Male germ cells employ specific metabolic pathways throughout their developmental *** a previous study,we discovered heightened expression of pyruvate kinase M(PKM),a pivotal glycolytic enzyme,in spermatogonia and *** gain deeper insights into PKM's roles in spermatogenesis,sperm function,and male fertility,we engineered a conditional-knockout mouse model(Pkm-vkO mice)to selectively disrupt the Pkm gene within germ *** maintaining regular testicular histology and sperm morphology,the male Pkm-vko mice were infertility,characterized by significant impairments in sperm motility and adenosine triphosphate(ATP)*** addition,Pkm-null spermatozoa exhibited similar deficits in protein tyrosine phosphorylation linked to capacitation,as well as compromised performance in in vitro fertilization *** conclude,PKM's presence is not obligatory for the entirety of spermatogenesis in male germ cells;however,it emerges as a critical factor influencing sperm motility and overall male fertility.

The northeastern Sichuan area in the northern Yangtze margin has unique Ediacaran geological records,especially the Doushantuo Formation(DST),and become a hot research area in recent ***,the Cryogenian-Ediacaran(C-E)boundary has not been precisely identified,which restricts the in-depth study of geological information during this crucial transitional period and is unfavorable for a systematic and complete understanding of the Yangtze Block and even the global paleogeographic *** study conducted stratigraphy,sedimentology,and chronostratigraphy to establish the stratigraphic framework and sedimentary evolution of the C-E transition strata in northeastern *** results showed that the Ediacaran sediments,without the cap dolomite,unconformably overlaid the Cryogenian sediments in the studied *** Member II of the DST,characterized by 50-160 m of red-green sandstone(approximately equivalent to the original Chengkou“Guanyinya Formation”),directly overlaid the Cryogenian sediments and displayed a 623±2.3 Ma maximum depositional age from the detrital zircon U-Pb *** stratigraphic correlations indicate that the C-E transition strata in northeastern Sichuan had a consistent lithological association and sedimentary sequence characteristics but differed from the Three ***,the upper Nantuo massive glacial diamictites transition to the icebergs rafted lonestone-bearing mudstones at the top,then change upward to DST barrier coast *** proposed DST of the northeastern Sichuan Basin was divided into three lithostratigraphic members without the regional Member I cap dolomite:(i)Member II purple-red,gray-green sandstone strata,(ii)MemberⅢblack mudstone strata,and(iii)Member IV P-Mn bearing *** the C-E transition,the study area experienced(i)the global deglaciation stage in the terminal Marinoan glaciation and(ii)the filling-leveling up stage with clastic rocks in the early ***,the early Ediacaran

采用热压烧结工艺制备了不同Cr_(3)C_(2)含量的石墨/Cr_(3)C_(2)共增强镍基复合材料,探究了不同Cr_(3)C_(2)含量的复合材料的微观结构、力学性能和摩擦性能。结果表明:热压烧结制备的镍基复合材料致密无孔隙,Cr_(3)C_(2)和石墨均匀分布于基体内部,异质界面结合良好。随着Cr_(3)C_(2)含量的增加,复合材料的割裂程度增加,硬度增加,但强度降低,断裂机制为脆性断裂。摩擦过程中,石墨具有良好的润滑作用,但Cr_(3)C_(2)造成磨粒磨损,且随着Cr_(3)C_(2)含量的升高,复合材料的摩擦系数和磨损率显著升高。

The lightweight shielding design of small reactors is a popular research *** on a small helium-xenon-cooled solid reactor,the effects of neutron and photon shielding sequence and the number of shielding layers on the radiation dose were first *** was found that when photons were shielded first and the number of shielding layers was odd,the radiation dose could be significantly *** reduce the weight of the shielding body,the relative thickness of the shielding layers was optimized using the genetic *** optimized scheme can reduce the radiation dose by up to 57%and reduce the weight by 11.84%.To determine the total thickness of the shielding layers and avoid the local optimal solution of the genetic algorithm,a series of formulas that describes the relationship between the total thickness and the radiation dose was developed through large-scale calculations.A semi-empirical and semi-quantitative lightweight shielding design method is proposed to integrate the above shielding optimization method that verified by the Monte Carlo ***,a code,SDIC1.0,was developed to achieve the optimized lightweight shielding design for small *** was verified that the difference between the SDIC1.0 and the RMC code is approximately 10%and that the computation time is shortened by 6.3 times.

With increasing demand for irrigation water, agricultural scientists and planners pay more attention to the utilization of diluted seawater as an alternative source for irrigation of crops. A greenhouse pot experiment was conducted to study how seawater stress(SS) affects growth, water content, cationic accumulation, and active ingredients in leaves of Aloe vera after 30 d of growth in nutrient media with 0%(control), 22%(22% SS), and 42%(42% SS) seawater stress. Results indicated the SS did not change dry biomass of leaves and stems, but gradually decreased biomass allocation to roots with increasing seawater stress. Na+and Cl-in A. vera plant did not increase obviously with a big increase in seawater percentage due to low transpiration of Aloe vera. 42% SS decreased N concentration in most plant organs, but did not change or increased P concentration. Seawater stress tended to decrease concentrations of K+and Ca2+in A. vera. However, seawater salinity tended to increase the concentrations of aloin concentration in top(young) and middle leaves, and there was no significant effect of both stresses on aloin concentration in base(old) leaves. The 42% SS treatment decreased polysaccharide concentrations only in the base leaves, but not in top and middle leaves. In summary, supplying suitably diluted seawater for 30 d could increase the qualities and value of A. vera, without substantial effects on shoot dry biomass production.

Hard carbons are widely investigated as potential anodes for lithium and sodium ion batteries owing to their internally well-tailored textures(closed pores and defects) and large microcrystalline interlayer spacing. The renewable biomass is a green and economically attractive carbon source to produce hard carbons. However, the chemical and structural complexity of biomass has plagued the understanding of evolution mechanism from organic precursors to hard carbons and the structure-property *** makes it difficult to finely tune the microstructure of biomass-derived hard carbons, thus greatly restricting their high-performance applications. Most recently, the optimal utilization and controllable conversion of biomass-derived biopolymers(such as starch, cellulose and lignin) at the molecular level have become a burgeoning area of research to develop hard carbons for advanced *** the principal source of carbonaceous materials is from biomass pyrolysis, we firstly overview the chemical structures and pyrolysis behaviors of three main biopolymers. Then, the controllable preparation of hard carbons using various physicochemical properties of biopolymers at the molecular level is systematically discussed. Furthermore, we highlight present challenges and further opportunities in this field. The Review will guide future research works on the design of sustainable hard carbons and the optimization of battery performance.

The kinetics of free radical decay in the polymerization of MMA initiated by AIBN was studied by means of ESR spectroscopy. It was found that the curves of radical decay are strongly associated with the reaction temperature, the initiator concentration and the solvent. In the case of the radical polymerization carried out at high temperature or in solution, the radical concentration first reached a maximum, then declined monotonously with reaction time. It was also found that the greater the amount of initiator or the higher the temperature, the more rapidly the radicals decay. When the bulk polymerization was implemented at a relatively low temperature, the curves of radical decay became more complicated, i.e., the radical concentration rapidly rose to a maximum, then dropped to a minimum, finally increased again with reaction time. Taking into account the diffusion effect, a semi-empirical equation is suggested to describe the kinetics of propagating radical decay.

Characterization and Differentiation into Adipocytes and Myocytes of Porcine Bone Marrow Mesenchymal Stem Cells

Bone marrow mesenchymal stem cells （BMSCs） could differentiate into various cell types including adipocytes and myocytes, which had important scientific significance not only in the field of tissue regeneration, but also in the field of agricultural science. In an attempt to exhibit the characterization and differentiation into adipocytes and myocytes of porcine BMSCs, we isolated and purified porcine BMSCs by red blood cell lysis method and percoll gradient centrifugation. The purified cells presented a stretched fibroblast-like phenotype when adhered to the culture plate. The results of flow cytometry analysis and immunofluorescence staining demonstrated that the isolated cells were positive for mesenchymal surface markers CD29, CD44 and negative for hematopoietic markers CD45 and the adhesion molecules CD31. Cells were induced to differentiate into adipocytes with adipogenic medium containing insulin, dexamethasone, oleate and octanoate. Oil Red O staining demonstrated that the porcine BMSCs successfully differentiated to adipocytes. Moreover, the findings of real-time PCR and Western blotting indicated that the induced cells expressed adipogenic marker genes （PPAR-y, C/EBP-c~, perilipin, aP2） mRNA or proteins （PPAR-3,, perilipin, aP2）. On the other hand, porcine BMSCs were induced into myoctyes with myogenic medium supplemented with 5-azacytidine, basic fibroblast growth factor, chick embryo extract and horse serum. Morphological observation by hochest 33342 staining showed that the induced cells presented as multi-nucleus muscular tube structure. And myogenic marker genes （Myf5, desmin） mRNA or proteins （MyfS, MyoD, myogenin, desmin） were found in the induced cells. In addition, the results of immunofluorescence staining revealed that myogenic marker （Myf5, MyoD, myogenin, desmin, S-MyHC） proteins was positive in the induced cells. Above all, these results suggested that the isolated porcine BMSCs were not only consistent with the characterization of mesenchyma
