Modularized construction is a new type of prefabricated building system with green environmental protection and excellent performance. There are few studies on the seismic performance of its key connection joint. This paper presents a new type of assembled connection joint for the high-rise modularized construction. Cyclic shear tests of full-scale joints were carried out, and the key indexes of their seismic performances including the hysteretic performance, ductility, and energy dissipation capacity were analyzed and obtained. The results show that the hysteresis loops of longitudinal and lateral cyclic shear tests were both plump in shapes. The ductility coefficients were 4.54 and 4.98, and the energy dissipation coefficients were 1.83 and 1.43, respectively. The test joint had good ductility and energy dissipation capacity. The positions of yield failure of specimens were mainly concentrated in the connection areas between the column and short beam or end-plate. The research can provide the technical reference for the seismic design and engineering application of related modularized constructions.

Background and Aims:With an increasing understanding of hepatitis B,the antiviral indications have been broadening *** evaluate the effectiveness of tenofovir alafena-mide(TAF)in chronic hepatitis B(CHB)patients with normal alanine aminotransferase(ALT)and detectable hepatitis B virus(HBV)DNA,those who are ineligible for broader anti-viral criteria from the Chinese CHB prevention guide(2019).Methods:A total of 117 patients were recruited and their data were collected from paper or electronic medical *** DNA and liver function were measured at baseline and throughout the 24-week *** effectiveness end-point was complete virological *** safety endpoint was the first occurrence of any clinical adverse event during the ***:Among the 117 patients,45 had normal ALT as well as detectable HBV DNA and they were not recommended for antiviral therapy according to Chinese Guidelines(2019).After TAF antiviral therapy,the rates of patients who achieved HBV DNAdetectable rates of HBV DNA in patients with low baseline viral load at 4,12 and 24 weeks were 92.3%,100%and 100%,while the rates of those with high baseline viral load were 68.2%,94.1%and 94.4%.Compared with 71.4%,94.4%and 94.7%in the high baseline group,the undetectable rates of HBV DNA at 4,12 and 24 weeks in the low baseline liver stiffness group were 85.7%,100%and 100%.There was no statistical significance among the above ***:CHB patients who had normal ALT and detectable HBV DNA and did not meet“CHB prevention guide(2019)”,could achieve complete virological response in 24 weeks after antiviral treatment by TAF.

AIM: To investigate the molecular mechanism for regulation of cholesterol metabolism by hepatitis C virus(HCV) core protein in Hep G2 ***: HCV genotype 1b core protein was cloned and expressed in Hep G2 cells. The cholesterol content was determined after transfection. The expression of sterol regulatory element binding protein 2(SREBP2) and the rate-limiting enzyme in cholesterol synthesis(HMGCR) was measured by quantitative real-time PCR and immunoblotting after transfection. The effects of core protein on the SREBP2 promoter and 3'-untranslated region were analyzed by luciferase assay. We used different target predictive algorithms, micro RNA(mi RNA) mimics/inhibitors, and site-directed mutation to identify a putative target of a particular mi ***: HCV core protein expression in Hep G2 cells increased the total intracellular cholesterol level(4.05 ± 0.17 vs 6.47 ± 0.68, P = 0.001), and this increase corresponded to an increase in SREBP2 and HMGCR m RNA levels(P = 0.009 and 0.037, respectively) and protein expression. The molecular mechanism studyrevealed that the HCV core protein increased the expression of SREBP2 by enhancing its promoter activity(P = 0.004). In addition, mi R-185-5p expression was tightly regulated by the HCV core protein(P = 0.041). Moreover, overexpression of mi R-185-5p repressed the SREBP2 m RNA level(P = 0.022) and protein expression. In contrast, inhibition of mi R-185-5p caused upregulation of SREBP2 protein expression. mi R-185-5p was involved in the regulation of SREBP2 expression by HCV core protein. CONCLUSION: HCV core protein disturbs the cholesterol homeostasis in Hep G2 cells via the SREBP2 pathway; mi R-185-5p is involved in the regulation of SREBP2 by the core protein.
