Multifunctional drug delivery systems(DDSs)have shown great prospects in overcoming the heterogeneous barrier of delivery drugs to the complex tumor microenvironment(TME).In this study,multifunctional AS/Ge-pNAB microgels with dual-active targeting,triple environment responsiveness,and fluorescence imaging capability were prepared through a straightforward procedure.This was aimed to improve the antitumor therapeutic application of gambogic acid(GA)based on the biological characteristics of TME.The microgels have a uniform double-layer structure with aptamer in the outer layer which helps in recognizing receptors on the tumor cells.The GA loaded nano-herb exhibited environment-responsive drug release profiles under acidic pH,reductant and high temperature.The nano-herb significantly improved the accumulation of GA in tumor sites through the synergistic combination of the enhanced permeability and retention effect and dual-ligand mediated internalization.Then,it accelerated intracellular drug release and killed tumor cells.Therefore,the nano-herb had specific therapeutic effects on the tumor in vitro and in vivo as they remarkably inhibited tumor growth while depicting optimal biosafety and lower levels of off-target toxicity.Overall,these findings demonstrate the great potential of the multifunctional AS/Ge-pNAB microgels for precisely targeted GA delivery and open a new avenue for the facile preparation of multifunctional DDSs.

The metal-based additive manufacturing(AM),also referred to as metal 3D printing,has drawn particular interest because it enables direct creation,aided by computationally-directed path design,of intricate components with site-specific compositions and geometrical requirements as well as low buy-to-fly ratios.During the last two decades,the objective of this revolutionary technology has been shifting from only“rapid prototyping”to advanced manufacturing of special high-end products or devices,which,in many aspects,outperform conventional manufacturing technologies.For fusion-based AM,significant progress has been achieved in understanding the processing window of macroscopic scales,non-equilibrium metallurgy of mesoscale scales,and grain evolution of microscopic scales.Although the versatile capacity of AM facilitates new avenues for discovering advanced materials and structures,their potential has still not been fully explored.Given the unique non-equilibrium solidification during the AM process,coarse columnar grains with strong textures are usually developed along the build direction,which downgrades the mechanical performance.To push the limits of this digital manufacturing,this review attempts to provide in-depth and comprehensive overviews of the recent progress in understanding the evolution and control of the as-built microstructure that has been made recently and the challenges encountered during the AM process.

Developing a localized and consistent model framework for climate loss and damage assessment is crucial for the policy-making of climate change mitigation and adaptation.This study introduces a comprehensive,multidisciplinary Integrated Assessment Model(IAM)framework for evaluating climate damage in China,utilizing BCC-SESM climate model and FUND sectoral climate damage model under the SSP2-RCPs scenario.Employing a bottom-up approach,the research estimates climate damage across eight major sectors,recalibrates sectoral climate damage functions and parameters for China,and elucidates distinctions among direct climate loss,market climate loss,and aggregate climate loss.The findings reveal that the total climate damage function for China follows a quadratic pattern in response to temperature rise.By 2050,the estimated climate damage is projected to be 5.4%,5.7%,and 8.2%of GDP under RCP2.6,RCP4.5,and RCP8.5,respectively.Additionally,both direct and market climate losses are projected to remain below 2%of GDP by 2050,while the aggregate climate loss could reach as high as 8.2%,which is predominantly attributed to non-market sectors.From a sectoral perspective,under the RCP8.5 scenario,human health damage constitutes the largest share(61.9%)of the total climate loss by 2050,followed by sea-level rise damage(18.6%).This study sheds lights on the adaptation policy that should attach importance to the non-market sectors,particularly focusing on human health and sea-level rise.

Lightweight aluminum(Al)alloys have been widely used in frontier fields like aerospace and automotive industries,which attracts great interest in additive manufacturing(AM)to process high-value Al parts.As a mainstream AM technique,laser-directed energy deposition(LDED)shows good scalability to meet the requirements for large-format component manufacturing and repair.However,LDED Al alloys are highly challenging due to their inherent poor printability(e.***.low laser absorption,high oxidation sensitivity and cracking tendency).To further promote the development of LDED high-performance Al alloys,this review offers a deep understanding of the challenges and strategies to improve printability in LDED Al alloys.The porosity,cracking,distortion,inclusions,element evaporation and resultant inferior mechanical properties(worse than laser powder bed fusion)are the key challenges in LDED Al alloys.Processing parameter optimizations,in-situ alloy design,reinforcing particle addition and field assistance are the efficient approaches to improving the printability and performance of LDED Al alloys.The underlying correlations between processes,alloy innovation,characteristic microstructures,and achievable performances in LDED Al alloys are discussed.The benchmark mechanical properties and primary strengthening mechanism of LDED Al alloys are summarized.This review aims to provide a critical and in-depth evaluation of current progress in LDED Al alloys.Future opportunities and perspectives in LDED high-performance Al alloys are also outlined.

在周代的礼乐文化中,诗歌、乐舞共同组成了保留、传播族群历史记忆的立体空间。雅乐概念的衍变标识着现实与历史的关联,其具体内涵展现了由仪式乐舞所建构的线性历史框架。其中的《大武》不仅是对周初历史的整体性和连续性表达,更揭示了仪式性活动与早期历史叙事的关联。周代乐师的“讽诵诗、奠世系”具有记忆族群世系、建构文化认同以及融合古今历史、填补记忆缺口的功能。随着礼乐制度崩坏,以及乐师主导的仪式性记忆模式向史官主导的文字性记忆模式的转变,原先附着于礼乐机制上的《诗》的乐义为后来孔子创作《春秋》所吸纳,这便是孟子所言“《诗》亡然后《春秋》作”的内涵。借助理解仪式性记忆与文字性记忆在早期文化发展中的关系,可以帮助我们重新看待此后学术发展中“口述”和“书写”两种知识传播途径,以及经学阐释在“重义”和“重事”上的两种偏好。

在我国,肝胆管结石病是临床常见病,肝胆管结石的高残石率、高复发率、高再手术率迄今仍是胆道外科的难题。因为肝胆管结石病多合并肝胆管狭窄、反复发作的胆管炎,晚期发展至肝硬化门静脉高压症,且是发生胆管癌的重要因素,诊断和治疗均非常困难。虽然规则性肝段、肝叶切除是治疗肝胆管结石病的首选方法,近年来,肝胆管结石病微创治疗领域已取得一系列进展和突破,但仍有部分肝胆管结石难以通过手术完全去除。涉及腹腔镜、机器人手术系统、内镜等众多微创技术,其中内镜技术同样发挥了重要的作用,弥补了单纯手术治疗的不足,为不能手术肝切除或多次手术后、合并肝硬化的残留和(或)复发性结石病人的治疗带来更多的选择。内镜技术的广泛应用为肝胆管结石病的治疗注入了新的活力,使得肝胆管结石的治疗方案更为多元化。

根据体外生命支持系统(ECLS)注册报告,体外膜肺氧合(ECMO)在新生儿严重呼吸衰竭支持的应用在全球于1992年达到顶峰。随着一氧化氮、肺表面活性物质的应用,以及高频通气等新型呼吸支持手段的出现,采用ECMO支持新生儿呼吸的病例数量逐渐减少,维持在约1000例/年。但这意味着需要ECMO支持的新生儿病情将会更重,对临床医师的要求更高。为了进一步规范新生儿体外生命支持技术,体外生命支持组织在2020年发布了新生儿呼吸衰竭的指南。指南对ECMO期间的技术、患儿管理及并发症处理等提出了建议,本文主要从患儿选择、支持模式、技术考虑、ECMO期间的管理及并发症处理等方面进行解读。

通过回顾2021年3月3日解放军总医院第八医学中心宏基因组学第二代测序技术(mNGS)检测到脑脊液EBV阳性合并干燥综合征的初发鼻咽癌患者1例的诊治过程,检索国内外数据库有关脑脊液EBV阳性的脑炎或脑膜炎、鼻咽癌相关报道,对所有患者的一般资料、首发症状、治疗及预后进行分析。脑脊液EBV阳性与自身免疫病、病毒性脑炎或脑膜炎、鼻咽癌相关,合并干燥综合征少见;脑脊液EBV阳性多在复发鼻咽癌患者中出现,初发鼻咽癌患者脑脊液EBV阳性鲜见报道。对于具有免疫背景、临床以头痛或颅神经受累为主要表现,或者疑诊鼻咽癌的患者,宜尽早行脑脊液mNGS或EBV DNA检测以评估危险分层,判断预后,制定个性化的治疗方案。

One-sixth of the currently known natural products containα,β-unsaturated carbonyl groups.Our previous studies reported a rare C-sulfonate metabolic pathway.Sulfonate groups were linked to theβ-carbon ofα,β-unsaturated carbonyl-based natural compounds through this pathway.However,the mechanism of this type of metabolism is still not fully understood,especially whether it is formed through enzyme-mediated biotransformation or direct sulfite addition.In this work,the enzyme-mediated and non-enzymatic pathways were studied.First,the sulfite content in rat intestine was determined by LC-MS/MS.The results showed that the amount of sulfite in rat intestinal contents was from 41.5 to 383μg·g^(-1),whereas the amount of sulfite in rat feed was lower than the lower limit of quantitation(20μg·g^(-1)).Second,the reaction kinetics of sulfite-andrographolide reactions in phosphate buffer solutions(pH 6-8)was studied.The half-lives of andrographolide ranged from minutes to hours.This was suggested that the C-sulfonate reaction of andrographolide was very fast.Third,the C-sulfonate metabolites of andrographolide were both detected when andrographolide and L-cysteine-S-conjugate andrographolide were incubated with the rat small intestine contents or sulfite,indicating that the sulfite amount in rat intestine contents was high enough to react with andrographolide,which assisted a significant portion of andrographolide metabolism.Finally,the comparison of andrographolide metabolite profiles among liver homogenate(with NADPH),liver S9(with NADPH),small intestine contents homogenate(with no NADPH),and sulfite solution incubations showed that the C-sulfonate metabolites were predominantly generated in the intestinal tract by non-enzymatic pathway.In summary,sulfite can serve as a substrate for C-sulfonate metabolism,and these results identified non-enzymatically nucleophilic addition as the potential mechanism for C-sulfonate metabolism of compounds containingα,β-unsaturated carbonyl moiety.
