First Report of Botryosphaeria dothidea Causing Sweet Osmanthus Leaf Dieback in China

Sweet osmanthus is one of the ten traditional famous flowers in *** occurrence of the diseases caused by fungi other than Botryosphaeria *** been reported mainly from China on sweet osmanthus.A leaf dieback of sweet osmanthus caused by Botryosphaeria *** found for the first time in 2007 in Nanning City,Guangxi,*** objectives of the present study were to isolate and characterize the causal organism of sweet osmanthus leaf *** fungus was isolated from the lesions of affected sweet osmanthus leaves and its pathogenicity to sweet osmanthus was confirmed using a detached-leaf-inoculation *** identification of the pathogen was carried out mainly based on the morphological characters and molecular analysis of internal transcribed spacer （ITS） *** morphological characters of the pathogenic isolate GHX6 were agreed with that of Botryosphaeria *** ITS sequence of the isolate was amplified with primers ITS1 and ITS4,and submitted to GenBank （accession ***368251）.Molecular analysis based on ITS sequence comparison between the isolate GHX6 and the other related fungi derived from GenBank supported that the causal agent of the sweet osmanthus leaf dieback belonged to Botryosphaeria *** is the first report of Botryosphaeria dothidea causing leaf dieback on sweet osmanthus in China.

Sensitivities of Phytophthora infestans to Metalaxyl, Cymoxanil, and Dimethomorph

The isolates of Phytophthora infestans on tomato in the Guangxi Zhuang Autonomous Region, China, were determined for the sensitivities to metalaxyl, cymoxanil and dimethomorph to give the basic information for integrating disease management. Sensitivities were tested by measuring the radial growth on agar medium amended with fungicide, compared with the floating-leaf-disk method. 239 isolates were collected from eight tomato growing areas during 2000-2006. The testing results indicated that the frequencies of sensitive, intermediate, and resistant isolates to metalaxyl were 42.26, 35.98, and 23.53%, respectively. Variations in sensitivities amongst isolates from different areas or different years were very high for metalaxyl. All isolates from Tianlin and Wuxuan were sensitive to metalaxyl, but the metalaxyl-resistant isolates predominated in Tianyang, with the frequency of 51.35%. The EC50 values of certain isolates from Tianyang were higher than 500 μg·mL^-1 and their resistance levels were over 100 000 folds. Cymoxanil has been used for nearly 10 years in Guangxi, and dimethomorph has been used for 5-6 years. However, there was no decrease in sensitivity of P. infestans populations and the sensitivities of the pathogen were nearly normally distributed. Hence, their mean ECs0 value [cymoxanil （0.1647 ±0.0255） μg·mL^-1, dimethomorph （0.0970 ± 0.0052） μg·mL^-1 could be used as the baseline sensitivities for monitoring the field resistance development. The comparison with the floating-leaf-disk method indicates that both the techniques provided equivalent results. These studies suggested that metalaxyl can be continuously applied in Tianlin, Wuxuan, and Nanning due to the resistant isolates that have not been found, while for those areas with resistant isolate, the use of metalaxyl should be reduced or alternated, and cymoxanil or dimethomorph was recommended for controlling late blight disease of tomato.

Gorham-Stout disease(GSD)is a sporadic chronic disease characterized by progressive bone dissolution,absorption,and disappearance along with lymphatic vessel infiltration in bone-marrow *** the osteolytic mechanism of GSD has been widely studied,the cause of lymphatic hyperplasia in GSD is rarely *** this study,by comparing the RNA expression profile of osteoclasts(OCs)with that of OC precursors(OCPs)by RNA sequencing,we identified a new factor,semaphorin 3A(Sema3A),which is an osteoprotective factor involved in the lymphatic expansion of *** to OCPs,OCs enhanced the growth,migration,and tube formation of lymphatic endothelial cells(LECs),in which the expression of Sema3A is low compared to that in *** the presence of recombinant Sema3A,the growth,migration,and tube formation of LECs were inhibited,further confirming the inhibitory effect of Sema3A on LECs in *** an LEC-induced GSD mouse model,the effect of Sema3A was examined by injecting lentivirus-expressing Sema3A into the tibiae in *** found that the overexpression of Sema3A in tibiae suppressed the expansion of LECs and alleviated bone loss,whereas the injection of lentivirus expressing Sema3A short hairpin RNA(shRNA)into the tibiae caused GSD-like *** staining further demonstrated that OCs decreased and osteocalcin increased after Sema3A lentiviral treatment,compared with the *** on the above results,we propose that reduced Sema3A in OCs is one of the mechanisms contributing to the pathogeneses of GSD and that expressing Sema3A represents a new approach for the treatment of GSD.

Chain Extender-induced Hydrogen Bonding Organization Determines the Morphology and Properties of Thermoplastic Polycarbonate Polyurethane

Thermoplastic polycarbonate polyurethanes(PCUs) are multiblock copolymers that have been applied for medical devices for long time. Aliphatic diols are common chain extenders(CE) involved in the composition of the hard segments of PCU. However, limited knowledge was discovered about how the chemical structure of CE affects the hydrogen bonding organization within PCUs and their mechanical *** investigate this problem, a group of PCUs were synthesized respectively by extending the polymer chain with 1,4-butanediol(BDO),aminoethanol(MEA), ethanediol(EO) as three kinds of chain extenders. Tiny differences in the CE chemical structure results in remarkable variations in phase separation, condensed morphologies, thermal and mechanical properties, which are characterized by Fourier transform infrared spectrometer, atomic force microscopy, small-angle X-ray scattering, differential scanning calorimetry, and tensile tests. The microstructural evolution during unilateral deformation and the different mechanism induced by the different CEs was probed and unveil by in situ wide-and small-angle X-ray diffraction. Symmetry of CE can improve the organization of the hydrogen bonding. The coherence strength of the urethane/urea group also plays a key role by comparing the two PCUs with ethanediol and aminoethanol.

Perfluorooctane sulfonate promotes the migration of colorectal cancer cells by inducing epithelial-mesenchymal transition

The potential association between colorectal cancer(CRC)and environmental pollutants is *** studies have found that some perfluoroalkyl acids,including perfluorooctane sulfonate(PFOS),induced colorectal tumors in experimental animals and promoted the migration of and invasion by CRC cells in vitro,but the underlying mechanism is ***,we investigated the effects of PFOS on the proliferation and migration of CRC cells and the potentialmechanisms involving activating the PI3K/Akt-NF-κB signal pathway and epithelial-mesenchymal transition(EMT).It was found that PFOS promoted the growth andmigration of HCT116 cells at non-cytotoxic concentrations and increased the mRNA expression of the migration-related angiogenic cytokines vascular endothelial growth factor(VEGF)and interleukin-8(IL-8).In a mechanistic investigation,the up-stream signal pathway PI3K/Akt-NF-κB was activated by PFOS,and the process was suppressed by LY294002(PI3K/Akt inhibitor)and BAY11-7082(NF-κB inhibitor)respectively,leading to less proliferation of HCT116 ***,matrix metalloproteinases(MMP)and EMT-related markers were up-regulated after PFOS exposure,and were also suppressed respectively by LY294002 and ***,the up-regulation of EMT markers was suppressed by a MMP inhibitor *** together,our results indicated that PFOS promotes colorectal cancer cell migration and proliferation by activating the PI3K/Akt-NF-κB signal pathway and epithelial-mesenchymal *** could be a potential toxicological mechanism of PFOS-induced malignant development of colorectal cancer.

Excess pore pressure behavior and evolution in deep coalbed methane reservoirs

An efficient data-driven global sensitivity analysis method of shale gas production through convolutional neural network

The shale gas development process is complex in terms of its flow mechanisms and the accuracy of the production forecasting is influenced by geological parameters and engineering parameters. Therefore, to quantitatively evaluate the relative importance of model parameters on the production forecasting performance, sensitivity analysis of parameters is required. The parameters are ranked according to the sensitivity coefficients for the subsequent optimization scheme design. A data-driven global sensitivity analysis(GSA) method using convolutional neural networks(CNN) is proposed to identify the influencing parameters in shale gas production. The CNN is trained on a large dataset, validated against numerical simulations, and utilized as a surrogate model for efficient sensitivity analysis. Our approach integrates CNN with the Sobol' global sensitivity analysis method, presenting three key scenarios for sensitivity analysis: analysis of the production stage as a whole, analysis by fixed time intervals, and analysis by declining rate. The findings underscore the predominant influence of reservoir thickness and well length on shale gas production. Furthermore, the temporal sensitivity analysis reveals the dynamic shifts in parameter importance across the distinct production stages.
