particles and the arm ratio between the wall morphology and the particle diameter.A comparison with the experimental results demonstrates that the particle resuspension model developed in this study accurately predicts the impact of flow velocity,particle size,and wall roughness on particle resuspension.

Dear Editor,This letter presents a distributed deep reinforcement learning(DRL) based approach to deal with the path following and formation control problems for underactuated unmanned surface vehicles(USVs). By constructing two independent actor-critic architectures,the deep deterministic policy gradient(DDPG) method is proposed to determine the desired heading and speed command for each USV.

As the core device of all-solid-state laser,infrared(IR)nonlinear optical(NLO)material plays a critical role in exploring new coherent lights by frequency conversion technologies,which are widely used in noninvasive medical diagnostics,spectroscopy,environmental monitoring and long-distance laser communication.To satisfy the growing demand of current laser technology in IR regions,high performance NLO materials with large second-harmonic generation(SHG)response,high laser-induced damage threshold(LIDT)and wide transparency range are highly expected.Recently,numerous efforts and advances have been achieved in sulfide system.This review highlights the progress of high-performance sulfide IR NLO materials with a good balance between strong SHG response(≥1.0×AgGaS_(2))and wide band gap(≥3.0 eV)since 2018.The chemical synthesis,crystal structure,optical properties and structure-property relationship of 13 high performance metal sulfide IR NLO materials are summarized and reviewed in detail.Finally,the potential opportunities and challenges in IR NLO materials are discussed.

Emerging memristors and applications in reservoir computing

Recently, with the emergence of ChatGPT, the field of artificial intelligence has garnered widespread attention from various sectors of society. Reservoir Computing (RC) is a neuromorphic computing algorithm used to analyze time-series data. Unlike traditional artificial neural networks that require the weight values of all nodes in the trained network, RC only needs to train the readout layer. This makes the training process faster and more efficient, and it has been used in various applications, including speech recognition, image classification, and control systems. Its flexibility and efficiency make it a popular choice for processing large amounts of complex data. A recent research trend is to develop physical RC, which utilizes the nonlinear dynamic and short-term memory properties of physical systems (photonic modules, spintronic devices, memristors, etc.) to construct a fixed random neural network structure for processing input data to reduce computing time and energy. In this paper, we introduced the recent development of memristors and demonstrated the remarkable data processing capability of RC systems based on memristors. Not only do they possess excellent data processing ability comparable to digital RC systems, but they also have lower energy consumption and greater robustness. Finally, we discussed the development prospects and challenges faced by memristors-based RC systems.
