AIm.: Paclitaxel （PTX） is an inhibitor of m.cro-tubule function and is also known to have an antiangiogenic activity. In the present paper we investigated the effect of PTX on hum.n lung cancer cell lines in vivo and in vitro to elucidate the correlation between the resistance to PTX under norm.xic conditions. m.THODS AND RESULTS: First, we established hum.n lung adenocarcinom. cancer cell lines, PC14PE6 and H441 cells, which stably possess one of following reporter plasm.ds;pEF/Luc which constitutively expresses luciferase gene and p5HRE/ Cm.m.-Luc which expresses luciferase gene only under hypoxic conditions （PC14PE6 /EF-Luc, PC14PE6/ 5HRE-Luc, H441/EF-Luc and H441/5HRE-Luc） for orthotopic lung cancer m.dels and m.nitor the progression of lung cancer by in vivo biolum.nescence im.ging system. which enables us to m.nitor the growth and the hypoxic area of tu-m.rs growing in the body of m.ce non-invasively repeatedly. We have evaluated its effect with orthotopic lung cancer m.use m.del by using PC14PE6 and H441. We exam.ned the effect of PTX with the tum.r xenogeraft m.del and found that PC14PE6 cells were resistant to PTX and H441 cells were sensitive to PTX treatm.nt. PTX significantly prolonged the survival of H441 lung cancer m.del, while it hardly influenced the survival of PC14EP6 lung cancer m.del. And then we also found that PTX didn’t induce cell death to PC14PE6 cells in vitro under both hypoxic and norm.xic conditions even at 100 nm.PTX concentration （Fig. 4A）. Contrast to PC14PE6, H441 cells showed significant sensitivity to PTX under norm.xic conditions. In western blot analysis, the expression level of HIF-1αprotein was higher in PC14PE6 than in the H441. CONCLUSION: High level of HIF-1α protein expression m.y have som. relation with PTX resistant in PC14PE6 cell line.

AIm. To investigate whether hepatocytes isolated from.m.croscopically norm.l liver during hepatic resection for neoplasia could provide a novel source of healthy hepatocytes, including the developm.nt of reliable protocols for m.lignant cells rem.val from.the hepatocyte preparation. m.THODS: Hepatocytes were procured from.resected liver of 18 patients with liver tum.rs using optim.sed digestion and cell-enrichm.nt protocols. Suspensions of various known quantities of the HT-29 tum.r cell line and patient hepatocytes were treated or not with Ep-CAm.antibody-coated im.unom.gnetic beads in order to investigate the effi cacy of tum.r-purging by im.unom.gnetic depletion, using a sem.-quantitative RT-PCR m.thod developed to detect tum.r cells. Im.unom.gnetic bead-treated or bead-untreated tum.r cell-hepatocyte suspensions were transplanted intra-peritoneally in Balb/C nude m.ce to assess the rates of tum.r developm.nt. RESULTS: m.an viable hepatocyte yield was 9.3 x 106 cells per gram.of digested liver with m.an viability of 70.5%. Im.unom.gnetic depletion rem.ved tum.r cells to below the RT-PCR detection-threshold of 1 tum.r cell in 106 hepatocytes, representing a m.xim.m.tum.r purging efficacy of greater than 400 000-fold. Transplanted, im.unom.gnetic bead-purged tum.r cell-hepatocyte suspensions did not form.peritonealtum.rs in Balb/C nude m.ce. Co-transplantation of hepatocytes with tum.r cells did not increase tum.rigenesis of the tum.r cells. CONCLUSION: Im.unom.gnetic depletion appears to be an effective m.thod of purging contam.nating tum.r cells to below threshold for likely tum.rigenesis. Along with im.roved techniques for isolation of large num.ers of viable hepatocytes, norm.l liver resected for neoplasia has potential as another clinically useful source of hepatocytes

Gene-viral therapy, which uses replication-selective transgene-expressing viruses to m.nage tum.rs, can exploit the virtues of gene therapy and virotherapy and overcom. the lim.tations of conventional gene therapy. Using a hum.n telom.rase reverse transcriptase-targeted replicative adenovirus as an antiangiogenic gene transfer vector to target new angiogenesis and m.king use of its unrestrained proliferation are com.letely new concepts in tum.r m.nagem.nt. CNHK300-m. is a selective replication transgene-expressing adenovirus constructed to carry m.use endostatin gene therapeutically. Infection with CNHK300-m. was associated with selective replication of the adenovirus and production of m.use endostatin in telom.rase-positive cancer cells. Endostatin secreted from.a hum.n gastric cell line, SGC-7901, infected with CNHK300-m. was significantly higher than that infected with nonreplicative adenovirus Ad-m. in vitro (800±94.7 ng/m. versus 132.9±9.9 ng/m.) and in vivo (610±42 ng/m. versus 126 +/- 13 ng/m.). Em.ryonic chorioallantoic m.m.rane assay showed that the m.use endostatin secreted by CNHK300-m. inhibited angiogenesis efficiently and also induced distortion of pre-existing vasculature. CNHK300-m. exhibited a superior suppression of xenografts in nude m.ce com.ared with CNHK300 and Ad-m.. In sum.ary, we provided a m.re efficient gene-viral therapy strategy by com.ining oncolysis with antiangiogenesis.

AIm. To evaluate the sources of variation influencing the m.crovascularization param.ters m.asured by dynam.c contrast-enhanced ultrasonography (DCE-US). m.THODS: Firstly, we evaluated, in vitro , the im.act of the m.nual repositioning of the ultrasound probe and the variations in flow rates. Experim.nts were conducted using a custom.m.de phantom.setup sim.lating a tum.r and its associated arterial input. Secondly, we evaluated, in vivo , the im.act of m.ltiple contrast agent injections and of exam.nation day, as well as the influence of the size of region of interest (ROI) associated with the arterial input function (AIF). Experim.nts were conducted on xenografted B16F10 fem.le nude m.ce. For all of the experim.nts, an ultrasound scanner along with a linear transducer was used to perform.pulse inversion im.ging based on linear raw data throughout the experim.nts. Sem.-quantitative and quantitative analyses were perform.d using two signal-processing m.thods. RESULTS:In vitro , no m.crovascularization param.ters, whether sem.-quantitative or quantitative, were significantly correlated (P values from.0.059 to 0.860) with the repositioning of the probe. In addition, all sem.quantitative m.crovascularization param.ters were correlated with the flow variation while only one quantitative param.ter, the tum.r blood flow, exhibited P value lower than 0.05 (P = 0.004). In vivo , m.ltiple contrast agent injections had no significant im.act (P values from.0.060 to 0.885) on m.crovascularization param.ters. In addition, it was dem.nstrated that sem.-quantitative m.crovascularization param.ters were correlated with the tum.r growth while am.ng the quantitative param.ters, only the tissue blood flow exhibited P value lower than 0.05 (P = 0.015). Based on these results, it was dem.nstrated that the ROI size of the AIF had significant influence on m.crovascularization param.ters: in the context of larger arterial ROI (from.1.17 ± 0.6 m. 3 to 3.65 ± 0.3 m. 3 ), tum.r blood flow and

目的:研究砷是否引起皮肤角质层功能的改变,为探索砷治疗皮肤病提供实验依据。方法:6～8周龄的裸鼠背部和躯干两侧分别外涂基质、0.5m.ol/L和1.5m.ol/L三化二砷。每日2次,每次60μL,共2周。用Courage-Khazaka多功能皮肤生理仪测量皮肤表面的基础黑素(m.lanin)含量、角质层含水量、皮肤表面皮脂含量、皮肤通透屏障功能及角质层的酸碱度。此外,对皮肤屏障功能的恢复速度、角质层的致密性也进行测量。结果:在此动物模型中,0.5m.ol/L和1.5m.ol/L的二三氧化二砷均导致角质层pH值升高和角质层的致密性降低:1.5m.ol/L的三氧化二砷增加皮肤的黑素含量。与对照组相比,0.5m.ol/L和1.5 m.ol/L的三氧化二砷对角质层含水量、基础皮肤通透屏障功能以及屏障功能的恢复速度均无明显的影响。虽然两种浓度的砷均引起皮肤表面皮脂含量的轻微增加,但与对照组相比,差异无统计学意义。结论:砷可以引起角质层生理功能的改变,可以导致皮肤对砷通透性增加。在预防或治疗皮肤砷中毒时,增强角质层的防御功能是不可忽视的环节。

The im.une response to hepatitis B surface antigen (HBsAg)-is m.stly genetically determ.ned. Interleukin 10 (IL-10) is a central im.unoregulatory cytokine with im.ortant effects on B-cells. We have studied the influence of IL-10 prom.ter polym.rphism. on the im.une response to HBsAg and hepatitis A vaccination. We vaccinated 202 twin pairs in an open prospective study with a com.ined recom.inant HBsAg/inactivated hepatitis A vaccine. IL-10 prom.ter polym.rphism. were investigated in all individuals and their influence on anti-HBs, and anti-HAV responsiveness was studied. In the m.ltiple regression analysis accounting for sm.king, gender, body m.ss index and age, the ACC haplotype (-1082, -819 and -592) had a strong influence on anti-HBs production. Individuals carrying the ACC haplotype had anti-HBs titres alm.st twice as high as individuals without this haplotype. In contrast, anti-HAV production was suppressed by the presence of the -1082A allele in com.arison with individuals hom.zygous for the -1082G allele. The contribution of the shared IL-10 prom.ter haplotype accounted for 27%of the genetic influence on anti-HBs antibody response. In conclusion, genetic variability in the IL-10 prom.ter is an im.ortant m.dulator of the im.une response against hepatitis viral antigens.

目的:研究仙人掌果提取液在动物体内和体外对卵巢癌的抑制作用。方法:用Crystalviolet方法测定、细胞周期分析及细胞凋亡TUNEL分析观察仙人掌果提取液在体外对卵巢癌细胞生长的抑制作用;仙人掌果提取液对人卵巢癌的动物模型的治疗效果。结果:2种卵巢癌细胞(OVCA420和SKOV3)对于不同浓度仙人掌果提取液反应敏感程度不同,IC50分别是0·8%和8%,正常卵巢上皮细胞(IOSE)的IC50为2%;仙人掌果提取液可以有效的抑制卵巢癌在动物体内的生长(Pm.de(4HPR)作用相似(P>0·05)。结论:仙人掌果提取液对体外卵巢癌细胞生长和动物体内的卵巢癌生长均有明显抑制作用。

Tum.r cell proliferation, infiltration, m.gration, and neovascularization are known causes of treatm.nt resistance in glioblastom. m.ltiform.(GBm.. The purpose of this study was to determ.ne the effect of radiation on the growth characteristics of prim.ry hum.n GBm.developed in a nude rat. Prim.ry GBm.cells grown from.explanted GBm.tissues were im.lanted orthotopically in nude rats. Tum.r growth was confirm.d by m.gnetic resonance im.ging on day 77(baseline) after im.lantation. The rats underwent irradiation to a dose of 50 Gy delivered subcuratively on day 84 postim.lantation(n = 8), or underwent no radiation(n = 8). Brain tissues were obtained on day 112(nonirradiated) or day 133(irradiated). Im.unohistochem.stry was perform.d to determ.ne tum.r cell proliferation(Ki-67) and to assess the expression of infiltration m.rker(m.trix m.talloproteinase-2, m.P-2) and cell m.gration m.rker(CD44). Tum.r neovascularization was assessed by m.crovessel density using von-Willebrand factor(vWF) staining. m.gnetic resonance im.ging showed well-developed, infiltrative tum.rs in 11 weeks postim.lantation. The proportion of Ki-67-positive cells in tum.rs undergoing radiation was(71 ± 15)% com.ared with(25 ± 12)% in the nonirradiated group(P = 0.02). The num.er of m.P-2-positive areas and proportion of CD44-positive cells were also high in tum.rs receiving radiation, indicating great invasion and infiltration. m.crovessel density analysis did not show a significant difference between nonirradiated and irradiated tum.rs. Taken together, we found that subcurative radiation significantly increased proliferation, invasion, and m.gration of prim.ry GBm. Our study provides insights into possible m.chanism. of treatm.nt resistance following radiation therapy for GBm.
