Neuromyelitis optica spectrum disorders are neuroinflammatory demyelinating disorders that lead to permanent visual loss and motor *** date,no effective treatment exists as the exact causative mechanism remains ***,experimental models of neuromyelitis optica spectrum disorders are essential for exploring its pathogenesis and in screening for therapeutic *** most patients with neuromyelitis optica spectrum disorders are seropositive for IgG autoantibodies against aquaporin-4,which is highly expressed on the membrane of astrocyte endfeet,most current experimental models are based on aquaporin-4-IgG that initially targets *** experimental models have successfully simulated many pathological features of neuromyelitis optica spectrum disorders,such as aquaporin-4 loss,astrocytopathy,granulocyte and macrophage infiltration,complement activation,demyelination,and neuronal loss;however,they do not fully capture the pathological process of human neuromyelitis optica spectrum *** this review,we summarize the currently known pathogenic mechanisms and the development of associated experimental models in vitro,ex vivo,and in vivo for neuromyelitis optica spectrum disorders,suggest potential pathogenic mechanisms for further investigation,and provide guidance on experimental model *** addition,this review summarizes the latest information on pathologies and therapies for neuromyelitis optica spectrum disorders based on experimental models of aquaporin-4-IgG-seropositive neuromyelitis optica spectrum disorders,offering further therapeutic targets and a theoretical basis for clinical trials.

As more and more devices in Cyber-Physical Systems(CPS)are connected to the Internet,physical components such as programmable logic controller(PlC),sensors,and actuators are facing greater risks of network attacks,and fast and accurate attack detection techniques are *** key problem in distinguishing between normal and abnormal sequences is to model sequential changes in a large and diverse field of time *** address this issue,we propose an anomaly detection method based on distributed deep *** method uses a bilateral filtering algorithm for sequential sequences to remove noise in the time series,which can maintain the edge of discrete *** use a distributed linear deep learning model to establish a sequential prediction model and adjust the threshold for anomaly detection based on the prediction error of the validation *** method can not only detect abnormal attacks but also locate the sensors that cause *** conducted experiments on the Secure Water Treatment(SWAT)and Water Distribution(WADI)public *** experimental results show that our method is superior to the baseline method in identifying the types of attacks and detecting efficiency.

Sinter is the core raw material for blast *** pressure,which is an important state parameter,affects sinter *** this paper,flue pressure prediction and optimization were studied based on the shapley additive explanation(SHAP)to predict the flue pressure and take targeted adjustment ***,the sintering process data were collected and processed.A flue pressure prediction model was then constructed after comparing different feature selection methods and model algorithms using SHAP+extremely random-ized trees(ET).The prediction accuracy of the model within the error range of±0.25 kPa was 92.63%.SHAP analysis was employed to improve the interpretability of the prediction *** effects of various sintering operation parameters on flue pressure,the relation-ship between the numerical range of key operation parameters and flue pressure,the effect of operation parameter combinations on flue pressure,and the prediction process of the flue pressure prediction model on a single sample were analyzed.A flue pressure optimization module was also constructed and analyzed when the prediction satisfied the judgment *** operating parameter combination was then *** flue pressure was increased by 5.87%during the verification process,achieving a good optimization effect.

Background:The prognostic significance of the chemokine receptor CCR7 in diffuse large B-cell lymphoma(DlBCl)has been reported ***,the detailed mechanisms of CCR7 in DlBCl,particularly regarding its interaction with lenalidomide treatment,are not fully ***:Our study utilized bioinformatics approaches to identify hub genes in SU-DHl-2 cell lines treated with lenalidomide compared to control *** data and clinical information from 122 patients with DlBCl were analyzed to assess the correlation of CCR7 and p-ERK1/2 expression with the prognosis of ***,in vitro and in vivo experiments were conducted to clarify the role of CCR7 in the response of DlBCl to lenalidomide ***:Our bioinformatics analysis pinpointed CCR7 as a hub gene in the context of lenalidomide treatment in ***,31.14%and 36.0%(44/122)of DlBCl cases showed positive expression for CCR7 and ERK1/2 respectively,establishing them as independent prognostic factors for adverse outcomes in DlBCl via multivariate Cox regression ***,our studies demonstrated that the external application of the protein CCl21 promoted proliferation,migration,invasion,and activation of the ERK1/2 pathway in SU-DHl-2 and OCI-lY3 cell lines with high levels of CCR7 *** effect was mitigated by CCR7 silencing through siRNA,application of ERK inhibitors,or lenalidomide *** vivo experiments reinforced the efficacy of lenalidomide,significantly reducing tumor growth rate,tumor mass,serum total lDH levels,and expression of CCR7 and p-ERK1/2 in a SUDHl-2 xenograft model in nude mice(plusion:Our study clarifies the potential role of the CCl21/CCR7/ERK1/2 axis in the therapeutic effects of lenalidomide in DlBCl treatment.

Tanycytes, specialized ependymal cells located in the hypothalamus, play a crucial role in the generation of new neurons that contribute to the neural circuits responsible for regulating the systemic energy balance. The precise coordination of the gene networks controlling neurogenesis in naive and mature tanycytes is essential for maintaining homeostasis in adulthood. However, our understanding of the molecular mechanisms and signaling pathways that govern the proliferation and differentiation of tanycytes into neurons remains limited. This article aims to review the recent advancements in research into the mechanisms and functions of tanycyte-derived neurogenesis. Studies employing lineage-tracing techniques have revealed that the neurogenesis specifically originating from tanycytes in the hypothalamus has a compensatory role in neuronal loss and helps maintain energy homeostasis during metabolic diseases. Intriguingly,metabolic disorders are considered early biomarkers of Alzheimer's disease. Furthermore,the neurogenic potential of tanycytes and the state of newborn neurons derived from tanycytes heavily depend on the maintenance of mild microenvironments, which may be disrupted in Alzheimer's disease due to the impaired blood–brain barrier ***, the specific alterations and regulatory mechanisms governing tanycyte-derived neurogenesis in Alzheimer's disease remain unclear. Accumulating evidence suggests that tanycyte-derived neurogenesis might be impaired in Alzheimer's disease, exacerbating neurodegeneration. Confirming this hypothesis, however, poses a challenge because of the lack of long-term tracing and nucleus-specific analyses of newborn neurons in the hypothalamus of patients with Alzheimer's disease. Further research into the molecular mechanisms underlying tanycyte-derived neurogenesis holds promise for identifying small molecules capable of restoring tanycyte proliferation in neurodegenerative diseases. The aim of this paper is to investigate the existence of solutions to the prescribing fractional q-curvature problem on S~n under some reasonable assumption of the laplacian sign at the critical point of prescribing curvature function K. Due to the lack of compactness, we choose to return to the basic elements of variational theory and study the deformation along the flow lines. The novelty of the paper is that we obtain the existence without assuming any symmetry and periodicity on K. In addition,to overcome the loss of compactness for high-order operator problem, we need more delicate estimates with the second order cases.
