Preparation and evaluation of a water-in-oil nanoemulsion drug delivery system loaded with salidroside

Salidroside(SAL)is a phenolic substance with high solubility and low permeability,which make it easy to cause the efflux effect of P-glycoprotein and degradation of intestinal flora,resulting in lower *** aim of this study was to develop and optimize a water-in-oil nanoemulsion of SAL(w/o SAL-N)to explore its suitability in oral drug delivery *** this work,SAL-N was successfully prepared by water titration method at K_(m)=1 to construct the pseudo-ternary phase *** characterization including the average viscosity,pH,refractive index,particle size,PDI,TEM,DSC,the content of SAL,and stability study were *** was evaluated for drug release in vitro and pharmacokinetic studies in *** optimized nanoemulsion formulation consisted of Labrafil M 1944 CS(63%),Span-80/Tween-80/EtOH(27%)and 200 mg·mL^(-1) SAL solution(SAL-SOL)(10%).Low viscosity and suitable pH were expected for the *** spherical morphology and nanoscale size of SAL-N enhanced the stability of the nanoemulsion *** vitro drug release showed that SAL-N had a better controlled release property than SAL-SOL at earlier time *** pharmacokinetic studies exhibited that SAL-N had significantly higher in t_(1/2)(2.11-fold),AUC_(0-48 h)(1.75-fold)and MRT0-48 h(2.63-fold)than SAL-SOL(Plivered via the oral *** can be seen w/o nanoemulsion is a strategy for the drug with polyphenols to delay the release,enhance oral absorption and reduce metabolic rate.

PEG-grafted PVA Membrane and Its Blood Compatibility

Preparation and blood compatibility of different shape polyvinyl alcohol（PVA） membrane were investigated. Firstly, the tabular and tubular[polytetrafluoroethylene（PTFE） capillary as supporter] PVA membranes were prepared; then, methoxy polyethylene glycol（mPEG） was grafted onto the surface of the PVA membranes. The effects of the shape, structure and properties of the membrane surface on blood compatibility were studied in detail. The experiment results show that mPEG modified PVA membranes, especially mPEG modified tubular membrane, could availably repel the adhesion of the platelets. In addition, the anticoagulant mechanism of mPEG with a steric repulsion effectiveness was confirmed further via different grafting methods.

Modification of Nanocomposites by Melting Intercalation of Polypropylene in Montmorillonite

The polypropylene wax modified by ultraviolet irradiation. The polypropylene-montmorillonite nanocomposiles were prepared by direct melting intercalation oj polypropylene powders. The structure of polypropylene , the polyproprlene irradiated, montmorillinote and polypropylene-montmorillonite composites were studied by XRD, 1R and DSC. The results show that the PP molecules can are oxidized during ultraviolet irradiation , melt polypropylene can intercalate into montmorillonite layer. As a result, the layered distance ( d0.01) of montmorillonite increases, and the melt absorption peak of polypropylene in layer is eliminated.

Peripheral nerve injury is a common neurological condition that often leads to severe functional limitations and *** on the pathogenesis of peripheral nerve injury has focused on pathological changes at individual injury sites,neglecting multilevel pathological analysis of the overall nervous system and target *** has led to restrictions on current therapeutic *** this paper,we first summarize the potential mechanisms of peripheral nerve injury from a holistic perspective,covering the central nervous system,peripheral nervous system,and target *** peripheral nerve injury,the cortical plasticity of the brain is altered due to damage to and regeneration of peripheral nerves;changes such as neuronal apoptosis and axonal demyelination occur in the spinal *** nerve will undergo axonal regeneration,activation of Schwann cells,inflammatory response,and vascular system regeneration at the injury *** damage to target organs can occur,including skeletal muscle atrophy and sensory receptor *** then provide a brief review of the research advances in therapeutic approaches to peripheral nerve *** main current treatments are conducted passively and include physical factor rehabilitation,pharmacological treatments,cell-based therapies,and physical ***,most treatments only partially address the problem and cannot complete the systematic recovery of the entire central nervous system-peripheral nervous system-target organ ***,we should further explore multilevel treatment options that produce effective,long-lasting results,perhaps requiring a combination of passive(traditional)and active(novel)treatment methods to stimulate rehabilitation at the central-peripheral-target organ levels to achieve better functional recovery.

Cereal endosperm comprises an outer aleurone and an inner starchy *** these two tissues have the same developmental origin,they differ in morphology,cell fate,and storage product accumulation,with the mechanism largely ***,we report the identification and characterization of rice thick aleurone 1(ta1)mutant that shows an increased number of aleurone cell layers and increased contents of nutritional factors including proteins,lipids,vitamins,dietary fibers,and *** identified that the TA1 gene,which is expressed in embryo,aleurone,and subaleurone in caryopses,encodes a mitochondrion-targeted protein with single-stranded DNA-binding activity named *** analyses revealed that the increased aleurone cell layers in ta1 originate from a developmental switch of subaleurone toward aleurone instead of starchy endosperm in the wild *** found that TA1/OsmtSSB1 interacts with mitochondrial DNA recombinase RECA3 and DNA helicase TWINKLE,and downregulation of REC A3 or TWINKLE also leads to ta1-like *** further showed that mutation in TA1/OsmtSSB1 causes elevated illegitimate recombinations in the mitochondrial genome,altered mitochondrial morphology,and compromised energy supply,suggesting that the OsmtSSB1-mediated mitochondrial function plays a critical role in subaleur one cell-fate determination in rice.

Cells are the fundamental units of biological systems and exhibit unique development trajectories and molecular features. Our exploration of how the genomes orchestrate the formation and maintenance of each cell, and control the cellular phenotypes of various organismsis, is both captivating and intricate. Since the inception of the first single-cell RNA technology, technologies related to single-cell sequencing have experienced rapid advancements in recent years. These technologies have expanded horizontally to include single-cell genome, epigenome,proteome, and metabolome, while vertically, they have progressed to integrate multiple omics data and incorporate additional information such as spatial sc RNA-seq and CRISPR screening. Single-cell omics represent a groundbreaking advancement in the biomedical field,offering profound insights into the understanding of complex diseases, including cancers. Here, we comprehensively summarize recent advances in single-cell omics technologies, with a specific focus on the methodology section. This overview aims to guide researchers in selecting appropriate methods for single-cell sequencing and related data analysis.

Rice endosperm consists of an outer aleurone layer and an inner starchy endosperm. In mature rice seeds,the aleurone consists of one layer of living cells that accumulate mainly storage proteins,lipids,iron and zinc,while the starchy endosperm comprising dead cells that accumulates mainly starch. In this study,we report the screening and identification of thick aleurone 1(ta1) mutant in rice. Instead of one layer of aleurone cells in the wild type,the mature ta1 seeds had 3 to 5 layers of aleurone cells. Nutritional analyses showed that the ta1 mature seeds had increased levels of storage proteins,lipids,iron and zinc content. Then we located TA1 at chromsome 5 through map based cloning approach. q RT-PCR result showed that TA1 is expressed mainly in mature pollens,shoot apical meristem,embryos and aleurone. Further genetic and biochemical analyses of TA1 may help to develop nutrientrich rice varieties and to establish a molecular network behind the differentiation of aleurone and starch endosperm in rice.
