Spatial confirmation of termite mounds as Bio-geo indicator for groundwater occurrences using ground magnetic survey: A case study from Perambalur Region of Tamil Nadu, India

Termite mounds are conventionally surmised as one of the best bio-geological indicator for groundwater occurrence in places where they inhibit.Ground magnetic survey was carried out to prove the assumption about termite mounds as an important indicator for groundwater exploration.Occurrences of 18 Termite mounds were mapped using handheld GPS to identify the suitable trend for magnetic survey.After considering all the criteria such as power lines,fences and fracture system,termite mound 2,12 and 18 were selected in the NNE-SSW trend,and the ground magnetic survey was conducted in ESE-WNW direction which is perpendicular to the trend of the termite mounds using Proton Precession Magnetometer.Totally,99 samples were collected with 5 m sampling interval and 50 m profile interval over an area of 22500 sq.m.The magnetic values varied from–7363 nT to 898 nT with the mean of–331 nT.Processed map of reduction to Equator indicates the presence of NNE-SSW and WNW-ESE magnetic breaks.Analytical signal map designated the presence of magnetic low in the WNW-ESE direction which coincides spatially with the magnetic breaks.The magnetic profile lines have also brought to light the structurally weak zones.Causative body depth range was estimated using power spectrum and Euler method which are from 120 m to 40 m and100 m,respectively.The present study appreciably brings out the spatial relationship between the termite mounds and the hydro-fractures.This confirms the assertion with regard to termite mounds as an effective tool for groundwater exploration.

Three dimensional(3D)echocardiogram enables cardiologists to visua-lize suspicious cardiac structures in detail.In recent years,this three-dimensional echocardiogram carries important clinical value in virtual surgical simulation.However,this 3D echocardiogram involves a trade-off difficulty between accu-racy and efficient computation in clinical diagnosis.This paper presents a novel Flip Directional 3D Volume Reconstruction(FD-3DVR)method for the recon-struction of echocardiogram images.The proposed method consists of two main steps:multiplanar volumetric imaging and 3D volume reconstruction.In the crea-tion of multiplanar volumetric imaging,two-dimensional(2D)image pixels are mapped into voxels of the volumetric grid.As the obtained slices are discontin-uous,there are some missing voxels in the volume data.To restore the structural and textural information of 3D ultrasound volume,the proposed method creates a volume pyramid in parallel with theflip directional texture pyramid.Initially,the nearest neighbors of missing voxels in the multiplanar volumetric imaging are identified by 3D ANN(Approximate Nearest Neighbor)patch matching method.Furthermore,aflip directional texture pyramid is proposed and aggregated with distance in patch matching tofind out the most similar neighbors.In the recon-struction step,structural and textural information obtained from differentflip angle directions can reconstruct 3D volume well with the desired ***-pared with existing 3D reconstruction methods,the proposed Flip Directional 3D Volume Reconstruction(FD-3DVR)method provides superior performance for the mean peak signal-to-noise ratio(40.538 for the proposed method I and 39.626 for the proposed method II).Experimental results performed on the cardi-ac datasets demonstrate the efficiency of the proposed method for the reconstruc-tion of echocardiogram images.

Energy management and packet delivery rate are the important factors in ad hoc networks.It is the major network where nodes share the information without administration.Due to the mobility of nodes,maximum energy is spent on transmission of packets.Mostly energy is wasted on packet dropping and false route discovery.In this research work,Fuzzy Based Reliable Load Balanced Routing Approach(RLRA)is proposed to provide high energy efficiency and more network lifetime using optimal multicast route discovery mechanism.It contains three phases.In first phase,optimal multicast route discovery is initiated to resolve the link failures.In second phase,the link quality is estimated and set to threshold value to meet the requirements of high energy efficiency.In third phase,energy model is shown to obtain total energy of network after transmission of packets.A multicast routing is established Based on path reliability and fault tolerant calculation is done and integrated with multicast routing.The routes can withstand the malicious issues.Fuzzy decision model is integrated with propose protocol to decide the performance of network lifetime.The network simulation tool is used for evaluating the RLRA with existing schemes and performance of RLRA is good compared to others.

Bartter syndrome is a group of autosomal recessive renal tubular disorders;it has two types of presentation:antenatal and classic.The antenatal type presents as severe unexplained polyhydramnios in the second trimester.This is due to fetal urinary losses of sodium,chloride,and potassium,leading to fetal polyuria.The classic type presents in the late neonatal or infancy stage,with dehydration,dyselectrolytemia,failure to thrive,and nephrocalcinosis.Antenatal scans are normal in such cases.Type I and II Bartter syndrome presents in the antenatal period,whereas type IV has a classic presentation.We describe an unusual case of type IVa Bartter syndrome presenting in the antenatal period,with severe polyhydramnios.The initial diagnosis was made based on amniotic fluid chloride levels and later confirmed by performing a genetic test.Genetic testing is important for confirming diagnosis and prognostication regarding the condition.

Despite the promise of immunotherapy such as the immune checkpoint inhibitors(ICIs)anti-PD-1 and anti-CTLA-4 for advanced melanoma,only 26%–52%of patients respond,and many experience grade III/IV immune-related adverse events.Motivated by the need for an effective therapy for patients non-responsive to clinically approved ICIs,we have developed a novel nanoimmunotherapy that combines locally administered Prussian blue nanoparticle-based photothermal therapy(PBNP-PTT)with systemically administered agonistic anti-CD137 monoclonal antibody therapy(aCD137).PBNP-PTT was administered at various thermal doses to melanoma cells in vitro,and was combined with aCD137 in vivo to test treatment effects on melanoma tumor progression,animal survival,immunological protection against tumor rechallenge,and hepatotoxicity.When administered at a melanoma-specific thermal dose,PBNP-PTT elicits immunogenic cell death(ICD)in melanoma cells and upregulates markers associated with antigen presentation and immune cell co-stimulation in vitro.Consequently,PBNP-PTT eliminates primary melanoma tumors in vivo,yielding long-term tumor-free survival.However,the antitumor immune effects generated by PBNP-PTT cannot eliminate secondary tumors,despite significantly slowing their growth.The addition of aCD137 enables significant abscopal efficacy and improvement of survival,functioning through activated dendritic cells and tumor-infiltrating CD8^(+)T cells,and generates CD4^(+)and CD8^(+)T cell memory that manifests in the rejection of tumor rechallenge,with no long-term hepatotoxicity.This study describes for the first time a novel and effective nanoimmunotherapy combination of PBNP-PTT with aCD137 mAb therapy for melanoma.

In recent years,carbon quantum dots(CQDs)have been of great enthrallment in the fluorescent probe field.Carbon precursors with interesting physicochemical properties may be derived from natural sources.In this research,for the first time,coconut water has been used as a source of carbon to prepare blue fluorescent CQDs without any modification and functionalization.The preparation of CQDs is very simple and cost-effective,when compared with other conventional techniques.The reaction parameters temperature and pH were varied to obtain monodispersed spherical CQDs with an average grain size of ∼5 nm.The CQDs exhibit blue emission with a wavelength of 487 nm with an excitation wavelength of 390 nm and are used for Fe^(3+) ions detection.Fe^(3+) ions have been observed to quench the fluorescence intensity of the CQDs than other heavy metals.In the presence of Fe^(3+) ions,the fluorescent CQDs are quenched due to the interaction of CQDs and Fe^(3+) ions.A spectroscopic result shows that Fe^(3+) ions can be observed within a concentration of 0-700μM,as well as the detection limit is 0.30μM.The CQDs provide a unique pathway for potential application in the environmental monitoring of heavy metal ions.

Conserved regions of Plasmodium vivax potential vaccine candidate antigens in Sri Lanka:Conscious in silico analysis of prospective conformational epitope regions

Objective:To do mapping and modeling of conformational B cell epitope regions of highly conserved and protective regions of three merozoitecandidate vaccine proteins of Plasmodium vivax(P.vivax),ie.merozoite purface protein-1(PvMSP-1),apical membrane antigen-1 domainⅡ(PvAMA1-DⅡ)and regionⅡof the Duffy binding protein(PvDBPⅡ).and to analyze the immunogenie properties of these predicted epitopes.Methods:3-D structures of amino acid haplotypes from Sri Lanka(available in GeneBank)of PvMSP-1_(19)(n=27),PvAMA1-DⅡ(n=21)and PvDBPⅡ(n=33)were modeled.SEPPA,selected as the best online server was used for conformational epitope predictions,while prediction and moodeling of protein structuve and properties related to immunogenicity was carried out with Geno3D server.SCRATCH Protein Server,NetSurfP Server and standaloneroftware,Genious 5.4.4.Results:SEPPA revealed that regions of predicted conformational epitopes formed 4 clusters in PvMSP-I_(19),and 3 clusters each in PvAMA1-DⅡand PvDBPⅡ,all of which displayed a high degree of hydrophilicity,contained solveut exposed residues,displayed high probability of antigenicity and showed positive antigenic propensity values,that indicated high degree of immunogenicity.Conclusions:Findings of this study revealed and confirmed that different parts of the sequences of each of the conserved regions of the three selected potential vaccine candidate antigens of P.vivax are important with regard to conformational epitope prediction that warrants further laboratory experimental invertigations in in vivo animal models.

Clostridium difficile(C.difficile)infection(CDI)is the leading identifiable cause of antibiotic-associated diarrhea.While there is an alarming trend of increasing incidence and severity of CDI in the United States and Europe,superimposed CDI in patients with inflammatory bowel disease(IBD)has drawn considerable attention in the gastrointestinal community.The majority of IBD patients appear to contract CDI as outpatients.C.difficile affects disease course of IBD in several ways,including triggering disease flares,sustaining activity,and in some cases,acting as an"innocent"bystander.Despite its wide spectrum of presentations,CDI has been reported to be associated with a longer duration of hospitalization and a higher mortality in IBD patients.IBD patients with restorative proctocolectomy or with diverting ileostomy are not immune to CDI of the small bowel or ileal pouch.Whether immunomodulator or corticosteroid therapy for IBD should be continued in patients with superimposed CDI is controversial.It appears that more adverse outcomes was observed among patients treated by a combination of immunomodulators and antibiotics than those treated by antibiotics alone.The use of biologic agents does not appear to increase the risk of acquisition of CDI.For CDI in the setting of underlying IBD,vancomycin appears to be more efficacious than metronidazole.Randomized controlled trials are required to clearly define the appropriate management for CDI in patients with IBD.

Structure-assisted reverse chemical discovery approaches are becoming attractive due to target based screening.However,Small molecules(SMs)identified by reverse chemical genetic approach require robust phenotyping assays to study the plant responses.We optimized a screening method in rice seedlings using methyl viologen(MV)induced oxidative stress under high light conditions.This method was suitable for studying the efficacy of SM targeting oxidative stress related genes.Structure-assisted drug designing approach was used to identify SM targeting DREB2A transcription factor(TF).Rice seedlings treated with pronetalol showed susceptible phenotype when exposed to oxidative stress.Pronetalol inhibited the DREB2A activity and suppressed the expression of its target LEA7 and HSFA3 genes under oxidative stress.The assay is quite simple and robust and can be expanded to high throughput screening method for functional validation of TF.
