The therapeutic efficiency of sonodynamic therapy(SDT)mainly depends on the presence of oxygen(O_(2))to generate harmful reactive oxygen species(ROS);thus,the hypoxic tumor microenvironment significantly limits the efficacy of SDT.Therefore,the development of oxygen-independent free radical generators and associated combination therapy tactics can be a promising field to facilitate the anticancer capability of SDT.In this study,a biomimetic drug delivery system(C-TiO_(2)/AIPH@PM)composed of an alkyl-radical generator(2,2′-azobis[2-(2-imidazolin-2-yl)propane]dihydrochloride,AIPH)-loaded C-TiO_(2) hollow nanoshells(HNSs)as the inner cores,and a platelet membrane(PM)as the outer shells is successfully prepared for synergistic SDT and oxygen-independent alkyl-radical therapy.The PM encapsulation can significantly prolong the blood circulation time of CTiO_(2)/AIPH@PM compared with C-TiO_(2)/AIPH while enabling C-TiO_(2)/AIPH@PM to achieve tumor targeting.C-TiO_(2)/AIPH@PM can efficiently produce ROS and alkyl radicals,which can achieve a more thorough tumor eradication regardless of the normoxic or hypoxic conditions.Furthermore,the generation of these radicals improves the efficiency of SDT.In addition,nitrogen(N_(2))produced due to the decomposition of AIPH enhances the acoustic cavitation effect and lowers the cavitation threshold,thereby enhancing the penetration of CTiO_(2)/AIPH@PM at the tumor sites.Both in vitro and in vivo experiments demonstrate that CTiO_(2)/AIPH@PM possesses good biosafety,ultrasound imaging performance,and excellent anticancer efficacy.This study provides a new strategy to achieve oxygen-independent free radical production and enhance therapeutic efficacy by combining SDT and free radical therapy.

Inhibitory Activity of Honeysuckle Extracts against Influenza A Virus In Vitro and In Vivo

Honeysuckle has been used in the treatment of influenza virus infection for thousands of years in China.However,its main active components and the functional mechanisms remain to be elucidated.Here,four honeysuckle extracts,including acids extract,flavonoids extract,total extract and acids-flavonoids mixture,were prepared to clarify the main active antiviral components.The cytopathic effect reduction assay showed that all the four extracts inhibited the replication of influenza viruses H1N1,H3N2 and the oseltamivir-resistant mutant strain H1N1-H275Y.The acids-flavonoids mixture had the strongest inhibitory effects in vitro with EC_(50) values of 3.8,4.1,and >20 lg/m L against H1N1,H3N2 and H1N1-H275Y,respectively,showing competitive antiviral activity with oseltamivir and ribavirin.Honeysuckle acids extract also showed the most significant antiviral activity in vivo.Oral administration of the acids extract at a dosage of 600 mg/kg/d effectively alleviated viral pneumonia,maintained body weight and improved the survival rate to 30% of the mice infected with a lethal dose of H1N1.The results of time-of-drug addition experiment and neuraminidase (NA) inhibition assay showed that honeysuckle extracts had a broad-spectrum inhibitory effect against influenza virus NAs.The flavonoid extract showed the strongest inhibitory effect on the NA of influenza virus H7N9 with an IC_(50) of 24.7 lg/m L.These results suggested that these extracts might exert their antiviral activity by suppressing the release of influenza viruses.Briefly,our findings demonstrate that acids and flavonoids extracts of honeysuckle are the major antiviral active components,and the acids extract has the potential to be developed into an antiviral agent against influenza virus,especially for oseltamivirresistant viruses.

The integrity of lysosomes is of vital importance to survival of tumor cells.We demonstrated that LW-218,a synthetic flavonoid,induced rapid lysosomal enlargement accompanied with lysosomal membrane permeabilization in hematological malignancy.LW-218-induced lysosomal damage and lysosome-dependent cell death were mediated by cathepsin D,as the lysosomal damage and cell apoptosis could be suppressed by depletion of cathepsin D or lysosome alkalization agents,which can alter the activity of cathepsins.Lysophagy,was initiated for cell self-rescue after LW-218 treatment and correlated with calcium release and nuclei translocation of transcription factor EB.LW-218 treatment enhanced the expression of autophagy-related genes which could be inhibited by intracellular calcium chelator.Sustained exposure to LW-218 exhausted the lysosomal capacity so as to repress the normal autophagy.LW-218-induced enlargement and damage of lysosomes were triggered by abnormal cholesterol deposition on lysosome membrane which caused by interaction between LW-218 and NPC intracellular cholesterol transporter 1.Moreover,LW-218 inhibited the leukemia cell growth in vivo.Thus,the necessary impact of integral lysosomal function in cell rescue and death were illustrated.

Naturally occurring molecules derived from higher plants, animals, microorganisms and minerals play an important role in the discovery and development of novel therapeutic agents. The identification of molecular targets is of interest to elucidate the mode of action of these compounds, and it may be employed to set up target-based assays and allow structure-activity relationship studies to guide medicinal chemistry efforts toward lead optimization. In recent years, plant-derived natural compounds possessing potential anti-tumor activities have been garnering much interest and efforts are underway to identify their molecular targets. Here, we attempt to summarize the discoveries of several natural compounds with activities against hematological malignancies, such as adenanthin, oridonin, gambogic acid and wogonoside, the identification of their targets, and their modes of actions.

In order to reduce cost and complexity,fiber-wireless(FiWi) networks emerge,combining the huge amount of available bandwidth of fiber networks and the flexibility,mobility of wireless networks.However,there is still a long way to go before taking fiber and wireless systems as fully integrated networks.In this paper,we propose a network visualization based seamless networking scheme for FiWi networks,including hierarchical model,service model,service implementation and dynamic bandwidth assignment(DBA).Then,we evaluate the performance changes after network virtualization is introduced.Throughput for nodes,bandwidth for links and overheads leaded by network virtualization are analyzed.The performance of our proposed networking scheme is evaluated by simulation and real implementations,respectively.The results show that,compared to traditional networking scheme,our scheme has a better performance.

Burr formation mechanism and morphological transformation critical conditions in grinding of nickel-based superalloy honeycomb cores

Nickel-based Superalloy Honeycomb(NBSH) is widely used in aerospace field due to the high temperature resistance, high specific strength and light weight. However, the NBSH is extremely prone to form burrs during processing. The unreasonable burr morphology causes insufficient welding strength or false welding between NBSH and skin, resulting in premature failure of the parts. Hence, the burr formation mechanism and morphological transformation critical conditions in grinding NBSH were investigated based on finite element method(FEM) and experiments. Furthermore, the orthogonal experiments were carried out to investigate the effects of grinding parameters on burr and obtain the suitable parameter intervals to restrain the flash burr formation. The results indicate that due to the absence of supporting material, the extruded material of edge transforms into the Fourth Deformation Zone(FDZ). The FDZ rotates a certain angle due to the extrusion of abrasive particle, which leads to the fact that the FDZ cannot be removed and remains on the honeycomb wall, thus forming micro burr. If the material removal per unit time is large, the chip is not removed from FDZ. The parts that should have been chips are bonded with micro burr to form a flash burr. To restrain the flash burr formation, the suitable intervals of grinding depth,grinding speed and feed speed are 5–15 μm, 60–70 m/min and 500–1000 mm/min, respectively.

Among various environmental problems,water pollution has drawn more and more attention.To develop an adsorbent with enhanced selectivity and high capacity for As(V)oxyanion,a novel lignin-based adsorbent was prepared by doping with N and modifying with Mg^(2+),using thiethylenetetramine(TETA)and MgCl_(2) as the source of N and Mg^(2+),respectively.N-doped lignin(NL)shows a high selectivity for As(Ⅴ),which is further enhanced by modifying with Mg^(2+)(MNL).MNL shows an excellent reusability.The adsorption isotherms and kinetics of MNL for As(Ⅴ)follow Langmuir isotherms model and pseudo-second-order kinetics model,respectively.The adsorption of MNL for As(Ⅴ)achieved a maximum adsorption capacity of 687.46 mg g^(−1) and the equilibrium was established within 30 min.This study offers a novel idea on the recycle of the waste of paper industry and provides a new technology for As(Ⅴ)adsorption from wastewater.

为了系统回顾我国高校实验室研究进展,以中国知网期刊数据库为来源,收集截至2022年4月发表的1316篇相关文献,利用CiteSpace和VOSviewer软件进行文献计量分析。结果显示,该领域发文量总体呈上升趋势。目前,该领域高产机构以浙江大学、上海交通大学和清华大学等为主,高产作者如冯建跃、彭华松等与高产机构相辅相成,其相互之间合作紧密。发文期刊主要为高校主办的专业刊物、学报或其它教育类期刊,其中《实验技术与管理》《实验室研究与探索》两本期刊影响力较大。实验室队伍建设、实验室开放、教学和科研实验室管理、高校实验室信息化等研究越来越重视。随着疫情新形势出现、信息化及双一流建设出台,高校实验室安全、管理和建设越来越注重体系构建。
