Nitric-oxide enriched plasma-activated water inactivates 229E coronavirus and alters antiviral response genes in human lung host cells

The ongoing pandemic caused by the novel coronavirus,SARS-CoV-2,is influencing global ***,there is a major threat of future coronaviruses affecting the entire world in a similar,or even more dreadful,***,effective and biocompatible therapeutic options against coronaviruses are urgently *** address this challenge,medical specialists require a well-informed and safe approach to treating human coronaviruses(HCoVs).Herein,an environmental friendly approach for viral inactivation,based on plasma technology,was considered.A microwave plasma system was employed for the generation of the high amount of gaseous nitric oxide to prepare nitric oxide enriched plasma-activated water(NO-PAW),the effects of which on coronaviruses,have not been reported to *** determine these effects,alpha-HCoV-229E was used in an experimental *** found that NO-PAW treatment effectively inhibited coronavirus infection in host lung cells,visualized by evaluating the cytopathic effect and expression level of spike ***,NO-PAW showed minimal toxicity towards lung host cells,suggesting its potential for therapeutic ***,this new approach resulted in viral inactivation and greatly improved the gene levels involved in host antiviral ***,our findings provide evidence of an initiation point for further progress toward the clinical development of antiviral treatments,including such coronaviruses.

MHD Stagnation Point Flow and Heat Transfer over a Permeable Surface

The steady two-dimensional, laminar flow of a viscous, incompressible, electrically conducting fluid near a stagnation point with heat transfer over a permeable surface in the presence of a uniform magnetic field is considered. Taking suitable similarity variables, the governing boundary layer equations are transformed to ordinary differential equations and solved numerically by Shooting method. The effects of the suction parameter, the magnetic parameter, the Prandtl number and the Eckert number are studied on the velocity and temperature distributions.

MHD Slip Flow past a Shrinking Sheet

An analysis is made for the steady two-dimensional laminar boundary layer flow of a viscous, incompressible, electrically conducting fluid near a stagnation point past a shrinking sheet with slip in the presence of a magnetic field. The governing boundary layer equations are transformed to ordinary differential equations by taking suitable similarity variables and solved numerically by Shooting method. The effects of the various parameters such as velocity ratio parameter, slip parameter, Prandtl number, Eckert number and magnetic parameter for velocity and temperature distributions have been discussed in detail through graphical representation.

Improvement in cryosurvival of buffalo bull (Bubalus bubalis) sperm by altering freezing rate within critical temperature range

Objective:To optimize the cryopreservation of buffalo bull semen by altering freezing rates within critical temperature range (4℃ to -60℃). Methods: A total of 20 ejaculates each from 5 Murrah buffalo bulls were cryopreserved using programmable biofreezer in 2 phases. In the 1st phase, 9 freezing rates were applied at -2, -5, -10, -20, -30, -40, -50, -60 or -4℃/min (control) from 4℃ to -15℃;at -40℃/min from -15℃ to -60℃. In the 2nd phase, a fixed freezing rate was applied at -30℃/min from 4℃ to -15℃. Six freezing rates were applied at -10, -20, -30, -40 (control), -50 or -60℃/min from -15℃ to -60℃. The freezing from -60℃ to -140℃ were fixed at -50℃/min in both the phases. Post thaw semen quality was assessed in terms of motility, viability, membrane integrity (hypo-osmotic swelling test), sperm abnormalities, and active mitochondria. Data were arc sine transformed and analyzed through one-way analysis of variance using SPSS software. Results: In the 1st phase, percent individual motility, progressive motility and viability were similar among various protocols. Percent hypo-osmotic swelling reactive sperm was higher with freezing at -30℃/min. In the 2nd phase, percent individual motility, viability and hypo-osmotic swelling reactive sperm was higher with freezing at -50℃/min. Sperm head abnormalities were lower at -30℃/min in the 1st phase, but were similar among the protocols of the 2nd phase. Percent active mitochondria were higher at -30℃/min in the 1st phase and at -50℃/min in the 2nd ***:The optimum post thaw semen quality of buffalo bull could be obtained by applying freezing rate at -30℃/min (4℃ to -15℃) and at -50℃/min (-15℃ to -140℃), followed by plunging of straws in into liquid nitrogen for storage.

Taking suitable similarity variables, the governing boundary layer equations are transformed to ordinary differential equations and solved numerically by a perturbation technique for a small magnetic parameter. The effects of various parameters such as unsteadiness parameter, velocity parameter, magnetic parameter, Prandtl number and Eckert number for velocity and temperature distributions along with local Skin friction coefficient and local Nusselt number have been discussed in detail through numerical and graphical representations.

Decadal glacial lake changes in the Koshi basin, central Himalaya, from 1977 to 2010, derived from Landsat satellite images

Changes in glacial lakes and the consequences of these changes, particularly on the development of water resources and management of glacial lake outburst flood(GLOF) risk, has become one of the challenges in the sustainable development of high mountain areas in the context of global warming. This paper presents the findings of a study on the distribution of, and area changes in, glacial lakes in the Koshi basin in the central *** on the number of glacial lakes and their area was generated for the years 1977, 1990, 2000, and 2010 using Landsat satellite images. According to the glacial lake inventory in 2010, there were a total of 2168 glacial lakes with a total area of 127.61 km^2 and average size of 0.06 km^2 in the Koshi basin. Of these,47% were moraine dammed lakes, 34.8% bedrock dammed lakes and 17.7% ice dammed lakes. The number of glacial lakes increased consistently over the study period from 1160 in 1977 to 2168 in 2010, an overall growth rate of 86.9%. The area of glacial lakes also increased from 94.44 km^2 in 1977 to 127.61 km^2 in 2010, a growth rate of 35.1%. A large number of glacial lakes in the inventory are small in size(≤ 0.1km^2). End moraine dammed lakes with area greater than 0.1 km^2 were selected to analyze the change characteristics of glacial lakes in the basin. The results show that, in 2010, there were 129 lakes greater than 0.1 km^2 in area; these lakes had a total area of 42.92km^2 in 1997, increasing to 63.28 km^2 in 2010. The distribution of lakes on the north side of the Himalayas(in China) was three times higher than on the south side of the Himalayas(in Nepal).Comparing the mean growth rate in area for the 33 year study period(1977-2010), the growth rate on the north side was found to be a little slower than that on the south side. In the present work,multi walled carbon nanotubes(MWCNT)reinforced magnesium(Mg)matrix composite was fabricated by friction stir processing(FSP)with an aim to explore its mechanical and electrochemical *** observations showed that the thickness of the produced composite layer was in the range of 2500μ*** resulted uniform distribution of CNT near the surface while agglomerated layers in the *** refinement of Mg achieved by FSP improved the hardness but significant enhancement in the hardness value was observed for FSPed MWCNT/Mg *** polarization studies revealed that the increase in corrosion current density was observed for MWCNT/Mg composite compared with grain refined Mg and pure Mg,implying the significance of secondary phase(MWCNT)in decreasing the corrosion resistance of the composite.

In VitroStudies on Sida Cordifolia Linn for Anthelmintic and Antioxidant Properties

The present study was undertaken to evaluate in-vitro antioxidant and anthelmintic activity of ethanolic and aqueous extract from whole plant Sida cordifolia Linn (Malvaceae). The antioxidant activities are evaluated by various antioxidant assays like α, α-Diphenyl-β-picrylhydrazyl (DPPH) free radical scavenging, total reducing power, nitric oxide scavenging and hydrogen peroxide scavenging. The various antioxidant activities were compared to standard antioxidants such as ascorbic acid. The antioxidant activity of ethanolic extract is almost quantitatively equivalent to that of the standards used, ascorbic acid. The further anthelmintic activity of whole plant is investigated through using Indian earthworm (Pheretima posthuma) showed that it is one of the most important local medicinal plants both for ritual and ethnomedical practices. Various concentrations of ethanol and aqueous extract (10, 20, 30, 40 mg/ml) of whole plant of Sida cordifolia Linn were tested in the bioassay, which involve determination of time of paralysis of the worms. Albendazole was included as reference standard. The most activity was observed with aqueous extract as compared to standard drug. The results from the above studies indicate that plant Sida cordifolia Linn. possesses potent antioxidant and anthelmintic activity.

Studies on Trans-Generational Transcriptional Silencing of cry1AcGene in Tobacco Transgenics

Developing transgenics that express high levels of Cry1Ac protein, and at the same time, are phenotypically normal, has not been an easy task to achieve. It has been routinely observed that most of the transgenic plants that survive, show no or extremely low levels of Cry1Ac protein. However, all of these plants do express the selectable marker, nptII gene. In the present study, we record an interesting observation of how one of the genes (cry1Ac) on a single T-DNA fragment is selectively silenced, keeping the expression of the other gene (nptII) intact. Further, this silenced state is inherited.

Fabrication and Characterizations of Mechanical Properties of Al-4.5%Cu/10TiC Composite by In-situMethod

Addition of reinforcement such as TiC, SiC, Al2O3, TiO2, TiN, etc. to Aluminium matrix for enhancing the mechanical properties has been a well established fact. In-situ method of reinforcement of the Aluminium matrix with ceramic phase like Titanium Carbide (TiC) is well preferred over the Ex-situ method. In the present investigation, Al-Cu alloy (series of 2014 Aluminium alloy) was used as matrix and reinforced with TiC using In-situ process. The Metal Matrix Composite (MMC) material, Al-4.5%Cu/10%TiC developed exhibited higher yield strength, ultimate strength and hardness as compared to Al-4.5%Cu alloy. Percentage increase in yield and ultimate tensile strengths were reported to be about 15% and 24% respectively whereas Vickers hardness increased by about 35%. The higher values in hardness indicated that the TiC particles contributed to the increase of hardness of matrix. Fractured surface of the tensile specimen of the composite material indicated presence of dimpled surface, indicating thereby a ductile type of fracture. During the fabrication of composite, reaction products such as Al3Ti, Al2Cu and Al3C4 were identified with various morphologies and sizes in metal matrix.
