To Analyze the Sensitivity of RT-PCR Assays Employing S Gene Target Failure with Whole Genome Sequencing Data during Third Wave by SARS-CoV-2 Omicron Variant

Introduction: Omicron is a highly divergent variant of concern (VOCs) of a severe acute respiratory syndrome SARS-CoV-2. It carries a high number of mutations in its spike protein hence;it is more transmissible in the community by immune evasion mechanisms. Due to mutation within S gene, most Omicron variants have reported S gene target failure (SGTF) with some commercially available PCR kits. Such diagnostic features can be used as markers to screen Omicron. However, Whole Genome Sequencing (WGS) is the only gold standard approach to confirm novel microorganisms at genetically level as similar mutations can also be found in other variants that are circulating at low frequencies worldwide. This Retrospective study is aimed to assess RT-PCR sensitivity in the detection of S gene target failure in comparison with whole genome sequencing to detect variants of Omicron. Methods: We have analysed retrospective data of SARS-CoV-2 positive RT-PCR samples for S gene target failure (SGTF) with TaqPath COVID-19 RT-PCR Combo Kit (ThermoFisher) and combined with sequencing technologies to study the emerged pattern of SARS-CoV-2 variants during third wave at the tertiary care centre, Surat. Results: From the first day of December 2021 till the end of February 2022, a total of 321,803 diagnostic RT-PCR tests for SARS-CoV-2 were performed, of which 20,566 positive cases were reported at our tertiary care centre with an average cumulative positivity of 6.39% over a period of three months. In the month of December 21 samples characterized by the SGTF (70/129) were suggestive of being infected by the Omicron variant and identified as Omicron (B.1.1.529 lineage) when sequence. In the month of January, we analysed a subset of samples (n = 618) with SGTF (24%) and without SGTF (76%) with Ct values Conclusions: During the COVID-19 pandemic, it took almost more than 15 days to diagnose infection and identify pathogen by sequencing technology. Molecular Dynamics(MD)simulation for computing Interatomic Potential(IAP)is a very important High-Performance Computing(HPC)*** simulation on particles of experimental relevance takes huge computation time,despite using an expensive high-end *** computing,a combination of the Field Programmable Gate Array(FPGA)and a computer,is proposed as a solution to compute MD simulation *** such heterogeneous computation,communication between FPGA and Computer is *** such MD simulation,explained in the paper,is the(Artificial Neural Network)ANN-based IAP computation of gold(Au_(147)&Au_(309))*** simulation calculates the forces between atoms and the total energy of the chemical *** work proposes the novel design and implementation of an ANN IAP-based MD simulation for Au_(147)&Au_(309) using communication protocols,such as Universal Asynchronous Receiver-Transmitter(UART)and Ethernet,for communication between the FPGA and the host *** improve the latency of MD simulation through heterogeneous computing,Universal Asynchronous Receiver-Transmitter(UART)and Ethernet communication protocols were explored to conduct MD simulation of 50,000 *** this study,computation times of 17.54 and 18.70 h were achieved with UART and Ethernet,respectively,compared to the conventional server time of 29 h for Au_(147) *** results pave the way for the development of a Lab-on-a-chip application.

Well productivity in the Asmari carbonate formation of southwest Iran has decreased in recent years as a result of production *** production rate must be maintained below 1500 STB/day to prevent water *** this study,a gas lift well is modeled using data from one of the producing wells of this *** analysis is performed using lift-gas injection rates and wellhead pressures at different reservoir pressures and water cut conditions to optimize *** aspects are considered to optimize the artificial gas injection rates at different tubing head pressures and water cut *** the lift-gas injection rate from 0.4 MMscf/day to 1 MMscf/day enhances the oil production rate by 37.71%and 43.89%for 10%and 30%water cut conditions,*** injection rates of 5.2 MMscf/day and 5.4 MMscf/day are determined to be economically optimal for 30%water cut with tubing head pressures of 260 psig and 270 psig,respectively.

Predictive active control of building structures using LQR and artificial intelligence

This study presents a neural network-based model for predicting linear quadratic regulator(LQR)weighting matrices for achieving a target response *** on the expected weighting matrices,the LQR algorithm is used to determine the various responses of the *** responses are determined by numerically analyzing the governing equation of motion using the state-space *** training a neural network,four input parameters are considered:the time history of the ground motion,the percentage reduction in lateral displacement,lateral velocity,and lateral acceleration,Output parameters are LQR weighting *** study the effectiveness of an LQR-based neural network(LQRNN),the actual percentage reduction in the responses obtained from using LQRNN is compared with the target percentage ***,to investigate the efficacy of an active control system using LQRNN,the controlled responses of a system are compared to the corresponding uncontrolled *** trained neural network effectively predicts weighting parameters that can provide a percentage reduction in displacement,velocity,and acceleration close to the target percentage *** on the simulation study,it can be concluded that significant response reductions are observed in the active-controlled system using ***,the LQRNN algorithm can replace conventional LQR control with the use of an active control system.

Colorimetric Sensor for Detection of Tabun Mimic Diethyl Cyanophosphonate

Herein,a colorimetric chemosensor （probe 7） was designed and synthesized for sensing diethyl cyanophosphonate （DCNP）,a Tabun nerve agent *** sensitive,selective,fast response and photostable probe 7 in dimethylsulfoxide （DMSO）responded to DCNP at low concentrations with visual naked eye color change from pink to pale yellow and displayed unresponsive behaviour against various interferences except for copper ions,which can be di?erentiated by adding ethylene diamine where the yellow color persisted in the case of DCNP but turned to pink in the case of copper ***,the yellow aqueous solution of probe 7 at pH 10 and 12 turned colorless against the response of *** limit of detection（LOD） of probe 7 was found to be 50 nmol/L in an aqueous basic solution at pH 10 for the detection of *** formation of the cyclized product was elucidated by1H and31P NMR nuclear magnetic resonance (1H NMR,31P NMR),Fourier transform infrared spectroscopy （FTIR）,and high-resolution mass spectrometry （HRMS） ***?lter paper and silica plate loaded with probe 7 were fabricated for real-time and on-site detection of DCNP.

Recent progress in carbonaceous materials based Z-scheme and Sscheme heterojunctions for photocatalytic clean energy generation

Carbonaceous materials including carbon nanotubes/nanofibers, graphene, graphene oxide, reduced graphene oxide,graphyne, graphdiyne, carbon quantum dots and fullerenes have gained considerable attention in the recent years for their unique properties such as high conductivity, excellent stability and biocompatibility. The integration of these carbonaceous materials into Zscheme and S-scheme heterojunctions has emerged as a transformative strategy to enhance the photocatalytic efficiency for energy conversion applications. This review delves into the fundamental principles of clean energy generation such as photocatalytic H2generation and CO2reduction, elucidating their respective mechanisms and advantages. Furthermore, various types of carbonaceous materials, their synthesis and construction of Z-scheme and S-scheme heterojunctions are discussed emphasizing their role in promoting charge separation, reducing recombination losses and extending the spectral response range. With the focus on solar fuel production,the recent advancements in carbonaceous based Z-scheme and S-scheme heterojunctions are discussed and summarized for photocatalytic H2generation and CO2reduction. Lastly, the current bottlenecks and challenges in the field of carbonaceous based photocatalysts are discussed with the valuable insights for future development in this particular field.
