An improved high performance liquid chromatography-tandem mass spectrometry（LC-MS/MS） method has been developed for sensitive and rapid determination of albendazole（ABZ） and its active metabolite,albendazole sulfoxide（ABZSO）,in the positive ionization *** method utilized solid phase extraction（SPE） for sample preparation of the analytes and their deuterated internal standards（ISs） from100 μL human *** chromatography was carried out on Hypurity C_（18） column using acetonitrile-2.0 mM ammonium acetate,pH 5.0（80：20,v/v） as the mobile *** assay exhibited a linear response over the concentration range of 0.200-50.0 ng/mL for ABZ and 3.00-600 ng/mL for *** recoveries of the analytes and ISs ranged from 86.03%-89.66%and 89.85%-98.94%,*** effect,expressed as IS-normalized matrix factors,ranged from 0.985 to 1.042 for the both *** method was successfully applied for two separate studies in healthy subjects using single dose of 400 mg conventional tablets and 400 mg chewable ABZ tablets,respectively.

Previously, we reported that Y_6, a new epigallocatechin gallate derivative, is efficacious in reversing doxorubicin(DOX)–mediated resistance in hepatocellular carcinoma BEL-7404/DOX cells. In this study, we evaluated the efficacy of Y_6 in reversing drug resistance both in vitro and in vivo by determining its effect on the adenosine triphosphate-binding cassette protein B1 transporter(ABCB1 or P-glycoprotein, P-gp). Our results showed that Y_6 significantly sensitized cells overexpressing the ABCB1 transporter to anticancer drugs that are ABCB1 substrates. Y_6 significantly stimulated the adenosine triphosphatase activity of ABCB1. Furthermore, Y_6 exhibited a higher docking score as compared with epigallocatechin gallate inside the transmembrane domain of ABCB1. In addition, in the nude mousetumor xenograft model, Y_6(110 mg/kg, intragastric administration), in combination with doxorubicin(2 mg/kg, intraperitoneal injection), significantly inhibited the growth of BEL-7404/DOX cell xenograft tumors, compared to equivalent epigallocatechin gallate. In conclusion, Y_6 significantly reversed ABCB1-mediated multidrug resistance and its mechanisms of action may result from its competitive inhibition of the ABCB1 drug efflux function.

A simple,rapid and sensitive ultra performance liquid chromatography-tandem mass spectrometry（UPLC-MS/MS） method has been developed for the simultaneous determination of cilostazol and its pharmacologically active metabolite 3,4-dehydro cilostazol in human plasma using deuterated analogs as internal standards（ISs）.Plasma samples were prepared using solid phase extraction and chromatographic separation was performed on UPLC BEH C18（50 mm × 2.1 mm.1.7 μm） *** method was established over a concentration range of 0.5-1000 ng/mL for cilostazol and 0.5-500 ng/mL for 3.4-dehydro ***- and inter-batch precision（%CV） and accuracy for the analytes were found within 0.93-1.88 and 98.8-101.7% for cilostazol and 0.91-2.79 and 98.0-102.7% for the metabolite *** assay recovery was within 95-97% for both the analytes and internal *** method was successfully applied to support a bioequivalence study of 100 mg cilostazol in30 healthy subjects.

Lemon oil (LO), also known as Citrus limonum is a highly volatile essential oil (EO) with potential therapeutic properties like anti-oxidative, anti-proliferative, anti-fungal and anti-cancerous. However, the efficacy of LO is limited due to its physiological factors such as high volatility, poor stability (particularly sensitive to sunlight) and quick degradability upon exposure. To overcome these challenges, we formulated lemon oil loaded nanoemulsion system (LO-NE) (oil-in-water), using aqueous titration method. The formulation comprised of lemon oil (LO), Tween 80 and ethanol as oil, surfactant and co-surfactant phases respectively. The existence zone of NE was established by constructing pseudo-ternary phase diagrams using different concentrations of LO, surfactant and co-surfactant (Smix). The quantitative estimation of LO was performed using a high throughput gas chromatography, revealing the presence of various compounds like Limonene, Alpha-Pinene and Linalyl acetate followed by the estimation of total phenolics and flavonoid content. The characterization of LO-NE indicated the particle size of 60 ± 2.5 nm along with the polydispersity index of 0.125 and zeta potential of −14.9 mV. The size range of the NE particles dispersed in the colloidal system was further verified by TEM micrograph which shows size range between 46.2 - 104.7 nm. All the anti-oxidant assays outcomes exhibited the higher activity of LO-NE in comparison to LO alone with lower IC50 values. The release kinetics statistical data showed that LO-NE had a sustained release and followed the Higuchi’s model in comparison to burst release of LO alone. Lastly, the stability analysis of the optimised formulation (LO-NE) and LO was estimated through antioxidant assay and subjecting them for thermodynamic stability after 6 months. A new technique using signal flow graph for conversion of ladder based filter into CFOA based filter has been proposed. The proposed technique converts the existing LC ladder based filter into CFOA in low pass and high pass configuration. The design of low pass filter and high pass filter has been realized using the proposed technique. The proposed configuration is implemented using CFOA as an active device and all the capacitors are grounded. Simulation has been carried out using simulation software I-cap. The simulation results have been demonstrated and discussed.

Imaging Review of Adolescent Tibial Tuberosity Fractures

Adolescent tibial tuberosity injuries are infrequent fractures usually seen in physically active adolescent males. Powerful contraction of the knee extensors by sudden acceleration or deceleration of the quadriceps muscle can result in avulsion fractures of the tibial tuberosity apophysis. In late puberty, as the growth plate closes, it is transiently replaced by fibrocartilaginous elements. This transition causes a period of weakened tensile strength, which predisposes the tibial tuberosity to traction injury. Classification of tibial tuberosity fractures includes types I-V with added A and B subsets to types I, II and III. Multidetector computed tomography (MDCT) is a useful tool to more accurately classify complex, higher grade adolescent tibial tuberosity avulsion fractures when compared to plain film. This aids in preoperative planning and, therefore, results in improved treatment and management.

目的:观察白内障合并翼状胬肉患者行联合手术后的疗效。方法:对22例(平均年龄:59.05±8.70岁)并发白内障和翼状胬肉(长度2-5mm)患者进行前瞻性单中心研究,随访时间为3mo~1a。测定术前和术后3mo的平均角膜曲率(Kmean)、平均散光和最佳矫正视力(LogMAR)。观察术前角膜曲率和翼状胬肉大小与预测误差的关系。结果:共18例(81.8%)患者PE1.00D的患者。手术前后眼轴长度无差异(P=0.77)。Kmean从术前42.994±1.536增加到术后43.324±1.479(P=0.105)。术后角膜散光由术前2.09±0.789D降至术后0.523±0.277D(P<0.05)。BCVA(LogMAR)从术前1.007±0.402显著提高到术后0.024±0.062(P<0.05)。角膜曲率变化与预测误差无相关性(r=-0.29,P=0.19)。翼状胬肉大小与预测误差无相关性(r=0.2997,P=0.17)。结论:超声乳化与折叠式人工晶状体植入联合自体结膜移植术的应用是一种安全有效的手术方法。

Recognizing the composition and modulation of the microbiome, a viable therapeutic tool for multi-targeted therapy is a new strategy that has recently been explored. Glucosamine (GS) is being studied for its prebiotic potential in addition to being the most abundant and naturally occurring amino monosaccharide. The current study focuses on glucosamine’s prebiotic potential by assessing the stability of various GS concentrations (1% - 5%) in the gastrointestinal tract (GIT) and its ability to be fermented by the gut microbiota. The results showed that GS stimulated the most growth in L. acidophilus even after a longer incubation time than B. bifidum and L. acidophilus growth was concentration-dependent, with maximum growth at 3% with a simultaneous decrease in pH (5.6 - 1.7). The decrease in GS concentration with time also represented the growth of bacterial species, demonstrating the species’ utilization of GS. Furthermore, at 3%, GS also represented the prebiotic index of 1.9. In addition, the concentration of GS in various simulated GIT fluids was estimated in both fast and fed conditions to examine GS stability at various levels in the gut. The results showed that GS remained unaffected and non-digestible in all of the simulated GIT fluids (salivary, gastric, intestinal, and colonic), but there was a slight decrease in GS concentration (2.8%) in the fasted state of gastric fluid due to low pH levels (1.6). As a result, the findings are conclusive and suggest that GS possesses prebiotic properties.
