Influence of vascular endothelial growth factor and radiation on gap junctional intercellular communication in glioblastoma multiforme cell lines

Glioblastoma multiforme （GBM） is a highly aggressive glial brain tumor with an unfavorable prognosis despite all current therapies including surgery, radiation and chemotherapy. One characteristic of this tumor is a strong synthesis of vascular endothelial growth factor （VEGF）, an angiogenesis factor, followed by pronounced vascularization. VEGF became a target in the treatment of GBM, for example with bevacizumab or the tyrosine kinase inhibitor axitinib, which blocks VEGF receptors. To improve patients＇ prognosis, new targets in the treatment of GBM are under investigations. The role of gap junctions in GBM remains un- known, but some experimental therapies affect these intercellular channels to treat the tumor. Gap junctions are composed of connexins to allow the transport of small molecules between adjacent cells through gap junc- tional intercellular communication （GJIC）. Based on data derived from astrocytes in former studies, which show that VEGF is able to enhance GJIC, the current study analyzed the effects of VEGF, radiation therapy and VEGF receptor blockade by axitinib on GJIC in human GBM cell lines U-87 and U-251. While VEGF is able to induce GJIC in U-251 cells but not in U-87 cells, radiation enhances GJIC in both cell lines. VEGF reocptor blockade by axitinib diminishes radiation induced effects in U-251 partially, while increases GJIC in U-87 cells. Our data indicate that VEGF and radiation are both modifying components of GJ1C in pathologic brain tumor tissue.

Does laser surface texturing really have a negative impact on the fatigue lifetime of mechanical components?

Laser surface texturing(LST)has been proven to improve the tribological performance of machine *** micro-scale patterns manufactured by LST may act as lubricant reservoirs,thus supplying oil when encountering insufficient ***,not many studies have investigated the use of LST in the boundary lubrication regime,likely due to concerns of higher contact stresses that can occur with the increasing surface *** study aims to examine the influence of LST on the fatigue lifetime of thrust rolling bearings under boundary lubrication.A series of periodic patterns were produced on the thrust rolling bearings,using two geometrically different designs,namely cross and dimple *** oil ISO VG 100 mixed with 0.05 wt%P of zinc dialkyldithiophosphate(ZDDP)was *** bearings with cross patterns reduce the wear loss by two orders of *** patterns not only retain lubricant in the textured pockets but also enhance the formation of an anti-wear *** tribofilm generation may be improved by the higher contact stresses that occur when using the textured ***,in contrast to the negative concerns,the ball bearings with cross patterns were instead found to increase the fatigue life by a factor of three.

The aim of the present study is to evaluate the effect of alloy processing and composition as well as the pH control and testing medium on the in vitro corrosion performance of Mg-Zn-Ca systems for biodegradable *** grain size and secondary phases were analyzed by optical microscopy,scanning electron microscopy,transmission electron microscopy,and X-ray *** kelvin probe force microscopy(SKPFM)was used to analyze the Volta potential values of the second *** corrosion performance of the three alloys was evaluated by electrochemical and hydrogen evolution methods inα-MEM with and without organic species(*** and inorganicα-MEM).Two strategies were followed to evaluate the influence of the pH on the corrosion behavior:daily solution replacement and CO_(2)flow based pH *** all the materials,the organic medium accelerates the corrosion *** pH maintained by CO_(2)flow through the medium results in considerably higher corrosion rates for all *** impact of pH is lesser on the as-cast alloys due to the barrier effect of the secondary phases,particularly pronounced in the Mg1Zn1Ca alloy which showed the lowest corrosion *** wrought Mg0.5Zn0.2Ca alloy that lacks the refined secondary phase network and exhibits high number of twins undergoes accelerated uniform corrosion under constant pH conditions.

Temperature-Dependent Photoconductance of Heavily Doped ZnO Nanowires

Ga-doped ZnO nanowires have been synthesized by a pulsed laser chemical vapor deposition method. The crystal structure and photoluminescence spectra indicate that the dopant atoms are well integrated into the ZnO wurtzite lattice. The photocurrent properties at different temperatures have been systematically investigated for nanowires configured as a three-terminal device. Among the experimental highlights, a pronounced semiconductor-to-metal transition occurs upon UV band-to-band excitation. This is a consequence of the reduction in electron mobility arising from the drastically enhanced Coulomb interactions and surface scattering. Another feature is the reproducible presence of two resistance valleys at 220 and 320 K upon light irradiation. This phenomenon originates from the trapping and detrapping processes in the impurity band arising from the native defects as well as the extrinsic Ga dopants. This work demonstrates that due to the dimensional confinement in quasi-one-dimensional structures, enhanced Coulomb interaction, surface scattering, and impurity states can significantly influence charge transport.

Corrosion Protection of AM50 Magnesium Alloy by Nafion/DMSO Organic Coatings

The effectiveness of the corrosion protection of Nafion/Dimethysulfoxid （DMSO） organic coatings for AM50 magnesium alloy prepared by simple immersion and heat treatment was investigated. Its corrosion resistance and morphologies of the Nafion/DMSO organic coatings were studied by electrochemical corrosion testing and optical microscopy. The results show that Nafion/DMSO organic coatings can improve the corrosion resistance of AM50 magnesium alloy effectively. Also, the corrosion resistance increases with the surface density of the organic coatings.

Two-Photon Direct Laser Writing Beyond the Diffraction Limit Using the Nanopositioning and Nanomeasuring Machine

In July 2023,the Center of Excellence in Respiratory Pathogens organized a two-day workshop on infectious diseases modelling and the lessons learnt from the Covid-19 *** report summarizes the rich discussions that occurred during the *** workshop participants discussed multisource data integration and highlighted the benefits of combining traditional surveillance with more novel data sources like mobility data,social media,and wastewater *** advancements were noted in the development of predictive models,with examples from various countries showcasing the use of machine learning and artificial intelligence in detecting and monitoring disease *** role of open collaboration between various stakeholders in modelling was stressed,advocating for the continuation of such partnerships beyond the pandemic.A major gap identified was the absence of a common international framework for data sharing,which is crucial for global pandemic ***,the workshop underscored the need for robust,adaptable modelling frameworks and the integration of different data sources and collaboration across sectors,as key elements in enhancing future pandemic response and preparedness.

NK cell detachment from target cells is regulated by successful cytotoxicity and influences cytokine production

Natural killer(NK)cells participate in early immune defenses against pathogens and tumors and play a major role as immune effector and regulatory *** NK cell-mediated elimination of an infected or cancerous cell is a highly regulated process that requires the formation of a cell contact,the establishment of an immunological synapse and the polarization and release of lytic ***,the detachment of NK cells from target cells is important for NK cells to bind and kill other cells in a process called serial ***,very little is known about this detachment ***,we show that NK detachment is directly connected to the successful killing of a target *** inhibition of killing due to reduced NK cell cytotoxicity or increased target cell resistance results in defective detachment and prolonged contact *** effect leads to sustained Ca^(2+) flux in NK cells and the hypersecretion of proinflammatory *** defective cytotoxicity with enhanced cytokine secretion via reduced detachment may explain inflammatory pathologies in several diseases.
