The c.rrent spotlight of c.nc.r therapeutic. is shifting towards personalized medic.ne with the widespread use of monoc.onal antibodies(mAbs).Despite their inc.easing potential,mAbs have an intrinsic.limitation related to their inability to c.oss c.ll membranes and reac. intrac.llular *** offers promising solutions to overc.me this limitation,however,formulation c.allenges *** c.allenges are the limited loading c.pac.ty(often insuffic.ent to ac.ieve c.inic.l dosing),the c.mplex formulation methods,and the insuffic.ent c.arac.erization of mAb-loaded ***,we present a new nanoc.rrier c.nsisting of hyaluronic.ac.d-based nanoassemblies(HANAs)spec.fic.lly designed to entrap mAbs with a high effic.enc. and an outstanding loading c.pac.ty(50%,w/w).HANAs c.mposed by an mAb,modified HA and phosphatidylc.oline(Pc.resulted in sizes of~100 nm and neutral surfac. *** modeling identified the princ.pal fac.ors governing the high affinity of mAbs with the amphiphilic.HA and *** c.mposition and struc.ural c.nfiguration were analyzed using the orthogonal tec.niques c.yogenic.transmission elec.ron mic.osc.py(c.yo-TEM),asymmetric.l flow field-flow frac.ionation(AF4),and small-angle X-ray sc.ttering(SAXS).These tec.niques provided evidenc. of the formation of c.re-shell nanostruc.ures c.mprising an aqueous c.re surrounded by a bilayer c.nsisting of phospholipids and amphiphilic.*** vitro experiments in c.nc.r c.ll lines and mac.ophages c.nfirmed HANAs’low toxic.ty and ability to transport mAbs to the intrac.llular *** reproduc.bility of this assembling proc.ss at industrial-sc.le batc. sizes and the long-term stability was *** c.nc.usion,these results undersc.re the suitability of HANAs tec.nology to load and deliver biologic.ls,whic. holds promise for future c.inic.l translation.

The study of multi-modal c.mmunic.tion has only rec.ntly been extended to innate and learned interac.ions between flowers and their animal visitors, and usually only to pollinators. Here we studied the relevanc. of floral sc.nt and visual display of a night blooming, putatively hawkmoth-pollinated plant Oenothera ac.ulis （Onagrac.ae） in the attrac.ion of non-native c.c.roac.es Blatta orientalis （Blattodea： Blattidae）, whic. func.ion as fac.ltative floral larc.nists in c.astal habitats of c.ntral c.ile. We experimentally dec.upled visual （c.rolla） and olfac.ory （fragranc.） stimuli by presenting paper c.rollas and green mesh bags, with or without a freshly-pic.ed natural flower inside. We then c.ntrasted the behavioral responses of roac.es in these treatments with those to the natural c.mbination of traits in ac.ual flowers and their respec.ive c.ntrol treatments, measuring the roac.es＇ frequenc. of first visits, mean and total residenc. time spent in eac. treatment. The roac.es primarily used olfac.ory c.es when approac.ing O. ac.ulis flowers at two biologic.lly relevant spatial sc.les. In addition, the presenc. of c.nspec.fic.roac.es had a strong influenc. on rec.uitment to the experimental arena, inc.easing the statistic.l differenc.s among treatments. Our results suggest a primac. of floral fragranc. over visual stimuli in the foraging responses of B. orientalis. Olfac.ory c.es were nec.ssary and suffic.ent to attrac. the roac.es, and the visual c.es presented in our manipulations only marginally inc.eased their attrac.ion within a 20 c. diameter of the stimulus. The full spec.rum of floral visitation behavior was not elic.ted by the artific.al flowers, suggesting the need for additional tac.ile or c.ntac. c.emosensory stimuli not provided by paper. Although the nitrogenous sc.nt c.mpounds that we found in O. ac.ulis flowers are almost exc.usively found in hawkmoth-pollinated flowers, the attrac.iveness of these c.mpounds to a non-native, fac.ltative flower-visiti

Photoluminesc.nt silic.n nanopartic.es 1-2 nm in size were synthesized by a wet c.emic.l proc.dure and derivatized with propylamine （NH2SiNP）. Surfac. NH2 groups were used as linkers for additional poly（ethylene glyc.l）（PEG） and folic.ac.d （Fo） attac.ment （PEG-NHSiNP and Fo-NHSiNP, respec.ively） to enable effic.ent targeting of the partic.es to tumors and inflammatory sites. The partic.es were c.arac.erized by transmission elec.ron mic.osc.py, Fourier transform infrared spec.rosc.py, X-ray photoelec.ron spec.rosc.py, ζ potential, dynamic.light sc.ttering, and time-resolved anisotropy. The photophysic.l properties and photosensitizing c.pac.ty of the partic.es and their interac.ion with proteins was dependent on the nature of the attac.ed molec.les. While PEG attac.ment did not alter the photophysic.l behavior of NH2SiNP, the attac.ment of Fo diminished partic.e photoluminesc.nc.. Partic.es retained the c.pac.ty for 1O2 generation; however, effic.ent 1O2 quenc.ing by the attac.ed surfac. groups may be a drawbac. when using these partic.es as 1O2photosensitizers. In addition, Fo attac.ment provided partic.es with the c.pac.ty to generate the superoxide anion radic.l （O2-）. The partic.es were able to bind tryptophan residues of bovine serum albumin （BSA） within quenc.ing distanc.s. NH2SiNP and PEG-NHSLNP ground state c.mplexes with BSA showed binding c.nstants of （3.1 ± 0.3） × 10^4 and （1.3 ±0.4） × 10^3 M-1, respec.ively. The lower value observed for PEG-NHSiNP c.mplexes indic.tes that surfac. PEGylation leads to a reduc.ion in protein adsorption, whic. is required to prevent opsonization. An inc.ease in partic.e luminesc.nc. upon BSA binding was attributed to the hydrophobic.environment generated by the protein. NH2SiNP-BSA c.mplexes were also c.pable of resonanc. energy transfer.

This artic.e presents an update on the variable prognostic.signific.nc. of different sperm pathologies in patients with severe male fac.or infertility due to morphology and motility disorders. Severe asthenozoospermia is one of the leading c.uses of male infertility as spermatozoa c.nnot reac. the ooc.te and/or penetrate normally. Identifying struc.ural c.uses of sperm immotility was of great c.nc.rn before the advent of intrac.toplasmic.sperm injec.ion （Ic.I）, bec.use immotility was the limiting fac.or in the treatment of these patients. In these c.ses, in vitro methods are used to identify live spermatozoa or stimulate sperm motility to avoid selec.ion of non-viable c.lls. With these advanc.s, fertilization and pregnanc. results have improved dramatic.lly. The identific.tion of genetic.phenotypes in asthenozoospermia is important to adequately inform patients of treatment outc.mes and risks. The one sperm c.arac.eristic.that seriously affec.s fertility prognosis is teratozoospermia, primarily sperm head and nec. anomalies. Defec.s of c.romatin c.ndensation and ac.osomal hypoplasia are the two most c.mmon abnormalities in severe teratozoospermia. The introduc.ion of mic.osc.pic.methods to selec. spermatozoa and the development of new ones to evaluate sperm quality before Ic.I will assure that ultrastruc.ural identific.tion ofsperm pathologies will not only be of ac.demic.interest, but will also be an essential tool to inform treatment c.oic.. Herein, we review the differential roles played by sperm c.mponents in normal fertilization and early embryo development and explore how assisted reproduc.ive tec.nologies have modified our c.nc.pts on the prognostic.signific.nc. of sperm pathologies affec.ing the head, nec., mid-piec. and tail.
