Fast and Effective Thermotherapy Treatment for In VitroVirus Eradication in Apple and pear Trees

Heat therapy followed by the isolation and in vitro culture of apical meristems is a suitable procedure for virus eradication. However, the period of heat treatment is usually long (28 - 50 days) and the yield of viable plants free of viruses after treatment is often low (plants. We used traditional Galician cultivars, six apple trees and two pear trees, infected with Apple chlorotic leaf spot virus (ACLSV) and Apple mosaic virus (ApMV). We combined heat therapy of in vitro shoots using a temperature gradient from 25℃to 40℃increasing 1℃per day for a shorter period of time (18 days) with the posterior isolation and culture of apical meristems. All DNA samples analyzed, obtained from plants developed from meristems, were 100% free of ApMV and almost 90% free of ACLSV. With this in vitro procedure combined we obtained a good yield of tested plants free of viruses. Our method is fast and effective and it could be also useful to eradicate these and other viruses in other fruit trees.

Analysis of Urban Morphology in Core Towns＇ Neighborhoods of Mexico： Santa Barbara＇s Neighborhood as a Case Study

Cities＇ urban morphology is the result of historic, cultural, political and social processes. The historical cores in Mexican cities have high cultural diversity, which also intermingle pre-Hispanic cultures with colonial forms, as well with modernity and post-modernity irruption. The case study is Santa Barbara＇s neighborhood in Toluca City, which was founded in 1524, and the case study was the first neighborhood in Toluca＇s historical core. At present time, this neighborhood is considered as one of the most dangerous places in the city＇s historical core, therefore, it has been abandoned. This paper will display the changes in urban morphology of the neighborhood through the years from 1877 to 2010. For site evaluation, the methodologies of Ashihara （1982） and Lynch＇s （1961） were used in order to analyze positive and negative spaces, as well as main street visual features, street and avenue directions, street circulation and street circulation path configuration. The results show that the focal nodes are a key factor for economic and social reactivation, with which, through urban activation of vacant lots and the traditional use of the streets as public space, is possible to generate centripetal development to restructure the neighborhood.

Cells encountering hypoxic stress conserve resources and energy by downregulating the protein synthesis. Here we demonstrate that one mechanism in this response is the translational repression of TOp mRNAs that encode components of the translational apparatus. This mode of regulation involves TSC and Rheb, as knockout of TSC1 or TSC2 or overexpression of Rheb rescued TOp mRNA translation in oxygen-deprived celts. Stress-induced translational repression of these mRNAs closely correlates with the hypophosphorylated state of 4E-Bp, a translational repressor. However, a series of 4E-Bp loss- and gain-of-function experiments disprove a cause-and- effect relationship between the phosphorylation status of 4E-Bp and the translational repression of TOp mRNAs under oxygen or growth factor deprivation. Furthermore, the repressive effect of anoxia is similar to that attained by the very efficient inhibition of mTOR activity by Torin 1, but much more pronounced than roptor or rictor knockouL Likewise, deficiency of raptor or rictor, even though it mildly downregulated basal translation efficiency of TOp mRNAs, failed to suppress the oxygen-mediated translational activation of TOp mRNAs. Finally, co-knockdown of TIA-1 and TIAR, two RNA-binding proteins previously implicated in translational repression of TOp mRNAs in amino acid-starved cells, failed to relieve TOp mRNA translation under other stress conditions. Thus, the nature of the proximal translational regulator of TOp m RNAs remains elusive.

＂Questionable＂ Behaviors and practices in Academic productivity in postgraduate Studies in Mexico

This paper reports the results acquired in a research work about ＂questionable＂ practices and behaviors in the academic production of researches and postgraduate social sciences and humanities studies of the programs that are appointed by the National program of Quality postgraduate Studies （pNpC, by its acronym in Spanish） in Mexico. Through a qualitative methodology, the authors interpreted some of the arguments that explain and/or justify certain practices in relation to doubled production, authorship, and coauthorship of academic products. In this paper, the authors present and analyze the results that they obtained after reviewing documents produced by professors and students of six postgraduate programs that are taught in two Mexican public universities. At the same time, the authors examine some of the practices that take place within said programs, given the institutional demands of improving finished studies efficiency. One of the hypotheses of this work is the demands that are imposed by external evaluations of academic processes on professors and their programs in order to reach desirable rates with the purpose of maintaining or increasing the levels of productivity, gives way to certain practices that must be analyzed. This work＇s theoretic framework is constituted by the contributions of career sociology and professional ethics.

Retinal and optic nerve diseases are degenerative ocular pathologies which lead to irreversible visual loss. Since the advanced therapies availability, cell-based therapies offer a new all-encompassing approach. Advances in the knowledge of neuroprotection, immunomodulation and regenerative properties of mesenchymal stem cells(MSCs) have been obtained by several preclinical studies of various neurodegenerative diseases. It has provided the opportunity to perform the translation of this knowledge to prospective treatment approaches for clinical practice. Since 2008, several first steps projecting new treatment approaches, have been taken regarding the use of cell therapy in patients with neurodegenerative pathologies of optic nerve and retina. Most of the clinical trials using MSCs are in Ⅰ/Ⅱ phase, recruiting patients or ongoing, and they have as main objective the safety assessment of MSCs using various routes of administration. However, it is important to recognize that, there is still a long way to go to reach clinical trials phase Ⅲ-Ⅳ. Hence, it is necessary to continue preclinical and clinical studies to improve this new therapeutic tool. This paper reviews the latest progress of MSCs in human clinical trials for retinal and optic nerve diseases.

In Vitropropagation and Recovery of Eight Apple and Two pear Cultivars Held in a Germplasm Bank

Eight apple (Malus x domestica) and two pear (pyrus communis) traditional cultivars of commercial interest were selected from the germplasm bank CIAM (Centro de Investigaciones Agrarias de Mabegondo, NW of Spain) one of the most important collections of The Iberian peninsula. In order to define the establishment protocol, apical buds 1.0 - 1.5 millimetre length of the selected cultivars were established in solid mineral medium Murashige and Skoog (MS) with modified vitamins (thiamine 10X) and supplemented with 1 mg·L-1 6-benzyladenine (BA), 0.2 mg·L-1 gibberellic acid (GA3) and 0.3 mg·L-1 of indole-3-butyric acid (IBA). The highest shoot initiation rates were achieved with apple cultivar “principe Grande” (55%) and “Barburiña” pear cultivar (50%);the lowest rate was obtained with apple cultivar “Camoesa” (11%). Initiation rates for shoots culture varied between 22% and 37% for the other cultivars. The cytokinins BA, zeatin, 2-isopentyladenine (2ip) and thidiazuron (TDZ) were tested at concentrations of 0, 0.25, 0.5 and 1 mg·L-1 in the absence or presence of IBA (0.1 mg·L-1) for the multiplication step. For all cultivars, the highest multiplication rates were obtained with medium supplemented with TDZ and also in media containing BA. The in vitro rooting success of shoots in medium with 0.1 mg·L-1 IBA without cytokinins, and the survival rate of plantlets acclimatized under greenhouse conditions depended on the genotype of the different cultivars. In vitro propagation of these ten traditional apple and pear cultivars in NW Spain has not previously been reported.
