A gain-of-function mutation in the fibroblast growth factor receptor 3 gene(FGFR3)results in achondroplasia(ACH),the most frequent form of *** activation of FGFR3 impairs bone formation and elongation and many signal transduction *** of new and relevant compounds targeting the FGFR3 signaling pathway is of broad importance for the treatment of ACH,and natural plant compounds are prime drug candidate ***,we found that the phenolic compound(-)-epicatechin,isolated from Theobroma cacao,effectively inhibited FGFR3’s downstream signaling *** analysis in an Fgfr3 mouse model showed that ciliary mRNA expression was modified and influenced significantly by the Indian hedgehog and pKA pathways.(-)-Epicatechin is able to rescue mRNA expression impairments that control both the structural organization of the primary cilium and ciliogenesis-related *** femurs isolated from a mouse model(Fgfr3^(Y367C/+))of ACH,we showed that(-)-epicatechin eliminated bone growth impairment during 6 days of ex vivo *** vivo,we confirmed that daily subcutaneous injections of(-)-epicatechin to Fgfr3^(Y367C/+) mice increased bone elongation and rescued the primary cilium defects observed in *** modification to the primary cilia promoted the typical columnar arrangement of flat proliferative chondrocytes and thus enhanced bone *** results of the present proof-of-principle study support(-)-epicatechin as a potential drug for the treatment of ACH.

Using presence signs to discriminate between similar species

The Lusitanian and the Mediterranean pine voles(Microtus lusitanicus Gerbe,1879 and Microtus duodecimcostatus de Selys-Longchamps,1839)are fossorial sister species and have an allopatric pattern of distribution in portugal,which includes a potential sympatry area in the centre of the *** present study aimed to determine the validity of using presence signs in the field for discrimination of the two species in an area of sympatry(Northern Alentejo)and the characteristics that achieve the best classification accuracy.A total of 175 trapping plots were sampled across the study *** to the set up of traps,ten presence signs were randomly selected for measure-ments of four variables:proportion of soil mounds,mean diameter of mounds,proportion of burrow openings and mean diameter of burrow *** the basis of a classification tree analysis,results showed that presence signs can be used to discriminate plots inhabited by one or the other species in the studied sympatry *** character-istic that most accurately enables species identification is the proportion of burrow openings:for every ten pres-ence signs found in a plot,if more than eight have an opening,then it is inhabited by ***(*** burrow openings with few or no mounds present);if eight or fewer have an opening,*** is present(*** mounds with few or no burrow openings).

participatory Early Warning Systems: Youth, Citizen Science,and Intergenerational Dialogues on Disaster Risk Reduction in Brazil

Building national people-centered early warning systems(EWS) is strongly recommended by the United Nations International Strategy for Disaster Reduction(UNISDR). Most of the scientific literature is critical of the conventional view of EWS as a linear model with a topdown approach, in which technological features are given more attention than human factors. It is argued that EWS should be people-centered, and used for risk prevention,with an emphasis on resilience, rather than only being triggered when a hazard occurs. However, both the UNISDR and the literature fail to say how a people-centered EWS should be built, and what steps are needed to put EWS into effect. This article examines the obstacles and measures required to promote people-centered EWS,with a focus on the situation in Brazil. After assessing the institutional vulnerability of EWS, we analyze some measures that can be taken to reduce institutional vulnerability,based on experiences with a participatory citizen science educational project that involved high school *** guidelines are developed for adopting a bottom-up approach towards achieving the four elements of EWS—risk knowledge, monitoring, communication of warnings,and response capability—with the help of school curricula.

A decrease of docosahexaenoic acid in testes of mice fed a high-fat diet is associated with impaired sperm acrosome reaction and fertility

Obesity is a major worldwide health problem that is related to most chronic diseases,including male *** to its wide impact on health,mechanisms underlying obesity-related infertility remain *** this study,we report that mice fed a high-fat diet(HFD)for over 2 months showed reduced fertility rates and increased germ cell apoptosis,seminiferous tubule degeneration,and decreased intratesticular estradiol(E2)and E2-to-testosterone ***,we also detected a decrease in testicular fatty acid levels,behenic acid(C22:0),and docosahexaenoic acid(DHA,22:6n-3),which may be related to the production of dysfunctional ***,we did not detect any changes in the frequency of seminiferous tubule stages,sperm count,or rate of in vitro ***,there was an increase in spontaneous and progesterone-induced acrosomal exocytosis(acrosome reaction)in spermatozoa from HFD-fed *** data suggest that a decrease in E2 and fatty acid levels influences spermatogenesis and some steps of acrosome biogenesis that will have consequences for ***,our results add new evidence about the adverse effect of obesity in male reproduction and suggest that the acrosomal reaction can also be affected under this condition.

Cost Optimisation for Minimizing the Visual Impact of Ornamental Stone Quarrying. A Case Study in Murcia Region

Quarrying of ornamental stone has adverse effects that are both visual and environmental. This paper aims to develop a methodology for minimising the costs associated with reducing the visual impact of ornamental stone quarrying. This study uses digital topographical maps of the study zone and a GpS and GIS application to calculate the extent of the area affected by quarrying activities for each altitude designated in the work plan and to calculate the extent of the potential visual impact. The results obtained applying the proposed methodology for the selected area suggested that the potential visual impact is minimal for an altitude of 520 metres, this being the optimal point for the observer. When altitude increases, the potential visual impact increases and the optimal point for the observer diminishes until the highest impact altitude (740 m) is readied. The optimal point that the exploitation should reach is that at which the values of the diagram generated by the (%) area of potential visual impact and area of exploitation (%) intersect. The methodology allows the optimal altitude to be determined for mining exploitations and helps assess the viability of a given exploitation from an environmental point of view.

Gender-Biased Regulation of Human IL-17-producing Cells <i>in Vitro</i>by peptides Corresponding to Distinct <i>HLA-DRB</i>1 Allele-Coded Sequences

Rheumatoid arthritis (RA) is associated with an HLA-DRB1-coded sequence motif called “shared epitope” (SE). To explore potential mechanisms of RA susceptibility, we analyze in vitro effect of peptides bearing different HLA-DR4 sequences on human peripheral blood-derived cells. Three 15-mer peptides were used: 65-79*0401 (HLA-DRB1*04:01- coded sequence SE motif, QKRAA);65-79*0402 (HLA-DRB1*04:02-coded sequence SE-negative motif, DERAA);65-79*0403 (HLA-DRB1*04:03-coded sequence SE-negative motif, QRRAE). We found that CD4 TH17 cells are regulated by peptide treatment with gender bias. In male-derived T cells, all peptide treatments significantly reduced TH17 cell differentiation in vitro when compared to no peptide treatment, and to female samples. TH17 differentiation in samples not treated with peptides, either in the presence or absence of TH17-polarizing cytokines, was higher in males than in females;however, in unfractionated pBMC after treatment with TH17 polarizing cytokines, IL-17A-positive cells were more abundant in females than in males. In addition, SE-positive females showed a significantly higher percentage of IL-17A-positive cells compared to SE-negative females. In conclusion, donor’s SE status and gender may both influence TH17 immune polarization.

New Reports of Spiders in Three Mexican Ant-Acacias

The richness of spider species on the foliage of Mexican ant-acacias in Los Tuxtlas (Veracruz), and Santiago pinotepa Nacional (Oaxaca) was documented. A total of 11 species, grouped into five families were found;in Los Tuxtlas five species representing two families were identified, whereas in Santiago pinotepa Nacional, the number of taxa was eight, belonging to five families. The richness of spiders in Santiago pinotepa Nacional was higher than that found in Los Tuxtlas. In both locations Nephila clavipes Linnaeus and Eustala illicita O. pickard-Cambridge, both from Araneidae, were found;these species had been previously reported as common inhabitants of Central American ant-acacias. The presence of E. illicita in Mexican myrmecophytic acacias was confirmed. A total of 10 species grouped into five families are reported for the first time inhabiting Mexican ant-acacias, increasing the richness of spider interactions documented in Mexico. This study showed that the occurrence of spiders in the Vachellia-pseudomyrmex mutualism system has been overlooked and is likely more common than what has been reported until now. This opens an opportunity for the study of the evolution of spider-ant-plant interactions.
