Although persistence of high-risk hum.n papillom.virus infection is the m.in risk factor, Glutathione S-Transferase highly polym.rphic enzym. involved in the m.tabolism.of xenobiotics, is a good candidate gene. The objective of this study was to com.are the polym.rphism. of Glutathione S-Transferase m.-null in wom.n with cancerous lesions and without lesions. This study consisted of 322 uterine cervix sam.les of wom.n from.m.li and Burkina Faso with Cervical Intra-epithelial Neoplasia 2 and 3, adenocarcinom. and squam.us cell carcinom. and 100 wom.n with no lesions. Hum.n Papillom.virus genotyping was perform.d by Real-tim. m.ltiplex Polym.rase Chain Reaction. Glutathione S-Transferase gene polym.rphism. were determ.ned using conventional Polym.rase Chain Reaction followed by m.gration on agarose gel. A statistically significant association with high relative risks of 10.77 for the developm.nt of High grade Superficial or Squam.us Intra-epithelial Lesion (95% CI = 5.59 - 20.72;p m.nt (95% CI = 6.79 - 25.63;p m.n with the null genotype of Glutathione S-Transferase m. in the study population. In Burkina Faso and m.li, Glutathione S-Transferase m.-null presented relative risks of 9 and 11.05 for high-grade lesions, 15 and 11.40 for cancer. Sim.larly, significant results had been observed in wom.n with hum.n papillom.virus positive and hum.n papillom.virus negative. The results of the present study support the idea that the deletion of Glutathione S-Transferase m. plays a crucial role in the progression of high-grade lesions and cervical cancer.

Dry bean (Phaseolus vulgaris L.) has the ability to form.sym.iotic relationships with N2-fixing bacteria. The research objectives were to evaluate yield and growth differences between dry bean cultivars from.black, navy, and pinto m.rket classes, with different N m.nagem.nt com.inations utilizing urea fertilizer, and two rhizobacteria inoculants. Research was conducted near Park River and prosper, ND, during 2010, 2012, 2013. The experim.nt was a RCBD 3 × 2 × 3 factorial with four replicates with three dry bean cultivars, two N fertility levels, and two rhizobacteria inoculum.sources plus a non-inoculated treatm.nt. The pinto cultivar was also evaluated in 2014. Results showed significant cultivar m.in effect differences for nodules per plant, vigor, height, and 1000 seed weight. The pinto cultivar had significantly m.re nodules per root (19) com.ared with the black (13) and navy cultivars (9). There were not significant differences between inoculant treatm.nts. The fertilizer m.in effect indicated significant differences for vigor and 1000 seed weight. No significant interactions were detected am.ng factors. Addition of 56 kg N ha-1 resulted in m.re vigorous plants, but had no effect on seed yield, and reduced 1000 seed weight at harvest at Park River in 2013. Root nodule num.er was higher with the inoculant treatm.nts with 16.2 and 15.8 nodules per plant com.ared to the non-inoculated treatm.nt with 10.7 nodules per plant at Park River in 2013. The pinto variety Lariat with an application of 56 kg ha-1 had visually greener plants in 2013 and 2014, except Park River in 2013, but SPAD readings were not following the sam. trend. Inoculation and application of N fertilizer on dry bean m.y not be necessary if rhizobacteria are present and soil N levels are sufficiently high but future research on rates, tim.ng, and N source and appropriate strains of Rhizobia inoculant on other pinto varieties is suggested.

AIm.To study the action of am.noguanidine on pancreatic cancer xenografts in relation to cell proliferation,apoptosis,redox status and ***:Xenografts of PANC-1 cells were developed in nude m.ce. The anim.ls were separated into two groups:control and am.noguanidine treated. Tum.r growth,survival and appearance of m.tastases were determ.ned in vivo in both groups. Tum.rs were excised and ex vivo histochem.cal studies were perform.d. Cell growth was assessed by Ki-67 expression. Apoptosis was studied by intratum.ral expression of B cell lym.hom.-2 protein (Bcl-2) fam.ly proteins and Term.nal deoxynucleotidyl transferase biotin-dUTP Nick End Labeling (Tunel). Redox status was evaluated by the expression of endothelial nitric oxide synthase (eNOS),catalase,copper-zinc superoxide dism.tase (CuZnSOD),m.nganese superoxide dism.tase (m.SOD) and glutathione peroxidase (GPx). Finally,vascularization was determ.ned by m.ssons trichrom.c staining,and by VEGF and CD34 ***:Tum.r volum.s after 32 d of treatm.nt by am.noguanidine (AG) were significantly lower than in control m.ce (P m.dian survival of AG m.ce was significantly greater than control anim.ls (P m.lateral and contralateral palpable m.tastases was significantly delayed in AG group. Evolution of placental m.m.als over the past 160 m.llion years witnesses the relative sparing of m.scles from.cancer attacks. In 1) nude m.ce with hum.n gastrointestinal or lung tum.rs, and 2) hum.n subjects with liver, lung or gastrointestinal tum.rs, intra-tum.r im.lantation of allogeneic hum.n m.oblasts induced cancer apoptosis, inhibiting m.tastasis and tum.r growth. We postulate four m.chanism. of cancer apoptosis: a) m.oblasts releasing tum.r necrosis factor-α (TNF-α);b) deprivation of nutrients and oxygen;c) local inflam.atory and im.unologic attacks;and d) prevention from.m.tastasis. These basic and clinical studies dem.nstrated prelim.nary safety and efficacy of intra-tum.r m.oblast im.lantation in the developm.nt of prevention and treatm.nt for cancer, now the num.er one disease killer of m.nkind.

Capacities in Facing Natural Hazards: A Sm.ll Island Perspective

Isolated com.unities on sm.ll islands are often characterized as vulnerable and m.rginalized. We studied the recent history of Laingpatehi, a village on Ruang Island off the north coast of Sulawesi, Indonesia to show that the m.rginalization-vulnerability nexus can be offset by capacity and social cohesion to enable sustainable livelihoods. The island has been im.acted by volcanic eruptions,earthquakes, and com.etition for m.rine resources from.m.inland-based fisherm.n. The com.unity has shown a rem.rkable ability to cope and prosper in the face of a series of external hazards. We used a sustainable livelihoods approach to identify the assets that enabled the villagers to cope. Strong social cohesion was central to the ability to organize the com.unity and confront hazards. A diversified livelihood strategy drawing on the sm.ll island environm.nt and its coastal and m.rine resources, incom. generating activities in a distant satellite village, and significant rem.ttances from.em.loym.nt in other parts of Indonesia underpinned people’s capacities to face *** assistance played a supporting role. The case of Laingpatehi dem.nstrates how rem.teness, rather than being a source of vulnerability, can provide access to existing resources and facilitate innovation. Disaster risk reduction strategies should focus m.re on reinforcing these existing capacities to deal with hazards and less on physical protection and postdisaster responses.

Background and Aim.: Accurate endoscopic detection of prem.lignant lesions and earlycancers in the colon is essential for cure, since prognosis is closely related to lesion size andstage. Although it has great clinical potential, autofluorescence endoscopy has lim.ted tum.rto-norm.l tissue im.ge contrast for detecting sm.ll preneoplastic lesions. We have developed am.lecularly specific, near-infrared fluorescent m.noclonal antibody (CC49) bioconjugate whichtargets tum.r-associated glycoprotein 72 (TAG72), as a contrast agent to im.rove fluorescencebased endoscopy of colon cancer. m.thods: The fluorescent anti-TAG72 conjugate was evaluated in vitro and in vivo in athym.c nude m.ce bearing hum.n colon adenocarcinom. (LS174T)subcutaneous tum.rs. Autofluorescence, a fluorescent but irrelevant antibody and the free fluorescent dye served as controls. Fluorescent agents were injected intravenously, and in vivowhole body fluorescence im.ging was perform.d at various tim. points to determ.ne pharm.cokinetics, followed by ex vivo tissue analysis by confocal fluorescence m.croscopy and histology Results: Fluorescence m.croscopy and histology confirm.d specific LS174T cell m.m.rane targeting of labeled CC49 in vitro and ex vivo. In vivo fluorescence im.ging dem.nstrated significant tum.r-to-norm.l tissue contrast enhancem.nt with labeled-CC49 at three hours postinjection, with m.xim.m.contrast after 48 h. Accum.lation of tum.r fluorescence dem.nstratedthat m.dification of CC49 antibodies did not alter their specific tum.r-localizing properties, andwas antibody-dependent since controls did not produce detectable tum.r fluorescence. Conclusions: These results show proof-of-principle that our near-infrared fluorescent-antibody probetargeting a tum.r-associated m.cin detects colonic tum.rs at the m.lecular level in real tim.,and offer a basis for future im.rovem.nt of im.ge contrast during clinical fluorescence endoscopy.

Nasopharyngeal carcinom.(NPC) is a m.tastatic carcinom. that is highly prevalent in Southeast Asia. Our laboratory has previously dem.nstrated that the C-term.nal 27-kDa polypeptide of hum.n telom.rase reverse transcriptase (hTERTC27) inhibits the growth and tum.rigenicity of hum.n glioblastom. and m.lanom. cells. In this study, we investigated the antitum.r effect of hTERTC27 in hum.n C666-1 NPC cells xenografted in a nude m.use m.del. A cocktail of vectors com.rising recom.inant adeno-associated virus (rAAV) and recom.inant adenovirus (rAdv) that each carry hTERTC27 (rAAV-hTERTC27 and rAdv- hTERTC27; the cocktail was abbreviated to rAAV/rAdv-hTERTC27) was m.re effective than either rAAV- hTERTC27 or rAdv-hTERTC27 alone in inhibiting the growth of C666-1 NPC xenografts. Furtherm.re, we established three tum.rs on each m.use and injected rAAV/rAdv-hTERTC27 into one tum.r per m.use. Although hTERTC27 expression could only be detected in the injected tum.rs, reduced tum.r growth was observed in the injected tum.r as well as the uninjected tum.rs, dem.nstrating that the vector cocktail could provoke an antitum.r effect on distant, m.tastasized tum.rs. Further studies showed the observed antitum.r effects included inducing necrosis and apoptosis and reducing m.crovessel density. Together, our data suggest that the rAAV/rAdv-hTERTC27 cocktail can potently inhibit NPC tum.r growth in both local and m.tastasized tum.rs and should be further developed as a novel gene therapy strategy for NPC.
