Engineered extracellular vesicles for targeted reprogramming of cancer-associated fibroblasts to potentiate therapy of pancreatic cancer

Pancreatic cancer is one of the deadly malignancies with a significant mortality rate and there are currently few therapeutic options for *** tumor microenvironment(TME)in pancreatic cancer,distinguished by fibrosis and the existence of cancer-associated fibroblasts(CAFs),exerts a pivotal influence on both tumor advancement and resistance to *** advancements in the field of engineered extracellular vesicles(EVs)offer novel avenues for targeted therapy in pancreatic *** study aimed to develop engineered EVs for the targeted reprogramming of CAFs and modulating the TME in pancreatic *** obtained from bone marrow mesenchymal stem cells(BMSCs)were loaded with miR-138-5p and the anti-fibrotic agent pirfenidone(PFD)and subjected to surface modification with integrinα5-targeting peptides(named IEVs-PFD/138)to reprogram CAFs and suppress their pro-tumorigenic ***α5-targeting peptide modification enhanced the CAF-targeting ability of ***-138-5p directly inhibited the formation of the FERMT2-TGFBR1 complex,inhibiting TGF-βsignaling pathway *** addition,miR-138-5p inhibited proline-mediated collagen synthesis by directly targeting the FERMT2-PYCR1 *** combination of miR-138-5p and PFD in EVs synergistically promoted CAF reprogramming and suppressed the pro-cancer effects of *** experiments using the orthotopic stroma-rich and patient-derived xenograft mouse models yielded promising *** particular,IEVs-PFD/138 effectively reprogrammed CAFs and remodeled TME,which resulted in decreased tumor pressure,enhanced gemcitabine perfusion,tumor hypoxia amelioration,and greater sensitivity of cancer cells to ***,the strategy developed in this study can improve chemotherapy *** IEVs-PFD/138 as a targeted therapeutic agent to modulate CAFs and the TME represents a promising therapeutic approach for pancreatic cancer.

Pervious concrete is a special type of concrete that is of high porosity and contains no or a small amount of fine aggregate,and it is an important basic material for sponge city *** presence of a large number of connected pores inside pervious concrete leads to a marked difference in durability failure mechanism compared with that of ordinary *** this study,the frost resistance and anti-clogging of pervious concrete were introduced in detail,and the methods to improve their performance were summarized *** cracking pattern of pervious concrete is influenced by geometric characteristics and three-dimensional morphological features of pores,resulting in its crack generation,development,and geometry being significantly different from those of ordinary concrete,thus leading to different freeze-thaw cycle *** addition,due to its different pore structure compared to ordinary concrete,three types of clogging mechanisms,affecting the long-term permeability of pervious concrete were elaborated systematically(i.e.,physical clogging,biological clogging,and chemical clogging).And the ways to improve the anti-clogging of pervious concrete are systematically presented from multiple ***,in order to broaden the engineering applications of pervious concrete,some research proposals are presented in this study.

Atomic-scale mechanism of effect of co-separation of elements at interface of ferrite and austenite on Cr-depleted zone of duplex stainless steel

Three-dimensional atom-probe tomography and first-principles calculation combined with density functional theory were used to study the effect of the co-segregation of different elements formed during the solidification process of S32205 duplex stainless steel on the Cr-depleted zone at the interface between ferrite and *** was found that the co-segregation of different elements formed during the solidification process of duplex stainless steel can also form Cr-depleted zone at the interface between ferrite and ***,Mo,Si,B,C and P atoms promote co-segregation with Cr atoms,which promotes the formation of Cr-depleted zone at the interface between ferrite and austenite in duplex stainless *** and Si strongly promote the segregation of Cr at the interface between ferrite and austenite,thereby promoting the formation of Cr-depleted zone.B,C and P elements also promote the segregation of Cr element at the interface between ferrite and austenite and the formation of Cr-depleted zone,but their effect is weaker than that of Mo and Si *** conclusions provide a new theoretical basis for improving the intergranular corrosion performance of duplex stainless steel.

Experimental study on secondary air mixing along the bed height in a circulating fluidized bed with a multitracer-gas method

A multitracer-gas method was proposed to study the secondary air(SA)mixing along the bed height in a circulating fluidized bed(CFB)using carbon monoxide(CO),oxygen(O_(2)),and carbon dioxide(CO_(2))as tracer *** were carried out on a cold CFB test rig with a cross-section of 0.42 m×0.73 m and a height of 5.50 *** effects of superficial velocity,SA ratio,bed inventory,and particle diameter on the SA mixing were *** results indicate that there are some differences in the measurement results obtained using different tracer gases,wherein the deviation between CO and CO_(2) ranges from 42%to 66%and that between O_(2) and CO_(2) ranges from 45%to 71%in the lower part of the fluidized ***,these differences became less pronounced as the bed height ***,the high solid concentration and fine particle diameter in the CFB may weaken the *** measurement results of different tracer gases show the same trends under the variation of operating *** superficial velocity and SA ratio and decreasing particle diameter result in better mixing of the *** effect of bed inventory on SA mixing is not monotonic.

Improving the accuracy of precipitation estimates in a typical inland arid area of China using a dynamic Bayesian model averaging approach

Oligomeric donor with appropriate crystallinity for organic solar cells

Improving the performance of all-small-molecule organic solar cells(ASM-OSCs)largely depends on the design and application of novel donors with appropriate *** molecular conjugation is an effective method for regulating molecular stacking and *** this work,we successfully designed and synthesized two novel acceptor-donor-donor-donor-acceptor(A-D-D-D-A)type oligomeric donors with three dithieno[2,3-d:2′,3′-d′]benzo[1,2-b:4,5-b']dithiophene(DTBDT)as the central unit,named as 3DTBDT-Cl and 3DTBDT,depending on with and without chlorine substitution on the thiophene side *** found that the introduction of chlorine atoms makes the blend films display stronger crystallinity but with large-scale phase separation morphology and more defects,which eventually leads to a power conversion efficiency(PCE)of only 10.83%,whereas the blend films based 3DTBDT with appropriate crystallinity achieved 13.74%*** with 3DTBDT-Cl/L8-BO,the 3DTBDT/L8-BO films exhibited a nanoscale bi-continuous interpenetrating network morphology with a smaller domain size and more suitable crystallinity,which guarantees the corresponding devices obtained more efficient exciton dissociation,efficient charge transport,reduced bimolecular recombination,and performed more balanced carrier *** results demonstrated that regulating the crystallinity of oligomeric donors to obtain the desired phase separation morphology in the blend films could facilitate further improving the performance of ASM-OSCs.

Distributed Stochastic Scheduling of Massive Backup Batteries in Cellular Networks for Operational Reserve and Frequency Support Ancillary Services

Base station(BS)backup batteries(BSBBs),with their dispatchable capacity,are potential demand-side resources for future power *** enhance the power supply reliability and post-contingency frequency security of power systems,we propose a two-stage stochastic unit commitment(UC)model incorporating operational reserve and post-contingency frequency support provisions from massive BSBBs in cellular networks,in which the minimum backup energy demand is considered to ensure BS power supply *** energy,operational reserve,and frequency support ancillary services are co-optimized to handle the power balance and post-contingency frequency security in both forecasted and stochastic variable renewable energy(VRE)***,we propose a dedicated and scalable distributed optimization framework to enable autonomous optimizations for both dispatching center(DC)and *** BS model parameters are stored and processed locally,while only the values of BS decision variables are required to upload to DC under the proposed distributed optimization framework,which safeguards BS privacy *** studies on a modified IEEE 14-bus system demonstrate the effectiveness of the proposed method in promoting VRE accommodation,ensuring post-contingency frequency security,enhancing operational economics,and fully utilizing BSBBs'energy and power ***,the proposed distributed optimization framework has been validated to converge to a feasible solution with near-optimal performance within limited ***,numerical results on the Guangdong 500 kV provincial power system in China verify the scalability and practicality of the proposed distributed optimization framework.

A modified 3D mean strain energy density criterion for predicting shale mixed-mode Ⅰ/Ⅲ fracture toughness

The fracture toughness of rocks is a critical fracturing parameter in geo-energy exploitation playing a significant role in fracture mechanics and hydraulic *** edge-notched disk bending(ENDB)specimens are employed to measure the entire range of mixed-modeⅠ/Ⅲfracture toughness of Longmaxi *** theoretically interpret the fracture mechanisms,this research first introduces the detailed derivations of three established fracture *** distinguishing the volumetric and distortional strain energy densities,an improved three-dimensional mean strain energy density(MSED)criterion is *** the critical volumetric to distortional MSED ratio decreases,the transition from tensiondominated fracture to shear-dominated fracture is *** results indicate that both peak load and applied energy increase significantly with the transition from pure mode I(i.e.,tension)to pure modeⅢ(i.e.,torsion or tearing)since mode-Ⅲcracking happens in a twisted manner and mode-Ⅰcracking occurs in a coplanar *** macroscopic fracture signatures are consistent with those of triaxial hydraulic *** average ratio of pure mode-Ⅲfracture toughness to pure mode-Ⅰfracture toughness is 0.68,indicating that the obtained mode-Ⅲfracture resistance for a tensionbased loading system is apparent rather than *** to the three mainstream fracture criteria,the present fracture criterion exhibits greater competitiveness and can successfully evaluate and predict mixed-modeⅠ/Ⅲfracture toughness of distinct materials and loading methods.

Assessing the energy release characteristics during the middle detonation reaction stage of aluminized explosives

Afterburning behind the detonation front of an aluminized explosive releases energy on the millisecond timescale,which prolong the release of detonation energy and the energy release at different stages also shows significant ***,at present,there are few effective methods for evaluating the energy release characteristics of the middle reaction stage of such explosives,which can have a duration of tens to hundreds of *** present work demonstrates an approach to assessing the midstage of an aluminized explosive detonation based on a water push test employing a high degree of *** this method,the explosive is contained in a steel cylinder having one end closed that is installed at the bottom of a transparent water *** detonation,the gaseous products expand in one direction while forcing water ahead of *** resulting underwater shock wave and the interface between the gas phase products and the water are tracked using an ultra-high-speed framing and streak *** shock wave velocity in water and the expansion work performed by the gaseous detonation products were calculated to assess the energy release characteristics of aluminized explosives such as CL-20 and RDX in the middle stage of the detonation *** the middle stage of the detonation process of these aluminized explosives,the aluminum reaction reduced the attenuation of shock waves and increased the work performed by gas phase products.A higher aluminum content increased the energy output while the presence of oxidants slowed the energy release *** work demonstrates an effective means of evaluating the performance of aluminized explosives.

The quality factor(Q factor)is a crucial performance parameter for *** this paper,a novel release method for highquality micro fused silica shell resonators with teeth-like tines with good surface morphology is *** method is based on femtosecond laser-assisted chemical ***,the optimal energy range of femtosecond laser modification is obtained through mechanism ***,the optimal parameters for a straight line and arc pattern are determined by optimizing the average output power,processing speed,and processing *** results demonstrate why edge breakage in rounded corners is easy under different ***,according to these conclusions,the processing is performed on a micro fused silica shell resonator with a Q factor exceeding 6 *** addition,subsurface damage is rare throughout the fabrication process,and the surface roughness of the released cross section reaches the nanometer *** improved Q factor helps suppress mechanical thermal noise and reduce zero bias and zero bias drift,constituting the primary method for enhancing the performance of the resonant gyroscope.
