Field scale interaction and nutrient exchange between surface water and shallow groundwater in the Baiyang lake region,North China Plain

Fertilizer input for agricultural food production, as well as the discharge of domestic and industrial water pollutants, increases pressures on locally scarce and vulnerable water resources in the North China Plain. In order to：（a） understand pollutant exchange between surface water and groundwater,（b） quantify nutrient loadings, and（c） identify major nutrient removal pathways by using qualitative and quantitative methods, including the geochemical model PHREEQC） a one-year study at a wheat（Triticum aestivum l.） and maize（Zea mays l.） double cropping system in the Baiyang lake area in Hebei Province, China, was undertaken. The study showed a high influence of low-quality surface water on the shallow aquifer. Major inflowing pollutants into the aquifer were ammonium and nitrate via inflow from the adjacent Fu River（up to 29.8 mg/l NH4-N and 6.8 mg/l NO3-N）, as well as nitrate via vertical transport from the field surface（up to 134.8 mg/l NO3-N in soil water）. Results from a conceptual model show an excess nitrogen input of about 320 kg/ha/a. Nevertheless,both nitrogen species were only detected at low concentrations in shallow groundwater,averaging at 3.6 mg/l NH4-N and 1.8 mg/l NO3-N. Measurement results supported by PHREEQC-modeling indicated cation exchange, denitrification, and anaerobic ammonium oxidation coupled with partial denitrification as major nitrogen removal pathways. Despite the current removal capacity, the excessive nitrogen fertilization may pose a future threat to groundwater quality. Surface water quality improvements are therefore recommended in conjunction with simultaneous monitoring of nitrate in the aquifer, and reduced agricultural N-inputs should be considered.

Quantities of comorbidities affects physical, but not mental health related quality of life in type 1 diabetes with confirmed polyneuropathy

BACKGROUND A large number of adults with long-term type 1 diabetes are affected by symmetrical peripheral neuropathy.These complications increase socioeconomic expenses and diminish the individual quality of life.The 36-Item Short Form Health Survey(SF-36)is a generic patient reported questionnaire,measuring mental and physical health related quality of life.We hypothesized that diabetic neuropathy would decrease physical and mental quality of life measured with SF-36,and that clinical appearance may be associated with the decline.AIM To investigate if diabetic neuropathy would decrease physical and mental quality of life measured with SF-36,and if clinical appearance may be associated with the decline.METHODS Forty-eight adults[age 50±9 years,10 females,disease duration 32(14-51)years]with verified diabetic symmetrical peripheral neuropathy and 21 healthy participants(age 51±6 years,6 females)underwent standardised nerve conduction testing and completed the SF-36 questionnaire.Furthermore,disease duration,number of comorbidities,both diabetes related and nondiabetes related,vibration perception threshold,number of hypoglycaemic events,HbA1c and administration way of insulin was notified.RESUlTS In comparison to healthy subjects,patients’mental composite score was not significantly diminished(51.9±8.9 vs 53.1±5.5,P=0.558),while the physical composite score was(46.3±11.7 vs 54.6±3.3,P=0.002).As expected,the overall physical health related symptoms in patients were associated to total number of comorbidities(Plation to diabetes(P=0.0002)and HbA1c(P=0.005)as well as comorbidities not related to diabetes(P=0.0006).CONClUSION The finding of this study emphasises the importance of focusing on quality of life in adults with diabetes and especially in those with multiple comorbidities as well as the possibility of HbA1c as a biomarker for severe complication.

Background: A marked decrease in malaria-related deaths worldwide has been attributed to the administration of effective antimalarials against Plasmodium falciparum. However, the continuous spread of P. falciparum resistance to anti-malarial drugs is raising a serious problem in controlling Malaria to the vulnerable children’s immune system. In recent studies, Plasmodium falciparum Kelch 13 propeller gene (Pfk13) has been reported to develop resistance to artemisinin in South Asia. In this study, we checked Plasmodium falciparum chloroquine resistance transporter gene (Pfcrt) involved in chloroquine (CQ) resistance. Method: In this study, archived 280 samples were collected from Alupe primary school children in Busia, Western Kenya from May, 2016 to November, 2016. Genomic DNA was extracted using the MightyPrep reagent. The samples were investigated for P. falciparum positivity out of which 67 of them tested positive giving a prevalence rate of 24%. The sixty-seven were subjected to PCR amplification for the molecular marker resistance to Pfcrt. After PCR amplification, the amplicons were purified and sequenced using Sanger Sequencing. The sequence data were analyzed using BioEdit software to identify point mutations. Results: 14 samples sequences were analyzed on Bioedit software giving the following amino acid changes F76C, Y66H, l70A, Y58C, T59V, V65I, P67l, T81l, Y60S, Y66S, P67T and I71F). New mutations have been reported at position 76 leading to an amino acid change, one of Pfcrt gold standard biomarkers. However, amino acid changes Y66H, l70A, Y58C, T59V, V65I, P67l, T81l, Y60S, Y66S, P67T and I71F are newly reported giving an increase in Pfcrt prevalence of concern from zero to 5.0%. A phylogenetic evolutionary relationship was constructed as shown below. Generally, the results showed a continuous resistance of P.falciparum to Pfcrt which calls for robust continuous monitoring and surveillance. Deployment of Pre-Industrial Autonomous Microbe Sensor in Saudi Arabia’s Injection Seawater System

Microbial growth in water injection systems can lead to many problems, including biofouling, water quality deterioration, injectivity loss, microbial corrosion, and reservoir formation damage. Monitoring of microbial activities is required in any mitigation strategy, enabling operators to apply and adjust countermeasures properly and in due time. In this study, the pre-industrial autonomous microbe sensor (AMS) was constructed with technical improvements from the prototype for increased sensitivity, durability, robustness, and maintainability. The pre-industrial AMS was lab validated, field proven, and deployed at critical locations of seawater injection network for automated detection of microorganisms under the Saudi Arabia’s harsh environment. An excellent correlation between AMS measurement data (fluorescence count) and actual count of microbial cell number under microscope was established (coefficient of determination, R2 > 0.99) for converting AMS fluorescence count to cell numbers (cell ml-1) in the injection seawater. The pre-industrial AMS only required monthly maintenance with solutions refill, and was able to cope with hot summer months even without protection in an air-conditioned shelter. The study team recommended wider deployment of the online AMS for real-time monitoring of bacteria numbers in the various strategic locations in Saudi Aramco’s complex seawater injection network, as an integral component of pipeline corrosion and leak mitigation program.

Human African trypanosomiasis (HAT), commonly known as sleeping sickness is one of the neglected tropical diseases (NTDs), which is fatal if left untreated. Its diagnosis is a challenge since the signs and symptoms of the primary phase are not specific, the existing diagnostic methods have low sensitivity and specificity, and the available drugs have some toxicity. New, robust, and cost-effective techniques are needed for the early identification of parasites. This study aimed to assess the sensitivity and specificity of two different types of polyclonal antibodies against T. b. gambiense using antigen detection ElISA. Polyclonal antibodies against the expressed proteins Tbg I2 and Tbg I17 were produced using New Zealand white rabbits. The antibody titer measured was greater than 32 g/l after the 3rd immunization for the expressed protein Tbg I2. For the expressed protein Tbg I17, the antibody titer measured was greater than 32 g/l after the 4th immunization. The sensitivity and specificity of the Tbg I2 polyclonal antibody confirmed with Polymerase Chain Reaction (PCR) as gold standard were respectively 89.5% and 80.6%, while for the Tbg I17 polyclonal antibody, the sensitivity and specificity were respectively 92.1% and 88.9%. The area under the curve for the Tbg I2 polyclonal antibody was 0.90 ± 0.032, while for the Tbg I17 polyclonal antibody, the area under the curve was 0.92 ± 0.0. The Tbg I17 polyclonal antibody produced in New Zealand white rabbits has good sensitivity and good specificity;it can be successfully used in the diagnosis of HAT.

【目的】探究非复制型尿嘧啶营养缺陷型弓形虫(NRTUAs)感染雏鸡脾脏的转录组学变化,明确NRTUAs对雏鸡脾脏先天性免疫的调节效果,为研发家禽抗弓形虫疫苗提供理论依据。【方法】分别以NRTUAs和磷酸盐缓冲液(PBS)感染14日龄雏鸡,感染后第3 d采集脾脏组织样品进行转录组测序,筛选出NRTUAs组与PBS组间的差异表达基因(DEGs),然后进行GO功能注释、KEGG信号通路富集及蛋白调控网络分析,并通过实时荧光定量PCR验证转录组测序的准确性。【结果】与PBS组相比,从感染NRTUAs的雏鸡脾脏中筛选出499个DEGs(286个为上调DEGs,213个为下调DEGs),且NRTUAs组中上调表达的DEGs多于下调表达的DEGs。GO功能注释分析发现,DEGs注释在生物过程(Biological process,BP)、细胞组分(Cellular component,CC)和分子功能(Molecular function,MF)三大类别的58个功能条目上,主要涉及端粒蛋白定位建立正调控、蛋白稳定、含伴侣蛋白T-复合物、内质网腔、内质网、内质网膜组成部分、纺锤中央区及未折叠蛋白结合等功能条目;KEGG信号通路富集分析显示,DEGs主要富集在内质网蛋白加工通路、RNA转运、细胞周期、钙信号通路、黏着斑、细胞衰老、神经活性配体—受体相互作用、细胞因子—细胞因子受体相互作用通路及氨基酸生物合成等通路中;蛋白互作网络分析结果表明,DEGs编码蛋白PlK1、CCNB1、CDK1、CDC20、CCNA2和NDC1均在细胞周期通路上调表达。实时荧光定量PCR验证结果显示,只有2个DEGs(CXCl12和VPREB3)呈上调表达,其余8个DEGs呈下调表达,与转录组测序结果基本一致。【结论】感染NRTUAs后雏鸡脾脏免疫相关基因表达上调,且多数DEGs富集在内质网蛋白合成与细胞周期等相关信号通路上,脾脏细胞活动明显增强,即通过加快建立细胞连接及启动先天性免疫反应而积极对抗弓形虫入侵。
